An eclectic encounter: ticks feeding on an electric eel and the untapped potential of natural history collections.

Parasites are quite likely the most diverse guild of species on earth. Nevertheless, they remain under-documented despite their impact on the health and ecology of nearly every other species. Natural history collections offer a powerful, underutilized resource for uncovering the hidden diversity of parasites, particularly by preserving specimens that reveal unexpected host-parasite associations. Ticks, although known for their broad host range among terrestrial vertebrates, have never been documented parasitizing fish. Here, the author reports the first known instance of ticks associated with

Natural sex reversal imparts permanent compositional changes to the swamp eel gonadal microbiome.

BACKGROUND: Microbial communities are increasingly recognized for their essential roles in the reproductive system. However, the microbial communities in healthy gonads-neither in the ovary nor the testis-have not been extensively explored, particularly with respect to sex differentiation. Sex reversal is a unique mode of sex differentiation that is a well-documented phenomenon in various animal species, with the swamp eel (Monopterus albus) being a notable example of a hermaphroditic species that undergoes natural female-to-male sex reversal. Thus, swamp eel offers a robust system for explori

The ontogenic gonadal transcriptomes provide insights into sex change in the ricefield eel Monopterus albus.

BACKGROUND: The ricefield eel is a freshwater protogynous hermaphrodite fish and has become an important aquaculture species in China. The s … , wnt4a, fgf16, and foxl2a may be closely associated with the initiation of sex change of ricefield eels. CONCLUSIONS: Collectively, results of the present study confirmed that the down-regulation of female-related genes, such as cyp19a1a, wnt4a, fgf16, and foxl2a, is important for the sex change of ricefield eels. More importantly, some novel genes, …

Nuclear and membrane progestin receptors in the European eel: Characterization and expression in vivo through spermatogenesis.

Characterization of all the progestin receptor genes (PRs) found in the European eel has been performed. There were five membrane PRs (mPRs): mPRα (alpha), mPRAL1 (alpha-like1), mPRAL2 (alpha-like2), mPRγ (gamma), mPRδ (delta) and two nuclear PRs (nPRs or PGRs): pgr1 and pgr2. In silico studies showed that the C and E(F) domains of Pgr are well conserved among vertebrates whereas the A/B domain is not. Phylogeny and synteny analyses suggest that eel duplicated pgr (pgr1 and pgr2) originated from the teleost-specific third whole genome duplication (3R). mPR phylogeny placed three eel mPRs toget

Chemically Contaminated Eel Fed to Pregnant and Lactating Mouse Dams Causes Hyperactivity in Their Offspring.

This study examined whether perinatal exposure to polluted eels (Anguilla anguilla L.) induces changes in the locomotor activity of offspring mice across lifespan (post-natal days (PNDs) 47 - 329), using the open field and the home cage activity tests. Dams were exposed during gestation and lactation, through diets enriched in eels naturally contaminated with pollutants including PCBs. Analysis of the eel muscle focused on the six non-dioxin-like (NDL) indicator PCBs (Σ6 NDL-PCBs: 28, 52, 101, 138, 153 and 180). Four groups of dams (n = 10 per group) received either a standard diet without eel…

Scp3 expression in relation to the ovarian differentiation in the protogynous hermaphroditic ricefield eel Monopterus albus.

Synaptonemal complex protein 3 (Scp3), which is encoded by scp3, is a meiotic marker commonly used to trace the timing of gonadal differentiation in vertebrates. In the present study, the ricefield eel scp3 cDNA was cloned, and a fragment encoding amino acids 49 to 244 was overexpressed. The recombinant Scp3 polypeptide was purified and used to generate a rabbit anti-Scp3 polyclonal antiserum. In adult ricefield eels, scp3 mRNA was predominantly detected in the gonads and faintly detected in discrete brain areas. In the gonads, Scp3 immunoreactivities were shown to be localized to the germ cel…

Accumulation of neurotoxic organochlorines and trace elements in brain of female European eel (Anguilla anguilla).

Xenobiotics such as organochlorine compounds (OCs) and metals have been suggested to play a significant role in the collapse of European eel stocks in the last decades. Several of these pollutants could affect functioning of the nervous system. Still, no information is so far available on levels of potentially neurotoxic pollutants in eel brain. In present study, carried out on female eels caught in Belgian rivers and canals, we analyzed brain levels of potentially-neurotoxic trace elements (Ag, Al, As, Cd, Co, Cr, Cu, Fe, Hg, MeHg, Mn, Ni, Pb, Sn, Sb, Zn) and OCs (Polychlorinated biphenyls, P…

Beta3-adrenoceptor in the eel (Anguilla anguilla) heart: negative inotropy and NO-cGMP-dependent mechanism.

