Study protocol of the Hungarian Longitudinal Study of Healthy Brain Aging (HuBA).

BACKGROUND AND PURPOSE: Neurocognitive aging and the associated brain diseases impose a major social and economic burden. Therefore, substantial efforts have been put into revealing the lifestyle, the neurobiological and the genetic underpinnings of healthy neurocognitive aging. However, these studies take place almost exclusively in a limited number of highly-developed countries. Thus, it is an important open question to what extent their findings may generalize to neurocognitive aging in other, not yet investigated regions. The purpose of the Hungarian Longitudinal Study of Healthy Brain Agi

T1 relaxation time is prolonged in healthy aging: a whole brain study.

BACKGROUND: : Measurement of tissue characteristics such as the longitudinal relaxation time (T1) provides complementary information to the volumetric and surface based structural analyses. We aimed to investigate T1 relaxation time characteristics in healthy aging via an exploratory design in the whole brain. The data processing pipeline was designed to minimize errors related to aging effects such as atrophy. METHODS: Sixty healthy participants underwent MRI scanning (28 F, 32 M, age range: 18-78, 30 young and 30 old) in November 2017-March 2018 at the Bilkent University UMRAM Center. Four …

Aging in psoriasis vulgaris: female patients are epigenetically older than healthy controls.

BACKGROUND: Psoriasis vulgaris is a skin autoimmune disease. Psoriatic patients have significantly lowered life expectancy and suffer from various comorbidities. The main goal of the study was to determine whether psoriatic patients experience accelerated aging. As accelerated aging might be the reason for the higher prevalence of comorbidities at lower chronological ages, we also wanted to investigate the relationship between aging and selected parameters of frequent psoriatic comorbidities - endocan, vascular endothelial growth factor and interleukin-17. Samples were obtained from 28 patient

Role of T1 mapping to evaluate brain aging in a healthy population.

PURPOSE: To investigate the relationship between healthy brain aging and T1 relaxation time obtained by T1 mapping. MATERIALS AND METHODS: A total of 211 (102 males, 109 females; age range: 20-89 years; mean age: 54 years) healthy volunteers underwent T1 mapping between July 2018 and January 2019. Regions of interest (ROIs) were placed on T1 maps in different anatomical regions, including the thalamus, putamen, globus pallidus, head of the caudate nucleus, nucleus accumbens, genu of the corpus callosum, and frontal lobe white matter (WM). Additionally, linear and quadratic regression analyses

Default Mode Network Lateralization and Memory in Healthy Aging and Alzheimer's Disease.

Lateralization of default mode network (DMN) functioning has been shown to change with age. Similarly, lateralization of frontal lobe function has been shown to decline in age. The impact of amyloid pathology and the progression of Alzheimer's disease (AD) on resting state lateralization has not been investigated. Due to the preferential involvement of the left hemisphere in verbal tasks, there may be a benefit to higher levels of left-lateralization in the performance of verbal memory tasks. Here we compared functional lateralization of the anterior and posterior DMN between four groups of pa

Sexual dimorphism in healthy aging and mild cognitive impairment: a DTI study.

Previous PET and MRI studies have indicated that the degree to which pathology translates into clinical symptoms is strongly dependent on sex with women more likely to express pathology as a diagnosis of AD, whereas men are more resistant to clinical symptoms in the face of the same degree of pathology. Here we use DTI to investigate the difference between male and female white matter tracts in healthy older participants (24 women, 16 men) and participants with mild cognitive impairment (21 women, 12 men). Differences between control and MCI participants were found in fractional anisotropy (FA

The effects of healthy aging on cerebral hemodynamic responses to posture change.

