De novo sequencing and analysis of the Ulva linza transcriptome to discover putative mechanisms associated with its successful colonization of coastal ecosystems.

BACKGROUND: The green algal genus Ulva Linnaeus (Ulvaceae, Ulvales, Chlorophyta) is well known for its wide distribution in marine, freshwater, and brackish environments throughout the world. The Ulva species are also highly tolerant of variations in salinity, temperature, and irradiance and are the main cause of green tides, which can have deleterious ecological effects. However, limited genomic information is currently available in this non-model and ecologically important species. Ulva linza is a species that inhabits bedrock in the mid to low intertidal zone, and it is a major contributor

A Natural qE- and qZ-Deficient Alga Retains Functional Plant-Like and Chlorophycean Violaxanthin De-Epoxidases.

Photosynthetic organisms protect themselves from excess light through non-photochemical quenching (NPQ), a process that safely dissipates excess light energy as heat. Current understanding of NPQ is largely drawn from a few model systems, leaving it unclear how universal NPQ mechanisms are across the green lineage. We characterize the slow, sustained NPQ of the green alga, Auxenochlorella protothecoides x symbiontica (UTEX 250-A). The Auxenochlorella genome lacks Light-Harvesting Complex Stress Related (LHCSR), one of the two proteins required for the fast energy-dependent quenching (qE) compo

The PSI-NDH supercomplex prevents chilling-induced PSI photoinhibition.

Chilling stress induces photosystem I (PSI) photoinhibition in cucumber (Cucumis sativus L.). This process is associated with insufficient activity of the chloroplast NADH dehydrogenase-like complex (NDH). However, whether the importance of NDH can be generalized to other species and how NDH function is impaired remain unclear. We first examined whether NDH is essential for PSI protection under chilling stress using an NDH-deficient rice (Oryza sativa L.) mutant. The NDH-deficient rice exhibited pronounced PSI photoinhibition under chilling stress. By contrast, rice expressing flavodiiron prot

Whole-Cell Glycerol-free Fluorescence Cryo-Spectroscopy Reveals Electronic Excitation Dynamics in the Diatom Cyclotella meneghiniana.

Diatoms possess a unique photosynthetic apparatus that adapts to constant, extreme changes in light intensity, enabling efficient light harvesting and photoprotection. Fluorescence spectroscopy studies at cryogenic temperatures can reveal details of the electronic excitation dynamics in these evolutionarily successful organisms. Whole-cell studies are highly valuable since they allow investigation of the photosynthetic machinery rather than separate complexes affected by isolation. In this study, we propose an approach that enables cryogenic fluorescence analysis of photosynthetic processes in

The effect of ozone on the yellowing process of magnesium-deficient clonal Norway spruce grown under defined conditions.

During two vegetation periods, young clonal spruce trees (Picea abies (L.) Karst.) with sufficient and poor magnesium (Mg) supply were exposed in the environmental chambers of the GSF phytotron to three levels of ozone (daily means: 18-22, 88-130, and 135-190 microg m(-3); 10% reduction at night). Previous year's needles were examined at 4-week intervals with respect to their contents of Mg, Ca, K, Mn, N, P, and chlorophyll (Chl), various parameters of Chl fluorescence, and the stability of the isolated light-harvesting Chl-a/b-protein complex LHC II. The needles of the two nutrition variants

Heat shock transcription factor-mediated thermal tolerance and cell size plasticity in marine diatoms.

Diatoms are a crucial component of marine ecosystems, recognized for their broad environmental adaptability and wide temperature tolerance. However, the molecular mechanisms underlying their adaptability to diverse temperatures are unknown. In this study, we discover that heat shock transcription factors (HSFs) are potentially important for thermal tolerance in diatoms. Our study focuses on PtHSF2, annotated as HSF2 in Phaeodactylum tricornutum's genome, which is ubiquitous in diatoms. Overexpression of PtHSF2 markedly enhances thermal tolerance and increases cell size; causes significant diff

Label-Free Quantitative Proteomic Analysis of Three Strains of Viscerotropic Leishmania Isolated from Patients with Different Epidemiological Types of Visceral Leishmaniasis in China.

BACKGROUND: There are three epidemiological types of visceral leishmaniasis in China, which are caused by Leishmania strains belonging to the L. donovani complex. The mechanisms underlying their differences in the population affected, disease latency, and animal host, etc., remain unclear. We investigated the protein abundance differences among Leishmania strains isolated from three types of visceral leishmaniasis endemic areas in China. METHODS: Promastigotes of the three Leishmania strains were cultured to the log phase and harvested. The protein tryptic digests were analyzed with liquid ch

Alleviation of CO2-Induced Reductions in Tomato Photosynthesis Under Deficit Irrigation by Purple Nonsulfur Photosynthetic Bacteria.

