Assessing Behavioral Risk Factors Driving Zoonotic Spillover Among High-risk Populations in Myanmar.

The increasing global emergence of zoonoses warrants improved awareness of activities that predispose vulnerable communities to greater risk of disease. Zoonotic disease outbreaks regularly occur within Myanmar and at its borders partly due to insufficient knowledge of behavioral risks, hindering participatory surveillance and reporting. This study employed a behavioral surveillance strategy among high-risk populations to understand the behavioral risks for zoonotic disease transmission in an effort to identify risk factors for pathogen spillover. To explore behavioral mechanisms of spillover

Viruses tipping the scales: The role of the CD4/CD8 ratio in determining viral outcome.

Here we review recent data on how animal viruses alter the T cell dynamics, and how stressed T cell populations alter viral disease outcomes. The CD4/CD8 ratio of T cells is regulated by the thymus well into adulthood, and determined by both host genetics and environmental exposures. Human and animal data now show many chronic viral infections interact with this ratio and Immune Health Grades, but this raises new questions and justifies new experimental systems. Immune Health Grades and the ratio generally can quantify subtle immunodeficiency for viral infections. Whether or not an animal reco

Functional and Pharmacological Comparison of Human, Mouse, and Rat Organic Cation Transporter 1 toward Drug and Pesticide Interaction.

Extrapolation from animal to human data is not always possible, because several essential factors, such as expression level, localization, as well as the substrate selectivity and affinity of relevant transport proteins, can differ between species. In this study, we examined the interactions of drugs and pesticides with the clinically relevant organic cation transporter hOCT1 (SLC22A1) in comparison to the orthologous transporters from mouse and rat. We determined Km-values (73 ± 7, 36 ± 13, and 57 ± 5 µM) of human, mouse and rat OCT1 for the commonly used substrate 1-methyl-4-phenylpyridinium

Producer perceptions of dairy calf management, behavior, and welfare.

Approaches for raising calves vary across commercial dairy farms and relate to behavioral opportunities and animal welfare. The objectives of this study were to evaluate how US dairy producers and calf managers perceive (1) the welfare implications of varying management practices (including social housing and milk allowance) and behaviors in dairy calves, and (2) aspects of the human-animal relationship in calf rearing and relationships with on-farm management and personal calf handling habits. Survey questions were primarily quantitative (e.g., Likert scales) and addressed how common calf man

Interferon-alpha and MxA inhibit BK polyomavirus replication by interaction with polyomavirus large T antigen.

INTRODUCTION: BK Polyomavirus (BKPyV) infection is a common complication in kidney transplant recipients and can result in poor outcomes and graft failure. Currently, there is no known effective antiviral agent. This study investigated the possible antiviral effects of Interferon alpha (IFNα) and its induced protein, MxA, against BKPyV. METHODS: In vitro cell culture experiments were conducted using human primary renal proximal tubular epithelial cells (HRPTECs). We also did animal studies using Balb/c mice with unilateral kidney ischemic reperfusion injury. RESULTS: Our results demonstrated

The Protective Effect of Social Reward on Opioid and Psychostimulant Reward and Relapse: Behavior, Pharmacology, and Brain Regions.

Until recently, most modern neuroscience research on addiction using animal models did not incorporate manipulations of social factors. Social factors play a critical role in human addiction: social isolation and exclusion can promote drug use and relapse, while social connections and inclusion tend to be protective. Here, we discuss the state of the literature on social factors in animal models of opioid and psychostimulant preference, self-administration, and relapse. We first summarize results from rodent studies on behavioral, pharmacological, and circuit mechanisms of the protective effec

Social and biological pathways linking relationships with companion animals to human well-being.

INTRODUCTION: Companion animals are widely recognized for their potential to support human mental health and well-being. These benefits may arise through changes in social relationships and physiological processes. This study examined whether dog and cat ownership and attachment promote closer social relationships and higher well-being, the relative magnitude of direct and indirect effects of companion animals on well-being, and whether physiological factors, including cortisol concentration and gut microbiota composition, are involved in these processes. METHODS: Two complementary studies we

Human Desmoglein-2 and Human CD46 Mediate Human Adenovirus Type 55 Infection, but Human Desmoglein-2 Plays the Major Roles.

Human adenovirus type 55 (HAdV55) represents an emerging respiratory pathogen and causes severe pneumonia with high fatality in humans. The cellular receptors, which are essential for understanding the infection and pathogenesis of HAdV55, remain unclear. In this study, we found that HAdV55 binding and infection were sharply reduced by disrupting the interaction of viral fiber protein with human desmoglein-2 (hDSG2) but only slightly reduced by disrupting the interaction of viral fiber protein with human CD46 (hCD46). Loss-of-function studies using soluble receptors, blocking antibodies, RNA i

Early-life risk factors for panic and separation anxiety disorder: insights and outstanding questions arising from human and animal studies of CO2 sensitivity.

Genetically informative studies showed that genetic and environmental risk factors act and interact to influence liability to (a) panic disorder, (b) its childhood precursor separation anxiety disorder, and (c) heightened sensitivity to CO2, an endophenotype common to both disorders. Childhood adversities including parental loss influence both panic disorder and CO2 hypersensitivity. However, childhood parental loss and separation anxiety disorder are weakly correlated in humans, suggesting the presence of alternative pathways of risk. The transferability of tests that assess CO2 sensitivity -

Schisandrol A and Schisandrol B from Schisandra chinensis: Drug Metabolism, Transporter Interactions, and Herb-Drug Interaction Potential.

