SnWO4-based nanohybrids with full energy transfer for largely enhanced photodynamic therapy and radiotherapy.

The "partial matching" between upconversion nanoparticle (UCNP) emission and absorption by photosensitizers (PSs) often leads to a theoretically reduced therapeutic efficiency in UC-based photodynamic therapy (PDT) strategies in which the chosen PSs have limited capabilities and are unable to utilize all the near-infrared-upconverted light. In this study, needle-like SnWO4 nanocrystals (SWs) with a broad UV-vis absorption region were synthesized to solve the problem. After covalent conjugation with UCNPs, all the UCNP-emitted light was effectively absorbed by SWs, triggering the type-I PDT pro

Design and assessment of novel artificial anal sphincter with adaptive transcutaneous energy transfer system.

This paper presents the in vitro assessment of a novel elastic scaling artificial anal sphincter system (ES-AASS) with an adaptive transcutaneous energy transfer system (TETS) for treatment of severe faecal incontinence (FI). The proposed adaptive TETS has a phase control, which can maintain the output voltage at ∼7 V across the full range of the coupling coefficient variation (from 0.09-0.31) during the whole process of charging with a phase shift of 177.5° to 79.1°. A maximum surface temperature of 42.2 °C was measured above the secondary coil during an energy transmission of 3.5 W in air. T…

Study on the fluorescence resonance energy transfer between CdS quantum dots and Eosin Y.

Water-soluble CdS quantum dots (QDs) were prepared using mercaptoacetic acid (TGA) as the stabilizer in an aqueous system. A fluorescence resonance energy transfer (FRET) system was constructed between water-soluble CdS QDs (donor) and Eosin Y (acceptor). Several factors that impacted the fluorescence spectra of the FRET system, such as pH (3.05-10.10), concentration of Eosin Y (2-80 mg/L) and concentration of CdS QDs (2-80 mg/L), were investigated and refined. Donor-to-acceptor ratios, the energy transfer efficiency (E) and the distance (r) between CdS QDs and Eosin Y were obtained. The resul…

Effect of nanoparticle dimensionality on fluorescence resonance energy transfer in nanoparticle-dye conjugated systems.

Fluorescence resonance energy transfer (FRET) involving a semiconductor nanoparticle (NP) acting as a donor, attached to multiple acceptors, is becoming a common tool for sensing, biolabeling, and energy transfer applications. Such nanosystems, with dimensions that are in the range of FRET interactions, exhibit unique characteristics that are related to the shape and dimensionality of the particles and to the spatial distribution of the acceptors. Understanding the effect of these parameters is of high importance for describing the FRET process in such systems and for utilizing them for differ…

Efficient radiative and nonradiative energy transfer from proximal CdSe/ZnS nanocrystals into silicon nanomembranes.

We demonstrate efficient excitonic sensitization of crystalline Si nanomembranes via combined effects of radiative (RET) and nonradiative (NRET) energy transfer from a proximal monolayer of colloidal semiconductor nanocrystals. Ultrathin, 25-300 nm Si films are prepared on top of insulating SiO(2) substrates and grafted with a monolayer of CdSe/ZnS nanocrystals via carboxy-alkyl chain linkers. The wet chemical preparation ensures that Si surfaces are fully passivated with a negligible number of nonradiative surface state defects and that the separation between nanocrystals and Si is tightly co…

Modeling Energy Transfer Efficiency in Collision-Induced Dissociation and Unfolding Using Molecular Dynamics Simulations.

Collisional activation is available ubiquitously in commercial mass spectrometers, but, due to differences in instrumentation, it can be dif … l activation. A key parameter in IonSPA is the "pseudo-atom mass", which controls the efficiency of energy transfer between an ion's vibrational modes and the gas particle on a collision-by-collision basis. Here, we use molecular dynamic (MD) simulations to study energy transfer efficiency for native charge states of three different protein ions. Energy transfer …

Self-Regulation of Resonance Energy Transfer to Electrochemiluminescence Response Tendency Conversion for Ultrasensitive Immunoassay.

The single-signal multistate conversion sensing mode not only improves detection sensitivity but also features a simple construction process and can effectively eliminate nonspecific signal interferences. The change of molecular structure and generation of new molecules are able to regulate the activation and inhibition of the resonance energy transfer (RET) mechanism more easily through the conversion of energy levels, thus realizing the on-off-on mode. In this work, a controlled release strategy was used to trigger the self-assembly of Bi2S3/BiOI heterojunctions via the release of Na2S, resu…

Localization of Apicomplexa Motor Myosin A with Axial Nanometric Precision Using Graphene Energy Transfer.

Apicomplexan parasites exhibit gliding motility, a fast locomotion mode, yet the precise localization of the migration motor myosin A (MyoA) remains unresolved because of limitations in light microscopy axial resolution, leading to two competing models: one placing MyoA in the cytoplasm and the linear model positioning it between the plasma membrane and the inner membrane complex (IMC), a doubled membrane underneath. To distinguish between these models, we applied graphene energy transfer (GET), a fluorescence-based axial ruler with nanometer precision, and determined the vertical position of …

Boron Enabled Directed [2+2]- and Dearomative [4+2]-Cycloadditions Initiated by Energy Transfer.

