A "Whirly" transcription factor is required for salicylic acid-dependent disease resistance in Arabidopsis.

Transcriptional reprogramming is critical for plant disease resistance responses; its global control is not well understood. Salicylic acid (SA) can induce plant defense gene expression and a long-lasting disease resistance state called systemic acquired resistance (SAR). Plant-specific "Whirly" DNA binding proteins were previously implicated in defense gene regulation. We demonstrate that the potato StWhy1 protein is a transcriptional activator of genes containing the PBF2 binding PB promoter element. DNA binding activity of AtWhy1, the Arabidopsis StWhy1 ortholog, is induced by SA and is req

Polymeric microneedle patches for rapid DNA extraction and molecular diagnosis of foliar pathogens of mung bean.

Global food security is threatened by crop losses due to plant diseases. Early detection of pathogens is essential for implementing effective management practices to mitigate the negative impacts of disease. On-site molecular diagnostic techniques have significantly advanced for rapid and accurate identification of pathogens, but DNA extraction remains a labour-intensive and time-consuming bottleneck. This study demonstrates the ability of polyvinyl alcohol/polyvinylpyrrolidone (PVA/PVP) microneedle patches to extract detectable amounts of bacterial and plant DNA from diseased leaves within 1

Subversion of the salicylic acid signaling pathway by the bipartite begomoviral protein BV1 promotes virus infection and vector preference to virus-infected plants.

The outbreak of vector-borne plant viruses entails both efficient in planta virus infection and productive vector-mediated transmission. Yet, the factors that concurrently regulate these two key viral traits remain understudied. Here we examine the role of viral proteins and the salicylic acid (SA) signaling pathway in modulating virus infection and vector preference to virus-infected plants. Infection of plants by the bipartite begomovirus, Sri Lankan cassava mosaic virus (SLCMV), dramatically induces the accumulation of SA, a positive regulator of antiviral defense. As countermeasures, SLCMV

A large, mobile pathogenicity island confers plant pathogenicity on Streptomyces species.

Potato scab is a globally important disease caused by polyphyletic plant pathogenic Streptomyces species. Streptomyces acidiscabies, Streptomyces scabies and Streptomyces turgidiscabies possess a conserved biosynthetic pathway for the nitrated dipeptide phytotoxin thaxtomin. These pathogens also possess the nec1 gene which encodes a necrogenic protein that is an independent virulence factor. In this article we describe a large (325-660 kb) pathogenicity island (PAI) conserved among these three plant pathogenic Streptomyces species. A partial DNA sequence of this PAI revealed the thaxtomin bios

Gibberellin 20-oxidase gene OsGA20ox3 regulates plant stature and disease development in rice.

Gibberellin (GA) 20-oxidase (GA20ox) catalyses consecutive steps of oxidation in the late part of the GA biosynthetic pathway. A T-DNA insertion mutant (17S-14) in rice, with an elongated phenotype, was isolated. Analysis of the flanking sequences of the T-DNA insertion site revealed that an incomplete T-DNA integration resulted in enhanced constitutively expression of downstream OsGA20ox3 in the mutant. The accumulation of bioactive GA(1) and GA(4) were increased in the mutant in comparison with the wild-type plant. Transgenic plants overexpressing OsGA20ox3 showed phenotypes similar to those

Modulation of heat shock factors accompanies salicylic acid-mediated potentiation of Hsp70 in tomato seedlings.

In plants, salicylic acid (SA) is a signalling molecule regulating disease resistance responses such as systemic acquired resistance (SAR) and the hypersensitive response (HR), and has been implicated in both basal and acquired thermotolerance. It has been shown that SA enhances heat-induced Hsp/Hsc70 accumulation in plants. To investigate the mechanism of how SA influences the heat shock response (HSR) in plants, tomato seedlings were treated with SA alone, heat shock, or a combination of both before analyses of hsp70 mRNA, heat shock factor (Hsf)-DNA binding, and gene expression of hsp70, hs

Plant responses to UV radiation and links to pathogen resistance.

