Removing Pb2+ ions from aqueous solution with freshwater cyanobacteria: Combined adsorption and intracellular precipitation.

This study investigated the removal of lead ions (Pb2+) from aqueous solutions by two freshwater cyanobacteria with distinct cellular inclusions: Synechococcus elongatus FACHB-410, which is rich in polyphosphate (polyP), and Cyanothece sp. PCC 7425, containing both polyP and amorphous calcium carbonate (iACC). Both strains were exposed to Pb2+ concentrations ranging from 1 to 50 mg/L and demonstrated high removal efficiencies exceeding 92%. The removal mechanism involved not only adsorption onto cell surfaces but also significant intracellular precipitation. The proportion of intracellular pre…

Systematic determination of disulfide bond reduction potentials reveals a nonequilibrium redox hierarchy in cyanobacteria.

Disulfide bonds act as reversible switches that regulate cellular function in nearly all organisms. Their behavior is set by the midpoint potential (Em), yet Em is known for only a small number of sites, mostly in purified proteins studied away from their natural partners. We developed a mass-spectrometry workflow that measures Em directly from native cell lysates. By equilibrating proteins of the cyanobacterium Synechocystis sp. PCC 6803 in defined redox buffers and reading out the oxidation state of individual cysteines, we obtained 368 Em values across the proteome and validated them agains

The calcium-binding protein CSE links Ca2+ signaling with cell-cell communication in cyanobacteria.

Multicellular cyanobacteria have evolved sophisticated cell-cell communication machinery to exchange, synchronize, and coordinate the efforts of individual cells. Analogous to gap junctions that have traditionally been regarded as a eukaryotic feature, multicellular cyanobacteria coordinate their cell-cell communication via septal junctions (SJs). However, the signals that regulate cell-cell communication and septal-junction assembly are largely unknown. Lately, calcium signaling has been implicated in regulating cell junctions in eukaryotes. We recently discovered a Ca2+-binding protein, CSE,…

Unraveling the mechanism of greenhouse gas emissions during allelochemical-triggered inhibition of cyanobacteria recovery.

Climate change amplifies cyanobacterial blooms, creating a feedback loop between warming and bloom intensity. This dynamic emphasizes the need to address greenhouse gas (GHG) emissions during in-situ cyanobacterial control, minimizing algicides use and its long-term climatic impacts. However, research on the dose-response of algicides on GHG emissions is limited. This study investigated the dose-dependent effects of artemisinin sustained-release algicides (ASAs) on GHG emissions and microbial dynamics during cyanobacteria recovery prevention. Microcosm experiments revealed that ASAs dosage res…

Acute Toxicity of Carbon Nanotubes, Carbon Nanodots, and Cell-Penetrating Peptides to Freshwater Cyanobacteria.

Synthetic non-metallic nanoparticles (NMNPs) such as carbon nanotubes (CNTs), carbon nanodots (CNDs), and cell-penetrating peptides (CPPs) have been explored to treat harmful algal blooms. However, their strain-specific algicidal activities have been rarely investigated. Here we determined their acute toxicity to nine freshwater cyanobacterial strains belonging to seven genera, including Microcystis aeruginosa UTEX 2386, M. aeruginosa UTEX 2385, M. aeruginosa LE3, Anabaena cylindrica PCC 7122, Aphanizomenon sp. NZ, Planktothrix agardhii SB 1810, Synechocystis sp. PCC 6803, Lyngbya sp. CCAP 144…

Dissolved organic nitrogen, dinoflagellates and cyanobacteria in two eutrophic lakes: Analysis by inferential modelling.

The Lake Suwa (Japan) has a history of non-N-fixing Microcystis blooms. Lake Kinneret (Israel) experienced multiannual periods of sole domination by the dinoflagellate Peridinium gatunense and periods dominated seasonally by P. gatunense or cyanobacteria. Extensive studies have been carried out in both lakes regarding the role of dissolved inorganic nitrogen and phosphorus as drivers of primary productivity. There is growing evidence that dissolved organic nitrogen (DON) compounds also influence not only biomass and structure of phytoplankton communities but also microcystin production. This s…

The rice field cyanobacteria Anabaena azotica and Anabaena sp. CH1 express vanadium-dependent nitrogenase.

