The study was performed in order to clarify whether granulocyte-macrophage colony stimulating factor (GM-CSF) and granulocyte colony stimulating factor (G-CSF) affect melatonin production and release. We have found that both factors (GM-CSF at doses of 10.0 and 100.0 micrograms/kg, and G-CSF at doses of 1.0, 10.0, and 100.0 micrograms/kg) stimulate melatonin secretion in rats in vivo. Positive correlations between tested doses of GM-CSF and G-CSF and plasma melatonin levels were observed (P < 0.01). The formation of T lymphocyte colonies was studied in 30 normal subjects and 12 patients with suspected abnormalities in immune function. The mean number of colonies per plate in normal subjects was 1.159 +/- 411 which represented a plating efficiency of 0.5-1.0%. Colony formation assays enable an assessment of the cells' ability to grow in colonies under in vitro conditions. Following the seeding and an incubation period, the cell colonies are stained using a crystal violet staining solution, and the resulting colonies are imaged and assessed. This method can be leveraged to investigate the effect of genetic modifications or the application of different drug compounds on colony growth. Here, we describe a protocol applied to Ewing sarcoma cell lines, as well as possible modifications.

OBJECTIVES: Research on historical populations in Europe finds that infectious disease epidemics appear to induce predictable cycles in age-specific mortality. We know little, however, about whether such cycles also occurred in less dense founder populations of North America. We used high-quality data on the Quebecois population from 1680 to 1798 to examine the extent to which age-specific mortality showed predictable epidemic cycles. We further examined whether environmental pressures-temperature, lack of precipitation, or crop failure-may have set the stage for the emergence of epidemics.

Bacterial variants of Staphylococcus aureus called small colony variants (SCVs) originate by mutations in metabolic genes, resulting in emergence of auxotrophic bacterial subpopulations.