The stimulatory effect of elevated CO2 (eCO2) on photosynthesis in most C3 crops under water deficit often declines over time due to photosynthetic acclimation. An exception occurs in plants inoculated with symbiotic nitrogen-fixing bacteria. Photosynthetic bacteria (PSB), specifically anoxygenic purple nonsulfur bacteria (Rhodopseudomonas palustris in this study), a group of nitrogen-fixing bacteria, are effective in enhancing crop photosynthesis. Therefore, this study investigated the synergistic effects of PSB and eCO2 in alleviating the effects of deficit irrigation and enhancing photosynt

Review of Mechanisms of Photodynamic and Photothermal therapy by Water-Soluble Conjugate Polymers and Their Applications in Drug and Gene Delivery.

Water-soluble conjugated polymers (WSCPs) represent an important class of functional macromolecules that combine the electronic advantages of π-conjugated polymer backbones with aqueous processability, biocompatibility, and tunable biointerfacial behavior. Unlike conventional small-molecule photosensitizers or inorganic nanomaterials, WSCPs offer polymeric design flexibility through controlled backbone engineering, side-chain functionalization, amphiphilicity, charge density, molecular weight, and nanoparticle self-assembly. These structural parameters govern their optical absorption, fluoresc

Par-3 modulates intestinal epithelial barrier function through regulating intracellular trafficking of occludin and myosin light chain phosphorylation.

BACKGROUND: Tight junctions play a critical role in the maintenance of intestinal barrier function. Partitioning-defective protein 3 (Par-3) can regulate intestinal barrier function through the modulation of tight junction assembly and cell polarity. However, the mechanisms are still not fully understood. METHODS: Adult C57BL/6 mice were treated with dextran sulfate sodium for 7 days, and segments of colon were harvested for immunofluorescent staining of Par-3. Caco-2 intestinal epithelial cells were treated with tumor necrosis factor α (TNF-α) for 24 h, and Par-3 expression was detected by W

The small helix-loop-helix protein NsrC is a regulator of phycobilisome biosynthesis.

Nitrogen is a key nutrient that influences primary productivity in both aquatic and terrestrial ecosystems. In cyanobacteria, its deficiency affects growth and phycobiliprotein biosynthesis and leads to degradation of phycobilisomes (PBSs). While the biosynthesis genes of phycobiliprotein are regulated by strong promoters, the molecular mechanisms underlying their regulation in response to nitrogen deficiency remain largely unknown. Using nondiazotrophic cyanobacterium Synechococcus elongatus PCC 7942 as a model system, we identified a novel PBS biosynthesis regulator, a small helix-loop-helix

Potato spindle tuber viroid infection enhances PEG-simulated drought stress tolerance in tomato by modulating photosynthesis and stress-responsive pathways.

In plant–pathogen interactions, infection typically leads to growth inhibition or even host mortality. However, under certain environmental conditions, some pathogens can paradoxically enhance host fitness in response to stress. Viroids, such as potato spindle tuber viroid (PSTVd), are circular single-stranded RNAs that cause plant diseases. In this study, PSTVd was used as a model to investigate adaptive host–pathogen interactions under polyethylene glycol (PEG)-simulated drought stress. Tomato (Solanum lycopersicum) plants were infected with PSTVd and then subjected to drought treatment usin

Engineering Synergistic Intra-/Intermolecular Conformational Locking via Side-Chain Branching: A β-Sheet-Inspired Strategy for Planar NIR-II Phototheranostic Aggregates.

Engineering stable noncovalent conformational locks (NoCL) network at the aggregate level is essential for developing phototheranostic aggregates (PTAs) with enhanced rigidity and light-harvesting ability, but remains a challenge due to complex intermolecular interactions. Inspired by β-sheet proteins, where branched side chains act as steric directors to program backbones into interlocked architectures, a side-chain isomerization strategy is proposed to manipulate analogous NoCL networks for constructing planar-structured NIR-II multimodal PTAs. Two isomeric pairs with linear (l-series) and b

Dissection of Genotype-Dependent Responses Reveals Leaf Proteome Signatures Associated with Maize Thermotolerance During Flowering Under Enclosure-Imposed Heat Stress.

