Root coverage using submarginal free mucosal grafts in conjunction with a retained gingival collar.

A modified technique using a submarginal free mucosal graft has been developed to aid in root coverage. The gingival collar of keratinized tissue is relaxed via a horizontal or semilunar incision and coronally placed. The periosteum is retained on the osseous structures of the recipient site. A free mucosal graft from the palate is sutured submarginally, while the dentist places tension on the gingival collar in an incisal direction. This method has been found, in 8 years of study, to be a predictable way for the periodontist to cover receded gingival tissue where esthetics or root sensitivity

Some considerations on the role of cementum in periodontal disease.

The protective role of cementum against root resorption is discussed in relation to buried teeth, orthodontic treatment, replantation and periodontal disease. The essential feature seems to be a layer of partially calcified precementum or vital cementoblasts. Root resorption close to the epithelial attachment, although not common, is probably the result of death of cementoblasts caused by irritants from bacterial plaque, and the presence of severe of prolonged inflammation. Cementum exposed in the pocket can be affected in different ways by these irritants, and rendered unacceptable for re-att…

Secreted adhesion molecules of Strongyloides venezuelensis are produced by oesophageal glands and are components of the wall of tunnels constructed by adult worms in the host intestinal mucosa.

The parasitic female of Strongyloides venezuelensis keeps invading the epithelial layer of the host intestinal mucosa. Upon invasion, it adheres to the surface of the intestinal epithelial cells with adhesion molecules secreted from the mouth. It has been demonstrated that S. venezuelensis are expelled from the intestine because mucosal mast cells inhibit the attachment of adult worms to the mucosal surface. In the present study, we generated specific antibodies against secreted adhesion molecules to investigate their function in vivo, because these molecules have been demonstrated only in vit

The Multivalent Adhesion Molecule SSO1327 plays a key role in Shigella sonnei pathogenesis.

Shigella sonnei is a bacterial pathogen and causative agent of bacillary dysentery. It deploys a type III secretion system to inject effector proteins into host epithelial cells and macrophages, an essential step for tissue invasion and immune evasion. Although the arsenal of bacterial effectors and their cellular targets have been studied extensively, little is known about the prerequisites for deployment of type III secreted proteins during infection. Here, we describe a novel S. sonnei adhesin, SSO1327 which is a multivalent adhesion molecule (MAM) required for invasion of epithelial cells

Effect of short chain fatty acids on human gingival epithelial cell keratins in vitro.

Hemidesmosomal attachment of the junctional epithelial cells to the tooth and the ability of the attached cells to divide are essential features of the healthy dentogingival junction. Short chain fatty acids are bacterial metabolites associated with gingival inflammation and periodontal pockets. In vitro, short chain fatty acids have been shown to inhibit epithelial cell division and increase the density of their keratin filaments. This study examined these keratin changes by making use of human gingival keratinocyte cultures, gel electrophoresis and Western blot. Short chain fatty acids, buty

Biofilm formation by Histophilus somni on 3D bovine respiratory tissue cultures.

The development of bacterial biofilms following infection involves bacterial attachment to tissues and maturation of the biofilm matrix (BM). However, studying the development of bacterial biofilms in vivo is difficult to follow. An established example of biofilm formation during host infection is bovine respiratory disease (BRD) due to Histophilus somni. To more accurately investigate in vivo biofilm development, three-dimensional bovine airway organoids comprised of bovine turbinate epithelial (BE) and pulmonary artery endothelial (BAE) cells embedded in type I collagen were developed. Cell-

Pathogenicity island-dependent activation of Rho GTPases Rac1 and Cdc42 in Helicobacter pylori infection.

Helicobacter pylori has been identified as the major aetiological agent in the development of chronic gastritis and duodenal ulcer, and it plays a role in the development of gastric carcinoma. Attachment of H. pylori to gastric epithelial cells leads to nuclear and cytoskeletal responses in host cells. Here, we show that Rho GTPases Rac1 and Cdc42 were activated during infection of gastric epithelial cells with either the wild-type H. pylori or the mutant strain cagA. In contrast, no activation of Rho GTPases was observed when H. pylori mutant strains (virB7 and PAI) were used that lack functi

Inhibition of CD44 N- and O-linked glycosylation decreases endometrial cell lines attachment to peritoneal mesothelial cells.

The attachment of endometrial epithelial cells (EECs) and endometrial stromal cells (ESCs) to peritoneal mesothelial cells (PMCs) with and without inhibition of N- and O-linked glycosylation, the viability of EECs and ESCs, and the expression of CD44 surface density were evaluated. Inhibition of CD44 N- and O-linked glycosylation by using tunicamycin and/or B-GalNAc statistically significantly inhibited endometrial cell attachment to peritoneal mesothelial cells, suggesting a role in establishment of early endometriotic lesions.

