For bone regeneration, matrix density is a critical determinant of scaffold performance, influencing both mechanical stiffness and biological accessibility, that is, how readily cells interact with and remodel the matrix. Here, we investigated how collagen matrix densification modulates stem cell mechanosensing in vitro and bone healing outcomes in vivo using density-tunable collagen microgels. Centrifugation-based collagen densification produced a sixfold increase in stiffness and a compact fibrillar architecture that enhanced early mechanosensitive signaling in human mesenchymal stem cells,

Engineering extracellular microenvironments to control stem cell fate remains a central challenge in regenerative medicine. Here, we develop ECM-mimetic cellular patches formed by the supramolecular assembly of laminin-derived, integrin-binding ligands. The resulting fibrillar networks exhibit well-defined molecular packing and nanoscale ligand distribution, enabling specific engagement of apical integrin β1 on mesenchymal stem cells. This controlled interface converts molecular assembly into hierarchical mechanotransduction, coordinating cytoskeletal remodeling, nuclear deformation, and chrom

Dynamic reciprocity (DR) refers to the ongoing, bidirectional interaction between cells and their microenvironment, specifically the extracellular matrix (ECM). The continuous remodeling of the ECM exerts mechanical force on cells and modifies biochemical mediators near the cell membrane, thereby initiating cell-signaling cascades that produce changes in gene expression and cell behavior. Cellular changes, in turn, affect the composition and organization of ECM components. These continuous interactions are the fundamental principle behind DR, and its critical role throughout development and ad

Adolescent idiopathic scoliosis (AIS) is the most prevalent pediatric spinal deformity. We previously demonstrated elongated cilia and an altered molecular mechanosensory response in AIS osteoblasts. The purpose of this exploratory study was to characterize the mechanosensory defect occurring in AIS osteoblasts. We found that cilia length dynamics in response to flow significantly differ in AIS osteoblasts compared to control cells. In addition, strain-induced rearrangement of actin filaments was compromised resulting in a failure of AIS osteoblasts to position or elongate in function of the b

Mechanotransduction translates forces into biological responses and regulates cell functionalities. It is implicated in several diseases, including laminopathies which are pathologies associated with mutations in lamins and lamin-associated proteins. These pathologies affect muscle, adipose, bone, nerve, and skin cells and range from muscular dystrophies to accelerated aging. Although the exact mechanisms governing laminopathies and gene expression are still not clear, a strong correlation has been found between cell functionality and nuclear behavior. New theories base on the direct effect of

Modulating inflammation within the tumor microenvironment (TME) has emerged as a crucial avenue in contemporary cancer therapy. However, the intricate signaling networks, mechanical heterogeneity, spatiotemporal drug gradients, and poor immune cell infiltration continue to hinder the effectiveness of conventional therapeutic strategies. Hydrogels, with tunable physicochemical properties and versatile biological functions, offer a promising platform for addressing these challenges. This review first unpacks the inflammatory pathways active in the TME and then provides a structured overview of h

BACKGROUND: Plant-parasitic nematodes (PPNs) locate host roots primarily through chemosensation, yet the sensory events that convert root contact into the stylet thrusts required for host penetration remain obscure. Root border cells (RBCs), terminally differentiated cells shed from the root cap, constitute the first point of physical contact between the root and invading nematodes, but whether and how RBCs trigger pre-invasion behaviors is unknown.
RESULTS: Here we showed that rice (Oryza sativa) RBCs induce preparasitic second-stage juveniles (pre-J2) of the rice root-knot nematode Meloidog

Pacinian corpuscles are the principal detectors of high-frequency vibrations in amniotes, enabling environmental awareness, the detection of fine textures, and the manipulation of tools and objects during exploration and foraging. Corpuscles consist of a multilayered outer core enclosing a sensory inner core, which contains lamellar Schwann cells enveloping the afferent terminal at its center. In the traditional model, the outer core acts as a mechanical filter, enabling only high-frequency vibrations to reach the afferent terminal. Here, we show that the afferent terminal, not the outer core,

Cadherin-11 has been proposed as a biomarker of kidney fibrosis, but whether it drives disease was unclear. Huffstater et al. show that genetic or antibody blockade of cadherin-11 limits fibrosis, preserves renal function, and prevents stiffening of the fibrogenic niche across 3 injury models. The authors argue that cadherin-11 affects fibroblasts, leading to their activation by reinforcing the cell's force-generating machinery.

