S-antigen specific T helper type 1 response is present in Behcet's disease.

PURPOSE: To investigate the frequency and phenotypic and functional characteristics of S-antigen (S-Ag) specific T cells in patients with Behcet's disease (BD). METHODS: Blood was taken from 23 active BD patients, 12 inactive BD patients, and 14 healthy controls. The clinical features of the patients were summarized. T cell response against 40 mixed S-Ag peptides was identified by interferon gamma (IFN-gamma) enzyme-linked immunospot assay (ELISPOT). CD69 and CD45RO were used to characterize the phenotype of S-Ag specific T cells. The functional property of S-Ag specific T cells was investiga

Killing me softly: chlamydial use of proteolysis for evading host defenses.

Chlamydial infections in humans cause severe health problems, including blinding trachoma and sexually transmitted diseases. Although the involved pathogenic mechanisms remain unclear, the ability to replicate and maintain long-term residence in the infected cells seems to significantly contribute to chlamydial pathogenicity. These obligate intracellular parasites maintain a delicate balance between exploiting and protecting their host: they occupy intracellular space and acquire nutrients from the infected cells, but at the same time they have to maintain the integrity of the host cells for t…

Clinicopathological characteristics of invasive gastric Helicobacter pylori.

Helicobacter pylori organisms have been observed deep within the stomach mucosa with an "intracellular" appearance, although the clinicopathological characteristics of such cases remain poorly understood. We analyzed 18 cases of deep mucosal H pylori and associated clinical (sex, age, history of H pylori infection, or proton pump inhibitor [PPI] use, medications, smoking, alcohol use, comorbidities, treatment response) and pathological (presence of lymphoid aggregates, intestinal metaplasia, PPI effect, active and/or chronic inflammation, quantity of invasive versus surface H pylori) character

Effects of oxidative and physical treatments on inactivation of Cylindrospermopsis raciborskii and removal of cylindrospermopsin.

The presence of toxic cyanobacterial blooms (or blue-green algae) in water bodies used either as drinking water or for recreational purposes may present serious health risks for the human population. In this study, the removal of the chemical toxin, cylindrospermopsin, via free chlorine, chlorine dioxide, monochloramine, permanganate, ozone, and UV irradiation was studied. Ozone and free chlorine were found to be highly effective for cylindrospermopsion removal while the other disinfectants were ineffective. Ozone and free chlorine were also determined to be highly effective for the inactivati

Steady-state water exchange in neural tissue is primarily passive and through the phospholipid bilayer.

Water molecules exchange incessantly across cell membranes and between intracellular compartments, but the dominant steady-state transport pathways, and whether they are active or passive, remain unclear. Low-field, high-gradient diffusion exchange spectroscopy (DEXSY) nuclear magnetic resonance (NMR) measurements on viable ex vivo neonatal mouse spinal cords show that water exchange is primarily passive. The apparent exchange rate constant (AXR) depends on osmotic conditions because it reflects multiple exchange pathways, each weighted by the exchanging compartments' volume fractions. A faste

On the characteristics of alcohol-induced liver enlargement and its possible hemodynamic consequences.

Chronic consumption of alcohol leads to an increase in liver weight, primarily due to an increase in hepatocyte volume. About 50-60% of such an increase is due to an increase in intracellular water. Accumulation of intracellular K+ osmotically accounts for about one half of the increase in intracellular water, while an increase in soluble proteins plays only a minor role in such an increase in cell volume. The increase in intracellular water is accompanied by a relative reduction in water in the extracellular space, probably due to compression of the extracellular volume by the enlarged hepato

Different Effects on Fluid Distribution between Tolvaptan and Furosemide in a Liver Cirrhosis Patient with Chronic Kidney Disease.

