Interaction of ferroptosis and cuproptosis in the perspective of pulmonary hypertension.

Copper (Cu) and iron (Fe) are essential trace elements that are involved in normal human metabolic processes. Disruption of their homeostasis contributes to disease pathogenesis through mechanisms such as cuproptosis and ferroptosis. Cuproptosis targets lipoylated proteins to disrupt mitochondrial respiration, whereas ferroptosis is driven by lipid peroxidation. These processes may independently or interactively exacerbate pulmonary hypertension (PH), a condition characterized by progressive pulmonary vascular remodeling, clinical manifestations of dyspnea, right-sided heart failure, and high

Mechanisms of copper metabolism and cuproptosis: implications for liver diseases.

Copper is an essential trace element in the human body, involved in various biological processes, including cell metabolism, nerve development, and immune function. Its homeostasis is vital for maintaining normal cellular functions, and disruptions in copper homeostasis can lead to a wide range of diseases. Cuproptosis is a copper ion-dependent form of programmed cell death that leads to abnormal oligomerization of lipoylated proteins and dysfunction of iron-sulfur cluster proteins in the mitochondrial tricarboxylic acid (TCA) cycle, thereby triggering intracellular oxidative stress and proteo

Photocatalytic Golgi Proteomics Reveals Palmitoylation-Regulated Golgiphagy.

The Golgi apparatus (GA) orchestrates protein modification, trafficking, and secretion through highly dynamic remodeling, yet its proteomic complexity remains difficult to resolve in living systems. Here, we report CAT-Golgi, a genetically independent and light-controlled photocatalytic proximity labeling strategy for in situ spatiotemporal mapping of the Golgi-associated proteome. Combining a cysteine-conjugated eosin photocatalyst (GolgiCat) with an aniline probe, CAT-Golgi enables rapid and precise protein labeling within minutes under mild green light, requiring no genetic manipulation and

zDHHC-Mediated S-Palmitoylation in Skin Health and Its Targeting as a Treatment Perspective.

S-acylation, which includes S-palmitoylation, is the only known reversible lipid-based post-translational protein modification. S-palmitoylation is mediated by palmitoyl acyltransferases (PATs), a family of 23 enzymes commonly referred to as zDHHCs, which catalyze the addition of palmitate to cysteine residues on specific target proteins. Aberrant S-palmitoylation events have been linked to the pathogenesis of multiple human diseases. While there have been advances in elucidating the molecular mechanisms underlying the pathogenesis of various skin conditions, there remain gaps in the knowledge

Lipid overload meets S-palmitoylation: a metabolic signalling nexus driving cardiovascular and heart disease.

S-palmitoylation has emerged as a critical integrator of lipid overload and cardiovascular dysfunction. Disordered lipid metabolism inundates endothelial cells, vascular smooth muscle cells and macrophages with triglyceriderich lipoproteins, oxidized LDL and saturated fatty acids, expanding the intracellular palmitoylCoA pool and perturbing redox balance. Protein Spalmitoylation, the reversible attachment of palmitate to cysteine residues, converts excess palmitoylCoA into broad alterations in signalling and membrane dynamics. The FASN-ACSL-ZDHHC axis channel excess fatty acids into palmitoylC

Palmitoylation in cardiometabolic diseases: From molecular mechanisms to clinical implications.

Cardiometabolic diseases (CMDs), encompassing both cardiovascular diseases and metabolic syndromes, are among the leading causes of global morbidity and mortality. Protein S-palmitoylation, a reversible lipid-based post-translational modification, has emerged as a crucial regulator of cellular function and homeostasis. Palmitoylation is catalysed by palmitoyl transferases (PATs), also known as ZDHHC (zinc finger Asp-His-His-Cys) proteins, while a series of deacylases mediate the process of depalmitoylation. By modulating protein hydrophobicity, stability, subcellular localization, enzymatic ac

Protein S-Palmitoylation as Potential Therapeutic Target for Dermatoses.

Protein S-palmitoylation is a pivotal yet poorly integrated research field in dermatology. This reversible post-translational lipid modification primarily occurs on cysteine residues and is principally catalyzed by zinc finger and Asp-His-His-Cys DHHC-domain containing proteins (zDHHCs). The S-palmitoylation/depalmitoylation cycle directly affects protein localization, trafficking, stability, and protein-protein interaction, thereby regulating a variety of signaling pathways, including those mediating inflammation and immune reaction. Accumulating evidence has indicated that S-palmitoylation r

Identification and validation of palmitoylation metabolism-related signature for liver hepatocellular carcinoma.

