Susceptibility of field populations of the diamondback moth, Plutella xylostella, to a selection of insecticides in Central China.

The diamondback moth (DBM), Plutella xylostella (L.) (Lepidoptera: Plutellidae), is a globally distributed and important economic pest. Chemical control is the primary approach to regulate populations of this pest. However, resistance to insecticides evolves following heavy and frequent use. Therefore, the insecticide resistance in field populations of P. xylostella collected from Central China from 2013 to 2014 was determined with a leaf-dipping method. Based on the results of the monitoring, P. xylostella has developed high levels of resistance to beta-cypermethrin (resistance ratio=69.76-33

Immuno-physiological adaptations confer wax moth Galleria mellonella resistance to Bacillus thuringiensis.

Microevolutionary mechanisms of resistance to a bacterial pathogen were explored in a population of the Greater wax moth, Galleria mellonella, selected for an 8.8-fold increased resistance against the entomopathogenic bacterium Bacillus thuringiensis (Bt) compared with a non-selected (suspectible) line. Defense strategies of the resistant and susceptible insect lines were compared to uncover mechanisms underpinning resistance, and the possible cost of those survival strategies. In the uninfected state, resistant insects exhibited enhanced basal expression of genes related to regeneration and a

Chemical composition and larvicidal activity of Zanthoxylum armatum against diamondback moth, Plutella xylostella.

The diamondback moth, Plutella xylostella (L.) (Lepidoptera: Yponomeutidae) is the most serious pest of cruciferous crops grown in the world causing economic yield loss. Several synthetic insecticides have been used against P. xylostella but satisfactory control was not achieved due to development of resistance to insecticides. Therefore, the present study was carried out to screen different fractions of Zanthoxylum armatum for their insecticidal activities against second instar larvae of P. xylostella. Results indicate, all the fractions showed activity to P. xylostella. However, n-hexane fra

Setae from the pine processionary moth (Thaumetopoea pityocampa) contain several relevant allergens.

BACKGROUND: Pine processionary larvae produce urticating hairs (setae) that serve for protection against predators. Setae induce cutaneous reactions in animals and humans. The presence of toxic or allergic mechanisms is a matter of debate. OBJECTIVES: To detect the presence of allergens in setae and to characterize them. MATERIALS AND METHODS: Setae extracts were characterized by gel staining and immunoblot, with sera from patients with immediate reactions and positive prick test reactions, as well as a rabbit antiserum raised against setae. Setae proteins were fractionated by high-performan

Industrial melanism in the peppered moth is not associated with genetic variation in canonical melanisation gene candidates.

Industrial melanism in the peppered moth (Biston betularia) is an iconic case study of ecological genetics but the molecular identity of the gene determining the difference between the typical and melanic (carbonaria) morphs is entirely unknown. We applied the candidate gene approach to look for associations between genetic polymorphisms within sixteen a priori melanisation gene candidates and the carbonaria morph. The genes were isolated and sequence characterised in B. betularia using degenerate PCR and from whole-transcriptome sequence. The list of candidates contains all the genes previous

Characterization of a newly established potato tuber moth (Phthorimaea operculella Zeller) cell line.

BACKGROUND & OBJECTIVE: Potato tuber moth (PTM), Phthorimaea operculella Zeller is a widely distributed, devastating pest of potatoes attacking the foliage and infest the tubers in both field and store causing serious economic damage. As application of PTM granulovirus (PTM-GV) has shown significant reduction in damage, attempts were made to develop a new cell line from this insect to grow PTM-GV for use as a biopesticide. METHODS: Approximately 100 mg of insect eggs were collected, surface sterilized and crushed gently in a boiling tube aseptically. The tissues were washed with physiological

Dopamine-synthesizing neurons include the putative H-cell homologue in the moth Manduca sexta.

The catecholamine dopamine (DA) plays a fundamental role in the regulation of behavior and neurodevelopment across animal species. Uncovering the embryonic origins of neurons that express DA opens a path for a deeper understanding of how DA expression is regulated and, in turn, how DA regulates the activities of the nervous system. In a well-established insect model, Manduca sexta, we identified the putative homologue of the embryonic grasshopper "H-cell" using intracellular techniques, laser scanning confocal microscopy, and immunohistochemistry. In both species, this neuron possesses four ax

Expression of opsin mRNA in normal and vitamin A deficient retinas of the sphingid moth Manduca sexta.

Two distinct opsin-encoding cDNAs, designated MANOP1 and MANOP2, were isolated as 3' fragments from the sphingid moth Manduca sexta. They were obtained by reverse transcription of retinal RNA and amplification with the polymerase chain reaction (PCR) using a degenerate primer designed to an amino-acid sequence conserved in arthropod opsins. The cDNA fragments labelled bands at approximately 1.8 kb on Northern blots of retinal RNA extracts. Levels of opsin message were compared in retinas from normal moths, whose diets were fortified with carotenoid precursors of the Manduca rhodopsin chromopho…

Improved, annotated reference genome for the highly polyphagous moth Hyphantria cunea (Fall webworm).