Increased incident ultraviolet (UV) radiation due to ozone depletion has heightened interest in plant responses to UV because solar UV wavelengths can reduce plant genome stability, growth, and productivity. These detrimental effects result from damage to cell components including nucleic acids, proteins, and membrane lipids. As obligate phototrophs, plants must counter the onslaught of cellular damage due to prolonged exposure to sunlight. They do so by attenuating the UV dose received through accumulation of UV-absorbing secondary metabolites, neutralizing reactive oxygen species produced by

Quantification of Saccharopolyspora rectivirgula in composting plants: assessment of the relevance of S. rectivirgula.

Exposure to bioaerosols in composting plants can lead to negative health effects on compost workers. Health complaints vary between cough, irritation of the eyes and the skin, sinusitis, or dyspnea among others. It is fact that compost materials harbor high concentrations of microorganisms, which were aerosolized during handling compost. Within the present study, total cell numbers between 3.4 × 10(4) and 1.6 × 10(8) cell counts per m(3) air were determined after 4',6-Diamidin-2-phenylindol DAPI staining in 124 samples from German composting plants. Special attention should be paid to some spe

Plants experiencing chronic internal exposure to ionizing radiation exhibit higher frequency of homologous recombination than acutely irradiated plants.

Ionizing radiation (IR) is a known mutagen responsible for causing DNA strand breaks in all living organisms. Strand breaks thus created can be repaired by different mechanisms, including homologous recombination (HR), one of the key mechanisms maintaining genome stability [A. Britt, DNA damage and repair in plants, Annu. Rev. Plant. Phys. Plant Mol. Biol., 45 (1996) 75-100; H. Puchta, B. Hohn, From centiMorgans to basepairs: homologous recombination in plants, Trends Plant Sci., 1 (1996) 340-348.]. Acute or chronic exposure to IR may have different influences on the genome integrity. DNA Damage-Induced Transcription of Transposable Elements and Long Non-coding RNAs in Arabidopsis Is Rare and ATM-Dependent.

Induction and mobilization of transposable elements (TEs) following DNA damage or other stresses has been reported in prokaryotes and eukaryotes. Recently it was discovered that eukaryotic TEs are frequently associated with long non-coding RNAs (lncRNAs), many of which are also upregulated by stress. Yet, it is unknown whether DNA damage-induced transcriptional activation of TEs and lncRNAs occurs sporadically or is a synchronized, genome-wide response. Here we investigated the transcriptome of Arabidopsis wild-type (WT) and ataxia telangiectasia mutated (atm) mutant plants 3 h after induction

DNA Metabarcoding Approach as a Potential Tool for Supporting Official Food Control Programs: A Case Study.

Food authentication significantly impacts consumer health and the credibility of Food Business Operators (FBOs). As European regulations mandate the verification of food authenticity and supply chain integrity, competent authorities require access to innovative analytical methods to identify and prevent food fraud. This study utilizes the DNA metabarcoding approach on meat preparations, sampled during an official control activity. It assesses animal and plant composition by amplifying DNA fragments of the 12S rRNA and trnL (UAA) genes, respectively. The results not only confirmed the declared

AtRTP5 negatively regulates plant resistance to Phytophthora pathogens by modulating the biosynthesis of endogenous jasmonic acid and salicylic acid.

Plants have evolved powerful immune systems to recognize pathogens and avoid invasions, but the genetic basis of plant susceptibility is less well-studied, especially to oomycetes, which cause disastrous diseases in many ornamental plants and food crops. In this research, we identified a negative regulator of plant immunity to the oomycete Phytophthora parasitica, AtRTP5 (Arabidopsis thaliana Resistant to Phytophthora 5), which encodes a WD40 repeat domain-containing protein. The AtRTP5 protein, which was tagged with green fluorescent protein (GFP), is localized in the nucleus and plasma membr

Anticancer activity of Ilex khasiana, a rare and endemic species of holly in Northeast India, against murine lymphoma.

Ilex khasiana Purkay. is a lesser-known species of holly (family Aquifoliaceae) that is endemic to Northeast India. Designated as critically endangered, the plant is used in the treatments of bacterial infections, cancer, intestinal helminthiasis, tuberculosis, and viral infections. A methanol extract of the leaves was prepared from which 16 different compounds were identified using gas chromatography-mass spectroscopy. An alkylated phenol, 2,6-di-tert-butylphenol, was the predominant compound. Acute toxicity test indicated that the plant extract was non-toxic even at the highest dosage tested

Physcomitrella patens DNA methyltransferase 2 is required for recovery from salt and osmotic stress.

