Use of Vascularized, Denervated Muscle Targets for Prevention and Treatment of Upper-Extremity Neuromas.

PURPOSE: Neuroma formation following upper-extremity peripheral nerve injury often results in persistent, debilitating neuropathic pain with a limited response to medical management. Vascularized, denervated muscle targets (VDMTs) offer a newly described surgical approach to address this challenging problem. Like targeted muscle reinnervation and regenerative peripheral nerve targets, VDMTs are used to redirect regenerating axons from an injured nerve into denervated muscle to prevent neuroma formation. By providing a vascularized muscle target that is reinnervated via direct neurotization, VD

mTORC1 and PKB/Akt control the muscle response to denervation by regulating autophagy and HDAC4.

Loss of innervation of skeletal muscle is a determinant event in several muscle diseases. Although several effectors have been identified, the pathways controlling the integrated muscle response to denervation remain largely unknown. Here, we demonstrate that PKB/Akt and mTORC1 play important roles in regulating muscle homeostasis and maintaining neuromuscular endplates after nerve injury. To allow dynamic changes in autophagy, mTORC1 activation must be tightly balanced following denervation. Acutely activating or inhibiting mTORC1 impairs autophagy regulation and alters homeostasis in denerva

Role of transforming growth factor-β1 in the process of fibrosis of denervated skeletal muscle.

In order to investigate the biological function of transforming growth factor-β1 (TGF-β1) during fibrosis in denervated skeletal muscle, we recruited sciatic nerve injury model of SD rats in which denervated gastrocnemius was isolated for analysis. At different time points after operation, denervated muscle was examined by several methods. Masson trichrome staining showed morphological changes of denervated skeletal muscle. Quantitative RT-PCR detected the rapid increase of TGF-β1 expression at mRNA level after nerve injury. It was found that a peak of TGF-β1 mRNA expression appeared one week

A potential indicator of denervated muscle atrophy: the ratio of myostatin to follistatin in peripheral blood.

Myostatin is a secreted negative regulator of muscle mass, and follistatin antagonizes the function of several members of the TGF-b family, including myostatin. Previously, myostatin expression was found to be closely associated with atrophy of the gastrocnemius muscle, showing a linear correlation, after sciatic nerve injury. In this study, we investigated the possibility of myostatin being an indicator of denervated muscle atrophy. ELISA was used to detect the concentration of myostatin and follistatin in sera collected from individual rats at different times after sciatic nerve crush. A str

Functional modulation of satellite cells in long-term denervated human laryngeal muscle.

OBJECTIVES/HYPOTHESIS: To evaluate the effects of long-term denervation on satellite cells (SCs) as myogenic stem cells in human posterior cricoarytenoid (PCA) muscle. STUDY DESIGN: Histological investigation of SCs and quantitative assessment of myoD and myogenin, which are two key myogenic regulatory factors. METHODS: According to the course of denervation, denervated PCA muscles of 58 patients who suffered from traumatic unilateral vocal cord paralysis were divided into four groups: group A (6-12 months, 15 cases), group B (13-24 months, 17 cases), group C (25-36 months, 14 cases), and gr

Juvenile parameningeal rhabdomyosarcoma in a dog causing unilateral denervation atrophy of masticatory muscles.

A 23-month-old, male, Labrador retriever dog with a history of slowly progressive right-sided atrophy of the masticatory muscles was submitted for necropsy. A highly invasive neoplasm which destroyed adjacent soft tissues including the right trigeminal nerve was found in the right side of the cranial cavity. Metastases to the liver were also present. Microscopical features of the neoplasm were compatible with those of rhabdomyosarcoma, embryonal type. This diagnosis was confirmed by immunohistochemical demonstration of desmin and muscle actin within tumour cells. In human patients, rhabdomyosa

Matrix metalloproteinases MMP-2 and MMP-9 in denervated muscle and injured nerve.

Nerve crush or axotomy results in a transient or longterm denervation accompanied by remodelling in nerve, muscle and neuromuscular junctions. These changes include an increased turnover of several extracellular matrix molecules and proliferation of Schwann cells in injured nerves. Given the role of matrix degrading metalloproteinases MMP-2 and MMP-9 (gelatinases-type IV collagenases) in extracellular matrix remodelling, we investigated their regulation and activation in denervated muscles and injured nerves in mice. For this, immunofluorescence using MMP-2 and MMP-9 antibodies was carried con

Cosmetic denervation of the muscles of facial expression with botulinum toxin. A dose-response study.

BACKGROUND: Botulinum toxin has been used for facial hemispasm, strabismus, and blepharospasm. Recently it has been advocated to treat the frown lines. We have extended this program to treatment of other muscles of facial expression. METHODS: Botulinum toxin is injected into the muscles of facial expression in two or three sessions to produce a temporary loss of muscle tone. A standard method of cooling, injection, and compression was developed to minimize pain and bruising. OBJECTIVE: To complete a dose-response study to document the optimum timing and amount of toxin needed for each muscle

Scanning electron microscopic study of denervated and reinnervated intrafusal muscle fibers in rats.

