Cutaneous Rosai-Dorfman disease with MAP2K1 mutation, initially mimicking an infection with parasitized histiocytes.

Rarely, Rosai-Dorfman disease (RDD) manifests exclusively in the skin, typically as nodules on the trunk and extremities. Recognition of characteristic histopathologic features enables diagnosis of RDD. A 55-year-old female presented with a 7-year history of cutaneous nodules involving the trunk and extremities. A prior skin biopsy specimen at a different institution had demonstrated a dense dermal lymphohistiocytic infiltrate with histiocytes containing GMS+ forms, favored to represent cryptococcal organisms, with a differential diagnosis including other infections with parasitized histiocyte…

Foamy histiocytes in a patient with visceral leishmaniasis after treatment with liposomal amphotericin B.

A seven-year-old patient was admitted to our hospital with the complaints of fever, malaise, and abdominal distention. In the bone marrow aspiration smears, leishmania amastigotes were observed. The patient was diagnosed as having visceral leishmaniasis (VL) and treated with liposomal amphotericin B (L-AMB). The authors report their observation of foamy histiocytes seen in the bone marrow aspiration smears of the patient with VL after L-AMB treatment. This effect of L-AMB needs to be recognized, since L-AMB may represent a further condition in addition to the other diseases that are associated…

Detection of large histiocytes in pap smears: role in the prediction of endometrial pathology?

OBJECTIVE: The objective of this study was to clarify the prognostic significance of large histiocytes in Papanicolaou smears. METHOD: During a 2-year period, 893 smears were assessed by the Cytology Laboratory of our Hospital. Twenty-one smears were reported including large histiocytes (2.4%). Colposcopy was also performed in all of these women. RESULTS: 9/21 of the women (43%) presented vaginocervicitis, whereas 3/21, 1/21, 1/21, 1/21 and 1/21 revealed endometrial polyps, complex hyperplasia with atypia, simple hyperplasia after tamoxifene use, submucosal myoma and endometrial carcinoma re…

Sea-blue histiocytes in idiopathic thrombocytopenic purpura. The first report from the Indian subcontinent.

Sea-blue histiocytes are described in the bone marrow of a 15-year-old male patient suffering from idiopathic thrombocytopenic purpura. Cytochemically the storage material in these cells appeared to be glycophospholipid. Lipid analysis of blood leucocytes and nucleated cells of bone marrow showed a significant increase in the phospholipids, triglycerides and cholesterol as compared to age-matched controls. Serum lipid analysis showed a mild increase of phospholipids as compared to age-matched controls.

Bronchial cytologic changes during cardiopulmonary bypass: iron transport by histiocytes in low flow states.

We previously reported (1) that tracheobronchial secretions of patients in hemorrhagic shock contained significantly more iron laden histiocytes than secretions of matched controls and demonstrated by histiologic studies in rats bled to hemorrhagic shock that these animals' lungs contained significantly more iron laden histiocytes than those of a control group. Two explanations appeared plausible: 1) abnormal hemoglobin breakdown products of systemic hypoperfusion during shock (2, 3, 5) may have accumulated in the pulmonary circulation where they were phagocytozed by pulmonary histiocytes, or …

Monocytes and histiocytes in cell cultures of cerebrospinal fluid. Morphology of cultured CSF cells.

A method of CSF cell culturing, based on observations of cultured cells isolated from 700 CSF specimens obtained for routine diagnostic procedures by lumbar puncture from patients who had no proven or suspected neoplastic disease, is described which enables the demonstration of proliferating mononuclear elements even when they are present in specimens with low cell count. Spread on surfaces of plastic and glass material, monocytes and histiocytes in CSF cell cultures can appear as polygonal or crescent shaped epitheloid cells, may assume spindle shapes, or transform into multinucleated giant c…

Immunophenotypic analysis of histiocytes involved in AIDS-associated Mycobacterium scrofulaceum infection: similarities with lepromatous lepra.

The present study reports a rare case of systemic M. scrofulaceum infection in an AIDS patient and analyses the inflammatory infiltrate in a lymph node by immunohistochemistry. Special emphasis is put on the histiocytes. The diffuse infiltrate consists mainly of large histiocytes that contain numerous bacilli. These cells display the phenotype of mature histiocytes and in addition coexpress the antigens recognized by RFD7 and RFD9, both markers of different subsets of histiocytes which have been reported to be co-expressed by the infected histiocytes in the infiltrate of lepromatous lepra. Int…

Regressing atypical histiocytosis: a cutaneous proliferation of atypical neoplastic histiocytes with unexpectedly indolent biologic behavior.

Regressing atypical histiocytosis (RAH) of skin is a cutaneous noduloulcerative proliferation of atypical neoplastic histiocytes with concomitant polymorphous inflammation, frequently pronounced epidermal hyperplasia, and an unexpectedly indolent biologic course. Spontaneous regression and recurrence without systemic spread were the course in follow-up periods of over six years. Histopathologically, characteristic-appearing atypical mononuclear and multinucleated "RAH" cells showed erythrophagocytosis as well as ultrastructural, surface marker, and enzyme cytochemical features indicating histi…

Intratubal Pseudopapillary Histiocytic Hyperplasia: A New Histologic Variant in the Spectrum of Histiocytic Lesions Involving the Fallopian Tube.

