Histological changes in relation to accumulation and elimination of inorganic and methyl mercury in gills of Labeo rohita Hamilton.

L. rohita was exposed to identical concentrations of inorganic and methyl mercury (HgCl2 and CH3HgCl) and the gills were studied for mercury bioaccumulation and histological changes. In methyl mercury exposed group the mercury level in the gills continuously increased til the end of the exposure period whereas the level started decreasing from the day 30 onwards in the other group even though the exposure was continued for 60 days. Histological changes were similar in inorganic and methyl mercury treated fish except the higher intensity observed in the latter treatment. Under depuration for 15

Acute exposure to methylmercury chloride induces fast changes in swimming performance, cognitive processes and oxidative stress of zebrafish (Danio rerio) as reference model for fish community.

Fishes are the first group of vertebrates that respond when the environment is contaminated with pollutants resulted from anthropogenic activities. The development of the toxicity tests is bringing new evidence about the toxicological effects of the pollutants upon the life forms. Behavioural abnormalities in the swimming performance and cognitive processes were well associated with the response of organisms to pollutants from environment. The aim of the paper was to study the behavioural changes of zebrafish (memory, swimming performances and aggression) and oxidative stress (superoxide dismu

Abiotic methylation of mercury in the aquatic environment.

Methylation of inorganic mercury in the aquatic environment has been considered to be largely the result of biological processes, primarily involving sulfate-reducing bacteria. However, these processes cannot account for all of the methylmercury that is formed naturally. A growing body of evidence suggests that chemical reactions represent another possible pathway for mercury methylation in the aquatic environment. In order to assess the abiotic contribution to mercury methylation in the water column, and specifically the conditions under which this contribution may be significant, the current

[Advances of toxicity evaluation of cinnabar and compatibility necessity analysis].

Cinnabars are widely used in the clinic and shows unique efficacy, and it has been used in treating diseases for thousands of years either single-use or combination with other traditional Chinese medicines. Cinnabar mainly contains mercury, which displays obvious hepatotoxicity and nephrotoxicity. The safety of cinnabar and its rational use caused extensive attention. However, there are some misunderstanding that cinnabar shows equivalent toxicity to the toxicity of mercury compounds such as mercuric chloride and methylmercury in safety evaluation of cinnabar, which in our view exaggerates the

Chemical demethylation of methylmercury by selenoamino acids.

A new chemical demethylation pathway for methylmercury under physiologically and environmentally relevant conditions is reported. The pathway involves the reaction between methylmercury and a selenoamino acid (L-selenocysteine, L-selenoglutathione, D,L-selenopenicillamine, or L-selenomethionine) via the formation of bis(methylmercuric)selenide and dimethylmercury as intermediates. The final degradation product is HgSe(s).

Geochemical mercury pools regulate diverse communities of hgcA microbes and MeHg levels in paddy soils.

Rice paddies are unique artificial wetlands generating methylmercury (MeHg), a highly potent neurotoxin. However, the impact of diverse mercury (Hg) pools on the Hg-methylating communities during rice growth is unclear. This study investigates soil treated with five mercury forms (HgCl2, α-HgS, β-HgS, nano-HgS, and Hg-DOM) at two levels (5 mg/kg and 50 mg/kg). The results showed a varying abundance of sulphate-reducing bacteria, Geobacteraceae, methanogens, and hgcA microbes in the soils across rice grown under different mercury treatments and concentrations. Soils treated with HgCl2, nano-HgS

N-Acetylcysteine Displaces Glutathionyl-Moieties from Hg2+ and MeHg+ to Form More Hydrophobic Complexes at Near-Physiological Conditions.

The anthropogenic release of Hg is associated with an increased human exposure risk. Since Hg2+ and MeHg+ have a high affinity for thiols, their interaction with L-glutathione (GSH) within mammalian cells is fundamentally involved in their toxicological chemistry and excretion. To gain insight into the interaction of these mercurials with multiple small molecular weight thiols, we have investigated their competitive interactions with GSH and N-acetylcysteine (NAC) at near-physiological conditions, using a liquid chromatographic approach. This approach involved the injection of each mercurial o

Determination of the Intracellular Complexation of Inorganic and Methylmercury in Cyanobacterium Synechocystis sp. PCC 6803.

Understanding of mercury (Hg) complexation with low molecular weight (LMW) bioligands will help elucidate its speciation. In natural waters, the rate of this complexation is governed by physicochemical, geochemical, and biochemical parameters. However, the role of bioligands involved in Hg intracellular handling by aquatic microorganisms is not well documented. Here, we combine the use of isotopically labeled Hg species (inorganic and monomethylmercury, iHg and MeHg) with gas or liquid chromatography coupling to elemental and molecular mass spectrometry to explore the role of intracellular bio

Microbial Biosynthesis of Thiol Compounds: Implications for Speciation, Cellular Uptake, and Methylation of Hg(II).

Cellular uptake of inorganic divalent mercury (Hg(II)) is a key step in microbial formation of neurotoxic methylmercury (MeHg), but the mechanisms remain largely unidentified. We show that the iron reducing bacterium Geobacter sulfurreducens produces and exports appreciable amounts of low molecular mass thiol (LMM-RSH) compounds reaching concentrations of about 100 nM in the assay medium. These compounds largely control the chemical speciation and bioavailability of Hg(II) by the formation of Hg(LMM-RS)2 complexes (primarily with cysteine) in assays without added thiols. By characterizing thes

Study on the simultaneous reduction of methylmercury by SnCl2 when analyzing inorganic Hg in aqueous samples.

