An "enhanced PET"-based dual-positive charges fluorescent probe to detect hypochlorous acid for monitoring the drug-damaged liver.

Drug-induced liver injury (DILI) is an acute inflammatory condition that poses a serious threat to human health. Hypochlorous acid (HClO), a reactive oxygen species, has been identified as a key biomarker for monitoring the progression of DILI. Herein, we rationally designed a phenothiazine-based fluorescent probe, DMPTC, featuring a "dual-positive-charge" structure. This design leverages an "enhanced photoinduced electron transfer (PET)" effect to simultaneously achieve high response sensitivity to HClO and superior mitochondrial targeting capability. As anticipated, DMPTC demonstrated excell

Hypochlorous acid-activatable two-photon excited NIR fluorescent probe with a high signal-to-background ratio for imaging osteoarthritis.

Osteoarthritis (OA) is a chronic joint disease, and early diagnosis of OA is crucial for its treatment, which is more beneficial for osteoarthritis patients to relieve symptoms, delay disease progression, and improve quality of life. Hypochlorous acid plays a "double-edged sword" role in OA, and can serve as a biomarker for OA. Here, based on the advantages of NIR fluorescent probe and two-photon fluorescent probe, a hypochlorous acid-activatable two-photon excited NIR fluorescent probe (TP-NIR-HClO) based on spirolactam ring-opening strategy was developed for imaging OA this paper. TP-NIR-HCl

Comparative efficacy of eyelid cleansing wipes, hypochlorous acid, and saline for blepharitis-associated ocular surface disease: a randomized trial.

Blepharitis and meibomian gland dysfunction (MGD) commonly cause ocular surface symptoms, but comparative evidence for commonly used eyelid-cleansing strategies remains limited. We compared the 4-week efficacy of preformulated eyelid cleansing wipes, hypochlorous acid-assisted cleansing, and saline-assisted cleansing in adults with blepharitis-associated ocular surface disease. In this single-center, randomized, assessor-masked, parallel-group trial, adults aged 18-70 years were assigned to one of three cleansing interventions for 4 weeks. The primary endpoint was the baseline-adjusted Week 4

GYY4137, a hydrogen sulfide donor, protects against endothelial dysfunction in porcine coronary arteries exposed to myeloperoxidase and hypochlorous acid.

BACKGROUND AND AIMS: Myeloperoxidase (MPO) and its principal reaction product hypochlorous acid (HOCl) are part of the innate immune response but are also associated with endothelial dysfunction, thought to involve a reduction in nitric oxide (NO) bioavailability. We aimed to investigate the effect of MPO and HOCl on vasorelaxation of coronary arteries and to assess directly the involvement of NO. In addition, we hypothesised that the slow release hydrogen sulfide (H2S) donor GYY4137 would salvage coronary artery endothelial function in the presence of MPO and HOCl. METHODS AND RESULTS: Contr

Distinct modes of cell death induced by different reactive oxygen species: amino acyl chloramines mediate hypochlorous acid-induced apoptosis.

Oxidants derived from inflammatory phagocytes compose a key element of the host immune defense system and can kill mammalian cells by one of several different mechanisms. In this report, we compare mechanisms of cell death induced in human B lymphoma cells by the inflammatory oxidants superoxide, H(2)O(2), and HOCl. The results indicate that the mode of cell death induced depends on the nature of the oxidant involved and the medium in which the cells are treated. When human Burkitt's lymphoma cells are exposed to superoxide anion, generated as a flux from xanthine and xanthine oxidase, the cel

Visualizing hypochlorous acid as a key oxidative stress biomarker in drug-induced liver injury with a red-emitting probe.

Drug-induced liver injury (DILI) is a common form of hepatotoxicity resulting from adverse drug reactions. Extensive studies indicate that reactive oxygen species (ROS) levels undergo significant alterations during the progression of DILI. Among various ROS, hypochlorous acid (HClO)-produced in substantial amounts during oxidative stress in liver injury-has emerged as a potential biomarker for DILI. In this work, we developed a fluorescent probe based on a benzopyran-derived fluorophore with diaminomaleonitrile as the HClO-specific recognition unit. Upon reaction with HClO, the probe emits int

Lanthanide complex-based probes for time-gated luminescence and 19F magnetic resonance imaging of hypochlorous acid in the liver.

Hypochlorous acid (HClO) is a highly reactive, short-lived oxidant in vivo whose fluctuating levels are closely linked to oxidative stress, inflammation, and tissue injury. However, its transient nature, low abundance, and interference from complex biological environments make precise detection as a biomarker of inflammatory diseases challenging. Here, we report a liver-targeted lanthanide complex probe, Ln(HAPP)3(DPBT) (Ln3+ = Eu3+, Gd3+), that functions as a dual-modal imaging probe for intrinsically background-free detection of HClO using time-gated luminescence (TGL) and 19F magnetic reson

Maintaining moderate levels of hypochlorous acid promotes neural stem cell proliferation and differentiation in the recovery phase of stroke.