Although histiocytic lesions of the fallopian tube are relatively rare compared to their epithelial counterparts, there exists a spectrum of histiocytic lesions involving the fallopian tube that are described under different terminologies dependent on the involved compartment of the fallopian tube. A common histologic denominator of all the hitherto reported tubal histiocytic lesions is the presence of sheets and clusters of histiocytes without any supportive connective tissue. The current study describes three cases of a heretofore-undescribed papillary histiocytic lesion in the lumen of the …

T-cell lymphomas of large cell type. A variety of malignant lymphomas: "histiocytic" and mixed lymphocytic-"histiocytic".

Clinical and morphologic features of seven T-cell lymphomas of the large cell type are described. The tumors were grouped into those with irregular (3 cases) and those with round and regular nuclei (4 cases). In both groups, variation in cell size, numerous histiocytes and vessels, and many mitoses were distinguishing features. In only 1 case in the round and regular nucleus group was there relatively little variation in cell size and a paucity of histiocytes. Abundant polyribosomes, long strands of rough endoplasmic reticulum, and lysosomal granules were prominent electron microscopic feature…

Successful Treatment of Histiocytic Sarcoma in Childhood.

Histiocytic sarcoma (HS) is a rare malignant neoplasm that belongs to malignant histiocytoses. HS primarily occurs in adult males and is exceptionally uncommon in pediatric populations. Extranodal regions represent the most common sites of HS manifestation. Diagnosis necessitates histopathological evidence of neoplasm, along with verification of cellular origin through specialized studies. Here, we describe a case of a three-year-old female toddler with HS in her calf that was successfully treated with the chemotherapy protocol of Langerhans Cell Histiocytosis (LCH)-IV, the International Colla

PDL1 Expression and Outcomes in Histiocytic Neoplasms.

INTRODUCTION: Histiocytic sarcoma (HS) and interdigitating dendritic cell sarcoma (IDCS) are both extremely rare and aggressive histiocytic neoplasms that are most often treated with lymphoma-based chemotherapy; however, there is growing use of immune checkpoint inhibitors (ICI) in the second- or further-line settings as prognosis is typically poor in these neoplasms. CASE PRESENTATIONS: We describe two cases: first, HS in a 60-year-old man with primary tumor in the right anterior lower limb, and second, IDCS in an 82-year-old man in the right para-parotid region after initial wide local exci

Histiocytic sarcoma of the spleen without thrombocytopenia.

One case of histiocytic sarcoma of the spleen is reported. The patient, a 54-year-old female, presented with a huge soft tissue mass with a clear border and pseudo capsule in the splenic parenchyma on CT. The density of the mass was uneven, multifocal cystic necrosis and irregular bleeding were observed inside, but no calcification could be seen. On contrast-enhancement scan, the solid component of tumor exhibited moderate progressive enhancement, and area of cystic necrosis exhibited no enhancement. In MRI, T1WI showed mixed low signal, and T2WI showed mixed slightly high signal. The patholog

Advancements in the understanding and management of histiocytic neoplasms.

Histiocytic neoplasms are rare diseases involving macrophages, dendritic cells, and monocytes. They include Langerhans cell histiocytosis (LCH), Erdheim-Chester disease (ECD), Rosai-Dorfman disease (RDD), juvenile xanthogranuloma (JXG), and histiocytic sarcoma. Histiocytic neoplasms are characterized by varied clinical courses and prognoses, necessitating a nuanced understanding of their classification, epidemiology, and clinical manifestations. Genetic studies have revealed somatic mutations, predominantly in the MAPK pathway, suggesting a clonal neoplastic nature. This review covers the curr

Histiocytic sarcoma of the palate: a case report.

Histiocytic sarcoma is a malignant proliferation of cells that exhibit morphological and immunophenotypic features of mature histiocytes. Owing to its rarity, its clinical features and standard treatment have not yet been established. This report describes a case of histiocytic sarcoma of the palate that developed in a 76-year-old man, the first report of an intraoral histiocytic sarcoma. An extended resection was performed; however, establishing the excision line was extremely difficult because assessing the tumour boundary on imaging was challenging and the tumour underwent dynamic gross mor…

Cutaneous Histiocytic Sarcoma With Cellular Cannibalism.

Cutaneous histiocytic sarcoma (HS) is a rare malignant tumor. An 82-year-old woman presented with a 4 × 2-cm irregular-shaped red nodule on the left posterior scalp. A biopsy specimen revealed sheets of pleomorphic atypical cells in the dermis and subcutis. A diagnosis of HS was made based on the results of a panel of immunohistochemical stains that revealed positivity of leukocyte common antigen, CD4, CD163, and HLA-DR. At the time of resection, the tumor grew rapidly to 12 × 6.5 × 5 cm in size in 2 months. The resected tumor comprised round, oval, plasmacytoid, and spindled cells. Signet-rin
