Effects of intratracheal Fusarium solani inoculation in immunocompetent mice.

Fusariosis is an infection that is caused by fungi of the Fusarium genus. It is the second most common fungus that is associated with human fungal infections, usually in immunocompromised individuals. The incidence of such infections has been increasing, including in immunocompetent hosts. Studies of host-pathogen interactions are scarce, and the pathophysiology of the disease is unknown. One limitation of such studies is the lack of adequate techniques for mammalian infection, in which no standardized protocols have been established with fungi with a focus on the respiratory tract. The aim of…

Fusarium species intramedullary spinal cord fungus ball: case report.

The Fusarium species are one of the most common opportunistic fungal infections occurring in immunocompromised patients and are associated with high morbidity and mortality. Common sites of infection include blood, skin, nasal passages, lungs, bone, and other visceral organs. There is a paucity of literature on Fusarium infections in the brain, and the true nature and extent of central nervous system involvement is not well described. To the authors' knowledge, there have been no reported cases of Fusarium infection of the spine. The authors report the case of a man with acute myeloblastic leu…

Keratitis secondary to Fusarium spp. in Spain 2012-2014.

OBJECTIVE: To describe the clinical-epidemiological characteristics of a case series of fungal keratitis associated with Fusarium spp.., in Spain during the years 2012 to 2014. METHODS: A retrospective study of a case series was conducted on a systematic random sample of 10% of patients identified in Health Centres of provincial capitals (n=250). The centres were asked whether they had been presented with cases of Fusarium spp. keratitis characterised by microbiological methods. Of the 23 centres that responded, 14 had detected cases, with 13 of them accepting to participate, and 11 of them c…

[Invasive fungal disease due to Scedosporium, Fusarium and mucorales].

The number of emerging organisms causing invasive fungal infections has increased in the last decades. These etiological agents include Scedosporium, Fusarium and mucorales. All of them can cause disseminated, virulent, and difficult-to treat infections in immunosuppressed patients, the most affected, due to their resistance to most available antifungal agents. Current trends in transplantation including the use of new immunosuppressive treatments, the common prescription of antifungal agents for prophylaxis, and new ecological niches could explain the emergence of these fungal pathogens. Thes…

Bilateral Fusarium oxysporum keratitis after laser in situ keratomileusis.

UNLABELLED: We report the successful management of a rare case of bilateral post-laser in situ keratomileusis (LASIK) Fusarium oxysporum keratitis and propose a therapeutic strategy. A 19-year-old white man with no systemic diseases was referred to our emergency service 3 days after microkeratome-assisted myopic bilateral LASIK correction. He complained of blurred-vision, photophobia, and ocular pain. Clinical findings (satellite lesions, hypopyon) suggested fungal keratitis. Flaps were immediately lifted and rinsed with povidone-iodine 10%, and intensive topical and systemic and combined anti…

A murine model of contact lens-associated fusarium keratitis.

PURPOSE: Fusarium solani and F. oxysporum were the causative organisms of the 2005/2006 outbreak of contact lens-associated fungal keratitis in the United States. The present study was an investigation of the ability of F. oxysporum grown as a biofilm on silicone hydrogel contact lenses to induce keratitis. METHODS: A clinical isolate of F. oxysporum was grown as a biofilm on lotrafilcon A contact lenses, and a 2-mm diameter punch was placed on the abraded corneal epithelium of either untreated or cyclophosphamide-treated C57BL/6 mice or of IL-1R1(-/-), MyD88(-/-), TLR2(-/-), or TLR4(-/-) mic…

Emerging moulds: Fusarium, Scedosporium and Zygomycetes in transplant recipients.

PURPOSE OF REVIEW: The incidence of non-Aspergillus mould infections in transplant recipients has increased over the past decade. This paper will review recent data regarding the epidemiology, clinical manifestations, treatment and the outcome of infections caused by Fusarium spp., Scedosporium spp. and the Zygomycetes in transplant recipients. RECENT FINDINGS: Although the reasons for the increased incidence of non-Aspergillus mould infections in transplant recipients are not very clear, changes in transplant procedures, resulting in more severe immunosuppression, seem to play a major role, …

Fungal pathogens of plants: deciphering the mechanisms of Fusarium wilt.

Fusarium oxysporum (Fo) is among the most dangerous soilborne pathogens, causing Fusarium wilt and root rots in over 100 plant species worldwide. Some pathogen strains can also infect immunocompromised animals and humans. Consequently, studying the molecular mechanisms associated with pathogen virulence and the plant immune response at different stages of disease development is of paramount importance. The process of host recognition by the pathogen and all stages of the infection process involve a wide repertoire of specific signaling molecules, effector proteins, receptor complexes, as well …

Characterization of a biofilm formed by Fusarium oxysporum on the human nails.

