Postoperative loss of perineuronal nets in the hippocampal CA1 region results in memory consolidation deficits.

Postoperative cognitive dysfunction (POCD) is a common postoperative neurological complication, with memory consolidation deficits being one of its prominent features. However, the mechanisms underlying postoperative memory consolidation deficits are still unclear. The perineuronal net (PNN) is a specialized extracellular matrix that predominantly surrounds parvalbumin (PV) interneurons and is closely associated with cognitive functions, including learning and memory. Theta oscillations in the hippocampal CA1 region are crucial for memory consolidation. However, the relationships among postope…

Synaptic weight dynamics underlying memory consolidation: Implications for learning rules, circuit organization, and circuit function.

Systems consolidation is a common feature of learning and memory systems, in which a long-term memory initially stored in one brain region becomes persistently stored in another region. We studied the dynamics of systems consolidation in simple circuit architectures with two sites of plasticity, one in an early-learning and one in a late-learning brain area. We show that the synaptic dynamics of the circuit during consolidation of an analog memory can be understood as a temporal integration process, by which transient changes in activity driven by plasticity in the early-learning area are accu

Dynamic alteration of intrinsic properties of the cerebellar Purkinje cell during the motor memory consolidation.

Intrinsic plasticity of the cerebellar Purkinje cell (PC) plays a critical role in motor memory consolidation. However, detailed changes in their intrinsic properties during memory consolidation are not well understood. Here, we report alterations in various properties involved in intrinsic excitability, such as the action potential (AP) threshold, AP width, afterhyperpolarization (AHP), and sag voltage, which are associated with the long-term depression of intrinsic excitability following the motor memory consolidation process. We analyzed data recorded from PCs before and 1, 4, and 24 h afte…

Functional MRI reveals brain-wide actions of thalamically-initiated oscillatory activities on associative memory consolidation.

As a key oscillatory activity in the brain, thalamic spindle activities are long believed to support memory consolidation. However, their propagation characteristics and causal actions at systems level remain unclear. Using functional MRI (fMRI) and electrophysiology recordings in male rats, we found that optogenetically-evoked somatosensory thalamic spindle-like activities targeted numerous sensorimotor (cortex, thalamus, brainstem and basal ganglia) and non-sensorimotor limbic regions (cortex, amygdala, and hippocampus) in a stimulation frequency- and length-dependent manner. Thalamic stimul…

Memory consolidation and inducible nitric oxide synthase expression during different sleep stages in Parkinson disease.

BACKGROUND: Parkinson disease (PD) is a neurodegenerative disease characterized by motor and nonmotor dysfunctions, which include sleep disturbances. Rapid eye movement (REM) sleep is associated with numerous physiologic changes such as memory consolidation. Compelling evidence suggests that nitric oxide (NO) is crucial to both sleep regulation and memory consolidation. In our study, we explored changes in biologic molecules during various sleep stages and the effects of sleep on memory consolidation in PD. METHODS: Ten PD patients and 14 volunteers without PD participated in our study. The g…

Systemic inhibition of mTOR kinase via rapamycin disrupts consolidation and reconsolidation of auditory fear memory.

The mammalian target of rapamycin (mTOR) kinase is a critical regulator of mRNA translation and is known to be involved in various long lasting forms of synaptic and behavioural plasticity. However, information concerning the temporal pattern of mTOR activation and susceptibility to pharmacological intervention during both consolidation and reconsolidation of long-term memory (LTM) remains scant. Male C57BL/6 mice were injected systemically with rapamycin at various time points following conditioning or retrieval in an auditory fear conditioning paradigm, and compared to vehicle (and/or anisom

Post-training amphetamine administration enhances memory consolidation in appetitive Pavlovian conditioning: Implications for drug addiction.

It has been suggested that some of the addictive potential of psychostimulant drugs of abuse such as amphetamine may result from their ability to enhance memory for drug-related experiences through actions on memory consolidation. This experiment examined whether amphetamine can specifically enhance consolidation of memory for a Pavlovian association between a neutral conditioned stimulus (CS-a light) and a rewarding unconditioned stimulus (US-food), as Pavlovian conditioning of this sort plays a major role in drug addiction. Male Long-Evans rats were given six training sessions consisting of …

Electroconvulsive shock and lidocaine reveal rapid consolidation of spatial working memory in the water maze.

Head trauma leading to concussion and electroconvulsive shock (ECS) in humans causes amnesia for events that occurred shortly before the injury (retrograde amnesia). The present experiment investigated the amnesic effect of lidocaine and ECS in 25 rats trained on a working memory version of the Morris water task. Each day, the escape platform was moved to a new location; learning was evidenced by a decrease in the latency to find the platform from the first to the second trial. "Consolidation" of this newly encoded spatial engram was disrupted by bilateral inactivation of the dorsal hippocampu

Longitudinal Alterations in Sleep EEG Biomarkers of Memory Consolidation in Middle-Aged and Older Adults.

The precise coordination of slow oscillations (SO) and sleep spindles during non-rapid eye movement (NREM) sleep supports memory consolidation and may serve as a sensitive marker of cognitive aging. However, longitudinal changes in their oscillatory dynamics in midlife and older age remain poorly understood. Using polysomnography with high-density EEG at two timepoints over ~2.5 years, we examined changes in local NREM slow wave (SW), sleep spindle (occurring in the 11-16 Hz sigma range), and SO-sigma coupling strength in cognitively unimpaired middle-aged to older adults at risk for Alzheimer…

Cannabidiol modulates contextual fear memory consolidation in animals with experimentally induced type-1 diabetes mellitus.

