OBJECTIVE: Cancer cell reprogramming is a potential tool to study cancer progression, disease pathology, and drug sensitivity. Prior to performing cancer reprogramming studies, it is important to evaluate the stemness predisposition of cells that will be reprogrammed. We performed a proof-of-concept study with chronic myeloid leukemia K562 cells in order to evaluate their tendency for cancer cell reprogramming.
MATERIALS AND METHODS: Expression of reprogramming factors, pluripotency markers, and tumor-suppressor genes was analyzed at gene and protein levels via real-time reverse transcription

Chronic myeloid leukemia is a clonal malignancy of hematopoietic stem cell that is characterized by the occurrence of t(9;22)(q34;q11.2) translocation, named Philadelphia chromosome. Ruxolitinib is a powerful Janus tyrosine kinase 1 and 2 inhibitor that is used for myelofibrosis treatment. DNA-histone connection mediates a wide range of genes that code methylation, demethylation, acetylation, deacetylation, ubiquitination, and phosphorylation enzymes. Epigenetic modifications regulate chromatin compactness, which plays pivotal roles in critical biological processes including the transcriptiona

Drug resistance to tyrosine kinase inhibitors (TKIs) is currently a clinical problem of chronic myelogenous leukemia (CML). Bcr-Abl protein depletion is considered as a way to overcome drug resistance to TKIs. In our study, Chk1 inhibitors, AZD7762 and MK-8776, had strong antitumor effects on CML cell line KBM5 and imatinib-resistant form KBM5T315I. Moreover, Chk1 inhibitors showed a strong cytotoxic effect on leukemia cells from primary CML and imatinib-resistance CML patients, but low cytotoxic effect on normal human mononuclear cells. Then, we found that Chk1 inhibitors induced apoptosis an

Advances in the design of targeted therapies for the treatment of chronic myeloid leukemia (CML) have transformed the prognosis for patients diagnosed with this disease. However, leukemic stem cell persistence, drug intolerance, drug resistance, and advanced-phase disease represent unmet clinical needs demanding the attention of CML investigators worldwide. The availability of appropriate preclinical models is essential to efficiently translate findings from the bench to the clinic. Here we review the current approaches taken to preclinical work in the CML field, including examples of commonly

Despite advances in the treatment of acute myeloid leukemia (AML), relapse and drug resistance frequently occur. Therefore, detailed mechanisms of refractoriness, including leukemia-initiating cell (LIC) biology, should be elucidated to treat AML. The self-degradative property of cytosolic macromolecules is central to autophagy and can contribute to homeostasis and stress response. Recent reports suggest the importance of autophagy in hematopoietic stem cells and various tumors. Thus, this study investigated the functional role of autophagy in AML maintenance and drug resistance using tamoxife

FOXP1 protein was firstly analyzed in normal tissues, and afterwards in different tumor tissues, mainly carcinoma and lymphoma. In B-cell malignancies, its role was well explored; its expression was shown to be connected with disease prognosis in certain B-non Hodgkin lymphomas. In this study, 16 bone marrow trephine samples from patients with no hematopoietic malignancies and 10 samples from peripheral blood of healthy individuals were immunostained with anti-FOXP1 antibody. Positive cells in bone marrows were not only lymphocytes, but also cells that are immunohistochemically positive for gl

INTRODUCTION: Chronic myeloid leukemia (CML) is a myeloproliferative disorder characterized by the translocation t(9;22), coding for the chimeric protein BCR-ABL. The development of BCR-ABL tyrosine kinase inhibitors (TKIs) has dramatically revolutionized and improved CML therapy. However, TKI-based therapy faces a major challenge: the insensitivity of CML leukemic stem cells (LSCs) to TKIs. In particular, while CML progenitor cells and differentiated cells are oncogene addicted, BCR-ABL tyrosine kinase is dispensable for CML LSC survival and maintenance. Notably, in CML, additional cellular m

The physiological hematopoietic niche located in bone marrow is a pluricellular structure whose components are now well identified. Within t … ed. Consequently, the occurrence of the so-called leukemic niche promotes both the proliferation of myeloid cells and the maintenance of quiescent leukemic stem cells. This bone marrow niche could also protect CML stem cells from tyrosine kinase inhibitors and probably contribute to their resistance towards targeted therapies.

Chronic neuropathic pain has become a real social issue, due to the difficulty of its treatment and by the major impairment to quality of life that it causes in every day behavior. Understanding neurobiological basis and pathophysiological causes of diverse painful syndromes constantly evolves and reports the complexity of its mechanisms. Unfortunately this complexity makes it difficult to discover effective treatments against chronic pain syndromes, in particular as regards peripheral neuropathic pains. Recent studies reveal that, during chronic peripheral neuropathy, inflammatory mediators (

Bone marrow-derived cells (BMDCs) are able to colonize the central nervous system (CNS) at sites of damage. This ability makes BMDCs an ideal cellular vehicle for transferring therapeutic genes/molecules to the CNS. However, conditioning is required for bone marrow-derived myeloid cells to engraft in the brain, which so far has been achieved by total body irradiation (TBI) and by chemotherapy (e.g. busulfan treatment). Unfortunately, both regimens massively disturb the host's hematopoietic compartment. Here, we established a conditioning protocol to target myeloid cells to sites of brain damag…