BACKGROUND: Fusarium spp. has been considered as an onychomycosis agent, but little is known about the etiopathogenesis of fusarial onychomycosis; thus, the objective of this study was to characterize the fungal-nail interaction and the consequences of the nail infection process by Fusarium oxysporum using the human nail, in an ex vivo model. METHODS: The kinetic of biofilm production and infection by F. oxysporum using the nail as the only nutritional source were evaluated by scanning electron microscopy, number of culturable cells, metabolic activity, characterization of extracellular matri…

Ungual hyalohyphomycosis caused by Fusarium proliferatum in an immunocompetent patient.

The patient was a 73-year-old healthy female farmer who had been treated with terbinafine for 25.5 months by a primary physician. She exhibited a discoloration and thickening of the right big toenail. She had no concomitant paronychia. Direct microscopy revealed chlamydoconidia and hyphae, and periodic acid-Schiff stained nail specimen showed septate hyphae. On the basis of these morphological features and gene analysis, the final diagnosis was ungual hyalohyphomycosis caused by Fusarium proliferatum. Topical application of 10% efinaconazole solution cured the disease in 10 months.

Molecular mechanism of resistance of Fusarium fujikuroi to benzimidazole fungicides.

Although carbendazim (MBC) and other benzimidazole fungicides have effectively controlled bakanae disease of rice (which is caused by Fusarium fujikuroi, F. proliferatum, and F. verticillioides) in the past, MBC resistance has become common. Previous research has shown that MBC resistance results from a mutation in the β1 -tubulin (β1 tub) gene in F. verticillioides. However, MBC resistance in F. fujikuroi, a predominant species in China, does not result from a mutation in the β1 tub. The molecular mechanism of F. fujikuroi resistance against benzimidazole fungicides is poorly understood. In t…

[Fusarium as a cause of onychomycosis resistant to common antifungal therapy].

INTRODUCTION: Fusarium species are common soil saprophytes and plant pathogens. Members of the genus have been frequently reported as etiologic agents of opportunistic infections in humans and animals. We report six cases of confirmed or suspected onychomycosis caused by members of the genus Fusarium (F. solani and F. oxysporum species complexes). MATERIAL AND METHODS: The isolates were identified by rDNA ITS sequencing analysis. The EMBL accession numbers for the ITS are HE974453-HE974458. A disk diffusion method was used for in vitro susceptibility testing. Comparison of disks (ITEST) and N…

Fusarium Solani: a causative agent of skin and nail infections.

Fusarium spp are non-dermatophytic hyaline moulds found as saprophytes and plant pathogens. Human infections are probably a result of various precipitating predisposing factors of impaired immune status. Immunocompetent individuals of late are also vulnerable to various unassuming saprophytic and plant pathogens. To stress the need to identify correctly and institute appropriate antifungal therapy in newly emerging human fungal infectious agents. Repeated mycological sampling of the skin and nails of the suspected fungal infection were processed as per the standard format including direct micr…

Effects of Fusarium mycotoxin butenolide on myocardial mitochondria in vitro.

Fusarium mycotoxin toxicosis has been implicated in the etiology of Keshan disease, an endemic mitochondrial cardiomyopathy prevailing in certain areas of China. Butenolide (4-acetamido-4-hydroxy-2-butenoic acid gamma-lactone) is one of the Fusarium mycotoxins which are frequently detected from cereal grains in endemic areas. A recent study indicates that this mycotoxin induces rat cardiotoxicity, but its effect on the myocardial mitochondria remains unclear. The present study is therefore undertaken to explore the toxic effect potential of butenolide on the myocardial mitochondria. Exposure o…

Virulence studies of Scedosporium and Fusarium species in Drosophila melanogaster.

We developed a model of infection, using 1 Fusarium species (F. moniliforme) and 2 Scedosporium species (S. apiospermum and S. prolificans), in wild-type and Toll-deficient Drosophila melanogaster flies by injecting them with a standardized conidia inoculum. Wild-type flies were resistant to infection with both S. apiospermum and S. prolificans but were susceptible to infection with F. moniliforme. When Toll-deficient flies were infected with any of the 3 pathogens, the result was acute infection and high mortality rates. Voriconazole mixed in fly food significantly protected flies infected wi…

Gangrenous necrosis of the diabetic foot caused by Fusarium acutatum.

Foot infections are common and serious complications of diabetic patients. We report the case of a 68-year-old patient with a diabetic foot infection that developed into a gangrenous necrosis. Fusarium spp. was isolated on two successive occasions with no other associated microorganisms. Histopathology demonstrated invasion of the fungus into the tisssue. These findings suggested an infection rather than colonization. A detailed morphological study showed that the isolate was Fusarium acutatum, which was confirmed by rDNA sequencing. This fungus is found only in Asia and has not been previousl…
