Activity of olive oil phenols on lymphomonocyte cytosolic calcium.

Phenols, present in the Mediterranean diet, have antioxidant properties and are free radical scavengers; however, the molecular mechanisms of their beneficial effects are not yet fully understood. The level of cytosolic calcium ([Ca2+]i) is an important signal also in nonexcitable cells, including immune cells, and regulates fundamental processes. In this paper, we determine [Ca2+]i in human lymphomonocytes incubated with two olive oil phenols: 3,4-(dihydroxyphenyl)ethanol and p-(hydroxyphenyl)ethanol. Both tested phenols increase [Ca2+]i in a dose-dependent way. This effect is antagonized by …

cAMP controls cytosolic Ca2+ levels in Dictyostelium discoideum.

BACKGROUND: Differentiating Dictyostelium discoideum amoebae respond upon cAMP-stimulation with an increase in the cytosolic free Ca2+ concentration ([Ca2+]i) that is composed of liberation of stored Ca2+ and extracellular Ca2+-influx. In this study we investigated whether intracellular cAMP is involved in the control of [Ca2+]i. RESULTS: We analyzed Ca2+-fluxes in a mutant that is devoid of the main cAMP-phosphodiesterase (PDE) RegA and displays an altered cAMP metabolism. In suspensions of developing cells cAMP-activated influx of extracellular Ca2+ was reduced as compared to wild type. Yet…

Salicylic acid induces a cytosolic Ca2+ elevation in yeast.

Cytosolic free calcium ion concentration ([Ca2+]cyt) after a salicylic acid (SA)-stimulus was monitored in cells of the yeast Saccharomyces cerevisiae expressing apoaequorin, which constitutes a Ca(2+)-sensitive luminescent protein, aequorin, when combined with coelenterazine. SA induced a transient [Ca2+]cyt elevation that was dependent on the concentration of SA and pH of the SA solution. The SA-induced [Ca2+]cyt elevation was not reduced in Ca(2+)-deficient medium, suggesting that Ca2+ was mobilized from an intracellular Ca2+ store(s). Benzoic acid, butyric acid and sorbic acid did not indu…

Characterization of the cytosolic estrogen receptor in rat skeletal muscle.

A charcoal assay and the synthetic estrogen [6,7-3H]R 2858 (17 alpha-ethynyl-11 beta-methoxyestradiol-17 beta) were used to show that rat skeletal muscle cytosol contains an estrogen receptor, which was characterized with regard to association and dissociation rate-constants at several temperatures. The degradation kinetics was also studied, and was more rapid in the absence than in the presence of ligand. Association, dissociation and degradation were all temperature-dependent. The apparent equilibrium dissociation constant (Kd), however, was not temperature-dependent, and was about 0.1-1.0 n…

Concentration of cytosolic estrogen receptors in patients with postmenopausal osteoporosis.

Although all postmenopausal women are relatively estrogen-deficient, osteoporosis develops in only some of them. This could be related to a greater-than-normal degree of postmenopausal failure in sex hormone production or to a decrease in biologic effect at target tissue sites. We compared the concentration of sex steroids in serum and the concentration of cytosolic estrogen receptors in one target tissue (obtained by biopsy of the uterine cervix) in 18 women with postmenopausal osteoporosis and in 18 age-matched postmenopausal control subjects. Serum androstenedione and estrone were not signi…

Improved cross-presentation enabled by fusion-dependent mini-proteins that efficiently reach the dendritic cell cytosol.

Cross-presentation is the process by which dendritic cells communicate to CD8 + T-cells the detection of exogenous foreign substances known as antigens. This process primes naïve CD8 + T-cells to eradicate antigen-expressing pathogens and tumors. It is well known that pathogens and tumors evade CD8 + T-cell immunity by reducing antigen uptake into the endocytic pathway and/or limiting endosomal escape of the antigen to the cytosol. Here we employ biophysical and cell biology tools to separately and quantitatively probe the efficiency of antigen uptake, internalization, display, and activity in…

Dual Roles of CCN5/WISP2 in Cytosol and Secretome for Maintaining Muscle Homeostasis and Preventing Sarcopenia.

BACKGROUND: Sarcopenia is a progressive and systemic skeletal muscle disorder characterized by the decline of muscle mass and function. Despite its early-stage, 'possible sarcopenia' has been emphasized for prompt intervention; there are currently no specific biomarkers for the diagnosis and treatment. METHODS: RNA sequencing of human skeletal muscle across sarcopenia stages identified CCN5. Adeno-associated virus (AAV) is intramuscularly injected into young C57BL/6J mice to knockdown CCN5 in skeletal muscle. Phenotypic alterations are assessed through behavioural testing, body composition an

A Chlamydial Plasmid-Dependent Secretion System for the Delivery of Virulence Factors to the Host Cytosol.

Chlamydia are obligate intracellular Gram-negative bacteria distinguished by a unique developmental biology confined within a parasitophorous vacuole termed an inclusion. The chlamydial plasmid is a central virulence factor in the pathogenesis of infection. Plasmid gene protein 4 (Pgp4) regulates the expression of plasmid gene protein 3 (Pgp3) and chromosomal glycogen synthase (GlgA), virulence factors secreted from the inclusion to the host cytosol by an unknown mechanism. Here, we identified a plasmid-dependent secretion system for the cytosolic delivery of Pgp3 and GlgA. The secretion syste…

The membrane-associated fraction of cyclase associate protein 1 translocates to the cytosol upon platelet stimulation.

