Peroxidation of brain lipids following cyanide intoxication in mice.

Lipid peroxidation of brain lipids as determined by the conjugated diene method was observed in mice following administration of sublethal doses of potassium cyanide (KCN). Conjugated diene production was dose- and time-dependent; 10 mg/kg KCN produced detectable levels of conjugated dienes at 30 min post cyanide, whereas, 15 mg/kg produced marked levels of conjugated dienes over a 10-60-min period after KCN. Pretreatment of mice with either diltiazem (600 micrograms/kg, i.v.) or allopurinol (25 mg/kg, i.v.) blocked the generation of conjugated dienes. These results suggest lipid peroxidation

Possible role of aldehydic lipid peroxidation products as chemoattractants.

Previous studies showed that the lipid peroxidation product 4-hydroxy-trans-2-nonenal (HNE) stimulates the rat neutrophil oriented migration in vitro within a micromolar range. Its effect is weak with respect to other known chemoattractants, but highly significant on the basis of a two-way analysis of variance. Other hydroxyalkenals were found to be chemotactic within a pico-micromolar range, and their chemotactic power was not correlated to their lipophilicity. The structural requirements for the chemotactic activity of the hydroxyalkenals were studied by testing the influence of alkanals and

Ascorbate-induced lipid peroxidation and the binding of [3H]dihydroalprenolol.

Sodium ascorbate addition caused a modest loss in the specific binding of [3H]dihydroalprenolol ([3H]DHA) in a rat cortical membrane preparation. Sodium ascorbate also caused a modest increase in lipid peroxide formation in the membrane preparation. Both the lipid peroxidation and the loss in [3H]DHA binding were greatly potentiated by the addition of low levels of iron (Fe2+). A kinetic analysis indicated that the iron/ascorbate caused a loss in the number of [3H]DHA binding sites but had no effect on the affinity of [3H]DHA for the binding site. Both the lipid peroxidation and the loss of [3

[Metabolism of lipid peroxidation products in multiple sclerosis patients].

Concentration of dienic conjugates and malonic dialdehyde (metabolites of lipid peroxidation) were examined in the blood and the cerebrospinal liquor of patients suffering from disseminated sclerosis, CNS tumours complicated with neurological pathology, and osteochondrosis with the secondary radical syndrome at D4--D5 and D5--S1 levels. In patients with disseminated sclerosis no malonic dialdehyde was found in the cerebrospinal liquor, while its blood level exceeded normal 1.5 to 2 times. After hormonal therapy the situation was found to be reverse: malonic dialdehyde appeared in the cerebrosp

[Effect of acute overheating of animals on the processes of lipid peroxidation].

In liver tissue, brain, lungs and skeletal muscles of rats, subjected to overheating, content of conjugated diens and lipid peroxides was distinctly increased. The decrease in the period of malon dialdehyde induction and in the antioxidative activity of lipids correlated with the alteration in content of peroxidation products. The overheating of animals was accompanied by a decrease in unsaturation of lipids in liver tissue, brain and skeletal muscles due to a decrease in amounts of arachidonic acid and to an increase in content of palmitinic and stearinic acids. In lipids of lungs the unsatur…

Melatonin and Lipid Peroxidation: Antioxidant Shield and Therapeutic Potential.

Melatonin, a highly conserved indoleamine produced by the pineal gland and also in the mitochondria of many, perhaps all, extrapineal tissues, has emerged as a powerful antioxidant molecule. This review explores its role in counteracting lipid peroxidation (LP), a process that damages cellular membranes through the oxidative degradation of lipids. LP is involved in numerous pathological conditions, including neurodegenerative diseases, cancer, cardiovascular disorders, and aging. The article discusses how melatonin prevents, mitigates, or even reverses LP-induced cellular damage by acting as b

Oxidative Stress and Skin Diseases: The Role of Lipid Peroxidation.

Lipid peroxidation (LPO) is a biochemical process through which lipids are subjected to a peroxidation reaction in the presence of free radicals. The process can cause alterations in biological membranes and the formation of substances harmful to the body that can form aggregates with proteins and nucleic acids. Malondialdehyde (MDA) and 4-hydroxynonenal (4-HNE) are the main products of LPO. These compounds have cytotoxic and genotoxic properties and contribute to the pathogenesis of various diseases. This research focuses on the correlation between LPO and skin diseases. For some skin disease

Lipid peroxidation-induced cell death in Rett syndrome.

