In-syringe solid-phase extraction coupled with UPLC-MS/MS for simultaneous determination of reducing sugars, animo acids and Amadori compounds in tobacco.

Considering the critical role of Maillard reaction-related compounds in determining the sensory and chemical stability of tobacco, this study develops an in-syringe SPE coupled with UPLC-MS/MS method for simultaneous determination of 43 Maillard reaction-related compounds in tobacco, covering reducing sugars (3), amino acids (17), Amadori compounds (17), and structurally related intermediates (6). The sample pretreatment utilizes HLB-packed self-assembled columns for SPE, followed by methanol elution. The extracted solution is then analyzed using a Titank C18 column (150×2.1 mm, 1.8 μm), with …

Quinoa protein-dextran conjugates as functional stabilizers for curcumin-loaded Nanoemulsions.

This study employed high-pressure microfluidization (HPM) to facilitate the Maillard reaction between quinoa protein (QP) and dextran (DX), systematically examining the effects of various pressures on the conjugate's physicochemical properties. Fourier transform infrared spectroscopy confirmed the formation of QP-DX conjugates, characterized by a new peak at 1149 cm-1 (covalent CN bond). Secondary and tertiary structure analyses revealed that HPM-assisted Maillard reaction partially unfolded QP molecules, enhancing conformational flexibility and interfacial properties. The QP-DX conjugate fabr…

Ultra-High Temperature Treatment and Storage of Infant Formula Induces Dietary Protein Modifications, Gut Dysfunction, and Inflammation in Preterm Pigs.

SCOPE: Ready-to-feed liquid infant formula is increasingly used for preterm infants when human milk is unavailable. These formulas are sterilized by ultra-high temperature treatment, but heating and storage may reduce bioactivity and increase formation of Maillard reaction products with potential negative consequences for immature newborns. METHODS AND RESULTS: Using preterm pigs as a model for sensitive newborn infants, the study tests the intestinal responses of feeding experimental liquid formula within 5 days. A pasteurized formula (PAST) with the same nutrient composition but less protei…

Effects of heat treatment, homogenization pressure, and overprocessing on the content of furfural compounds in liquid milk.

BACKGROUND: Sterilization of milk is aimed at killing the microorganisms present. There are three main sterilization methods commonly used in milk processing: high temperature and short time (HTST) pasteurization, ultrahigh pasteurization (UP), and ultrahigh temperature (UHT) sterilization. The Maillard reaction is of special interest in studying the effect of heat treatment on milk quality. Furfural compounds are one of the typical intermediates of the Maillard reaction, which have safety risks related to mutagenic and genotoxic effects. The furfural compounds content is directly related to t…

Differences and mechanisms of color deterioration in three types of ready-to-eat shellfishes during storage.

Ready-to-eat (RTE) abalones, scallops and oysters were prepared through a process of cooking, drying, vacuum packaging, and high-temperature sterilization, and were subjected to accelerated storage. Upon storage, the three RTE shellfishes all showed color deterioration, as indicated by darker color, decreased L* and W* values, and increased a* value. In contrast, the color deterioration of RTE oysters was more pronounced. Meanwhile, oxidation reactions occurred during storage, which were manifested as increased peroxide value, thiobarbituric acid reactive substances value and aldehyde content.

Impact of Grape Seed Extract on Flavor and Functionality of Pea Protein Extrudates and Patties.

Consumer interest in plant-based proteins is increasing, but undesirable off-flavors from Maillard reaction and lipid oxidation processes hinder their broader acceptance. Our previous work showed that proanthocyanidin-rich plant extracts like grape seed extract (GSE) could reduce the formation of these off-flavors in aqueous plant protein solutions. This study investigated the effects of GSE addition pre- and post-extrusion at two levels (0.02% and 1.0% w/w) on off-flavor development in pea protein extrudates and patties. We hypothesized that GSE addition at different processing phases would i…

Protein glycation and in vivo distribution of human lens fluorescence.

Glycated proteins formed by the Maillard reaction were measured by furosine determination in human normal lenses and in senile and diabetic cataracts. Furosine, an hydrolysis product of fructose-lysine adduct formed in the early stages of the Maillard reaction, was measured by high performance liquid chromatography (HPLC). Furosine levels in diabetic cataracts were found to be 3 to 4 times higher than those observed for senile cataracts. The increased glycation levels both in cortex and nucleus were related to the increase of fluorescence determined in vitro by fluorometry and in vivo by Schei…

Beyond the browning index: RGB radiomics enables non-destructive prediction of fresh-cut "Red Fuji" apple browning and enzyme activities.

Fresh-cut apples undergo rapid enzymatic browning, which compromises visual quality and causes substantial postharvest losses. Most RGB vision approaches rely on handcrafted color/texture descriptors that are sensitive to acquisition conditions and provide limited linkage to the underlying enzyme dynamics. This study proposes a radiomics-enhanced RGB imaging framework to non-destructively quantify browning progression and predict key browning-related enzymatic activities in fresh-cut "Red Fuji" apples. Apple slices were imaged from 0 to 24 h after cutting using a custom high-resolution RGB set

Improvement of Robusta coffee aroma with l-leucine powder.

BACKGROUND: l-Leucine powder (LP) was added to green Robusta coffee beans in order to reduce the difference in flavour between Robusta and Arabica coffee. l-Leucine was selected as an additive based on the Maillard reaction. The pre-treatment method conducted in this study was a short soaking (M1) or spraying procedure (M2), then LP was added at varying levels up to 3% (w/w, 30 g kg-1 ). All samples were roasted (240 °C for 15 min) and extracted using an espresso machine. Volatile compounds were analysed by solid-phase microextraction-gas chromatography-mass selective detection. RESULTS: Thir…

Molecular Mechanisms Underlying the Synergistic Regulation of Glucose and Clay Minerals on Polyphenol-Maillard Mediated Abiotic Humification.

The synergistic effects of glucose (Glu) concentration and clay mineral type (kaolinite [Kao], montmorillonite [Mon]) on abiotic humification via the polyphenol-Maillard reaction remain poorly understood. To address these scientific challenges, a series of controlled, sterile batch experiments was conducted. Specifically, a glucose concentration gradient (0, 0.03, 0.06, 0.12, and 0.24 mol/L) was established; Kao and Mon were separately introduced as mineral catalysts; and the Maillard reaction was facilitated in the presence of catechol and glycine under strictly abiotic conditions to preclude…

Isolation and identification of a novel Maillard pigment, beer-furpipate, from dark beer.

Beer color is primarily formed through the Maillard reaction. Our research focused on identifying low-molecular-weight pigments in beer, which led to the detection of an unidentified pigment (X) with an absorption maximum at around 400 nm in the basic fraction of beer samples. We found that the amount of compound X increased more than 30 times when fructose was added to dark beer and subsequently autoclaved. We successfully isolated compound X from 10 L of the heated beer using various chromatographic procedures. The mass spectrometry showed that the molecular formula of compound X is C16H17O4…