BACKGROUND: Protein S-palmitoylation is a reversible posttranslational modification widely involved in tumor progression. Nevertheless, the function of palmitoylation metabolism in prognosis and tumor microenvironment characteristics in liver hepatocellular carcinoma (LIHC) patients is not fully understood. METHODS: mRNA and clinical data of LIHC patients were obtained from the TCGA and ICGC databases. Consensus clustering was used to construct palmitoylation metabolism-related clusters. Univariate Cox and Lasso regression analyses were employed to establish a palmitoylation metabolism-relate

Increasing brain palmitoylation rescues behavior and neuropathology in Huntington disease mice.

Huntington disease (HD) damages the corticostriatal circuitry in large part by impairing transport of brain-derived neurotrophic factor (BDNF). We hypothesized that improving vesicular transport of BDNF could slow or prevent disease progression. We therefore performed selective proteomic analysis of vesicles transported within corticostriatal projecting neurons followed by in silico screening and identified palmitoylation as a pathway that could restore defective huntingtin-dependent trafficking. Using a synchronized trafficking assay and an HD network-on-a-chip, we found that increasing brain

Putative Role of Protein Palmitoylation in Cardiac Lipid-Induced Insulin Resistance.

In the heart, inhibition of the insulin cascade following lipid overload is strongly associated with contractile dysfunction. The translocation of fatty acid transporter CD36 (SR-B2) from intracellular stores to the cell surface is a hallmark event in the lipid-overloaded heart, feeding forward to intracellular lipid accumulation. Yet, the molecular mechanisms by which intracellularly arrived lipids induce insulin resistance is ill-understood. Bioactive lipid metabolites (diacyl-glycerols, ceramides) are contributing factors but fail to correlate with the degree of cardiac insulin resistance i

Palmitoyltransferase ZDHHC19 regulates histone-to-protamine exchange during spermiogenesis in mice.

Spermiogenesis, the final phrase of spermatogenesis, involves replacing histones by protamines, a process known as histone-to-protamine exchange, which is crucial for chromatin condensation. While this exchange is essential, the precise mechanisms remain incompletely understood. In this study, we discovered that the palmitoyltransferase ZDHHC19 functions as a key regulator of mouse spermiogenesis. Loss of Zdhhc19 leads to male infertility and abnormal sperm morphology. Zdhhc19-deficient sperm exhibit impaired histone-to-protamine exchange, leading to retention of histones and misdistribution o

S-palmitoylation in bone biology: emerging insights and therapeutic potential.

S-Palmitoylation, the reversible covalent attachment of palmitate to cysteine residues, is a dynamic post-translational lipid modification that regulates the localization, stability, and function of a wide array of proteins. Although traditionally linked to membrane trafficking and signaling, recent preliminary studies have begun to reveal its potential role in bone biology, underscoring its significant research value. This review highlights current advances in detection methodologies-particularly the use of 3H labeling and acyl-biotin exchange (ABE) techniques-that have enabled more precise m

S-Palmitoylation during Retinoic Acid-Induced Neuronal Differentiation of SH-SY5Y Neuroblastoma Cells.

S-Palmitoylation is the covalent attachment of C14:0-C22:0 fatty acids (mainly C16:0 palmitate) to cysteines via thioester bonds. This lipid modification is highly abundant in neurons, where it plays a role in neuronal development and is implicated in neurodegenerative diseases, such as Alzheimer's disease, Parkinson's disease, and Huntington's disease. The knowledge of S-palmitoylation in neurodevelopment is limited due to technological challenges in analyzing this highly hydrophobic protein modification. Here, we used two orthogonal methods, acyl-biotin exchange (ABE) and lipid metabolic lab

Irritable Bowel Disease: Think Malonic and Methylmalonic Aciduria (CMAMMA)!

Combined malonic and methylmalonic aciduria (CMAMMA; OMIM 614265) is a rare disorder of mitochondrial fatty acid synthesis caused by pathogenic variants in the ACSF3 gene, which encodes malonyl-CoA synthetase. The clinical presentation is highly heterogeneous and may include seizures, memory impairment, psychiatric manifestations, and cognitive decline. We describe an adult patient with long-standing unexplained fatigue and irritable bowel syndrome who remained undiagnosed for 15 years. Subsequent blood and urine analyses revealed elevated methylmalonic acid levels, and molecular genetic testi

Inhibition of the Sp1/CTR1 axis attenuates cuproptosis and alleviates axonal injury following intracerebral hemorrhage.