Fall webworm (Hyphantria cunea) is a widespread, highly polyphagous moth in the family Erebidae, whose native range spans much of North America and invasive range includes Asia and Europe. The species uses over 600 plant species as a larval host, making it among the most generalized insect herbivores described. Its variable host use, wide range, and genetic diversity make fall webworm an attractive emerging model system for the study of diet breadth, but studies have been limited by the lack of a high-quality annotated reference genome. Here we report an annotated, chromosome-scale genome of m

Functional Analysis of PxylPBP2 Responding to Repellent Activity of Natural Pyrazine Against Diamondback Moth.

2,3-dimethyl-6-(1-hydroxy)-pyrazine showed significant repellent activity to diamondback moth (Plutella xylostella), the PxylPBP2 was considered as the potential molecular target, though this has not been validated. In this study, the repellent rate was significantly decreased by about 20% when the PxylPBP2 was silenced using RNAi. There was obvious interaction between 2,3-dimethyl-6-(1-hydroxy)-pyrazine and PxylPBP2, with a Kd (Dissociation constant) value of 7.78 μmol/L via microscale thermophoresis, while the value was changed to 4.45 μmol/L when the PxylOR31 (Pxyl, Plutella xylostella; OR3

Transcriptomic analysis of male diamondback moth antennae: Response to female semiochemicals and allyl isothiocyanate.

Female semiochemicals and allyl isothiocyanate (AITC) attract moths, and the moths use odorant-degrading enzymes (ODEs) to break down the excess odor. By identifying antennae-specific ODEs, researchers have established the molecular foundation for odorant degradation and signal inactivation in insects. This enables further exploration of new pest control methods. Currently, the degradation of female semiochemicals and AITC has received limited attention, inspiring this study to identify target ODEs in diamondback moths through transcriptome analysis. Sequencing of antennae from male adults (MA…

Shedding light with harmonic radar: Unveiling the hidden impacts of streetlights on moth flight behavior.

One of the most dramatic changes occurring on our planet is the ever-increasing extensive use of artificial light at night, which drastically altered the environment to which nocturnal animals are adapted. Such light pollution has been identified as a driver in the dramatic insect decline of the past years. One nocturnal species group experiencing marked declines are moths, which play a key role in food webs and ecosystem services such as plant pollination. Moths can be easily monitored within the illuminated area of a streetlight, where they typically exhibit disoriented behavior. Yet, little…

Identification of the sex pheromone of the tree infesting Cossid Moth Coryphodema tristis (Lepidoptera: Cossidae).

The cossid moth (Coryphodema tristis) has a broad range of native tree hosts in South Africa. The moth recently moved into non-native Eucalyptus plantations in South Africa, on which it now causes significant damage. Here we investigate the chemicals involved in pheromone communication between the sexes of this moth in order to better understand its ecology, and with a view to potentially develop management tools for it. In particular, we characterize female gland extracts and headspace samples through coupled gas chromatography electro-antennographic detection (GC-EAD) and two dimensional gas

Site-specific cassette exchange systems in the Aedes aegypti mosquito and the Plutella xylostella moth.

Genetically engineered insects are being evaluated as potential tools to decrease the economic and public health burden of mosquitoes and agricultural pest insects. Here we describe a new tool for the reliable and targeted genome manipulation of pest insects for research and field release using recombinase mediated cassette exchange (RMCE) mechanisms. We successfully demonstrated the established ΦC31-RMCE method in the yellow fever mosquito, Aedes aegypti, which is the first report of RMCE in mosquitoes. A new variant of this RMCE system, called iRMCE, combines the ΦC31-att integration system

Cutting wild grapevines as a cultural control strategy for grape berry moth (Lepidoptera: Tortricidae).

A 3-yr field study was conducted at commercial grape farms to evaluate cutting wild grapevines as a cultural control strategy for grape berry moth, Paralobesia viteana (Clemens). At each farm, wild grapevines were cut in the woods adjacent to one vineyard for control of P. viteana, whereas the comparison vineyard received no such cutting. Both vineyards received a standard broad-spectrum insecticide program for control of P. viteana and other vineyard insect pests. Monitoring with pheromone traps showed no differences between treatments in the total number of male moths trapped in both woods a…

Do anthropogenic transports facilitate stored-product pest moth dispersal? A molecular approach.

Stored-product moths cause large economic damage in food processing industries and storage facilities. Control of indoor pests is currently dealt with locally, and control strategies seldom include different mills or cooperative industries in joint efforts to reduce infestations. In colder climates where conditions hinder flight dispersal of stored-product moths, we hypothesize that human transport between mills will facilitate dispersal. Albeit considered intuitive, this hypothesis has so far never been tested. Male moths from three mills (populations) in southern Sweden and Denmark were coll…