Neuroendocrine regulation of cardiac function involves a population of three types of beta-adrenoceptors (ARs). In various mammalian species, beta1- and beta2-AR stimulation produces an increase in contractility; whereas beta3-AR activation mediates negative inotropic effects. At the moment, nothing is known about the physiological role of beta3-AR in fish. Using an isolated working heart preparation, we show that a beta3-AR selective agonist BRL(37344) (0.1-100 nmol l(-1)) elicits a dose-dependent negative inotropism in the freshwater eel Anguilla anguilla. This effect was insensitive to the

Patterns of white muscle activity during terrestrial locomotion in the American eel (Anguilla rostrata).

Eels (Anguilla rostrata) are known to make occasional transitory excursions into the terrestrial environment. While on land, their locomotor kinematics deviate drastically from that observed during swimming. In this study, electromyographic (EMG) recordings were made from white muscle at various longitudinal positions in eels performing undulatory locomotion on land to determine the muscle activity patterns underlying these terrestrial movements. As during swimming, eels propagate a wave of muscle activity from anterior to posterior during terrestrial locomotion. However, the intensity of EMG …

Effect of low temperature on starvation-survival of the eel pathogen Vibrio vulnificus biotype 2.

At present, no reports exist on the isolation of the eel pathogen Vibrio vulnificus biotype 2 from water samples. Nevertheless, it has recently been demonstrated that this biotype can use water as a route of infection. In the present study, the survival of this pathogen in artificial seawater (ASW) microcosms at different temperatures (25 and 5 degrees C) was investigated during a 50-day period, with biotype 1 as a control, V. vulnificus biotype 2 was able to survive in the culturable state in ASW at 25 degrees C in the free-living form, at least for 50 days, entering into the nonculturable st

Mechanisms of acid release in isolated gas gland cells of the European eel Anguilla anguilla.

Mechanisms of acid production and of acid release have been analyzed in isolated gas gland cells of the eel swimbladder using a cytosensor microphysiometer. Incubation of isolated cells with oxamic acid caused a dose-dependent decrease in the rate of proton release. At the highest oxamic acid concentration used (20 mmol/l), proton release was reduced by approximately 40%; incubation with sodium fluoride (10 mmol/l) or removal of glucose from the extracellular medium caused 60 and 80% reduction, respectively. NaCN had little effect on proton secretion. Proton release of isolated gas gland cells

Establishment and application of a detection method for chinese rice-field eels rhabdovirus (CrERV) using the RPA-CRISPR/Cas12a System.

The Chinese rice-field eel rhabdovirus (CrERV) is an emerging pathogen that causes hemorrhagic disease in Chinese rice-field eels (Monopterus albus), leading to epidemic outbreaks, mass mortality, and considerable economic losses in aquaculture. Thus, the development of rapid and reliable diagnostic tools for on-site detection is urgently needed to address this issue. In this study, we established an RPA-CRISPR/Cas12a-based assay for CrERV detection, which exhibited superior sensitivity, specificity, and stability. The assay achieved a detection limit of 10¹ copies/µL. Specificity testing conf…

Ciguatoxin extracted from poisonous moray eels Gymnothorax javanicus triggers acetylcholine release from Torpedo cholinergic synaptosomes via reversed Na(+)-Ca2+ exchange.

Ciguatoxin (CTX) (0.1 pM to 10 nM) added to a suspension of Torpedo synaptosomes incubated in Ca(2+)-free medium caused no detectable acetylcholine (ACh) release. However, subsequent addition of Ca2+ caused a large ACh release that depended on time of exposure, dose of CTX and on [Ca2+]. Tetrodotoxin completely prevented CTX-induced Ca(2+)-dependent ACh release. Simultaneous blockade of Ca2+ channel subtypes by FTX, a toxin extracted from the venom of the spider Agelenopsis aperta, omega-conotoxin and Gd3+ did not prevent ACh release caused by CTX, upon addition of Ca2+. These results suggest

Protein expression patterns and metal metabolites in a protogynous hermaphrodite fish, the ricefield eel (Monopterus albus).

BACKGROUND: The ricefield eel Monopterus albus undergoes a natural sex change from female to male during its life cycle, and previous studies have shown the potential mechanisms of this transition at the transcriptional and protein levels. However, the changes in protein levels have not been fully explored, especially in the intersexual stage. RESULTS: In the present study, the protein expression patterns in the gonadal tissues from five different periods, the ovary (OV), early intersexual stage gonad (IE), middle intersexual stage gonad (IM), late intersexual stage gonad (IL), and testis (TE