Isolated muscle spindles from lower lumbrical muscles of rats were used to study the 3-dimensional organization of intrafusal structures by scanning electron microscopy following (a) complete denervation, (b) reinnervation after a single crush lesion of the sciatic nerve, or (c) reinnervation after transection and immediate suture of this nerve. One week after complete denervation, previous sites of intrafusal motor endplates were transformed into sarcolemmal ovoid bulges. These bulges persisted in denervated muscle spindles up to 12 weeks. Regenerated motor nerve endings were detected on intr

Ultrastructural and histochemical study of the neuromuscular junctions in the denervated intrinsic laryngeal muscle of the cat.

Morphological changes and acetylcholinesterase (AchE) activity in the neuromuscular junctions (NMJ) of the normal and denervated posterior cricoarytenoid muscle (PCA muscle) of the cat were studied. Two days after denervation, the nerve terminals at the NMJ had almost disappeared. Six weeks after denervation, intensity of AchE activity at the former junctional site (FJS) was unchanged histochemically. At this stage, primary synaptic clefts were distorted and the Schwann's cells covered the FJS. Fourteen weeks after denervation, AchE activity at the FJS had decreased in contrast to that of the

[Functional electrostimulation of the denervated posticus muscle in an animal experiment: histo- and biochemical results].

Histochemical and biochemical investigations have been carried out in 2 sheep following unilateral recurrent laryngeal nerve paralysis and direct electrical stimulation of the denervated posticus muscles. The stimulation effect was determined histochemically (standard ATP-ase staining) and compared with the fibre pattern of normal posticus muscles. In addition, one-dimensional gel electrophoresis of myosin heavy-chain isoforms was carried out and correlated with the histochemical results. A dramatic increase of type I fibres could be seen after long-term low-frequency direct electrical stimula

Tomographic evaluation of size and X-ray density of normal and denervated human muscles.

Neuromuscular disease will lead to size changes, degeneration or destruction of muscle fibres. However, fat or connective tissue infiltration occurs and this prevents the use of simple measures to assess wasting. Computerized tomography will allow accurate assessment of this wasting. Five normal subjects and five patients with total traumatic denervation of radial or peroneal origin were examined. Denervated muscle was shown to have lower than normal cross-section and X-ray density. These reductions were not paralleled in circumference or limb volume. Recovery could be followed by tomography.

Effects of denervation and colchicine treatment on the chloride conductance of rat skeletal muscle fibers.

Membrane potentials, cable parameters, and component resting conductances were measured in extensor digitorum longus (EDL) muscle fibers from adult rats in vitro at 24 degrees C, after 15 to 18 days of denervation by nerve section, and at seven to ten days following epineural injection of 100 to 450 mug of colchicine in the peroneal nerve. The denervated muscles were paralyzed throughout the experimental period, whereas the colchicine-treated preparations showed no clinical paralysis except for the first day or two. The EDL from the untreated side served as a control. Both the denervated and c

Relationship between muscle FNDC5, FGF21 mRNA, miR-150 and bone loss after sciatic denervation.

It has been proposed that muscle influences adjacent bones, but the mechanisms by which muscle regulates bone physiology are not fully understood. Sciatic denervation is an experimental model that induces both skeletal muscle atrophy and osteoporosis. Therefore, the aim of this study was to analyze, in this animal model, expression of myokines in the atrophic soleus and its possible effect on changes in tibia microarchitectural parameters in 9-12 weeks old male rats. For this purpose, soleus insulin-like growth factor-1 (IGF-1), interleukin-6 (IL-6), fibronectin type III domain-containing 5 (F

T-cell immunosuppression in sepsis is augmented by sciatic denervation-induced skeletal muscle atrophy.

Skeletal muscle atrophy is a known risk factor for immunosuppressive conditions and for a poor prognosis in sepsis. However, its immunopathology has not been experimentally elucidated. This study investigated the effects of skeletal muscle atrophy on the immunopathology of sepsis. Male C57BL/6J mice were subjected to sciatic denervation (DN) and caecal ligation and puncture (CLP) to induce muscle atrophy or sepsis. The macrophages, myeloid-derived suppressor cells (MDSC), and T-cells in peritoneal and spleen were analysed using flow cytometry. DN-induced muscle atrophy did not affect macrophag

Therapeutic potential of omaveloxolone in counteracting muscle atrophy post-denervation: a multi-omics approach.

BACKGROUND: Muscle atrophy caused by denervation is common in neuromuscular diseases, leading to loss of muscle mass and function. However, a comprehensive understanding of the overall molecular network changes during muscle denervation atrophy is still deficient, hindering the development of effective treatments. METHOD: In this study, a sciatic nerve transection model was employed in male C57BL/6 J mice to induce muscle denervation atrophy. Gastrocnemius muscles were harvested at 3 days, 2 weeks, and 4 weeks post-denervation for transcriptomic and proteomic analysis. An integrative multi-om

Altered m6A RNA methylation governs denervation-induced muscle atrophy by regulating ubiquitin proteasome pathway.

BACKGROUND: Denervation-induced muscle atrophy is complex disease involving multiple biological processes with unknown mechanisms. N6-methyladenosine (m6A) participates in skeletal muscle physiology by regulating multiple levels of RNA metabolism, but its impact on denervation-induced muscle atrophy is still unclear. Here, we aimed to explore the changes, functions, and molecular mechanisms of m6A RNA methylation during denervation-induced muscle atrophy. METHODS: During denervation-induced muscle atrophy, the m6A immunoprecipitation sequencing (MeRIP-seq) as well as enzyme-linked immunosorbe