Schisandrol A (Sch A) and Schisandrol B (Sch B) are structurally related dibenzocyclooctadiene lignans from Schisandra chinensis, but their drug-metabolism and interaction profiles are not interchangeable. This review critically evaluates their absorption, distribution, metabolism, transporter interactions, nuclear-receptor regulation, and herb-drug interaction potential, with particular emphasis on the distinction between purified compounds and multi-lignan botanical preparations. Oral exposure is strongly formulation dependent. In rats, purified Sch B shows substantially lower absolute bioav

Cannabinoids and General Anesthetics: Revisiting Molecular Mechanisms of Their Pharmacological Interactions.

Cannabis has been used for recreation and medical purposes for more than a millennium across the world; however, its use's consequences remain poorly understood. Although a growing number of surgical patients are regular cannabis consumers, little is known regarding the pharmacological interactions between cannabis and general anesthetics; consequently, there is not a solid consensus among anesthesiologists on the perioperative management of these patients. The existing evidence about the molecular mechanisms underlying pharmacological interactions between cannabinoids and anesthetic agents, b

The monkeypox diagnosis, treatments and prevention: A review.

The world is currently dealing with a second viral outbreak, monkeypox, which has the potential to become an epidemic after the COVID-19 pandemic. People who reside in or close to forest might be exposed indirectly or at a low level, resulting in subclinical disease. However, the disease has lately emerged in shipped African wild mice in the United States. Smallpox can cause similar signs and symptoms to monkeypox, such as malaise, fever, flu-like signs, headache, distinctive rash, and back pain. Because Smallpox has been eliminated, similar symptoms in a monkeypox endemic zone should be treat

Assessment of Survival and Replication of Brucella spp. in Murine Peritoneal Macrophages.

Brucella spp. are bacteria that naturally infect a variety of domesticated and wild animals leading to abortions and infertility, and these bacteria are also capable of causing debilitating human infections, which often result from human exposure to infected animals and animal products. The brucellae are intracellular pathogens that reside in host cells, including macrophages and dendritic cells, and it is paramount for the pathogenesis of Brucella that the bacteria are able to survive and replicate in these host cells. The methods outlined in this chapter can be employed to study the interact

Recent Advances in Understandings Towards Pathogenesis and Treatment for Intrauterine Adhesion and Disruptive Insights from Single-Cell Analysis.

Intrauterine adhesion is a major cause of menstrual irregularities, infertility, and recurrent pregnancy losses and the progress towards its amelioration and therapy is slow and unsatisfactory. We aim to summarize and evaluate the current treatment progress and research methods for intrauterine adhesion. We conducted literature review in January 2020 by searching articles at PubMed on prevention and treatment, pathogenesis, the repair of other tissues/organs, cell plasticity, and the stem cell-related therapies for intrauterine adhesion. A total of 110 articles were selected for review. Uterin

Norovirus Infection: Replication, Manipulation of Host, and Interaction with the Host Immune Response.

Noroviruses (NoVs) belong to the Caliciviridae family of viruses and are responsible for causing the majority of gastroenteritis outbreaks worldwide. In the past decade, research on NoV biology has intensified because of the discovery of murine NoV and subsequently the first cell culture system and small animal model for NoV replication and pathogenesis. In this review, we discuss the current literature on NoV biology, focusing particularly on NoV replication and the interaction between NoV and the host immune response. Understanding the NoV replication cycle and its interaction with cellular

One Health and sex and gender-related perspective in the ecosystem: Interactions among drivers involved in the risk of leptospirosis in Europe. A scoping review.

Leptospirosis has a complex transmission, involving rodents and many species of domestic and wild animals. Carrier animals spread leptospires, contaminating soil and water, the main sources of human infection. The risk of infection is modulated by socio-economic factors, environment and host animals and has changed, historically linked to agriculture but now prevalent in recreational environments. Leptospirosis also reveal gender-specific exposure patterns that determine infection risks. Emphasizing the interconnectedness of humans, animals, and the environment, the One Health approach highlig

The effects of oxytocin on social behavior and eye gaze: Insights from dog-human partnership.

Oxytocin (Oxt) plays a crucial role in forming stable social bonds and promoting social interactions in both humans and animals. During the long history of domestication, dogs have evolved advanced social communication, empathy, and a remarkable ability to perceive and respond to human emotions. Given the effective and intimate social interactions between dogs and humans, the laboratory Beagle dogs have been utilized to model mental disorders such as autism spectrum disorder (ASD). Indeed, ASD-associated Shank3 mutant dogs show impaired social interaction with both conspecifics and heterospeci

Land use and land cover changes influence the rate of accidents involving venomous animals.

Direct human-nature interactions can have positive and negative impacts on people's health and well-being, and there is an increasing trend of some negative direct human-nature interactions in many parts of the world. Every year, hundreds of thousands of people suffer accidents with venomous animals around the world, including fatal accidents. However, the drivers of the increase in the number of accidents remain unclear. In this study, we evaluated whether land use and land cover changes affect the rate of accidents involving venomous animals in Brazil. We recorded the rate of accidents per c