Platelets undergo profound shape changes upon adhesion to damaged blood vessel walls that are mediated by reorganisation of the actin cytosk … cell cortex and the periphery of actin nodules. Upon subcellular fractionation most CAP1 was found cytosolic but part associated to the membrane fraction in an actin-independent manner. Interestingly, upon stimulation with thrombin a significant proportion of the membrane-associated CAP1 translocates to the cytosol. This relocalisation was prevented by p …

Distinct subcellular localization in the cytosol and apicoplast, unexpected dimerization and inhibition of Plasmodium falciparum glyoxalases.

The ubiquitous glyoxalase system removes methylglyoxal as a harmful by-product of glycolysis. Because malaria parasites have drastically increased glycolytic fluxes, they could be highly susceptible to the inhibition of this detoxification pathway. Here we analysed the intracellular localization, oligomerization and inhibition of the glyoxalases from Plasmodium falciparum. Glyoxalase I (GloI) and one of the two glyoxalases II (cGloII) were located in the cytosol of the blood stages. The second glyoxalase II (tGloII) was detected in the apicoplast pointing to alternative metabolic pathways. Usi…

Serum estradiol-17 beta, progesterone and respective uterine cytosol receptor concentrations in bitches with spontaneous pyometra.

The role of serum estradiol-17 beta (E(2)) and progesterone (P(4)) in relation to uterine estrogen (ER) and progesterone receptors (PR) was investigated in canine cystic endometrial hyperplasia-pyometra (CEH-P). Blood and uterine samples were collected pre- and post-ovariohysterectomy, respectively, from 54 bitches presenting spontaneous CEH-P and 25 healthy control bitches. Competitive enzyme immunoassays (EIA) and enzyme ligand immunoassays (ELIA) were applied to estimate serum hormones and uterine cytosol active receptors, respectively. Animals were classified in the stages of first half of…

Interaction of chlorpromazine with low molecular mass ion-transporting ATPase modulator proteins from rat brain cytosol.

Two low molecular mass proteins (13 kDa which inhibits Na+,K(+)-ATPase and 12 kDa which modulates Ca2+, Mg(2+)- and Ca(2+)-ATPases), purified from rat brain cytosol form complexes with chlorpromazine (CPZ) on incubation. The conformational characteristics of the proteins and their complex have been studied by comparing the fluorescence and CD spectra. The tryptophan fluorescence data show that the inhibitor-CPZ complex does not quench the fluorescence of NA+,K(+)-ATPase significantly. CD spectra indicate that the structure of the inhibitor is changed on formation of the complex. The inhibitor-…

[Role of hormones in maintaining the sex-differentiated level of estrogen receptors in rat liver cytosol].

A study was made of the endocrine mechanisms of the formation and maintenance of a sex-differentiated level of estrogen receptors (ER) in rat liver cytosol. The administration of testosterone-propionate (TP) at a dose of 3 mg for 3 days was shown to cause a significant decrease in the concentration of ER in the liver of gonadectomized animals to the level in intact male rats. In a week after the discontinuation of TP, a complete restoration of the basal level of receptors was observed. Neonatal and prepubertal administration of TP to gonadectomized male rats at early stages of ontogenesis made…

Syncytial therapeutics: Receptor-specific and direct-to-cytosol biologic drug delivery mediated by measles fusion complex.

This work explores cell-cell fusion mediated by measles virus (MeV) as a potential new cell therapy modality that achieves direct-to-cytosol (DTC) drug delivery. MeV induces receptor-mediated fusion at the cell surface via its hemagglutinin (H) and fusion glycoproteins (F), bypassing endocytic membrane transport, and enabling direct cytosolic mixing between a fusogenic donor and host target cell. Fusion of this type gives rise to large syncytia formed by the inclusion of additional target cells over time. Fusion receptor specificity was first examined in CHO "non-target" and CHO "target" cells…

The effect of ionic redistributions on the microwave dielectric response of cytosol water upon glucose uptake.

The sensitivity of cytosol water's microwave dielectric (MD) response to D-glucose uptake in Red Blood Cells (RBCs) allows the detailed study of cellular mechanisms as a function of controlled exposures to glucose and other related analytes like electrolytes. However, the underlying mechanism behind the sensitivity to glucose exposure remains a topic of debate. In this research, we utilize MDS within the frequency range of 0.5-40 GHz to explore how ionic redistributions within the cell impact the microwave dielectric characteristics associated with D-glucose uptake in RBC suspensions. Specific…

Exosomes induce endolysosomal permeabilization as a gateway by which exosomal tau seeds escape into the cytosol.

Degradation products of the extracellular pathogen Streptococcus pneumoniae access the cytosol via its pore-forming toxin.

UNLABELLED: Streptococcus pneumoniae is a leading pathogen with an extracellular lifestyle; however, it is detected by cytosolic surveillance systems of macrophages. The innate immune response that follows cytosolic sensing of cell wall components results in recruitment of additional macrophages, which subsequently clear colonizing organisms from host airways. In this study, we monitored cytosolic access by following the transit of the abundant bacterial surface component capsular polysaccharide, which is linked to the cell wall. Confocal and electron microscopy visually characterized the loca…

How ricin and Shiga toxin reach the cytosol of target cells: retrotranslocation from the endoplasmic reticulum.

A number of protein toxins bind at the surface of mammalian cells and after endocytosis traffic to the endoplasmic reticulum, where the toxic A chains are liberated from the holotoxin. The free A chains are then dislocated, or retrotranslocated, across the ER membrane into the cytosol. Here, in contrast to ER substrates destined for proteasomal destruction, they undergo folding to a catalytic conformation and subsequently inactivate their cytosolic targets. These toxins therefore provide toxic probes for testing the molecular requirements for retrograde trafficking, the ER processes that prepa…