Intracerebral hemorrhage (ICH) causes severe neurological deficits, and secondary neuronal injury is a major barrier to recovery. Cuproptosis, a copper-dependent form of regulated cell death, has been linked to metabolic stress, but its role in axonal injury after ICH remains unclear. Here, we used in vivo rat ICH models and in vitro ICH-like neuronal injury models to examine changes in copper homeostasis, cuproptosis-associated molecular alterations, and axonal injury following ICH. Copper levels were markedly increased in perihematomal brain tissue and injured neurons, accompanied by reduced

Pseudomonas aeruginosa and Staphylococcus aureus Display Differential Proteomic Responses to the Silver(I) Compound, SBC3.

The urgent need to combat antibiotic resistance and develop novel antimicrobial therapies has triggered studies on novel metal-based formulations. N-heterocyclic carbene (NHC) complexes coordinate transition metals to generate a broad range of anticancer and/or antimicrobial agents, with ongoing efforts being made to enhance the lipophilicity and drug stability. The lead silver(I) acetate complex, 1,3-dibenzyl-4,5-diphenylimidazol-2-ylidene (NHC*) (SBC3), has previously demonstrated promising growth and biofilm-inhibiting properties. In this work, the responses of two structurally different ba

Small ubiquitin-related modifiers: A novel and independent class of autoantigens in primary biliary cirrhosis.

Serum autoantibodies against components of nuclear dots (anti-NDs), namely PML and Sp100, are specifically detected in 20% to 30% of patients with primary biliary cirrhosis (PBC). Although anti-ND antibodies are nonpathogenic, the mechanisms that lead to this unique reactivity are critical to understanding the loss of immune tolerance in PBC. Importantly, Sp100 and PML are both covalently linked to small ubiquitin-related modifiers (SUMOs). Therefore, we investigated whether SUMO proteins are independent autoantigens in PBC and studied 99 PBC sera samples for reactivity against NDs, PML, and S

Bimetallic nanozyme-mediated dual ferroptosis/cuproptosis synergy potentiates immunotherapy in bladder cancer.

Immunotherapy for bladder cancer remains limited by immunosuppressive tumor microenvironment (TME) and low immunogenicity. Here, we developed a TME-responsive bimetallic porous nanozyme, Fe/Cu-HPC@GOx/PEG, to synergistically induce dual ferroptosis/cuproptosis and amplify antitumor immunity. The nanozyme integrates Fe-Cu bimetal catalytic centers within a hierarchical porous carbon framework, enabling glucose oxidase (GOx)-triggered H₂O₂ generation and subsequent Fenton/Fenton-like reactions. In vitro studies demonstrated that the nanozyme effectively depleted glutathione, inactivated GPX4, an

GSH-Responsive Nanoparticles Enhance Hepatocellular Carcinoma Immunotherapy Through Synergistic Effects of Cuproptosis and PI3K Inhibitor Combination.

Cuproptosis, an emerging form of programmed cell death, is capable of inducing mitochondrial dysfunction. Moreover, the PI3K-AKT-mTOR signaling pathway contributes to tumor cell progression by reprogramming mitochondrial morphology and function. In this study, we have designed copper complex nanoparticles (NPCu) and PI3K-AKT-mTOR inhibitor Alpelisib nanoparticles (NPALP) that enhance the efficacy of cuproptosis-based therapies. NPCu triggers mitochondrial dysfunction and promotes the aggregation of lipoylated dihydrolipoamide S-acetyltransferase (DLAT), while NPALP inhibits the PI3K-AKT-mTOR s

Metabolic underpinnings of cuproptosis.

Cuproptosis is a recently defined form of regulated cell death (RCD) driven by copper-induced mitochondrial proteotoxic stress. Distinct from other RCD pathways such as apoptosis and ferroptosis, cuproptosis arises when copper binds to lipoylated metabolic enzymes in mitochondria, leading to their aggregation, loss of iron-sulfur cluster proteins, and metabolic collapse. This discovery establishes a mechanistic link between metal homeostasis and mitochondrial metabolism, positioning cuproptosis as a "cell-sabotage" pathway in which essential metabolic processes become self-destructive when dys
