Glanders, a major zoonotic disease declared eradicated in China, still faces the risk of external reintroduction. The complement fixation test (CFT) is a key serological diagnostic method for glanders. To address the limitations of conventional CFT, such as high reagent consumption, cumbersome titration procedures, and frequent occurrence of atypical partial hemolysis leading to ambiguous interpretation, this study developed a more economical, user-friendly, and clearly interpretable micro-complement fixation test (mCFT). According to the WOAH guidelines and Chinese agricultural industry stand…

Burkholderia mallei is the etiologic agent of glanders. It is difficult to diagnose this zoonotic disease in its early stages. Some methods such as the complement fixation test (CFT) cause some problems for veterinary authorities and financial losses to animal owners due to false-positive results. The mallein test requires appropriate laboratory equipment and skilled personnel. To quickly and accurately diagnose the disease, especially in areas where animals cannot be kept, new methods (such as the Western blot test [WBT]) should be used to identify the disease. This study designed and optimiz…

BACKGROUND: Glanders is a contagious and fatal zoonotic disease of solipeds caused by the Gram-negative bacterium Burkholderia (B.) mallei. Although regulations call for culling of diseased animals, certain situations e.g. wild life conservation, highly valuable breeding stock, could benefit from effective treatment schemes and post-exposure prophylaxis.
RESULTS: Twenty three culture positive glanderous horses were successfully treated during a confined outbreak by applying a treatment protocol of 12 weeks duration based on the parenteral administration of enrofloxacin and trimethoprim plus s…

Criteria for the evaluation of the plasmocoagulase activity of natural isolates and mutant strains of the causative agents of glanders and melioidosis were worked out, which made it possible to subdivide them by this sign into pathogens with high, moderate and low activity. Plasmocoagulase produced by pathogenic Burkholderia was shown to be a thermolabile enzyme, comparatively stable with respect to the action of such chemico-biological agents as hydrogen peroxide and chloramine.

In multiple experiments on golden hamsters, highly sensitive to glanders, the method of the transfer of antibacterial resistance was worked out. The most positive effect was obtained after the subcutaneous injection of saline-suspended marrow cells, obtained from white rats, infected with Burkholderia mallei C5 in a single inoculation and passaged through noninfected white rats 5 times. Out of 60 golden hamsters, treated with such material and subsequently (on day 21) injected with 1.0 x 10(2) B. mallei cells, 42 animals survived (70%).

MICs and MBcCs of minocycline, doxycycline, methacycline and chlortetracycline for 8 strains of Pseudomonas mallei were determined. The chemotherapeutic efficacy of minocycline and doxycycline was studied on golden hamsters and their efficacy indices were estimated in comparison with those of chlortetracycline in the prophylaxis and treatment of experimental malleus. Minocycline was shown to be the most efficient drug in the treatment of malleus. Doxycycline in a dose of 0.25 mg/kg practically had the same efficacy as chlortetracycline in a dose of 25 mg/kg. Methacycline was inefficient.

Combined bacteriological, pathomorphological, luminescent-serological, and electron microscopic studies were used to elucidate some aspects of pathogenesis of glanders in animals with different susceptibility to this infection. The animals highly susceptible to glanders (golden hamsters) were shown to develop the infection of the sepsis type with granulomas of the exudative nature. In the animals highly susceptible to glanders the bacteria were proved experimentally to parasitize in a rod-shaped form within macrophages of the spleen, liver, and lungs as well as extracellularly. In white mice r…

The authors examined the ultrastructure of the causative agents of glanders and melio idosis. It was revealed that the structure of their cell wall and of the cytoplasmic membrane was characteristic of Gram negative bacteria. The cytoplasm of both types of the causative agents showed the presence of ribosomes, membrane structure, nucleoid, and also osmiophilic and osmiophobic inclusions.

Sixty male BALB/c mice were inoculated intraperitoneally with either a sublethal or a lethal dose of Burkholderia mallei China 7 strain, the … indicate that mice are susceptible to B. mallei infection and may serve as an appropriate model for glanders infection in a resistant host such as human beings. Additionally, by immunoelectron microscopy, we showed the presence of type I O-antigenic polysaccharide (capsular) antigen surrounding B. mallei.

UNLABELLED: Glanders is a contagious zoonotic disease caused by Burkholderia mallei that primarily affects horses, donkeys, and mules. It remains endemic in parts of Asia, Africa, the Middle East, and South America, where increasing case reports highlight the need for accessible diagnostic tools. Conventional diagnostic methods are limited by high costs and specialized laboratory requirements, whereas lateral flow assays (LFAs) offer practical, user-friendly, and rapid alternatives. In this study, we developed three LFAs targeting antibodies against the B. mallei antigens Hcp1, GroEL, and whol…

BACKGROUND: Glanders is a rare disease that has been eradicated in many countries but may be difficult to diagnose due to its nonspecific symptoms. This disease, which can be highly fatal if left untreated, is caused by a bacterium called Burkholderia mallei. Humans can get the disease through contact with infected animals, such as horses. Over time, various treatment strategies have been proposed for this disease, and attempts haveeven been made to develop a vaccine, but thus far, no effective vaccine has been developed to prevent it.
