Accumulation of formaldehyde causes motor deficits in an in vivo model of hindlimb unloading.

During duration spaceflight, or after their return to earth, astronauts have often suffered from gait instability and cerebellar ataxia. Here, we use a mouse model of hindlimb unloading (HU) to explore a mechanism of how reduced hindlimb burden may contribute to motor deficits. The results showed that these mice which have experienced HU for 2 weeks exhibit a rapid accumulation of formaldehyde in the gastrocnemius muscle and fastigial nucleus of cerebellum. The activation of semicarbazide-sensitive amine oxidase and sarcosine dehydrogenase induced by HU-stress contributed to formaldehyde gener…

Pulsatility Index as a Novel Parameter for Perfusion in Mouse Model of Hindlimb Ischemia.

BACKGROUND/AIMS: In clinical settings, the pulsatility index (PI) has become a widely used tool for monitoring obstetrics or other vascular diseases. It is based on the maximum Doppler shift waveform derived from ultrasonography. However, it remains unclear whether the PI levels are correctly predicted from the perfusion in mouse model of hindlimb ischemia. METHODS: To explore the relationship between PI and perfusion, we generated a unilateral hindlimb ischemia model in 8-week-old C57BL/6 male mice by ligation of the right common iliac artery and femoral artery. These mice were monitored wit…

MRI of iron oxide nanoparticle-labeled ADSCs in a model of hindlimb ischemia.

Adipose-derived stem cells (ADSCs) exhibit tremendous potential for repair of ischemic diseases. However, studies on the fate, migration, di … DSCs were evaluated in vitro. Labeled and non-labeled GFP-ADSCs were injected into a mouse model of hindlimb ischemia, and 3T magnetic resonance imaging (MRI) was acquired. The synthesized DMSA-NPs efficiently labeled the GFP-ADSCs in vitro and in vivo without affecting cell viability, proliferation, cell cycle, and multi-differentiation capacity. The MRI …

Role of the parasympathetic nervous system in cardioprotection by remote hindlimb ischaemic preconditioning.

This investigation was designed to determine the participation of the vagus nerve and muscarinic receptors in the remote ischaemic precondit … entricular end-diastolic pressure. In the rIPC group, the rabbits were subjected to three cycles of hindlimb ischaemia (5 min) and reperfusion (5 min), and the same protocol as that used in non-rIPC group was then repeated. In order to evaluate the afferent neural pathway during the rIPC protocol we used two groups, one in which the femoral and sciatic ner …

Neuromuscular electrical stimulation of the hindlimb muscles for movement therapy in a rodent model.

Neuromuscular electrical stimulation (NMES) can provide functional movements in people after central nervous system injury. The neuroplastic effects of long-term NMES-induced repetitive limb movement are not well understood. A rodent model of neurotrauma in which NMES can be implemented may be effective for such investigations. We present a rodent model for NMES of the flexor and extensor muscles of the hip, knee, and ankle hindlimb muscles. Custom fabricated intramuscular stimulating electrodes for rodents were implanted near identified motor points of targeted muscles in ten adult, female Lo…

Muscarinic control of the excitability of hindlimb motoneurons in chronic spinal-transected salamanders.

The excitability of spinal motoneurons (MNs) is regulated by acetylcholine via the activation of muscarinic receptors. The objective of the present study was to determine whether this cholinergic modulation of MN excitability is altered following a chronic spinal cord transection. Juvenile salamanders (Pleurodeles waltlii) were spinally transected at the mid-trunk level, and patch-clamp recordings from hindlimb MNs in spinal cord slices were performed 9-30 days after transection, with and without bath application of muscarine (20 mum). Our results showed that the input-output relationship was …

Independent development of sensory and motor innervation patterns in embryonic chick hindlimbs.