Mercury (Hg) is among the most concerned contaminants in the world. It has three major chemical forms in the environment, including Hg0, Hg2+, and methylmercury (MeHg). Due to their differences in toxicity, mobility, and bioavailability, speciation analysis is critical for understanding Hg cycling and fate in the environment. SnCl2 reduction-atomic fluorescence spectrometry detection is the most commonly used method for analyzing inorganic Hg. However, it should be noted that MeHg may also be reduced by SnCl2, which would result in the overestimation of inorganic Hg. In this study, the reducti

[Surveillance of Total Mercury and Methylmercury Concentrations in Retail Fish].

Most fish samples contain methylmercury, that the concentrations very greatly according to the fish species. To avoid the adverse health effects of methylmercury while retaining the benefits provided by fish consumption, it is important to select suitable fish species and to control the amount of the fish intake. We surveyed the concentrations of total mercury and methylmercury in 210 retail fish samples classified into 19 fish species by using validated analytical methods. The results of this survey were as follows. The total mercury and methylmercury concentrations were higher than 1 mg/kg i

The toxicology of mercury: Current research and emerging trends.

Mercury (Hg) is a persistent bio-accumulative toxic metal with unique physicochemical properties of public health concern since their natural and anthropogenic diffusions still induce high risk to human and environmental health. The goal of this review was to analyze scientific literature evaluating the role of global concerns over Hg exposure due to human exposure to ingestion of contaminated seafood (methyl-Hg) as well as elemental Hg levels of dental amalgam fillings (metallic Hg), vaccines (ethyl-Hg) and contaminated water and air (Hg chloride). Mercury has been recognized as a neurotoxica

Regulatory volume decrease in primary astrocyte cultures: relevance to methylmercury neurotoxicity.

Because of the multiple and varied roles of astrocytes in brain homeostasis, primary cultures of astrocytes from neonatal rat brains have proven to be an excellent model for the study of in vitro cell functions and control mechanisms. In addition, their ability to preferentially sequester a number of heavy metals, such as methylmercury, has lead to intense research on their potential to modulate heavy metal-induced dysfunction. In the present review we briefly discuss the mechanisms associated with astrocytic swelling, an early and prominent event in brain injury, followed by a description on

Alkyl Mercury-Induced Toxicity: Multiple Mechanisms of Action.

There are a number of mechanisms by which alkylmercury compounds cause toxic action in the body. Collectively, published studies reveal that there are some similarities between the mechanisms of the toxic action of the mono-alkyl mercury compounds methylmercury (MeHg) and ethylmercury (EtHg). This paper represents a summary of some of the studies regarding these mechanisms of action in order to facilitate the understanding of the many varied effects of alkylmercurials in the human body. The similarities in mechanisms of toxicity for MeHg and EtHg are presented and compared. The difference in m

Should ebselen be considered for the treatment of mercury intoxication? A minireview.

Mercury is a ubiquitous environmental contaminant and can be found in inorganic (Hg0, Hg+ and Hg2+) and organic forms (chiefly CH3Hg+ or MeHg+). The main route of human, mammals and bird exposure occurs via predatory fish ingestion. Occupational exposure to Hg0 (and Hg2+) can also occur; furthermore, in gold mining areas the exposure to inorganic Hg can also be high. The toxicity of electrophilic forms of Hg (E+Hg) is mediated by disruption of thiol (-SH)- or selenol (-SeH)-containing proteins. The therapeutic approaches to treat methylmercury (MeHg+), Hg0 and Hg2+ are limited. Here we discuss

Sources and Toxicity of Mercury in the San Francisco Bay Area, Spanning California and Beyond.

This report synthesizes and evaluates published scientific literature on the environmental occurrence and biomagnification of mercury with emphasis on the San Francisco Bay Area (SFBA), California. Mercury forms various compounds, well known for their toxicity in humans and environmental ecosystems. Elemental mercury is transported and distributed by air, water, and sediments. Through the metabolic processes of algae and bacteria, mercury is converted into organic compounds, such as methylmercury (MeHg), which then bioaccumulates up through trophic levels. In fish, it is found primarily in ske

Microbial community function and methylmercury production in oxygen-limited paddy soil.

Methylmercury is a neurotoxic compound that can enter rice fields through rainfall or irrigation with contaminated wastewater, and then contaminate the human food chain through the consumption of rice. Flooded paddy soil has a porous structure that facilitates air exchange with the atmosphere, but the presence of trace amounts of oxygen in flooded rice field soil and its impact on microbial-mediated formation of methylmercury is still unclear. We compared the microbial communities and their functions in oxygen-depleted and oxygen-limited paddy soil. We discovered that oxygen-limited paddy soil

Discovery of a transalkylation mechanism--identification of ethylmercury+ at a tetraethyllead-contaminated site using sodiumtetrapropylborate, GC-AED and HPLC-AFS.

Although organolead as a gasoline additive is banned in most countries, contamination by organolead compounds is still present. Little is known about transformation reactions of organolead compounds and especially transalkylation reactions with other metals. Laboratory experiments to clarify transalkylation reactions between organolead and inorganic mercury, and investigations of sites, where organolead compounds were emitted, are reported. Under laboratory conditions, inorganic mercury is ethylated to ethylmercury+ in presence of tetraethyllead. These transalkylations take place very fast and

Open focused microwave-assisted sample preparation for rapid total and mercury species determination in environmental solid samples.

This paper describes rapid, simple microwave-assisted leaching/ digestion procedures for total and mercury species determination in sediment samples and biomaterials. An open focused microwave system allowed the sample preparation time to be dramatically reduced to only 24 min when a power of 40-80 W was applied. Quantitative leaching of methylmercury from sediments by HNO(3) solution and complete dissolution of biomaterials by an alkaline solution, such as 25% TMAH solution, were obtained. Methylmercury compounds were kept intact without decomposition or losses by evaporation. Quantitative re…
