Role of gangliosides in tumour progression: a molecular target for cancer therapy?

In a number of patients with tumours of either neuroectodermal or epithelial origin, polysialylated gangliosides (e.g. GD3) are over-expressed. The mechanism of ganglioside over-expression may be different for the two classes of tumour and could represent distinct secondary genetic mutations or epigenetic changes affecting the enzymes (transferases and/or hydrolases) controlling the metabolic interconversions of these gangliosides. Tumour cells of neuroectodermal origin (e.g. melanomas and brain tumours) are known to produce and shed polysialylated gangliosides, whereas paracrine signal(s) fro

[The quality practice of molecular-genetic testing in the era of molecular target therapy in chronic myelogenous leukemia].

The advent of tyrosine kinase inhibitors as molecular target therapy has resulted in a marked change in the laboratory process for the diagnosis and therapeutic monitoring of chronic myelogenous leukemia. This includes defining the molecular typing of BCR-ABL1 to establish the diagnosis, a quantitative and/or high quality assay for minimal residual disease to evaluate the molecular response, and mutation analysis and chromosomal examination to assess its resistance to inhibitors. These processes should be used where appropriate for each patient. In the ongoing development and clinical use of n

The role of ERK signaling pathway in pituitary adenomas: from molecular mechanisms to targeted therapy.

ERK, a core member of the MAPK family, operates through the classical Ras/Raf/MEK/ERK signaling cascade. Abnormal activation of this pathway is closely associated with malignant tumor progression and represents a critical target in anticancer drug development. In this article, we systematically describe the regulatory network of the ERK pathway, including negative feedback mechanisms, cross-pathway interactions, non-coding RNAs, and epigenetic regulation, with a particular focus on ERK's dual role-promoting both cell proliferation and apoptosis-in pituitary adenomas compared to other tumors. A

European Association of Neuro-Oncology guideline on molecular testing of meningiomas for targeted therapy selection.

Meningiomas are the most common primary intracranial tumors of adults. For meningiomas that progress or recur despite surgical resection and radiotherapy, additional treatment options are limited due to a lack of proven efficacy. Meningiomas show recurring molecular aberrations, which may serve as predictive markers for systemic pharmacotherapies with targeted drugs or immunotherapy, radiotherapy, or radioligand therapy. Here, we review the evidence for a predictive role of a wide range of molecular alterations and markers including NF2, AKT1, SMO, SMARCE1, PIK3CA, CDKN2A/B, CDK4/6, TERT, TRAF

Modulation of Hsp90 function in neurodegenerative disorders: a molecular-targeted therapy against disease-causing protein.

Abnormal accumulation of disease-causing protein is a commonly observed characteristic in chronic neurodegenerative disorders such as Alzheimer's disease, Parkinson's disease, and polyglutamine (polyQ) diseases. A therapeutic approach that could selectively eliminate would be a promising remedy for neurodegenerative disorders. Spinal and bulbar muscular atrophy (SBMA), one of the polyQ diseases, is a late-onset motor neuron disease characterized by proximal muscle atrophy, weakness, contraction fasciculations, and bulbar involvement. The pathogenic gene product is polyQ-expanded androgen recep

Molecular Targeted Therapy for Chronic Non-Bacterial Mastitis: New Insights into Pathophysiology and Therapeutic Strategies.

Chronic Non-bacterial Mastitis (CNBM) is a benign and heterogeneous breast disease whose etiology is not yet fully understood and primarily includes plasma cell mastitis and idiopathic granulomatous mastitis. Its management is challenged by limited therapeutic choices, poor treatment outcomes, and considerable adverse effects. The emergence of molecular targeted therapy has presented new alternatives and hope for the treatment of this condition. The marked dysregulation of critical inflammatory pathways, including NF-κB, JAK-STAT, and MAPK, observed during the onset and progression of CNBM, su…

Sialic Acid in Neurodegenerative and Psychiatric Disorders: From Molecular Regulation to Targeted Nanocarrier-Based Therapy.