OBJECTIVES: In view of the neuroprotective characteristic of cannabidiol (CBD) and its beneficial action on aversive memory in non-diabetic animals, we aimed to investigate in animals with experimentally induced type-1 diabetes mellitus (T1DM) whether CBD treatment would be able to impair the contextual fear memory consolidation, its generalisation and whether the effect would be lasting. We also investigated the CBD effect on anxiety-like responses. METHODS: After T1DM induction, animals received single or more prolonged treatment with CBD and were submitted to the contextual fear conditioni…

The effects of passive and active administration of heroin, and associated conditioned stimuli, on consolidation of object memory.

Mode of administration (i.e., active vs passive) could influence the modulatory action that drugs of abuse exert on memory consolidation. Similarly, drug conditioned stimuli modulate memory consolidation and, therefore, acquisition and extinction of this conditioned response could also be influenced by mode of drug administration. Exploring these questions in male Sprague-Dawley rats, Study 1 assessed memory modulation by post-training 0, 0.3 and 1 mg/kg heroin injected subcutaneously in operant chambers (i.e., drug conditioned context). Study 2 asked a similar question but in rats trained to …

Posterior parietal cortex is critical for the encoding, consolidation, and retrieval of a memory that guides attention for learning.

Within most contemporary learning theories, reinforcement prediction error, the difference between the obtained and expected reinforcer value, critically influences associative learning. In some theories, this prediction error determines the momentary effectiveness of the reinforcer itself, such that the same physical event produces more learning when its presentation is surprising than when it is expected. In other theories, prediction error enhances attention to potential cues for that reinforcer by adjusting cue-specific associability parameters, biasing the processing of those stimuli so t

Absence of systems consolidation of fear memories after dorsal, ventral, or complete hippocampal damage.

We examined the effects of dorsal, ventral, or complete damage to the hippocampus on long-term retention of a Pavlovian conditioned fear response to a tone and a context paired with foot shock. Rats received a fear conditioning episode, in which a tone and context or context-alone were paired with foot shock. Two days or 12 weeks later, they received sham, dorsal, ventral, or complete NMDA-induced damage of the hippocampus. During a retention test conducted 2 weeks after surgery, the sham control rats exhibited high levels of freezing in the context and in the presence of the tone. Rats with d

Consolidation improves the learning of new meanings for known words but not necessarily their integration into semantic memory.

Consolidation is essential to the integration of novel words into the mental lexicon; however, its role in learning new meanings for known words remains unclear. This old-form-new-meaning learning is very common, as when one learns that "skate" is also a type of fish in addition to its familiar roller- or ice-skating meaning. To address consolidation effects for new meanings, we compared the behavioral and ERP measures on new and original meanings tested 24 hours after learning with words tested immediately after learning. Semantic judgments of both new and original meanings benefitted from th

Role of protein synthesis and DNA methylation in the consolidation and maintenance of long-term memory in Aplysia.

Previously, we reported that long-term memory (LTM) in Aplysia can be reinstated by truncated (partial) training following its disruption by reconsolidation blockade and inhibition of PKM (Chen et al., 2014). Here, we report that LTM can be induced by partial training after disruption of original consolidation by protein synthesis inhibition (PSI) begun shortly after training. But when PSI occurs during training, partial training cannot subsequently establish LTM. Furthermore, we find that inhibition of DNA methyltransferase (DNMT), whether during training or shortly afterwards, blocks consoli

Beyond memory consolidation? Varied effects of post-training heroin on drug place conditioning and escape learning.

To test the hypothesis that opiates reinforce self-administration through modulation of memory consolidation, three experiments in male Sprague-Dawley rats explored whether post-training heroin administration could facilitate drug place conditioning and escape learning in the Barnes maze. Immediately following heroin- (1 mg/kg; Experiment 1) or cocaine-compartment (20 mg/kg; Experiment 2) conditioning sessions, rats received vehicle, 1 mg/kg heroin, or 1 mg/kg heroin in combination with 3 mg/kg naloxone. All groups were tested after one pairing (Test 1) and again after three additional pairing…

Exposure to broadband noise during non-REM sleep impairs hippocampal sharp-wave ripples and memory consolidation.

Sleep is critical for consolidating recent experiences into memories. A key mechanism underlying this process is hippocampal sharp-wave ripples (SWRs). Although SWR-dependent memory consolidation is often viewed as an "offline" process, sounds heard during sleep are processed by a highly active auditory system that projects to medial temporal lobe structures, providing a pathway by which sounds may modulate hippocampal activity. While processing salient sounds during sleep can be adaptive, whether ongoing processing of environmental sounds interferes with SWR-dependent memory consolidation rem…

Noradrenergic infraslow rhythm during sleep is the critical link between heart-rate dynamics and memory consolidation.

Recent work shows that the brain's arousal system remains active during sleep, with rhythmic locus coeruleus (LC) activity shaping sleep architecture and supporting memory consolidation. The LC releases norepinephrine (NE) in infraslow (~0.02 Hz) bouts that gate NREM sleep spindles. Here, we demonstrate that heart rate (HR) fluctuations during NREM are tightly phase-locked to these NE rhythms, identifying the LC as a key driver of very-low-frequency HR variability (VLF-HRV), an understudied autonomic signal. Using optogenetics, transient LC inhibition blunts HR slowing, whereas LC activation p…
