Demonstration of myoglobin in formalin-fixed renal sections by immunoperoxidase technic.

The use of an immunohistochemical technic offers a very sensitive method to detect myoglobin in formalin-fixed, paraffin-embedded tissues. Thus, the technic is suitable for retrospective studies. Myoglobin can be demonstrated at the brush border and apical cytoplasm of the tubular epithelium of proximal tubules. Granular substances and casts in the lumens also react positively with antimyoglobin. Our findings confirm the results of a previous animal study, showing that myoglobin is filtered through glomeruli and reabsorbed by the proximal tubules.

The detection of myoglobin in urine and its application to the diagnosis of myocardial infarction.

An indirect haemaglutination method was developed for the immunochemical detection of myoglobin in human urine. Myoglobin was found in the urine of 84% of 44 patients with acute myocardial infarction. In contrast, it was detected in the urine of 2.7% of 147 control subjects and in none of a control group of 15 patients who had received intramuscular injections. In patients with myocardial infarction, myoglobin was detected in most but not all urine specimens collected between 10 and 50 hours after the onset of chest pain. The results suggest that the detection of myoglobin in urine may be a va…

Myoglobin/sol-gel film modified electrode: direct electrochemistry and electrochemical catalysis.

Direct electrochemical and electrocatalytic behavior of myoglobin (Mb) immobilized on carbon paste electrode (CPE) by a silica sol-gel film derived from tetraethyl orthosilicate was investigated for the first time. Mb/sol-gel film modified electrodes show a pair of well-defined and nearly reversible cyclic voltammetric peaks for the Mb Fe(III)/Fe(II) redox couple at about -0.298 V (vs Ag/AgCl) in a pH 7.0 phosphate buffer solution. The formal potential of the Mb heme Fe(III)/Fe(II) couple shifted linearly with pH with a slope of 52.4 mV/pH, denoting that an electron transfer accompanies single…

Myoglobin inhibits proliferation of cultured human proximal tubular (HK-2) cells.

Following nephrotoxic injury, renal repair is dependent on tubular regeneration. In the case of myoglobinuric acute renal failure (ARF), persistence of myoglobin within tubular cells, or sublethal injury sustained at the height of exposure to it, might retard this process. To test this hypothesis, a human proximal tubular cell line (HK-2) was cultured for 24 hours in the absence or presence of clinically relevant myoglobin concentrations (0.5, 1, 2, 4 mg/ml). Immediately following myoglobin removal, lethal cell injury (vital dye uptake), lipid peroxidation, and DNA damage (alkaline unwinding a…

The amino acid sequence of alligator (Alligator mississippiensis) myoglobin. Phylogenetic implications.

The amino acid sequence of myoglobin from cardiac muscle of the American alligator (Alligator mississippiensis) was established by alignment of overlapping peptides and dansyl-Edman degradation. The chain initiating methonine seems to be retained. Thus, this molecule has 154 amino acid residues rather than 153 as in other tetrapod myoglobins. Maximum parsimony analysis indicated that alligator myoglobin diverges more from both bird and mammal myoglobins than these latter do from each other. The phylogenetic implications are discussed with respect to a possible diphyletic origin for diapsid rep…

Detecting muscle strain in professional football players through plasma myoglobin levels.

BACKGROUND: Muscle issues such as soreness, cramps, and strains are common consequences of sports exercise, making it critical to accurately identify the nature of muscle discomfort. It is important for not only medical professionals but also athletes and sports enthusiasts to independently identify and manage the risk of muscle strain. METHODS: This prospective observational study explores the effectiveness of measuring blood myoglobin (MB) levels as a method for detecting muscle strains among athletes. The study involved 30 male professional footballers monitored over one season. MB levels …

Myoglobin expression by alternative transcript in different mesenchymal stem cells compartments.