In recent years, the exploration of molecular and cellular mechanisms underlying central nervous system (CNS) disorders has expanded beyond classical neurotransmitter- and receptor-based approaches toward a more integrated view including immune, metabolic, and glycosylation processes. Among these, sialic acid and its derivatives have emerged as critical regulators of neuronal communication, immune modulation, and synaptic plasticity. Their involvement ranges from maintaining neurochemical homeostasis under physiological conditions to contributing to the onset and progression of neurodegenerati

The Type I Interferon Axis in Systemic Autoimmune Diseases: From Molecular Pathways to Targeted Therapy.

Type I interferons (IFN-I) are pivotal effectors of innate immunity and constitute a central axis of host defense against pathogens. Sensing of exogenous or endogenous nucleic acids by pattern-recognition receptors-exemplified by Toll-like receptors-triggers transcriptional induction of IFN-I. Engagement of the heterodimeric IFN-I receptor on nucleated cells reprograms cellular states via canonical Janus kinase-signal transducer and activator of transcription (JAK-STAT) signaling as well as STAT-independent, noncanonical pathways. This axis is tempered by multilayered regulatory mechanisms, in

Targeting CD148 for Antithrombotic Therapy: Functional and Molecular Evaluation of AKB-9778.

CD148 is the primary receptor-type protein tyrosine phosphatase (PTP) regulating platelet activation, and its inhibition has been proposed as a novel anti-thrombotic strategy with potentially lower bleeding risk than current therapies. However, selective and potent inhibitors of CD148 are currently lacking. AKB-9778 (razuprotafib), a potent inhibitor of the closely related vascular endothelial-PTP (VE-PTP), has also been reported to inhibit CD148. This study investigated the effect of AKB-9778 on in vitro whole-blood thrombogenicity, examined the molecular basis of its interaction with the CD1

Molecular Drivers of Prostate Cancer Metastasis: Emerging Targets for Precision Therapy.

Prostate cancer (PCa) stands as one of the primary cancer diseases affecting male health and represents the principal reason behind death when metastasis develops during late-stage disease. The spread of prostate cancer from its initial site demands intricate molecular interactions through signaling pathways which permit cancer cells to break their attachment to the original tumor mass and form secondary tumor sites in distant organs. Prostate cancer metastasis depends heavily on three main molecular pathways which include the PI3K/AKT signaling cascade along with Wnt/β-catenin pathway and epi

Molecular Pathogenesis and Targeted Therapies in Eosinophilic Granulomatosis with Polyangiitis: An Updated Review.

Eosinophilic granulomatosis with polyangiitis (EGPA) is a rare systemic vasculitis characterized by asthma, eosinophilia, and necrotizing inflammation of small- to medium-sized vessels. Accumulating evidence indicates that EGPA is a polygenic and heterogeneous disorder comprising distinct antineutrophil cytoplasmic antibody (ANCA)-defined endotypes with divergent genetic backgrounds, immune pathways, and clinical phenotypes. Its pathogenesis reflects the convergence of epithelial-alarmin signaling, type 2 inflammation, eosinophil effector mechanisms, and B-cell/autoantibody responses, with mye

Role of SIRT1 in sepsis-induced encephalopathy: Molecular targets for future therapies.

Sepsis induces neuroinflammation, BBB disruption, cerebral hypoxia, neuronal mitochondrial dysfunction, and cell death causing sepsis-associated encephalopathy (SAE). These pathological consequences lead to short- and long-term neurobehavioural deficits. Till now there is no specific treatment that directly improves SAE and its associated behavioural impairments. In this review, we discuss the underlying mechanisms of sepsis-induced brain injury with a focus on the latest progress regarding neuroprotective effects of SIRT1 (silent mating type information regulation-2 homologue-1). Targeted Therapies for Lung Cancer Patients With Oncogenic Driver Molecular Alterations.

Lung cancer has traditionally been classified by histology. However, a greater understanding of disease biology and the identification of oncogenic driver alterations has dramatically altered the therapeutic landscape. Consequently, the new classification paradigm of non-small-cell lung cancer is further characterized by molecularly defined subsets actionable with targeted therapies and the treatment landscape is becoming increasingly complex. This review encompasses the current standards of care for targeted therapies in lung cancer with driver molecular alterations. Targeted therapies for EG