BACKGROUND: During maize anthesis, heat stress severely limits productivity-particularly under humid conditions where high humidity suppresses transpirational cooling, forcing tissues to endure direct thermal load. METHODS: Using field enclosures to impose enclosure-imposed humid heat shock (EHS), we screened 135 maize inbred lines for flowering-stage yield resilience, using grain weight per ear at maturity under EHS relative to the corresponding control (CK) condition as the primary selection criterion. Based on this screen, we selected two tolerant (R025, R100) and two sensitive (R133, R135

Generation of high-expressing transposon-based stable pools to produce (1 + 1) and (2 + 1) bispecific common light chain antibodies.

Bi- and multispecific antibodies can deliver effective protein-based therapeutics by enabling novel biology. Transient expression of these complex asymmetric antibody formats may become a bottleneck in providing sufficient amounts of high-quality material for preclinical discovery and developability assessment to support candidate selection activities. In contrast, transposase-based stable cell line generation is being recognized as a robust and rapid path to enable the evaluation of product quality from stable pool derived material. Transposase-based systems feature a semi-targeted integratio

Kinesin light chain 1 interacts with NS1 and is a susceptibility factor for dengue virus infection in mosquito cells.

A hallmark of the dengue virus (DENV) infection is the manipulation of host cell membranes, lipid trafficking and lipid droplets (LD), all cellular functions that depend on the cytoskeleton and the cytoplasmic streaming system. We previously reported the interaction between the DENV non-structural (NS1) protein and members of the kinesin motor complex in the Aedes albopictus cell line C6/36. In this work, we present evidence indicating that the protein kinesin light chain 1 (KLC1) is indeed a susceptibility factor for the DENV replicative cycle in mosquito cells. The interaction between NS1 an

Interleukin-26 Production in Human Primary Bronchial Epithelial Cells in Response to Viral Stimulation: Modulation by Th17 cytokines.

Interleukin (IL)-26 is abundant in human airways and this cytokine is involved in the local immune response to a bacterial stimulus in vivo. Specifically, local exposure to the toll-like receptor (TLR) 4 agonist endotoxin does increase IL-26 in human airways and this cytokine potentiates chemotactic responses in human neutrophils. In addition to T-helper (Th) 17 cells, alveolar macrophages can produce IL-26, but it remains unknown whether this cytokine can also be produced in the airway mucosa per se in response to a viral stimulus. Here, we evaluated whether this is the case using primary bro

Recurrent laryngeal nerve regeneration using an oriented collagen scaffold containing Schwann cells.

OBJECTIVES/HYPOTHESIS: Regeneration of the recurrent laryngeal nerve (RLN), which innervates the intrinsic laryngeal muscles such that they can perform complex functions, is particularly difficult to achieve. Synkinesis after RLN neogenesis leads to uncoordinated movement of laryngeal muscles. Recently, some basic research studies have used cultured Schwann cells (SCs) to repair peripheral nerve injuries. This study aimed to regenerate the RLN using an oriented collagen scaffold containing cultured SCs. STUDY DESIGN: Preliminary animal experiment. METHODS: A 10-mm-long autologous canine cerv

Beyond conduction impairment: Unveiling the profound myocardial injury in left bundle branch block.

BACKGROUND: Left bundle branch block (LBBB) represents a frequently encountered conduction system disorder. Despite its widespread occurrence, a continual dilemma persists regarding its intricate association with underlying cardiomyopathy and its pivotal role in the initiation of dilated cardiomyopathy. The pathologic alterations linked to LBBB-induced cardiomyopathy (LBBB-CM) have remained elusive. OBJECTIVE: This study sought to investigate the chronologic dynamics of LBBB to left ventricular dysfunction and the pathologic mechanism of LBBB-CM. METHODS: LBBB model was established through m

Complete genome sequence data of Acidisoma sibiricum DSM 21000T with PHA pathway and a putative aerobic anoxygenic photosynthesis gene cluster.

Acidisoma sibiricum DSM 21000T, the type strain of an obligately aerobic, acidophilic, and psychrotolerant bacterium, was isolated from a Sphagnum-dominated peat bog in the Bakchar region of western Siberia, Russia. Here we report the complete genome sequence data of this type strain. Genomic DNA was sequenced using both Illumina NovaSeq X (paired-end, 2 × 150 bp) and PacBio Revio platforms. Hybrid assembly yielded a complete genome consisting of one chromosome (3925,831 bp) and two plasmids, pAS1 (577,300 bp) and pAS2 (19,537 bp), with a total genome size of 4522,668 bp and a sequencing cover