Anabaena azotica FACHB-118 and Anabaena sp. CH1, heterocystous cyanobacteria isolated from Chinese and Taiwanese rice fields, expressed vanadium-containing nitrogenase when under molybdenum deficiency. This is the second direct observation of an alternative nitrogenase in cyanobacteria. The vanadium nitrogenase-specific genes vnfDG are fused and clustered in a phylogenetic tree next to the corresponding genes of Methanosarcina. The expression of vnfH in cells cultured in Mo-free medium and of nifH in Mo-grown cells was shown for the first time by sequencing cDNA derived from cultures of A. azo…

Adaptive Multi-Scale Fusion Enhanced RT-DETR for Efficient Cyanobacteria Detection in Microscopic Images.

Accurate and efficient detection of cyanobacteria in microscopic images is important for automated water-quality monitoring, but remains challenging because of complex aquatic backgrounds, large scale variation, and uneven sample quality. This study proposes an adaptive multi-scale fusion enhanced RT-DETR framework for cyanobacteria detection. The baseline RT-DETR-R18 is improved by incorporating the SeFaster module for efficient feature extraction, the high-level screening-feature fusion pyramid network for semantic-guided multi-scale fusion, and the Wise-IoU loss for more stable localization…

Multi-level osmoadaptation strategies of filamentous cyanobacteria within oxygenic photogranules under high salinity stress.

Filamentous cyanobacteria (FC) are key structural and functional components of oxygenic photogranules (OPGs) for saline wastewater treatment. However, how FC maintain cellular osmotic balance while supporting community stability under salinity stress remains poorly understood. Here, we integrated long-term reactor operation, physiological characterization, and multi-omics analyses to elucidate the multi-level osmoadaptation strategies of dominant FC within OPGs. Despite stepwise increases in salinity, OPGs maintained stable pollutant removal and showed enhanced FC growth, polysaccharide (PS) p…

Cyanobacteria from marine oxygen-deficient zones encode both form I and form II Rubiscos.

Cyanobacteria are highly abundant in the marine photic zone and primary drivers of the conversion of inorganic carbon into biomass. To date, all studied cyanobacterial lineages encode carbon fixation machinery relying upon form I Rubiscos within a CO2-concentrating carboxysome. Here, we report that the uncultivated anoxic marine zone (AMZ) IB lineage of Prochlorococcus from pelagic oxygen-deficient zones (ODZs) harbors both form I and form II Rubiscos, the latter of which are typically noncarboxysomal and possess biochemical properties tuned toward low-oxygen environments. We demonstrate that …

Characterizing A21: Natural Cyanobacteria-Based Consortium with Potential for Steroid Bioremediation in Wastewater Treatment.

Microalga-bacteria consortia are increasingly recognized for their effectiveness in wastewater treatment, leveraging the metabolic synergy between microalgae and bacteria to enhance nutrient removal and overall treatment efficiency. These systems offer a sustainable approach to addressing pollutants such as nitrogen and phosphorus. However, their potential in removing specific contaminants like steroid hormones is less explored. In this study, a natural microbial consortium, A21, has been characterized and isolated from primary sewage treatment in Madrid and its potential for bioremediation of

Degradative solvent extraction of cyanobacteria: From reaction kinetics to potential organic matter evolution mechanism.

This study proposed a new continuous lumped reaction kinetics model to accurately reveal the control mechanism of cyanobacteria at each stage of degradative solvent extraction and discussed the potential evolution mechanism of organic matter. Results showed that degradation solvent extraction successfully separated nitrogen and phosphorus from cyanobacteria. The solute has high carbon and volatile contents, is almost ash-free, and forms a phosphorus-rich residue. The lowest fitting degree of the continuous lumped reaction model kinetics was 94.5%, suggesting that this model worked well. The de…

Synthesis and Activity of 1,2,3-Triazole Aminopyrimidines against Cyanobacteria as PDHc-E1 Competitive Inhibitors.

