Hepatic focal nodular hyperplasia: specific findings at dynamic contrast-enhanced US with carbon dioxide microbubbles.

Dynamic contrast material-enhanced ultrasonography (US) with intraarterial infusion of carbon dioxide microbubbles was performed for four cases of histologically proved focal nodular hyperplasia (FNH) in four patients and for 167 cases of various hepatic nodules in 144 patients. No complications due to dynamic US were observed in any of the 148 patients. All FNH nodules were less than 3 cm in diameter. Consistent specific findings of FNH were not obtained with US, computed tomography, magnetic resonance imaging, radiocolloid scanning, or angiography in the four cases of FNH. In contrast, the c…

Effects of Pressure Cycling, Concentration, and Excitation Amplitude on Ambient Pressure Sensitivity of Perfluorobutane Microbubbles.

OBJECTIVE: The high ambient pressure sensitivity of the subharmonic response from contrast microbubbles makes it a promising means for noninvasive diagnosis of vascular pressures, i.e., subharmonic aided pressure estimation (SHAPE). Here, we investigated the effects of concentration and multiple pressure cycles on the subharmonic response of Sonazoid, a commercially available microbubble with a perfluorobutane gas core. METHODS: We performed a pressure sensitivity study at 3 MHz excitation frequency, 100-700 kPa peak negative pressures (PNP), and ambient overpressures of 10-20 kPa (75-150 mm …

Macrophage-Mimicking Biomimetic Microbubbles for Early Ultrasound Molecular Imaging of Acute Rejection in Heart Transplantation.

Acute rejection (AR) is an important cause of heart graft injury and failure. Accurate diagnosis of AR is essential to achieving early intervention and improving the prognosis of patients. Current clinical monitoring of AR requires repeated invasive endocardial biopsies, and there is an urgent need to develop a noninvasive diagnostic technique. As antigen-presenting cells, macrophages play an important role in adaptive immunity, such as the early activation of T cells in AR, and are considered to be key drivers of AR. Macrophage membrane biomimetic microbubbles (MB-Mφ) inherit a variety of fun…

Selective Photochemical Conversion of Carbon Dioxide to Formic Acid at Gas-Water Interface of Microbubbles.

We report a selective photochemical conversion of carbon dioxide (CO2) to formic acid (HCOOH, FA) at the gas-water interface (GWI) of microbubbles. The microbubbles with an average diameter of 42 μm are produced by passing CO2 gas through a porous thermoplastic bubbler immersed in an aqueous solution of the copper(II)-phenanthroline complex [Cu(Phen)2]2+. The average FA production rate at room temperature is found to be 47.5 μM h-1 for 5 mM of [Cu(Phen)2]2+. When 5 mM iodide (I-) is added to the system, the FA production rate increases to a maximum value of 63.8 μM h-1. We also demonstrated th…

Multi-modality imaging assessment of microbubbles and cerebral emboli in left ventricular pulsed field ablation.

BACKGROUND: Pulsed field ablation (PFA) may have a superior safety profile compared to other technologies, but it has the potential to cause gaseous microbubbles (MB), which may be associated with cerebral emboli. Limited relative safety data has been published regarding PFA in the left ventricle (LV). METHODS: Healthy and chronic myocardial infarction (MI) swine underwent PFA (monopolar, biphasic, 25 Amps) in the LV using an irrigated focal catheter under intra-cardiac echocardiography (ICE) guidance for MB monitoring. Two control swine received air MBs through the lumen of the ablation cath…

Accelerating thrombolysis using a precision and clot-penetrating drug delivery strategy by nanoparticle-shelled microbubbles.

Conventional thrombolytic drugs for vascular blockage such as tissue plasminogen activator (tPA) are challenged by the low bioavailability, off-target side effects and limited penetration in thrombi, leading to delayed recanalization. We hypothesize that these challenges can be addressed with the targeted and controlled delivery of thrombolytic drugs or precision drug delivery. A porous and magnetic microbubble platform is developed to formulate tPA. This system can maintain the tPA activity during circulation, be magnetically guided to the thrombi, and then remotely activated for drug release

The use of cationic microbubbles to improve ultrasound-targeted gene delivery to the ischemic myocardium.

We synthesized a cationic microbubble (CMB) with the aim of enhancing its DNA-carrying capacity to improve targeted gene transfection of the ischemic heart for cardiac regeneration. We previously reported that ultrasound-targeted microbubble destruction (UTMD) employing the commercial Definity microbubble (MB) successfully transfected genes into rodent hearts, but the transfection efficiency was modest. We synthesized a CMB and compared its DNA-carrying capacity and reporter gene transfection efficiency with the Definity MB. The CMB bound 70% more plasmid DNA than the Definity MB. UTMD-mediate

Paclitaxel and rose bengal loaded microbubbles for the ultrasound targeted chemo-sonodynamic therapy of pancreatic cancer.

Despite significant advances in cancer treatment over the past five decades, survival outcomes for pancreatic cancer have remained largely unchanged. The effectiveness of chemotherapy as a treatment for pancreatic cancer is limited by the dense, protective tumour stroma, which impedes drug delivery. Ultrasound-targeted microbubble destruction (UTMD) has emerged as a promising strategy for enhancing the delivery of chemotherapy agents to solid tumours. In this study, we report the development and evaluation of a novel microbubble (MB) formulation, ST-001, which incorporates paclitaxel chemother

Enhancement of non-invasive trans-membrane drug delivery using ultrasound and microbubbles during physiologically relevant flow.

Sonoporation has been associated with drug delivery across cell membranes and into target cells, yet several limitations have prohibited further advancement of this technology. Higher delivery rates were associated with increased cellular death, thus implying a safety-efficacy trade-off. Meanwhile, there has been no reported study of safe in vitro sonoporation in a physiologically relevant flow environment. The objective of our study was not only to evaluate sonoporation under physiologically relevant flow conditions, such as fluid velocity, shear stress and temperature, but also to design ult

Effects of focused ultrasound and microbubbles on the vascular permeability of nanoparticles delivered into mouse tumors.

Ultrasound sonication with microbubbles (MBs) was evaluated for enhancement of the release of nanoparticles from vasculature to tumor tissues. In this study, tumor-bearing Balb/c mice were insonicated with focused ultrasound (FUS) in the tumors after the injection of MBs (SonoVue) and then lipid-coated quantum dot (LQD) nanoparticles (130 +/- 25 nm) were injected through the tail vein. We studied the effects of the injected MB dose (0-300 microL/kg), sonication duration (0-300 s) and treatment-procedure sequence on the accumulation of nanoparticles in the tumors 24 h after the treatment and th…