Aging is associated with an increased incidence of orthostatic hypotension, impairment of the baroreceptor reflex and lower baseline cerebra … icant declines in rCBF, THC and HbO(2), and an increase in Hb, across the age continuum (p < 0.01). Healthy aging did not alter postural changes in frontal cortical rCBF (p = 0.23) and was associated with a smaller magnitude of decline in HbO(2) (p < 0.05) during supine-to-standing posture change. We conclude that healthy aging does not alter postural changes i …

Differential patterns of implicit emotional processing in Alzheimer's disease and healthy aging.

Implicit memory for emotional facial expressions (EFEs) was investigated in young adults, healthy old adults, and mild Alzheimer's disease (AD) patients. Implicit memory is revealed by the effect of experience on performance by studying previously encoded versus novel stimuli, a phenomenon referred to as perceptual priming. The aim was to assess the changes in the patterns of priming as a function of aging and dementia. Participants identified EFEs taken from the Facial Action Coding System and the stimuli used represented the emotions of happiness, sadness, surprise, fear, anger, and disgust.

Different Grey Matter Microstructural Patterns in Cognitively Healthy Versus Typical Ageing Healthy Versus Typical Brain Ageing.

Ageing is a complex phenomenon affecting a wide range of coexisting biological processes. The homogeneity of the studied population is an essential parameter for valid interpretations of outcomes. The presented study capitalises on the MRI data available in the Human Connectome Project-Aging (HCP-A) and, within individuals over 55 years of age who passed the HCP-A section criteria, compares a subgroup of 37 apparently neurocognitively healthy individuals selected based on stringent criteria with 37 age and sex-matched individuals still representative of typical ageing but who did not pass the

Age-Related Alterations in Swallowing in Healthy Aged Rodents: A Systematic Review.

Age-related decline in swallowing function is increasingly recognized as a clinical concern in aging populations. To better understand the biological mechanisms underlying functional and structural changes associated with normal aging, rodent models provide a controlled experimental platform. This review systematically examined age-related changes in swallowing function in healthy rodents, compared with that of young adults. A comprehensive literature search was conducted in PubMed, Embase, and the Cochrane Library. Twenty-nine articles were ultimately included in this review. Age-related alte

Mitochondria-derived peptides in healthy ageing and therapy of age-related diseases.

Mitochondrial-derived peptides (MDPs) are small bioactive peptides encoded by mitochondrial DNA and involved in various stress-protecting mechanisms. To date, eight mitochondrial-derived peptides have been identified: MOTS-c sequence is hidden in the 12 S rRNA gene (MT-RNR1), and the other 7 (humanin and small humanin-like peptides 1-6) are encoded by the 16 S rRNA (MT-RNR2) gene. While the anti-apoptotic, anti-inflammatory and cardioprotective activities of MDPs are well described, recent research suggests that MDPs are sensitive metabolic sensors, closely connected with mtDNA mutation-associ

The Healthy Ageing Questionnaire Index: Validation in the Singapore Longitudinal Ageing Study.

INTRODUCTION: Healthy ageing (HA) indices typically use full questionnaire, performance- or blood-based assessment of functional ability which are time-consuming and resource-intensive. We developed and validated a simple and brief Healthy Ageing Questionnaire (HAQ) index with comparable measurement accuracy. METHODS: The 15-item HAQ (scored 0-100) was developed using data of 500 participants in the Singapore Study of Successful Ageing (SSOSA), a sub-cohort of the Singapore Longitudinal Ageing Study (SLAS-2). Its construct, concurrent, and predictive validity were evaluated in 2,161 participa

Default mode network functional connectivity is associated with intention interference in healthy aging.

Prospective memory (PM), the ability to fulfil an intended action, is important in daily life. However, completed intentions can interfere with subsequent behaviors, known as the aftereffect. The neural basis of this phenomenon remains unclear, particularly from a network perspective. Based on cognitive theories, the present study aimed to examine the underlying neural mechanisms, focusing on the default mode network (DMN) and the frontoparietal network (FPN) to resolve previously inconsistent findings. Thirty-six middle-to-older adults completed a memory-for-intention task, with outdated cues
