[Outline of the pathology of the Langerhans cell system].

About 100 biopsy specimens, most of them from the skin, were used for comparisons between high alpha-D-mannosidase activity and the presence of Langerhans' cells identified according to the electronoptic picture. Agreement between the two was absolute so that alpha-D-mannosidase could be rated as a reliable index of Langerhans' cells not only in histiocytosis X but also in a number of non-tumour and other cancerous processes in the skin. -- Contact couples (apposition) of Langerhans' cells and lymphocytes were often found in as well as outside the epidermis in mycosis fungoides just as in skin

Langerhans cells as targets of rabies virus immune evasion.

Rabies virus (RABV) is a neurotropic virus of the genus Lyssavirus that causes a nearly always fatal encephalitis in humans once it reaches the central nervous system. Dog bites are a major transmission route of RABV, and following superficial skin exposure, RABV first encounters epidermal Langerhans cells (LCs), yet their role in early infection remains poorly understood. Here, we show that exposure of immature human epidermal LCs to RABV leads to RABV association with cells in the absence of productive viral replication. RABV-exposed LCs display reduced cytokine and type I IFN responses and …

A pilot study on corneal Langerhans cells in keratoconus.

PURPOSE: To report the density and morphology of cells that are analogous to corneal Langerhans cells and their associations in keratoconus. MATERIALS AND METHODS: This prospective cross-sectional study included a convenience sample of keratoconus subjects aged between 18-65 years. Corneal topography, assessment of ocular symptoms, tear variables, corneal sensitivity, in-vivo confocal microscopy were performed. The number of Langerhans cells were manually counted and averaged across three central corneal images. Cell morphology was graded on a 0-3 scale, where grade 3 indicates cells with lon…

Langerhans cells and their role in oral mucosal diseases.

Dendritic cells are arguably the most potent antigen-presenting cells and may be the only cells capable of initiating the adaptive immune response. The epithelial residents of dendritic cells are Langerhans cells, which serve as the "sentinels" of the mucosa, altering the immune system not only to pathogen entry but also of tolerance to self antigen and commensal microbes. Oral mucosal Langerhans cells are capable of engaging and internalizing a wide variety of pathogens and have been found responsive to nickel in patients with nickel allergies, oral Candida species, oral lichen planus, lichen…

Epidermal Langerhans cells tune skin reactivity to contact allergens.

Allergic contact dermatitis is a common disorder that has fascinated dermatologists and immunologists for decades. Extensive studies of contact sensitivity reactions in mice established a mechanistic paradigm that has been revisited in recent years, and the involvement of Langerhans cells (LCs), a population of epidermal dendritic cells, in immune responses to epicutaneously applied antigens has been questioned. In this issue of the JCI, Gomez de Agüero et al. describe an elegant series of experiments that implicate LCs in tolerance induction, positioning these cells as key regulators of immun…

Langerhans cells in mice with severe combined immunodeficiency (SCID).

The existence of Langerhans cells in the stratified squamous epithelium of severely combined immunodeficient (SCID) mice was studied by enzyme histochemistry, immunohistochemistry and electron microscopy. Considerable numbers of ATPase-positive Langerhans cell were observed in the epidermis (ear and hind limb) as well as in the stratified squamous mucosal epithelium (forestomach and tongue) of SCID mice. The cells also expressed Ia antigen. This indicates that, in spite of the lack of functional T and B lymphocytes, the antigen-presenting function of Langerhans cell was retained in SCID mice. …

Langerhans cells in normal and pathological vocal cord mucosa.

The Langerhans cells in samples of histologically normal and pathological vocal cord mucosa were counted after identification using S-100 polyclonal antibody. Langerhans cells were commonly seen in vocal cord polyp epithelium but were infrequent in normal cord mucosa. They were also identified in samples of squamous carcinoma, severe dysplasia and chronic inflammation.

[Langerhans cells and corneal neovascularization in experimental alkali burns].

Experimental alkali-burns rabbit corneas present a neovascularisation. Six weeks later the corneas have been processed for ATPase Cytochemistry, allowing the observation of Langerhans cells. We observed an increase in the number of Langerhans cells that was statistically significant in alkali-burned corneas when compared to unburned corneas in the peripheral and midperipheral cornea.

Lymphocytes and Langerhans cells in the normal human cornea.

The distribution of B- and T-lymphocytes, of OKIa positive cells, and of HLA-ABC antigens in the normal cornea was investigated using monoclonal antibodies. The lymphocytes and Langerhans cells are present mainly in the well-vascularized limbic region but also occur albeit in small number in the center of the cornea. HLA-ABC antigens are strongly expressed on the epithelial cells of the cornea and the limbus.

The pathological significance of Langerhans cells in oral cancer.