A 73-year-old man with liver cirrhosis and advanced chronic kidney disease was admitted to our hospital due to bilateral lower leg edema and appetite loss. Furosemide to treat fluid retention markedly decreased extracellular water compared with intracellular water, but the addition of tolvaptan equally decreased both with a greater diuretic response than furosemide alone. Furthermore, tolvaptan administration increased the plasma colloid osmotic pressure, which might facilitate the shift of fluid from the extravascular space to the intravascular space. This is the first case showing different

[Interepithelial lymphocytes in the pathogenesis and morphogenesis of atrophic gastritis (electron-microscopic study)].

Electron microscopic studies of fundal gastrobiopsies from patients with atrophic gastritis have been performed. Fat degeneration in all epitheliocytes and release of myelin-like figures (residual bodies) in the interepithelial space of the mucosa were observed. Permanent contact of the residual bodies possessing both antigenic properties and specific structural signs of biological membranes with lymphocytes in the intracellular space of the epithelial layer and phagocytosis of myelin figures by lymphocytes were detected. Peculiarities of plasma cells in the interepithelial position are descri…

Simulated microgravity reduces intracellular-free calcium concentration by inhibiting calcium channels in primary mouse osteoblasts.

Calcium homeostasis in osteoblasts plays fundamental roles in the physiology and pathology of bone tissue. Various types of mechanical stimuli promote osteogenesis and increase bone formation elicit increases in intracellular-free calcium concentration in osteoblasts. However, whether microgravity, a condition of mechanical unloading, exerts an influence on intracellular-free calcium concentration in osteoblasts or what mechanisms may underlie such an effect are unclear. Herein, we show that simulated microgravity reduces intracellular-free calcium concentration in primary mouse osteoblasts. I

Autowave distribution of nitric oxide and its endogenous derivatives in biosystems strongly enhances their biological effects: A working hypothesis.

It is hypothesized that in cells producing nitric oxide (NO), NO and its endogenous derivatives (low-molecular S-nitrosothiols and dinitrosyl iron complexes (DNIC) with thiol-containing ligands) can move in the intracellular space not only by diffusion but also in an autowave mode. This hypothesis is based on the previously obtained data on autowave distribution of DNIC with glutathione following application of a drop of a solution of Fe(2+)+glutathione onto the surface of a thin layer of a S-nitrosoglutathione solution. The appearance of autowaves is conditioned by a self-regulating self-sust…

Ultrastructural markers of tubular transport in experimental diabetes insipidus.

Stereological analysis of changes in tubular transport markers in diabetes insipidus has been undertaken. Intracellular spaces and basal infolded channels were considered as the markers of water transport, while mitochondrial metabolic steady states were considered as the active transport markers. Transmission electron microscopic observations revealed morphometric differences in the surface area and volume of intercellular space and basal infolded channels in the distal tubules. Stereological markers of mitochondrial metabolic states demonstrated significant differences in the distal tubules …

Phosphodiesterase 5 (PDE5) restricts intracellular cGMP accumulation during enterotoxigenic Escherichia coli infection.

Diarrhea caused by enterotoxigenic Escherichia coli (ETEC) has a continuing impact on residents and travelers in underdeveloped countries. Both heat-labile (LT) and heat-stable (ST) enterotoxins contribute to pathophysiology via induction of cyclic nucleotide synthesis, and previous investigations focused on intracellular signal transduction rather than possible intercellular second messenger signaling. We modeled ETEC infection in human jejunal enteroid/organoid monolayers (HEM) and evaluated cyclic nucleotide pools, finding that intracellular cAMP was significantly increased but also underwe

Purinergic Signaling: A Common Path in the Macrophage Response against Mycobacterium tuberculosis and Toxoplasma gondii.

Immune responses are essential for the protection of the host against external dangers or infections and are normally efficient in the clearance of invading microbes. However, some intracellular pathogens have developed strategies to replicate and survive within host cells resulting in latent infection associated with strong inflammation. This excessive response can cause cell and tissue damage and lead to the release of the intracellular content, in particular the nucleotide pool, into the extracellular space. Over the last decade, new studies have implicated metabolites from the purinergic p

Polarographic assay and intracellular distribution of superoxide dismutase in rat liver.

