[Isolation of protoplasts from vegetable tissues using extracellular lytic enzymes from fusarium oxysporum f.sp. melonis].

Fusarium oxysporum f.sp. melonis, a pathogen of melon (Cucumis melo L.), was grown in shaken cultures at 26 degrees C in a mineral salts medium containing glucose, xylan and apple pectin as carbon sources. The extracellular enzymic complex obtained from these cultures showed lytic activity on plant tissues, causing maceration of melon fruits, potato tubers and carrot roots. Protoplasts were isolated from melon fruits when the maceration was carried out under appropriate osmotic conditions. This fact suggest a possible relationship between the enzymes produced by Fusarium oxysporum f.sp. meloni…

Hydrogen sulfide is required for salicylic acid-induced chilling tolerance of cucumber seedlings.

Salicylic acid (SA) and hydrogen sulfide (H2S) have been proved to be multifunctional signal molecules to participate in the response of plants to abiotic stresses. However, it is still unclear whether there is interaction between SA and H2S in response to chilling intensity of cucumber seedlings. Here, we found SA was sensitive to chilling intensity. Under normal condition, NaHS (H2S donor) or removing endogenous H2S with hypotaurine (HT, a specific scavenger of H2S) and DL-propargylglycine (PAG, a specific inhibitor of H2S) has no effect on endogenous SA level; however, SA induced endogenous

Ligno-Suberized Fruit Periderm as a Mechanically Reinforced Biomacromolecular Composite.

Plant surfaces are biomacromolecular composites whose organization determines barrier performance. We examined the transition from a cutin-dominated cuticle in common cucumber to a ligno-suberized periderm in Sikkim cucumber. Multiscale microscopy and histochemistry resolved a developmental shift from a smooth epidermis to a multilayered phellem characterized by distinct suberin lamellae and lignin deposition. GC-MS-based profiling revealed that periderm formation involves a shift from C16/C18 cutin monomers to an enrichment in >C20 very-long-chain aliphatics (fatty acids, alcohols, hydroxy ac

Tasting of traditional Polish fermented cucumbers: Microbiology, morpho-textural features, and volatilome.

In the present study, naturally fermented and unpasteurized cucumbers (Cucumis sativus L.) collected from 4 producers located in different regions of Poland were studied. The fermented cucumbers were characterized by significant nutritional features in terms of polyphenols content and antioxidant activity. Microbiological analyses revealed active bacterial populations of lactococci, thermophilic cocci, lactobacilli, and coagulase-negative cocci. The microbiological characterization of cucumber and brine samples through metataxonomic analysis allowed the dominant species to be detected, being L…

Hydrogen sulfide acts downstream of methane to induce cucumber adventitious root development.

Previous results have shown that hydrogen sulfide (H2S), mainly catalyzed by l-cysteine desulfhydrase (DES) in plants, triggers adventitious rooting. The objective of this study was to test whether H2S is involved in methane (CH4)-induced adventitious root development in cucumber explants. First, we observed that the activities of DES, endogenous H2S production, and thereafter adventitious root development were induced by CH4 and NaHS (an H2S donor). Some responses were sensitive to hypotaurine (HT; a scavenger of H2S), showing that endogenous H2S production and adventitious rooting were obvio

Estimation of Phenylalanine Ammonia-Lyase Activity for Assessing Resistance of Muskmelon Against Fusarium oxysporum f. sp. melonis Infection.

Phenylalanine ammonia-lyase is a key enzyme in the phenylpropanoid pathway, responsible for the conversion of L-phenylalanine into trans-cinnamic acid. This step is crucial for the biosynthesis of lignin, phytoalexins, and other phenolic compounds that play a significant role in plant defense responses. This chapter describes a detailed protocol for the extraction and quantitative estimation of phenylalanine ammonia-lyase activity in muskmelon (Cucumis melo L.) tissues in response to Fusarium oxysporum f. sp. melonis (FOM) infection. The method enables researchers to compare phenylalanine ammo…

Internal stress controls tendril writhing dynamics in climbing plants.

We present experimental results on the writhing dynamics of cucumber plant tendrils subjected to an axial traction force. At low forces, the tendrils' curvature saturates exponentially in time at a limiting value. At forces higher than the critical threshold, the tendril fails to coil but develops an intrinsic coiling-prone stress, corresponding to an intrinsic residual curvature, visible if the load is released. Growth-field assessments indicate that curvature arises from differential growth. To understand these experiments we develop an autotropic morphoelastic growth model with a bi-strip g

Saponins of North Atlantic Sea Cucumber: Chemistry, Health Benefits, and Future Prospectives.