The functional role of Langerhans cells (LCs) in oral cancer was studied, based on a quantitative analysis of LCs in squamous cell carcinomas arising from the tongue. LCs were identified by their positiveness for S100-protein and other cytological characters based on PAP method. LCs increased in number in the cancer region and showed an intimate relationship with lymphocytes in terms of their number and distribution. Moreover, in those cases with many LCs, clusters consisting of LCs and lymphocytes were often found to combine into "hydropic degeneration-like" lesions as seen in allergic diseas…

Epidermal Langerhans cells in mycosis fungoides and Sézary syndrome.

The purpose of the study was to examine whether apposition between epidermal Langerhans cells and atypical lymphoid cells is a constant finding in mycosis fungoides (MF) and whether it also occurs in the Sézary syndrome (SS). Biopsies of 8 patients with MF and 2 patients with SS were examined by transmission electron microscopy. In both cases of SS and in 7 cases of MF appositions between the 2 types of cells were observed. The appositions were less frequently seen in stage III MF lesions than in MF stage II. The cell membranes of the Langerhans cells and the apposed atypical lymphoid cells fo…

A rare presentation of langerhans cell histiocytosis tonsil infiltration: review of the literature: atypical presentation of langerhans cell histiocytosis.

MedImmune Vaccines (formerly Aviron) has developed a cold-adapted live influenza virus vaccine [FluMist] that can be administered by nasal spray. FluMist is the first live virus influenza vaccine and also the first nasally administered vaccine to be marketed in the US. The vaccine will be formulated to contain live attenuated (att) influenza virus reassortants of the strains recommended by the US Public Health Service for each 'flu season. The vaccine is termed cold-adapted (ca) because the virus has been adapted to replicate efficiently at 25 degrees C in the nasal passages, which are below n

Langerhans cell hyperplasia of the skin mimicking Langerhans cell histiocytosis: a report of two cases in children not associated with scabies.

Langerhans cells histiocytosis (LCH) affecting the skin most commonly has clinical and histopathologic diagnostic features. We are reporting two examples of Langerhans cell (LC) hyperplasia recognized in the skin biopsies of two children initially interpreted as LCH. The first was an 8-year-old boy finally interpreted as having an atypical type of contact dermatitis, while the second, an 8-year-old girl, was assumed to have Pytiriasis lichenoides et varioliformis acuta. None showed evidences of scabies. Both presented spongiotic dermatitis with numerous CD1a+ cells. As more cases of LC hyperpl…

Presence of langerhans cells, regulatory T cells (Treg) and mast cells in asymptomatic apical periodontitis.

Asymptomatic Apical Periodontitis is essentially an inflammatory disease of microbial aetiology. Association and function of the cell components involved, or specific inductive factors and growth mediators associated with development, maintenance and resolution of the periapical lesions are still unknown. The objective of this study was to evaluate the concentration of Regulatory T cells (FoxP3+; Treg), Langerhans cells (CD1a+; LC) and mast cells in asymptomatic apical periodontitis. 73 cases were selected: 30 periapical granulomas, 29 radicular cysts and 14 residual cysts. All groups were sub…

Factor XIIIa+ dendritic cells and S-100 protein+ Langerhans' cells in adult periodontitis.

OBJECTIVE: The purpose of this study was to evaluate the presence of factor XIIIa+ dendritic cells and S-100 protein+ Langerhans' cells in the gingival epithelium and connective tissue of periodontal pockets, before and after non-surgical periodontal therapy. BACKGROUND: The microbial flora in periodontal pockets provokes complex immune reactions. Dendritic cells play a critical role in primary and secondary immune responses and are considered as antigen-presenting cells. Factor XIIIa positive dendritic cells and S-100 protein positive Langerhans' cells identified by immunoreactivity against

Langerhans Cells Prevent Autoimmunity via Expansion of Keratinocyte Antigen-Specific Regulatory T Cells.

Langerhans cells (LCs) are antigen-presenting cells in the epidermis whose roles in antigen-specific immune regulation remain incompletely understood. Desmoglein 3 (Dsg3) is a keratinocyte cell-cell adhesion molecule critical for epidermal integrity and an autoantigen in the autoimmune blistering disease pemphigus. Although antibody-mediated disease mechanisms in pemphigus are extensively characterized, the T cell aspect of this autoimmune disease still remains poorly understood. Herein, we utilized a mouse model of CD4+ T cell-mediated autoimmunity against Dsg3 to show that acquisition of Dsg…

Concurrent Mastoid Cellulitis and Langerhans Cells Histiocytosis:
A Challenging Diagnosis.

Langerhans cell histiocytosis (LCH) is a rare proliferative disorder, which commonly arises in the bone and may involve other systems. To date, the diagnosis of temporal bone LCH remains a challenge as it may masquerade as a common ear infection. We report a case of a child who presented to us with persistent bilateral ear discharge for four months and was not responding to treatment. Her condition subsequently worsened, with clinical features and radiological findings suggestive of mastoid cellulitis. Nevertheless, further histopathology study revealed LCH.
