Regulation and function of Zip4, the acrodermatitis enteropathica gene.

The SLC39A (solute carrier 39A) [ZIP (Zrt-Irt-like protein)] family consists of 14 members which are thought to control zinc uptake into the cytoplasm. Among these, ZIP4 is known to be particularly important for zinc homoeostasis. Mutations in this gene cause acrodermatitis enteropathica, a rare recessive-lethal human genetic disorder. In the present paper, our studies of the regulation and function of the mouse Zip4 gene are briefly reviewed. Mouse Zip4 is expressed at highest levels in tissues involved in absorption of dietary or maternal zinc, and the gene and protein are dynamically regula

[Characterization of novel genes regulating adipocyte differentiation].

The mid- and late stages of adipocyte differentiation are known to be regulated by transcription factors such as peroxisome proliferator-activated receptor (PPAR)gamma and CCAAT-box/enhancer binder protein (C/EBP) families. However, events in the early stage of adipocyte differentiation remain largely unknown. To gain insights into the molecular mechanisms underlying the beginning of adipocyte differentiation, we have isolated 102 genes, which are induced at the beginning of the differentiation of mouse 3T3-L1 preadipocytes, using the polymerase chain reaction (PCR)-subtraction method. Of thes

Transcriptional regulation of the Drosophila glial gene repo.

reversed polarity (repo) is a putative target gene of glial cells missing (gcm), the primary regulator of glial cell fate in Drosophila. Transient expression of Gcm is followed by maintained expression of repo. Multiple Gcm binding sites are found in repo upstream DNA. However, while repo is expressed in Gcm positive glia, it is not expressed in Gcm positive hemocytes. These observations suggest factors in addition to Gcm are required for repo expression. Here we have undertaken an analysis of the cis-regulatory DNA elements of repo using lacZ reporter activity in transgenic embryos. We have f

Denitrification genes regulate Brucella virulence in mice.

Brucella is the causative agent of the zoonotic disease brucellosis, which is endemic in many parts of the world. Genome sequencing of B. suis and B. melitensis revealed that both are complete denitrifiers. To learn more about the role of denitrification in these animal pathogens, a study of the role of denitrification in the closely related B. neotomae was undertaken. In contrast to B. suis and B. melitensis, it was found that B. neotomae is a partial denitrifier that can reduce nitrate to nitrite but no further. Examination of the B. neotomae genome showed that a deletion in the denitrificat

Lamins: building blocks or regulators of gene expression?

Intermediate filament (IF) proteins are the building blocks of cytoskeletal filaments, the main function of which is to maintain cell shape and integrity. The lamins are thought to be the evolutionary progenitors of IF proteins and they have profound influences on both nuclear structure and function. These influences require the lamins to have dynamic properties and dual identities--as building blocks and transcriptional regulators. Which one of these identities underlies a myriad of genetic diseases is a topic of intense debate.

Retinoid regulation of the phosphoenolpyruvate carboxykinase gene in liver.

The cytosolic PEPCK gene is a model gene for assessing retinoid regulation of liver-specific genes encoding enzymes of carbohydrate metabolism. In vivo, we have demonstrated that the PEPCK gene is inhibited by vitamin A deficiency. Specifically, under conditions of food deprivation, induction of the PEPCK gene is inhibited in the vitamin A deficient mouse. Inhibition of the PEPCK gene by vitamin A deficiency is reversed by all-trans or 9-cis retinoic acid (RA) treatment. In a transgenic mouse model, a -460 and -355 bp PEPCK promoter fragment confers susceptibility to inhibition by vitamin A de

Bacillus anthracis genetics and virulence gene regulation.

The Bacillus anthracis genome consists of an approximately 5.3-Mb chromosome and two plasmids, pXO1 (182 kb) and pXO2 (96 kb). Genetic analysis has focused primarily on the structural genes for the anthrax toxin proteins, pagA, lef, and cya, the biosynthetic genes for capsule synthesis, capB, capC, and capA, and a gene associated with depolymerization of capsule, dep. The three toxin genes are located at distinct loci on pXO1, while the cap and dep genes are arranged in an apparent operon on pXO2. Additional genes that may play a role in B. anthracis virulence include the germination operon ge

Regulation of cytochrome P450 gene transcription by phenobarbital.

The ability of phenobarbital to induce levels of drug metabolism in mammals has been known for over 40 years. However, the molecular mechanisms underlying increased expression of the genes of the key enzyme in drug metabolism, cytochrome P450, have not been elucidated, primarily because in vitro model systems in which the induction could be studied were not available. Transfected primary cultured hepatocytes, transfection of liver in situ, and transgenic mice now provide suitable models for phenobarbital induction. In this review, progress toward understanding the mechanism of phenobarbital in

Regulation of virulence gene expression in pathogenic Listeria.