Previous studies suggest that sensory axon outgrowth is guided by motoneurons, which are specified to innervate particular target muscles. Here we present evidence that questions this conclusion. We have used a new approach to assess the pathfinding abilities of bona fide sensory neurons, first by eliminating motoneurons after neural crest cells have coalesced into dorsal root ganglia (DRG) and second by challenging sensory neurons to innervate muscles in a novel environment created by shifting a limb bud rostrally. The resulting sensory innervation patterns mapped with the lipophilic dyes DiI

Increases in gastric acidity in response to electroacupuncture stimulation of the hindlimb of anesthetized rats.

Acupuncture treatment has been used as physiotherapy to treat gastrointestinal disorders in traditional Oriental medicine. In the present study, we selected anesthetized animals as the subjects of this experiment in order to eliminate emotional factors, and investigated the responses to acupuncture stimulation at only a single point by changing animal conditions. 1) There was marked enhancement of gastric acid secretion by electroacupuncture stimulation of the hindlimb based on pH values of the perfusate. However, the enduring response after the momentary initial decrease suggested the existen…

Pungent principles of ginger (Zingiber officinale) are thermogenic in the perfused rat hindlimb.

Crude extracts of both fresh and dry ginger induced the perfused rat hindlimb to consume oxygen in association with increases in perfusion pressure and lactate production. The principles responsible for these observations, the gingerols and shogaols, were isolated and tested for relative thermogenic activity. The gingerol homologues possessed greater molar potency than their shogaol counterparts. (6)-Gingerol was the most potent principle isolated, causing a mean maximal increase in oxygen consumption of 1.4 +/- 0.1 mumol/g/h (21%), an increase in lactate efflux of 4.7 +/- 0.6 mumol/g/h (87%) …

The interaction of vasodilating drugs and sympathetic blockade in normal and ischemic canine hindlimbs.

The purpose of this study was to determine if intraarterial vasodilating drugs could augment the vasodilation produced by sympathetic blockade, such as occurs during epidural anesthesia. Papaverine (2 mg/min), nitroglycerin (1 microgram/kg/min), aand saline were infused into the femoral artery before and after lumbar sympathectomy in six arterially isolated canine hindlimbs. Femoral blood flow was controlled with a perfusion circuit at baseline (80 ml/min), half-baseline (40 ml/min), and low (5 ml/min) flow rates so that hindlimb mean arterial pressure served as an index of peripheral vasodila…

Optical measurement of perfused rat hindlimb muscle with relation of the oxygen metabolism.

Optical measurements of myoglobin (Mb) and cytochromes (Cyts) in the isolated rat hindlimb muscle perfused with cell-free medium were performed at 35 and 15 degrees C under various oxygen supply, in the relation to the oxygen metabolism. Molar ratio of Cyt a + a3, Cyt b, Cyt c + c1, and Mb, oxygen affinity of Mb (P50), and the thermodynamic parameter (delta H degree) of Mb oxygenation in cyanide-perfused muscle were similar to those reported. The "apparent P50" of Mb in cyanide-free muscle was almost two orders larger in magnitude than P50 in the presence of cyanide. O2 uptake by the perfused …

Differentiation of neurogenic vasoconstrictor responses in skin and skeletal muscle of dog hindlimb.

Femoral blood flow was measured electromagnetically in chloralose-anaesthetised dogs pretreated with atropine and pancuronium, during pregan … sponses in timecourse and amplitude. It is suggested that skin and muscle resistance vessels in the hindlimb receive their primary vasoconstrictor nerve supply via peripheral pathways that can be distinguished by the sensitivity to hexamethonium of their ganglionic synapses, and that the timecourse of constrictor responses in skeletal muscle is longer than …

Area specific reflexes from normal and supernumerary hindlimbs of Xenopus laevis.

Two area specific reflexes elicited by natural stimulation of different regions of the hindlimbs of Xenopus laevis have been identified. Light or intense mechanical stimulation of the foot evokes reflex activity in the ipsilateral knee flexor nerve; moderate pressure applied to the calf evokes reflex activity predominantly in the ipsilateral knee extensor nerve. The reflex responses have been recorded electrophysiologically to overcome the limitations of behavioral observations for determining the presence of activity in particular muscles. Normal area specific reflexes are elicited in the nor…

Development and characterization of rat hindlimb ischemia models mimicking peripheral arterial disease severity.