Osteoarthritis (OA) is increasingly recognized as a mechanically driven whole-joint disease in which abnormal mechanotransduction initiates a cascade of mitochondrial dysfunction, chronic inflammation, and progressive cartilage degeneration. Among the mechanosensitive molecules identified to date, Piezo1 has emerged as a key mechanosensitive regulator linking pathological mechanical loading to intracellular calcium signaling and downstream cellular responses. Growing evidence indicates that persistent Piezo1 activation promotes mitochondrial calcium overload, excessive reactive oxygen species

Skeletal muscle development, homeostasis, and repair rely on mechanical signals. Piezo1, a mechanosensitive cation channel, translates membrane lipid cues into intracellular responses. This review summarizes Piezo1's roles in skeletal muscle physiology and pathology. Physiologically, Piezo1 mediates calcium influx to regulate myogenesis, satellite cell quiescence/activation, myofibrillar protein synthesis/metabolism, and microvascular homeostasis. Pathologically, Piezo1 exhibits a dual nature: its downregulation drives disuse atrophy via the Piezo1/KLF15/IL-6 axis, while aberrant activation in

The myelin sheath is a multilayered glial membrane that surrounds axons providing insulation and rapid propagation of action potentials and contributing to structural support in both the central nervous system (CNS) and the peripheral nervous system (PNS). Therefore, myelin formation plays a crucial role in maintaining the integrity, connectivity, and functionality of the nervous system, as well as neuronal regeneration after injury or damage. Myelination in early developmental stages requires the molecular and mechanical exogenous cues provided mainly by the axons of the neurons, coupling the

Cytosolic phospholipase A2 (cPLA2) controls some of the most powerful inflammatory lipids in vertebrates by releasing their metabolic precursor, arachidonic acid, from the inner nuclear membrane (INM). Ca2+ and INM tension (TINM) are thought to govern the interactions and activity of cPLA2 at the INM. However, as compensatory membrane flow from the contiguous endoplasmic reticulum (ER) may prevent TINM, the conditions permitting nuclear membrane mechanotransduction by cPLA2 or other mediators remain unclear. To test whether the ER buffers TINM, we created the genetically encoded, Ca²⁺-insensit

Transmembrane channel-like protein 1 (TMC1) forms the pore of the mechanotransduction channel in cochlear and vestibular hair cells, converting mechanical stimuli from sound and head movements into electrochemical signals. Recent evidence supports a dimeric structure for TMC1, with each monomer harboring an independent ion-conducting pore. The p.(M654V) variant, in which methionine 654 is substituted with valine, is associated with non-syndromic autosomal recessive deafness. In the present work, we used molecular dynamics (MD) simulations to compare the structural and biophysical properties of

Cells exhibit remarkable sensitivity to the mechanical properties of their surrounding matrix, particularly stiffness changes, a phenomenon known as cellular mechanotransduction. In vivo, tissues exhibit a wide range of stiffness, from kilopascals (kPa) to megapascals (MPa), which can alter with aging and disease. Traditional cell culture methods employ plastic substrates with stiffness in the gigapascal range, which does not accurately mimic the physiological conditions of most biological tissues. Therefore, employing substrates that can be engineered to span a wide range of stiffnesses, clos

The ovary is a dynamic organ where mechanical forces profoundly regulate follicular development, oocyte maturation, and overall reproductive function. These forces, originating from the extracellular matrix (ECM), granulosa and theca cells, and ovarian stroma, influence cellular behavior through mechanotransduction, translating mechanical stimuli into biochemical responses. This review explores the intricate interplay between mechanical cues and ovarian biology, focusing on key mechanosensitive pathways such as Hippo signaling, the PI3K/AKT pathway, and cytoskeletal remodeling, which govern fo

This study investigates the structural organization of telocytes (TCs) in the quail beak, focusing on their association with mechanoreceptors like Herbst and Ruffini corpuscles. By exploring these features, the study aim to expand the understanding of TCs' role in mechanosensation and sensory modulation. Paraffin sections stained with Hematoxylin and Eosin revealed TCs surrounding both Herbst and Ruffini corpuscles, as well as nerve fibers. Similar findings observed using Mallory Trichrome staining, which highlighted TCs around these sensory structures. Methylene Blue staining further confirme

Hair cells of the inner ear are particularly sensitive to changes in mitochondria, the subcellular organelles necessary for energy production in all eukaryotic cells. There are over 30 mitochondrial deafness genes, and mitochondria are implicated in hair cell death following noise exposure, aminoglycoside antibiotic exposure, as well as in age-related hearing loss. However, little is known about the basic aspects of hair cell mitochondrial biology. Using hair cells from the zebrafish lateral line as a model and serial block-face scanning electron microscopy, we have quantifiably characterized

Adipose tissue is a crucial regulator in maintaining cardiovascular homeostasis by secreting various bioactive products to mediate the physiological function of the cardiovascular system. Accumulating evidence shows that adipose tissue disorders contribute to several kinds of cardiovascular disease (CVD). Furthermore, the adipose tissue would present various biological effects depending on its tissue localization and metabolic statuses, deciding the individual cardiometabolic risk. Crosstalk between adipose and myocardial tissue is involved in the pathophysiological process of arrhythmogenic r