Dynamic interactions between host and pathogen are characteristic of infections caused by intracellular bacteria. This has favoured the evolution of highly effective control systems by which these pathogens regulate the expression of different virulence factors during sequential steps of the infection process. In the case of the facultative intracellular bacterium Listeria monocytogenes, these steps involve internalization by eukaryotic cells, lysis of the resulting phagosome, replication as well as movement within the host cytoplasm, direct cell-to-cell spread, and subsequent lysis of a doubl

Transcriptional regulation of the murine TCR zeta gene.

The TCR zeta chain plays a significant role in the assembly of the receptor complex and in coupling antigen recognition to the intracellular signal transduction apparatus. Since the zeta protein level is considered the limiting factor for receptor assembly in mature T cells, aberrant expression of the zeta chain affects both receptor structure and function. To understand the regulatory mechanisms controlling zeta gene expression, we characterized the 5' flanking region of the gene. Our analysis reveals the existence of at least three regions within the -784 to +121 fragment involved in the tra

Regulation of kappa immunoglobulin gene transcription in vitro.

An in vitro transcription system has been established with a whole-cell extract from the human Burkitt's lymphoma, Daudi, cell line. The B cell extract has been compared with a HeLa cell extract in an effort to study lymphocyte-specific regulatory factors of kappa light chain gene transcription. Both extracts were capable of transcribing Vk genes and other RNA polymerase II dependent genes. Alpha-amanitin at [0.1 micrograms/ml] completely inhibited the accumulation of transcripts in both systems. At low DNA template concentrations the kappa intronic enhancer stimulated Vk promoter transcriptio

Regulation of gene expression in the Schistosoma mansoni female.

The maturation of females of S. mansoni is a process which depends on the presence of male parasites. Preliminary results indicated that the males may secret hormones, possibly steroid or steroid-like compounds, which regulate the expression of genes directly connected to events pertaining to oogenesis. Because the eggs are important agents in the pathology of schistosomiasis, it is of interest to elucidate the biochemical events associated to this developmental stage. The present work describes the efforts to characterize proteins which regulate the expression of a gene encoding a major prote

Regulation of neuropeptide gene expression by steroid hormones.

Steroid hormones modify several brain functions, at least in part by altering expression of particular genes. Of interest are those genes that are involved in cell-cell communication in the brain, for instance neuropeptide genes and genes that code for enzymes involved in synthesis of neurotransmitters. Steroid regulation of mRNA levels for several genes has been reported, including the genes coding for the neuropeptides vasopressin, corticotropin releasing factor, luteinizing hormone-releasing factor, pro-opiomelanocortin; somatostatin, preproenkephalin, and the enzyme tyrosine hydroxylase. [Regulator of expression of the brachypodism-H gene in mice].

Extracts were prepared from the whole limb buds of the 11 day-old mouse embryos and from the postaxial or preaxial parts of the 12 day-old embryo limb buds. Effects of these extracts on the growth of the 13 day-old bpH/bpH embryo tibia rudiments were studied in vitro. It was shown that the limb buds of the 11- and 12 day-old mouse embryos contain a factor regulating bpH gene expression in differentiating cartilage cells. This factor is present in the postaxial part of the limb bud and is absent in its preaxial one. When the extract from the limb bud postaxial parts of the 12 day-old +/+ embryo

Intertwining genome organisation and gene regulation in Leishmania.

Leishmania are trypanosomatid parasites whose unorthodox genomes challenge classical views of how DNA replication and gene expression are regulated in eukaryotes. Within this unconventional genomic context, chromosome replication, constitutive polycistronic transcription, chromatin architecture, and noncoding RNAs emerge not as discrete regulatory layers but as mutually dependent determinants of gene expression and genome stability. Recent advances have also uncovered an unexpected interconnection between Leishmania's evolutionarily divergent replication stress response machinery and its pecul

The evolution of gene regulation in mammalian cerebellum development.

Gene regulatory changes are considered major drivers of evolutionary innovations, including the cerebellum's expansion during human evolution, yet they remain largely unexplored. In this study, we combined single-nucleus measurements of gene expression and chromatin accessibility from six mammals (human, bonobo, macaque, marmoset, mouse, and opossum) to uncover conserved and diverged regulatory networks in cerebellum development. We identified core regulators of cell identity and developed sequence-based models that revealed conserved regulatory codes. By predicting chromatin accessibility acr
