Microbes and microbial enzymes for cyanide degradation.

Cyanide is an important industrial chemical produced on a grand scale each year. Although extremely toxic to mammalian life, cyanide is a natural product generated by fungi and bacteria, and as a result microbial systems have evolved for the degradation of cyanide to less toxic compounds. The enzymes which utilize cyanide as a substrate can be categorized into the following reaction types: substitution/addition, hydrolysis, oxidation, and reduction. Each of these categories is reviewed with respect to the known biochemistry and feasibility for use in treatment of cyanide containing wastes.

Extraadrenal adrenaline formation by two separate enzymes.

Adrenaline (A) is synthesized in the adrenal medullae by the enzyme phenylethanolamine-N-methyltransferase (PNMT). After surgical removal of the adrenal medullae tissue A levels ranged from 22% of control in the heart to 125% of control in the liver. Use of a novel assay to measure tissue A formation revealed that many tissues can synthesize A using PNMT and another enzyme that N-methylates both noradrenaline and dopamine. These enzymes are non-neuronal, inducible and synthesize a major fraction of tissue and urine A.

Inhibition of fibrinolytic enzymes by thrombin inhibitors.

Thrombin inhibitors have recently advanced to the stage of preclinical testing as anticoagulants. However, little is known about the effects of these inhibitors on the enzymes of the fibrinolytic system. In the present study we evaluated the effect of two protein and two synthetic inhibitors of thrombin on tissue plasminogen activator (tPA), urokinase, and plasmin. We found that hirudin inhibited the amidolytic activity of plasmin but had no effect on tPA or urokinase. Antithrombin III inhibited plasmin and urokinase but had no effect on tPA. D-Phe-Pro-Arg-CH2Cl inhibited plasmin and tPA but h…

Naltrexone: lack of effect on hepatic enzymes.

A number of studies have established the clinical efficacy of naltrexone in the treatment of opiate addiction. However, questions have been … trexone package insert, the benefit of admitting patients with the sole problem of elevated hepatic enzymes generally exceeds the risk.

The induction of microsomal electron transport enzymes.

Liver endoplasmic reticulum contains as NADPH-dependent electron transport complex where the family of hemeproteins, termed cytochrome P-450, serve as catalysts for the oxidation of a variety of different organic chemicals. The content and inventory of the types of cytochrome P-450 is readily modified following in vivo treatment of animals with 'inducing agents' such as barbiturates, steroids and polycyclic hydrocarbons. Recent studies have applied the methods of molecular biology to evaluate changes in the transcription and translation of genomic information occurring concomitant with the ini

CSF enzymes in lacunar and cortical stroke.

Cerebrospinal fluid enzyme levels of creatine kinase (CK), lactate dehydrogenase (LDH), glutamate oxaloacetate transaminase (GOT) and angiotensin converting enzyme (ACE) were studied in 40 acute stroke patients comprising 20 lacunar strokes and 20 cortical strokes. A marked elevation of at least one of the enzymes CK, GOT or LDH was seen in 80% of cases of cortical strokes. No elevation was seen in lacunar stroke with CK, GOT or ACE and only a slight elevation with LDH. Within the cortical group, there was a correlation between the site, size of infarction seen on CT scan and enzyme level. The…

The oxygen sensing characteristics of microsomal enzymes.

The microsomal electron transport complex, in particular the segment associated with cytochrome P-450 function, is both qualitatively and quantitatively an important contributor to the cellular respiration of tissues such as liver. Although our knowledge is still limited, it is apparent that oxygen plays a pivitol role in dictating the mode of substrate hydroxylation or the generation of hydrogen peroxide. As illustrated in Figure 7, current evidence suggests that hydrogen peroxide is formed by the dismutation of the superoxide anion resulting from the dissociation of oxycytochrome P-450. Of i