Frondosides are the major saponins (triterpene glycosides) of the North Atlantic sea cucumber (Cucumaria frondosa). Frondosides possess amphiphilic characteristics due to the presence of various hydrophilic sugar moieties and hydrophobic genin (sapogenin). Saponins are abundant in holothurians, including in sea cucumbers that are widely distributed across the northern part of the Atlantic Ocean. Over 300 triterpene glycosides have been isolated, identified, and categorized from many species of sea cucumbers. Furthermore, specific saponins from sea cucumbers are broadly classified on the basis

Pythium deliense Causing Cottony Leak on Pickling Cucumber Fruit in South Carolina, U.S.A.

Cottony leak, a soft rot on fruit of cucurbit crops, is a concern for pickling cucumber (Cucumis sativus cv. Gershwin) growers in South Carolina and other states. Fruit with typical symptoms and signs of cottony leak were sampled from five commercial fields in 2022 and 2023. Most isolates (28) recovered from symptomatic fruit were identified as Pythium deliense, and the remaining five isolates were P. aphanidermatum, based on sequences of the mitochondrially encoded cytochrome c oxidase subunit 1 gene. P. deliense also was recovered from stem lesions on two seedlings. In pathogenicity tests, P…

Comparative hyperparameter optimization of object detection models for precision monitoring of cucumber beetles and similar insects on yellow sticky cards.

Computer vision presents a great opportunity for improving pest monitoring in agriculture, particularly for yellow sticky traps, a critical component in IPM. However, despite the growing interest in applying object detection models for insect identification, insect datasets present unique challenges, and approaches for fine-tuning model parameters to achieve reliable performance remain limited. This study explores the influence of fine tuning three key hyperparameters (learning rate, optimizer type and batch size) on the performance of two popular object detection models (YOLO and RT-DETR), in

Growth-promoting effects of Aspergillus Elegans and the dark septate endophyte (DSE) Periconia macrospinosa on cucumber.

Cucurbits are subject to a variety of stresses that limit their sustainable production, despite their important role in ensuring food security and nutrition. Plant stress tolerance can be enhanced through fungal endophytes. In this study, two endophytes isolated from wild plant roots, were tested to determine their effect on the growth promotion of cucumber (Cucumis sativus L.) plants. The phylogenetic analysis revealed that the designated isolates were Aspergillus elegans and Periconia macrospinosa. The results of the Plant Growth Promoting Fungal (PGPF) tests showed that both Aspergillus ele…

Cucumber NUCLEAR FACTOR-YC2/-YC9 target translocon component CsTIC21 in chloroplast photomorphogenesis.

Light signals promote photomorphogenesis and photosynthesis, allowing plants to establish photoautotrophic growth. Chloroplasts are organelles responsible for photosynthesis in which light energy is converted into chemical energy and stored as organic matter. However, how light regulates chloroplast photomorphogenesis remains unclear. Here, we isolated a cucumber (Cucumis sativus L.) mutant albino seedling (as) from an ethyl methane sulfonate mutagenesis library with an albino phenotype. Map-based cloning revealed that the mutation occurred in a component of cucumber translocon at the inner me…

Hydrogen sulfide and nitric oxide are involved in melatonin-induced salt tolerance in cucumber.

Hydrogen sulfide (H2S) is a novel gaseous signaling molecule in response to adversity stress. Melatonin (MT) is a multifunctional molecule that plays an important role in regulating plant stress resistance. However, the interactions between H2S and MT are still unknown. Therefore, the role of H2S in MT-induced salt tolerance was elucidated in this study by measuring the antioxidant defense system and photosynthetic characteristics of cucumber. In addition, the crosstalk among H2S, NO, and mitogen-activated protein kinase (MAPK) was investigated. Results showed that MT induced the production of

[Hydrogen sulfide acted as a downstream signal was involved in the regulation of salicylic acid on photosynthesis of cucumber seedlings under low temperature and low light intensity].

Both salicylic acid (SA) and hydrogen sulfide (H2S) play an important role in regulating plant growth and development and physiological metabolism under abiotic stresses. As signal molecules, the interaction between them in regulating cucumber photosynthesis under low temperature and low light is still unclear. Here, we examined the regulation and interaction of SA and H2S on photosynthesis in cucumber seedlings under low temperature (8 ℃/5 ℃, day/night) and low light (100 μmol·m-2·s-1). Seedlings were foliar-sprayed with SA, sodium sulfide (NaHS, H2S donor), and their scavenger or biosynthesi

A case of vitiligo cured with cucumber and sulfur.

This study investigates a 42-year-old man who used sulfur powder adhered to cucumber slices to successfully self-treat a vitiligo condition. The treatment has resulted in no recurrence of the disease for 21 years. We analyzed the mechanism of this folk prescription for vitiligo, concluding that the success of the self-treatment may be mainly associated with hydrogen sulfide (H2 S). The antibacterial activity of pentathionic acid (H2 S5 O6 ) and the antioxidant activity of cucumber might also play a role in the treatment.