Rett Syndrome (RTT) is a rare neurodevelopmental disorder, primarily affecting girls (1:10,000 live births), largely caused by mutations in the X-linked gene MECP2, an epigenetic regulator encoding for the methyl-CpG binding protein 2 (MeCP2). Recent evidence links ferroptosis, an iron-dependent cell death characterized by lipid peroxide accumulation, to neurodegenerative and neurodevelopmental disorders like autism. Several RTT hallmarks, including redox imbalance, excess labile iron, increased lipid peroxidation, and impaired antioxidant enzyme activity, align with ferroptosis characteristic

Lysosomal lipid peroxidation mediates immunogenic cell death.

Cancer cells rely on lysosome-dependent degradation to recycle nutrients that serve their energetic and biosynthetic needs. Despite great interest in repurposing the antimalarial hydroxychloroquine as a lysosomal inhibitor in clinical oncology trials, the mechanisms by which hydroxychloroquine and other lysosomal inhibitors induce tumor-cell cytotoxicity remain unclear. In this issue of the JCI, Bhardwaj et al. demonstrate that DC661, a dimeric form of chloroquine that inhibits palmitoyl-protein thioesterase 1 (PPT1), promoted lysosomal lipid peroxidation, resulting in lysosomal membrane perme

Protein adducts with lipid peroxidation products in patients with psoriasis.

Psoriasis, one of the most frequent immune-mediated skin diseases, is manifested by numerous psoriatic lessons on the skin caused by excessive proliferation and keratinization of epidermal cells. These disorders of keratinocyte metabolism are caused by a pathological interaction with the cells of the immune system, including lymphocytes, which in psoriasis are also responsible for systemic inflammation. This is accompanied by oxidative stress, which promotes the formation of lipid peroxidation products, including reactive aldehydes and isoprostanes, which are additional pro-inflammatory signal

Lipid peroxidation, DNA damage, and apoptosis in dengue fever.

Interplay between the apoptosis, DNA damage, and oxidative stress as a host response to dengue viral infections remains unclear. Peripheral blood mononuclear cells (PBMCs) were isolated from 60 dengue infected patients, 20 patients with febrile illness other than dengue (OFI) and 10 non-febrile illness (NFI) patients. DNA damage in the PBMCs was assessed using single cell gel electrophoresis and stages of apoptosis underwent by the PBMCs were studied by Annexin-PI staining using flow cytometry. Plasma levels of malondialdehyde levels were estimated using thiobarbituric acid assay. Dengue infec

The onset of lipid peroxidation in rheumatoid arthritis: consequences and monitoring.

Several epidemiological studies propose the association of rheumatoid arthritis (RA) with oxidative stress. The aim of this study was to estimate the possible onset of systemic lipid peroxidation in RA patients and its relevance for pathophysiology and monitoring of RA. Seventy-three patients with RA and 73 healthy subjects were included in the study. Lipid peroxidation was estimated by the measurement of 4-hydroxynonenal (4-HNE), 4-hydroxyhexenal, malondialdehyde, acrolein, crotonaldehyde, 4-oxononenal, and isoprostanes (8-isoPGF(2α)) levels. Cytosolic phospholipase A(2) (cPLA(2)), platelet-a

Oxidative and reductive metabolism of lipid-peroxidation derived carbonyls.

Extensive research has shown that increased production of reactive oxygen species (ROS) results in tissue injury under a variety of pathological conditions and chronic degenerative diseases. While ROS are highly reactive and can incite significant injury, polyunsaturated lipids in membranes and lipoproteins are their main targets. ROS-triggered lipid-peroxidation reactions generate a range of reactive carbonyl species (RCS), and these RCS spread and amplify ROS-related injury. Several RCS generated in oxidizing lipids, such as 4-hydroxy trans-2-nonenal (HNE), 4-oxo-2-(E)-nonenal (ONE), acrolei

Oxidative stress and lipid peroxidation are upstream of amyloid pathology.

Oxidative stress is a common feature of the aging process and of many neurodegenerative disorders, including Alzheimer's disease. Understanding the direct causative relationship between oxidative stress and amyloid pathology, and determining the underlying molecular mechanisms is crucial for the development of more effective therapeutics for the disease. By employing microdialysis technique, we report local increase in the amyloid-β42 levels and elevated amyloid-β42/40 ratio in the interstitial fluid within 6h of direct infusion of oxidizing agents into the hippocampus of living and awake wild