Cyanobacteria harmful algal blooms are of global concern, but all currently available algicides in the market are nonselective and have potential side effects on nontarget species. In the present work, two series of compounds (4 and 6) comprising 16 novel 1,2,3-triazole aminopyrimidines were rationally designed and synthesized as control agent for cyanobacteria. Our design focus was the inhibiting cyanobacteria by inhibition against pyruvate dehydrogenase complex E1 (PDHc-E1). Compounds 4 and 6 showed potent inhibition against Escherichia coli PDHc-E1 (IC50 = 4.13-23.76 μM) and also strong alg…

Biomonitoring genotoxicity and cytotoxicity of Microcystis aeruginosa (Chroococcales, cyanobacteria) using the Allium cepa test.

Water pollution caused by toxic cyanobacteria is a problem worldwide, increasing with eutrophication. Due to its biological significance, genotoxicity should be a focus for biomonitoring pollution owing to the increasing complexity of the toxicological environment in which organisms are exposed. Cyanobacteria produce a large number of bioactive compounds, most of which lack toxicological data. Microcystins comprise a class of potent cyclic heptapeptide toxins produced mainly by Microcystis aeruginosa. Other natural products can also be synthesized by cyanobacteria, such as the protease inhibit…

Survival of cyanobacteria and mitigation of Fe(II) toxicity effects in a silica-rich Archean ocean.

Banded iron formations (BIF) were deposited abundantly between 2.7-2.4 Ga from iron- and silica-rich oceans, with cyanobacterial oxygen (O2) as a possible oxidant for Fe(II)(aq) oxidation and Fe(III) oxyhydroxide precipitation. However, toxic reactive oxygen species (ROS) from Fe(II)/O2 interactions might have inhibited cyanobacterial growth, contributing to the delay between cyanobacterial evolution (>3.0 Ga) and the Great Oxidation Event (2.5 Ga). Here, we explored the impact of Fe(II)(aq) and SiO2(aq) on Synechococcus sp. PCC 7002. High Fe(II)(aq) ( > 500 µM) increased ROS formation, but el…

The Ability of Airborne Microalgae and Cyanobacteria to Survive and Transfer the Carcinogenic Benzo(a)pyrene in Coastal Regions.

Air pollution has been a significant problem threatening human health for years. One commonly reported air pollutant is benzo(a)pyrene, a dangerous compound with carcinogenic properties. Values which exceed normative values for benzo(a)pyrene concentration in the air are often noted in many regions of the world. Studies on the worldwide spread of COVID-19 since 2020, as well as avian flu, measles, and SARS, have proven that viruses and bacteria are more dangerous to human health when they occur in polluted air. Regarding cyanobacteria and microalgae, little is known about their relationship wi…

Cyanobacteria Microcystis aeruginosa Contributes to the Severity of Fish Diseases: A Study on Spring Viraemia of Carp.

Fish are exposed to numerous stressors in the environment including pollution, bacterial and viral agents, and toxic substances. Our study with common carps leveraged an integrated approach (i.e., histology, biochemical and hematological measurements, and analytical chemistry) to understand how cyanobacteria interfere with the impact of a model viral agent, Carp sprivivirus (SVCV), on fish. In addition to the specific effects of a single stressor (SVCV or cyanobacteria), the combination of both stressors worsens markers related to the immune system and liver health. Solely combined exposure re…

Engineering a synthetic pathway in cyanobacteria for isopropanol production directly from carbon dioxide and light.

Production of alternate fuels or chemicals directly from solar energy and carbon dioxide using engineered cyanobacteria is an attractive method to reduce petroleum dependency and minimize carbon emissions. Here, we constructed a synthetic pathway composed of acetyl-CoA acetyl transferase (encoded by thl), acetoacetyl-CoA transferase (encoded by atoAD), acetoacetate decarboxylase (encoded by adc) and secondary alcohol dehydrogenase (encoded by adh) in Synechococcus elongatus strain PCC 7942 to produce isopropanol. The enzyme-coding genes, heterogeneously originating from Clostridium acetobutyli…