Enteropathogenic E. coli shows delayed attachment and host response in human jejunum organoid-derived monolayers compared to HeLa cells.

Enteropathogenic E. coli (EPEC) attaches to host intestinal epithelial cells, resulting in severe illness and diarrhoea. Different cell models have been used to study EPEC infection, but a direct comparison of infection of cell lines and primary cells is lacking. In this study, we compare EPEC infection in primary epithelial cells with HeLa cells. Jejunal organoid-derived cells contain differentiated intestinal epithelial cells and form a tight monolayer. Upon infection, they retain integrity over longer time periods than HeLa cells. Attachment of EPEC to host cells and innate immune response

The regenerative potential of the periodontal ligament.

The current decline in the incidence of dental caries indicates that patients will retain most of their dentition. Reconstruction or regeneration of the entire attachment apparatus, including the periodontal ligament, cementum, and bone, is an attainable therapeutic goal. Clinical treatments designed to reattach connective tissue to exposed root surfaces commonly result in the formation of a long junctional epithelial attachment. This attachment is probably not an effective barrier to bacterial toxins and may allow recurrent pocket formation. To enhance reattachment of connective tissue, root-…

Surgical management of invasive cervical resorption using resin-modified glass ionomer cement.

Invasive cervical resorption is an external resorption that begins below the epithelial attachment. It is caused primarily by dental trauma, orthodontic treatment, or dental bleaching. This case report involved an invasive Class III cervical resorption resulting from trauma to the superior right central incisor. Root canal treatment was followed by surgical intervention. The resorptive defect was debrided, and part of the tooth was restored with resin-modified glass ionomer cement. Postoperative follow-up revealed complete healing and healthy gingival attachment.

Ultrastructural aspects of feeding and secretion-excretion by the equine parasite Strongylus vulgaris.

Light, scanning, and transmission electron microscopy were employed to provide further data on the putative origins of the immunogenic secretory-excretory product (ESP) of Strongylus vulgaris (Looss 1900). The sharply delineated but superficial attachment to the equine caecum by the mouth leaves behind an oval area devoid of epithelial cells. Attachment does not extend deeply enough to reach the muscularis mucosa layer of the equine intestine. The progressive digestion of the ingested plug of tissue (epithelial cells, blood cells and mucous) was visualized. The coelomocytes, floating cells and

Cryptosporidium parvum invasion of biliary epithelia requires host cell tyrosine phosphorylation of cortactin via c-Src.

BACKGROUND & AIMS: Cryptosporidium parvum invasion of epithelia requires polymerization of host cell actin at the attachment site. We analyzed the role of host cell c-Src, a cytoskeleton-associated protein tyrosine kinase, in C. parvum invasion of biliary epithelia. METHODS: In vitro models of biliary cryptosporidiosis using a human biliary epithelial cell line were used to assay the role of c-Src signaling pathway in C. parvum invasion. RESULTS: c-Src and cortactin, an actin-binding protein and a substrate for c-Src, were recruited to the parasite-host cell interface during C. parvum invasi

Attachment of Neisseria meningitidis to human mucosal surfaces: influence of pili and type of receptor cell.

The factors that determine attachment of meningococci predominantly to the mucosa of the nasopharynx rather than to other mucosal surfaces are unknown. Isolates of Neisseria meningitidis from the nasopharynx of carriers and from patients with meningococcal disease were found to be heavily piliated. Isogenic piliated and nonpiliated meningococcal clones were derived from blood and cerebrospinal fluid isolates. Meningococci with pili consistently attached to human nasopharyngeal cells in greater numbers than meningococci without pili. Meningococci treated with trypsin or mechanical shear forces

The significance of podoplanin expression in human inflamed gingiva.

Podoplanin, a transmembrane glycoprotein, has been considered to be expressed specifically by lymphatic endothelial cells. However, recent studies have shown that the protein is expressed in a variety of normal as well as neoplastic tissues, and that its expression might be related to cell migration and invasion. In this study, we examined podoplanin expression in inflamed gingival tissues using an immunohistochemical method. Positive immunoreactivity for podoplanin was found in the cell membrane and cytoplasm of basal cells of oral gingival epithelium when severe inflammatory cell infiltratio

Human bronchial epithelial cells secrete laminin 5, express hemidesmosomal proteins, and assemble hemidesmosomes.

Epithelial cells attach to the basement membrane through adhesive contacts between the basal cells of the epithelium and the proteins of the extracellular matrix (ECM). The hemidesmosome (HD) is a specialized cell-ECM contact, that mediates the attachment of the epithelial cell basal surface to the ECM. In bronchial epithelial cells, the protein components that constitute the HD have not been demonstrated. Using immunohistochemical techniques, we determined that normal human bronchial epithelial (NHBE) cells express the HD cell surface integrin alpha6beta4 and produce laminin 5, the ECM protei