A strategy for the photosensitized [2+2]-cycloaddition between styrenyl dihaloboranes and unactivated allylamines to access cyclobutylboronates with control of stereochemistry and regiochemistry is presented. The success of the reaction relies on the temporary coordination between in situ generated dihaloboranes and amines under mild reaction conditions. In addition, cyclobutanes with varying substitution patterns have been prepared using N-heterocycles as directing group. Manipulation of the C-B bond allows for the synthesis of a diverse class of cyclobutanes from simple precursors. Moreover,

All-quantum dot based Förster resonant energy transfer: key parameters for high-efficiency biosensing.

While colloidal quantum dots (QDs) are commonly used as fluorescent donors within biosensors based on Förster resonant energy transfer (FRET), they are hesitantly employed as acceptors. On the sole basis of Förster theory and the well-known behaviour of organic dyes, it is often argued that the QD absorption band over the UV-visible range is too wide. Discarding these preconceptions inherited from classical fluorophores, we experimentally examine the FRET process occurring between donor and acceptor CdTe QDs and provide a mathematical description of it. We evidence that the specific features o…

Measuring Interactions Between Tau and Aggregation Inducers with Single-Molecule Förster Resonance Energy Transfer.

Tau is an intrinsically disordered protein implicated in the pathogenesis of Alzheimer's disease and other neurodegenerative disorders. Here we describe the application of single-molecule Förster resonance energy transfer (smFRET) for the characterization of the interactions between tau and polyphosphate, an intracellular polymer that accelerates tau aggregation. We describe the design of tau constructs, purification and fluorescent labeling of tau, and details of acquisition and analysis of smFRET data. The protocols provided here outline an approach that may be applied to the study of other …

Upconversion nanoparticle-based fluorescence resonance energy transfer assay for Cr(III) ions in urine.

A novel assay of chromium(III) ion based on upconversion fluorescence resonance energy transfer was designed and established. Lysine-capped NaYF(4):Yb/Er upconversion nanoparticles (UCNPs) and dimercaptosuccinic acid-capped gold nanoparticles (AuNPs) were used as the energy donor and acceptor, respectively. They were bound together via electrostatic interaction, resulting in the quenching of the fluorescence of UCNPs by AuNPs. Chromium(III) ions can specifically and strongly interact with dimercaptosuccinic acid that was modified on the surface of AuNPs, leading to the separation of AuNPs from…

Donor-acceptor systems: energy transfer from CdS quantum dots/rods to Nile Red dye.

We demonstrate strong evidence of shape-dependent efficient resonance energy transfer between CdS quantum dots (QDs) and quantum rods (QRs) (donor) to Nile Red dye (acceptor). We also report a simple solution-based method for the preparation of high quality CdS QDs and CdS QRs at relatively low temperature. The observed quenching of PL intensities are 78.8 % and 63.8 % for CdS QDs and QRs, respectively in the presence of Nile Red dye. The calculated energy-transfer efficiencies are 45 % and 19 % from QDs and QRs to dyes, respectively. The energy transfer varies with changing the shape of the n…

Rapid detection of enterotoxigenic clostridium perfringens by real-time fluorescence resonance energy transfer PCR.

Clostridium perfringens is one of the etiologic agents of gas gangrene that can occur when a wound is contaminated with soil. Type A C. perfringens can cause foodborne and nonfoodborne gastrointestinal illnesses due to an enterotoxin (CPE) produced by some strains during sporulation. We developed a quantitative real-time PCR assay based on fluorescence resonance energy transfer hybridization chemistry that targets the C. perfringens-specific phospholipase C (plc) gene and the enterotoxigenic gene (cpe) with the LightCycler and the Ruggedized Advanced Pathogen Identification Device (R.A.P.I.D.)…

Fluorescence resonance energy transfer in CdSe/ZnS-DNA conjugates: probing hybridization and DNA cleavage.

Nucleic-acid-functionalized CdSe/ZnS quantum dots (QDs) were hybridized with the complementary Texas-Red-functionalized nucleic acid. The hybridization was monitored by following the fluorescence resonance energy transfer from the QDs to the dye units. Treatment of the QD/dye DNA duplex structure with DNase I resulted in the cleavage of the DNA and the recovery of the fluorescence properties of the CdSe/ZnS QDs. The luminescence properties of the QDs were, however, only partially recovered due to the nonspecific adsorption of the dye onto the QDs. Similarly, nucleic-acid-functionalized Au nano…

A peptide-based fluorescence resonance energy transfer assay for Bacillus anthracis lethal factor protease.

A fluorescence resonance energy transfer assay has been developed for monitoring Bacillus anthracis lethal factor (LF) protease activity. A fluorogenic 16-mer peptide based on the known LF protease substrate MEK1 was synthesized and found to be cleaved by the enzyme at the anticipated site. Extension of this work to a fluorogenic 19-mer peptide, derived, in part, from a consensus sequence of known LF protease targets, produced a much better substrate, cleaving approximately 100 times more efficiently. This peptide sequence was modified further on resin to incorporate donor/quencher pairs to ge…

Hypothalamic energy balance gene responses in the Sprague-Dawley rat to supplementation of high-energy diet with liquid ensure and subsequent transfer to chow.

Energy dense, high fat, high sugar, foods and beverages in our diet are a major contributor to the escalating global obesity problem. Here, we examine the physiological and neuroendocrine effects of feeding rats a solid high-energy (HE) diet with or without a liquid supplement (Ensure) and the consequence of subsequently transferring animals back to chow (C). Outbred Sprague-Dawley rats were fed C until 49-56 days of age, and then transferred a HE diet for 3 weeks before allocation to one of two weight-matched groups. Over the next 10 weeks, one group remained on HE diet, whereas the other had