DNA methyltransferase 2 (DNMT2) unlike other members of the cytosine DNA methyltransferase gene family has dual substrate specificity and it methylates cytosines in both the DNA and transfer RNA (tRNA). Its role in plants, however, has remained obscure to date. In this study, we demonstrate that DNMT2 from Physcomitrella patens accumulates in a temporal manner under salt and osmotic stress showing maximum accumulation during recovery, i.e. 24 h after plants are transferred to normal growth medium. Therefore, to study its role in stress tolerance, we generated PpDNMT2 targeted knockout plants (

Cloning of a cDNA from Arabidopsis thaliana homologous to the human XPB gene.

The human gene XPB, defective in xeroderma pigmentosum patients complementation group B, encodes a DNA helicase involved in several DNA metabolic pathways, including DNA repair and transcription. The high conservation of this gene has allowed the cloning of homologs in various species, such as mouse, yeast and Drosophila. Not much information on the molecular basis of nucleotide excision repair in plants is available, but these organisms may have similar mechanisms to other eukaryotes. A homolog of XPB was isolated in Arabidopsis thaliana by using polymerase chain reaction (PCR) with degenerat

RAD4 and RAD23/HMR Contribute to Arabidopsis UV Tolerance.

In plants, exposure to solar ultraviolet (UV) light is unavoidable, resulting in DNA damage. Damaged DNA causes mutations, replication arrest, and cell death, thus efficient repair of the damaged DNA is essential. A light-independent DNA repair pathway called nucleotide excision repair (NER) is conserved throughout evolution. For example, the damaged DNA-binding protein Radiation sensitive 4 (Rad4) in Saccharomyces cerevisiae is homologous to the mammalian NER protein Xeroderma Pigmentosum complementation group C (XPC). In this study, we examined the role of the Arabidopsis thaliana Rad4/XPC h

Anticancer activity with physical property analysis of Mollugo cerviana (L). Ser whole plant extract on hepatocellular carcinoma cell lines (HepG2).

Hepatocellular carcinoma is a rapidly advancing malignancy and remains a leading cause of cancer-related mortality. Recently, plant-based pharmaceuticals have garnered increased attention compared to synthetic medications due to their non-toxic nature and accessibility. Mollugo cerviana, a prevalent weed in the Mollugnaceae family, is recognised for its antioxidant and anti-inflammatory properties. The ethanolic extract of the plant was evaluated for its cytotoxic effects on hepatocarcinoma cell lines (HepG2). The vitality of the treated cells was evaluated using the MTT test. The induction of

Mre11 deficiency in Arabidopsis is associated with chromosomal instability in somatic cells and Spo11-dependent genome fragmentation during meiosis.

The Mre11/Rad50/Nbs1 complex is involved in many aspects of chromosome metabolism. Aberrant function of the complex is associated with defects in the DNA checkpoint, double-strand break repair, meiosis, and telomere maintenance. In this article, we report the consequences of Mre11 dysfunction for the stability of mitotic and meiotic chromosomes in Arabidopsis thaliana. Although plants homozygous for a T-DNA insertion in a conserved region of the MRE11 gene are viable, they exhibit growth defects and are infertile. Analysis of mitotic chromosomes prepared from the mutant plants revealed abundan

Self-DNA acts as an immunostimulant for common bean (Phaseolus vulgaris) that induces defense against pests and diseases and increases seed yield.

Treating plants with plant-derived DNA triggers a strong and usually highly self/nonself-specific immune response. Several studies reported an induction of early signaling responses, resistance gene expression, and increased resistance to certain biotic stressors for self-DNA-treated Arabidopsis and also for crops like bean, lettuce, tomato, maize, or peach. These results indicate that self-DNA might bear potential as a tool for future strategies for a preventive biological pest control. However, it remains to be studied how DNA treatment affects different major defense hormones, to which degr