1. A polarographic assay of superoxide (O2--) dismutase (EC 1.15.1.1) activity is described, in which the ability of the enzyme to inhibit O2---dependent sulphite oxidation, initiated by xanthine oxidase activity, is measured. The assay was used in a study of the intracellular distribution of superoxide dismutase in rat liver. Both cyanide-sensitive cupro-zinc dismutase (92% of the total activity) and cyanide-insensitive mangano-dismutase (8%) were measured. 2. Rat liver homogenates contained both particulate (16%y and soluble (84%) dismutase activity. The particulate activity contained both t

Selective retention and degradation of molecules with a single mutant alpha1(I) chain in the Brtl IV mouse model of OI.

We investigated the secretion, matrix incorporation and interactions of molecules with one and two mutant alpha1(I) collagen chains in the Brtl IV murine model for Osteogenesis Imperfecta, carrying a Gly-349 to Cys substitution in one col1a1 allele. We detected a significant deviation from the expected 25 and 50% content of the molecules with no (37-46%) and one (26-40%) mutant chains in skin and bone as well as in fibroblast and osteoblast cell culture media. Steady-state labeling with (35)S-Cys demonstrated incomplete secretion of the mutant collagen in cell culture, particularly molecules c

Tetrasulfide-bridged virus-like particles enable host-sparing cascade membrane-metabolic disruption for drug-resistant bacterial keratitis.

Drug-resistant bacterial keratitis remains difficult to treat because anatomical barriers and biofilm-associated tolerance restrict antibact … disrupts membrane barriers, driving antibacterial intervention from the bacterial exterior into the intracellular space. Within the reductive bacterial intracellular milieu, the tetrasulfide-bridged framework is cleaved, glutathione is depleted, and sulfur overload is induced, thereby undermining intracellular bacterial defenses. Mild near-infrared irradiation provid …

Ca2+ influx via the Na+/Ca2+ exchanger is enhanced in malignant hyperthermia skeletal muscle.

Malignant hyperthermia (MH) is potentially fatal pharmacogenetic disorder of skeletal muscle caused by intracellular Ca(2+) dysregulation. NCX is a bidirectional transporter that effluxes (forward mode) or influxes (reverse mode) Ca(2+) depending on cellular activity. Resting intracellular calcium ([Ca(2+)]r) and sodium ([Na(+)]r) concentrations are elevated in MH susceptible (MHS) swine and murine muscles compared with their normal (MHN) counterparts, although the contribution of NCX is unclear. Lowering [Na(+)]e elevates [Ca(2+)]r in both MHN and MHS swine muscle fibers and it is prevented b

Growth-limiting intracellular metabolites in yeast growing under diverse nutrient limitations.

Microbes tailor their growth rate to nutrient availability. Here, we measured, using liquid chromatography-mass spectrometry, >100 intracellular metabolites in steady-state cultures of Saccharomyces cerevisiae growing at five different rates and in each of five different limiting nutrients. In contrast to gene transcripts, where approximately 25% correlated with growth rate irrespective of the nature of the limiting nutrient, metabolite concentrations were highly sensitive to the limiting nutrient's identity. Nitrogen (ammonium) and carbon (glucose) limitation were characterized by low intrace

IDPology of the living cell: intrinsic disorder in the subcellular compartments of the human cell.

Intrinsic disorder can be found in all proteomes of all kingdoms of life and in viruses, being particularly prevalent in the eukaryotes. We conduct a comprehensive analysis of the intrinsic disorder in the human proteins while mapping them into 24 compartments of the human cell. In agreement with previous studies, we show that human proteins are significantly enriched in disorder relative to a generic protein set that represents the protein universe. In fact, the fraction of proteins with long disordered regions and the average protein-level disorder content in the human proteome are about 3 t