JOURNAL/nrgr/04.03/01300535-202503000-00029/figure1/v/2024-06-17T092413Z/r/image-tiff It has been shown clinically that continuous removal of ischemia/reperfusion-induced reactive oxygen species is not conducive to the recovery of late stroke. Indeed, previous studies have shown that excessive increases in hypochlorous acid after stroke can cause severe damage to brain tissue. Our previous studies have found that a small amount of hypochlorous acid still exists in the later stage of stroke, but its specific role and mechanism are currently unclear. To simulate stroke in vivo, a middle cerebral

Inactivation of SARS-CoV-2 and influenza A virus by dry fogging hypochlorous acid solution and hydrogen peroxide solution.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), the causative agent of coronavirus disease 2019 (COVID-19), is transmitted mainly by droplet or aerosol infection; however, it may also be transmitted by contact infection. SARS-CoV-2 that adheres to environmental surfaces remains infectious for several days. We herein attempted to inactivate SARS-CoV-2 and influenza A virus adhering to an environmental surface by dry fogging hypochlorous acid solution and hydrogen peroxide solution. SARS-CoV-2 and influenza virus were air-dried on plastic plates and placed into a test chamber for i

Extracellular Matrix Oxidised by the Granulocyte Oxidants Hypochlorous and Hypobromous Acid Reduces Lung Fibroblast Adhesion and Proliferation In Vitro.

Chronic airway inflammation and oxidative stress play crucial roles in the pathogenesis of chronic inflammatory lung diseases, with airway inflammation being a key driving mechanism of oxidative stress in the lungs. Inflammatory responses in the lungs activate neutrophils and/or eosinophils, leading to the generation of hypohalous acids (HOX). These HOX oxidants can damage the extracellular matrix (ECM) structure and may influence cell-ECM interactions. The ECM of the lung provides structural, mechanical, and biochemical support for cells and determines the airway structure. One of the critica

Symptom-based assessment during prolonged low-concentration atomization of hypochlorous acid in occupied hospital rooms: A controlled pilot study.

INTRODUCTION: Information on health effects during prolonged low-concentration atomization of hypochlorous acid in occupied hospital rooms is limited. METHODS: We conducted a single-centre controlled pilot study in occupied four-bed hospital rooms. Rooms were assigned non-randomly to atomization with hypochlorous acid solution (24 enrolled patients) or tap water (16 enrolled patients). Participants and nurses conducting the symptom assessments were blinded to the atomized solution. Atomization was performed for 12 h/day over a 30-day room-level intervention period, with a target airborne avai

Dual-response mitochondria-targeted fluorescent probe for Hypochlorous acid and viscosity: Applications in liver injury diagnosis and environmental monitoring.

Hypochlorous acid (HClO) is widely used in disinfection yet poses severe threats to environmental safety and human health, while abnormal cellular microenvironmental viscosity is closely linked to oxidative stress-mediated liver pathogenesis. Herein, a mitochondria-targetable, dual-channel near-infrared fluorescent probe TPTY-Q-HClO was rationally constructed for simultaneous and selective sensing of HClO and viscosity. The probe employs dimethylthiocarbamate as the HClO-specific recognition site and a quaternized quinoline moiety for mitochondrial targeting, with viscosity response governed b

A near-infrared fluorescent probe based on photostable Si-rhodamine for imaging hypochlorous acid during lysosome-involved inflammatory response.

Hypochloric acid (HClO) is mainly distributed in acidic lysosomes of phagocytes and closely associated with numerous physiological and pathological processes, especially inflammatory response. Fluorescent probe has become an important tool for imaging HClO in lysosomes, but suffered from interference from autofluorescence in vivo, phototoxicity to biosamples and photobleaching phenomenon due to their short-wavelength excitation and emission. Unfortunately, up to now, no near-infrared (NIR) lysosome-targetable fluorescent probe has been reported for imaging HClO. In this paper, a near-infrared

Development of an endoplasmic reticulum-lipid droplet network-targetable ratiometric fluorescent probe for evaluating hypochlorous acid fluctuations during oxidative stress.

Synchronous and highly sensitive detection of hypochlorous acid (HOCl) dynamics in living cells is of fundamental significance for understanding intracellular oxidative stress processes. In this work, we present a phenothiazine-based ratiometric fluorescent probe, ER-LDs, designed for the rapid and selective sensing of HOCl with subcellular targeting abilities. Spectroscopic studies demonstrated that ER-LDs exhibits an ultra-sensitive ratiometric fluorescence response to HOCl within 60 s, accompanied by a distinct 110 nm hypsochromic emission shift from 560 nm (yellow) to 450 nm (blue). Comparative Efficacy of Hypochlorous Acid and Mafenide Acetate in Reducing Infection Rates in Postoperative Burn Patients: A Retrospective Cohort Study.

Infection is a leading cause of mortality in patients with third-degree burns, contributing to over 75% of burn-related deaths. Despite the widespread use of hypochlorous acid (HOCl) and mafenide acetate (MA) as wet dressings in burn care, direct comparisons of their effectiveness are limited. Thus, this study evaluates the relative efficacy of HOCl and MA in managing infections and improving survival outcomes in postoperative burn patients. This retrospective study included adults with second and third-degree burns who underwent excision and grafting between January 2012 and February 2024 at