Chronic myeloid leukemia is a neoplasia resulting from a translocation between chromosomes 9 and 22 producing the BCR-ABL hybrid known as the Philadelphia chromosome (Ph). In chronic myeloid leukemia a proliferation of malignant myeloid cells occurs in the bone marrow due to excessive tyrosine kinase activity. In order to maintain homeostasis, natural killer cells, by means of receptors, identify the major histocompatibility complex on the surface of tumor cells and subsequently induce apoptosis. The NKG2D receptor in the natural killer cells recognizes the transmembrane proteins related to ma…

We previously demonstrated that severe hypoxia inhibits growth of Chronic Myeloid Leukemia (CML) cells and selects stem cells where BCR/Abl(protein) is suppressed, although mRNA is not, so that hypoxia-selected stem cells, while remaining leukemic, are independent of BCR/Abl signaling and thereby refractory to Imatinib-mesylate. The main target of this study was to address the effects of the proteasome inhibitor Bortezomib (BZ) on the maintenance of stem or progenitor cells in hypoxic primary cultures (LC1), by determining the capacity of LC1 cells to repopulate normoxic secondary cultures (LC

BACKGROUND AND PURPOSE: Early detection of resistance development is crucial for imatinib-based treatment in chronic myeloid leukaemia (CML) patients. We aimed to distinguish metabolic markers of cell resistance to imatinib.
EXPERIMENTAL APPROACH: Two human imatinib-sensitive CML cell lines: LAMA84-s and K562-s, and their resistant counterparts: LAMA84-r and K562-r (both resistant to 1 microM imatinib), and K562-R (5 microM) were analysed by nuclear magnetic resonance spectroscopy to assess global metabolic profiling, including energy state, glucose and phospholipid metabolism.

Fanconi anaemia (FA) is a genetically heterogeneous chromosome instability syndrome characterised by bone marrow failure and congenital anomalies. Although an increasing number of reports suggest that reversion mosaicism noted in peripheral blood lymphocytes (PBLs) is associated with mild haematopoietic failure in FA, myeloid cells are rarely directly examined. We here report a patient with prolonged mild pancytopenia in whom proliferation of revertant cells was detected in mature myeloid cells but not in PBLs. While this patient had inherited heterozygous mutations, 2546delC and 3720-3724del,…

A 63-year-old man with refractory anemia with excess of blasts in transformation exhibited erythroid hyperplasia, dyserythropoiesis, a del(20q) abnormality, susceptibility to bacterial infections, and a relatively short survival. Phagocytosis of erythrocytes by blast cells was observed. Erythrophagocytosis was also seen in myeloid cells, including promyelocytes, myelocytes, metamyelocytes and neutrophils. Neither the monocytes nor the macrophages showed erythrophagocytosis. This is the first report of erythrophagocytosis by blasts and myeloid cells in a patient with myelodysplastic syndrome.

Cells from a clone of mouse myeloid leukemic cells that can be induced to differentiate in vitro to mature cells by the normal macrophage- and granulocyte-inducing protein, MGI-2, were injected into mouse embryos at 10 days gestation. The leukemic cells, derived from an SJL/J mouse, were microinjected into the placentae of C3HeB fetuses and the viable progeny were tested for chimerism by analysis of glucose phosphate isomerase isozymes. Fifty-five percent out of 201 viable progeny died prior to weaning due to tumors derived from the injected cells. Of the remaining 91 apparently healthy adult

Acute myeloid leukemia (AML) is a hematological malignancy characterized by impaired hematopoiesis and aggressive progression, with limited treatment options and poor prognosis, especially in elderly patients. Metabolic reprogramming, particularly enhanced glycolysis, is a hallmark of AML and contributes to disease progression and therapy resistance. Upstream stimulatory factor 1 (USF1) regulates metabolism, the cell cycle, and oncogenesis in solid tumors, whereas its role in AML remains unclear. Herein, we investigated the role and underlying mechanism of USF1 in regulating USP25 transcriptio

BACKGROUND: Myeloid-derived suppressor cells (MDSCs) play a pivotal role in shaping the immunosuppressive tumor microenvironment, thereby driving tumor progression and contributing to resistance to therapy. The surface molecules expressed on MDSCs serve not only as identification markers but also as critical regulators of their immunosuppressive functions. Identifying surface markers that are both widely expressed and functionally significant on MDSCs is essential for the development of effective immunomodulatory therapies.
METHODS: A comprehensive analysis of publicly available single-cell R

Sjögren's Disease (SjD) is a chronic autoimmune disease characterized by exocrine glandular lymphocyte infiltration and functional damage, with a complex pathogenesis and immune cell imbalance as a key driver. Myeloid-derived suppressor cells (MDSCs), as heterogeneous immature myeloid cells, play an immunosuppressive function by secreting molecules such as arginase 1, nitric oxide, and reactive oxygen species, and play a dual role in tumor immune escape and autoimmune disease regulation. In recent years, studies have shown that the frequency of MDSCs increases abnormally in SjD patients and an…