Peripheral arterial disease (PAD) is a progressive atherosclerotic disorder that reduces blood flow to the lower extremities, leading to skeletal muscle degeneration, functional decline, and limb loss in advanced stages. To enable rigorous preclinical evaluation of regenerative therapies across disease stages, we developed and quantitatively characterized three rat hindlimb ischemia models representing graded arterial insufficiency: iliac artery ligation (Iliac), femoral artery ligation and excision (Femoral), and combined femoral plus popliteal artery ligation and excision (Fem/Pop). Laser Do…

Short term treatment of secondary lymphedema with hyaluronidase injections reduces mouse hindlimb lymphedema.

Lymphedema is a common complication following breast cancer treatment with axillary lymphadenectomy and radiotherapy. Currently, there is no curative treatment for this disease, hence there is a need for new therapeutic suggestions. The aim of this study was to investigate the effect of hyaluronidase (HYAL) injections after inducing hindlimb lymphedema in 36 female C57BL/6 mice. HYAL injections were administered every second day for 14 days in three groups: (1) HYAL for 1 week followed by saline for 1 week, (2) HYAL for 2 weeks, and (3) saline injections for 2 weeks. Volume of the lymphedema l…

Popliteal Vascular Lymph Node Resection in the Rabbit Hindlimb for Secondary Lymphedema Induction.

Lymphedema is a common condition often associated with cancer and its treatment, which leads to damage to the lymphatic system, and current treatments are mostly palliative rather than curative. Its high incidence among oncologic patients indicates the need to study both normal lymphatic function and pathologic dysfunction. To reproduce chronic lymphedema, it is necessary to choose a suitable experimental animal. Attempts to establish animal models are limited by the regenerative capacity of the lymphatic system. Among the potential candidates, the rabbit hindlimb is easy to handle and extrapo…

NADPH Oxidase Accounts for Changes in Cerebrovascular Redox Status in Hindlimb Unweighting Rats.

OBJECTIVE: The roles of cerebrovascular oxidative stress in vascular functional remodeling have been described in hindlimb-unweighting (HU) rats. However, the underlying mechanism remains to be established. METHODS: We investigated the generation of vascular reactive oxygen species (ROS), Nox2/Nox4 protein and mRNA levels, NADPH oxidase activity, and manganese superoxide dismutase (MnSOD) and glutathione peroxidase-1 (GPx-1) mRNA levels in cerebral and mesenteric smooth muscle cells (VSMCs) of HU rats. RESULTS: ROS production increased in cerebral but not in mesenteric VSMCs of HU rats compa…

Hindlimb stretching alters locomotor function after spinal cord injury in the adult rat.

BACKGROUND: Stretching is a widely accepted standard-of-care therapy following spinal cord injury (SCI) that has not been systematically studied in animal models. OBJECTIVE: To investigate the influence of a daily stretch-based physical therapy program on locomotor recovery in adult rats with moderate T9 contusive SCI. METHODS: A randomized treatment and control study of stretching in an animal model of acute SCI. Moderate SCIs were delivered with the NYU Impactor. Daily stretching (30 min/day, 5 days/wk for 8 weeks) was provided by a team of animal handlers. Hindlimb function was assessed u…

Hindlimb paralytic effects of prodynorphin-derived peptides following spinal subarachnoid injection in rats.

Dynorphin A-(1-17) acts through non-opioid mechanisms to produce dose-related neurological deficits following injection into the lumbar spinal subarachnoid space in rats. Hindlimb motor function was examined following subarachnoid injection of dynorphin A fragments and other opioid peptides derived from prodynorphin to establish: (1) which portion(s) of the dynorphin A molecule cause hindlimb motor dysfunction, and (2) whether these paralytic actions are shared by other opioids (dynorphin B, alpha-neo-endorphin, and beta-neo-endorphin) derived from the same promolecule. To minimize the influen…