BACKGROUND: The metabolic phenotype of stem cells is increasingly recognized as a hallmark of their pluripotency with mitochondrial and oxygen-related metabolism playing a not completely defined role in this context. In a previous study, we reported the ectopic expression of myoglobin (MB) in bone marrow-derived hematopoietic stem/progenitor cells. Here, we have extended the analysis to mesenchymal stem cells (MSCs) isolated from different tissues. METHODS: MSCs were isolated from human placental membrane, mammary adipose tissue and dental pulp and subjected to RT-PCR, Western blotting and ma…

Monitoring myoglobin by capillary zone electrophoresis with end-column amperometric detection.

Capillary zone electrophoresis was employed for the determination of myoglobin in human urine using end-column amperometric detection with a carbon fiber microelectrode at a constant potential of 1.80 V vs. saturated calomel electrode (SCF). The optimum conditions of separation and detection are: 3.73 x 10-4 mol/L sodium diethyl malonyl urea (barbitone sodium), 1.34 x 10-4 mol/L HCl for the buffer solution, 20 kV for separation voltage, 5 kV and 5 s for injection voltage and injection time, respectively. The limit of detection is 4.4 x 10-8 mol/L or 84 amole signal to noise (S/N = 2). The rela…

Real-Time Dual Sensing of cTnI and Myoglobin via Printable Graphitized Electrodes.

Acute myocardial infarction (AMI) has emerged as a serious global health concern. According to the World Health Organization (WHO), health authorities report approximately 100 million cases annually worldwide, with about 9 million associated mortalities. As AMI can affect individuals regardless of their age or prior health status, making early and pre-symptomatic detection is crucial for clinical intervention. Targeting AMI biomarkers such as cardiac troponin I (cTnI) and myoglobin is therefore essential for timely diagnosis and treatment. Hence, the development of ultra-sensitive and highly s…

Horse heart mini- and full length-myoglobin: pH effects on CO binding.

Mini-myoglobin (mini-HH-Mb) is a proteolytic fragment of horse heart myoglobin (HH-Mb) comprising residues 32-139, grossly corresponding to the central exon of the HH-Mb gene, which encodes residues 31-105. Unlike HH-Mb, which displays a single exponential for both CO association and CO dissociation kinetics, mini-HH-Mb shows a biphasic kinetic behavior for both processes, indicating the presence of at least two distinct conformations which are in a very slow (or no) equilibrium with each other. Between pH 2 and 12, CO association to both species of mini-HH-Mb shows two proton-linked transitio…

Reactions of ferric hemoglobin and myoglobin with hydrogen sulfide under physiological conditions.

Ferric hemoglobin (metHb) and myoglobin (metMb), present at low levels in vivo, have been recently found to oxidize hydrogen sulfide (H2S) in excess, thus potentially contributing to removal of toxic H2S in blood and heart, respectively. Here, we present a kinetic and thermodynamic study of the reaction of metHb and metMb with H2S under physiological conditions, i.e. at low H2S concentrations and with protein in excess of H2S. We show here that both proteins react with sub-stoichiometric H2S:heme ratios following two processes: a fast reversible binding of H2S to ferric heme that prevails at h…

Picosecond dynamics of photoexcited DNO-bound myoglobin probed by femtosecond vibrational spectroscopy.

Like nitric oxide (NO), nitroxyl (HNO), a reduced form of NO, plays many biologically important roles including neurological function and vascular regulation. Although HNO is unstable in aqueous solution, it is exceptionally stable on binding to ferrous myoglobin (Mb) to form MbHNO. Various experimental and theoretical investigations has been carried out to unveil the structure of the active site and binding characteristics of MbHNO that can explain its functioning mechanism and the origin of its unusual stability. However, the binding dynamics of HNO to Mb, as well as the photochemical and ph…

Myoglobin cast nephropathy in a kidney transplant patient with normal creatine kinase.

Delayed graft function in kidney transplant recipients is a known complication associated with increased risk of acute rejection and reduced transplant survival after 1 year. There are multiple risk factors, including prolonged cold ischemia time, donor age, and cause of donor's death. Major causes of delayed graft function are acute kidney injury in the donor, often from prolonged terminal ischemia, reflected by acute tubular injury in the recipient. However, the differential diagnosis of delayed graft function includes acute rejection, recurrence of the primary glomerular diseases, and other

Inherently distorted heme as a novel tool for myoglobin-based oxygen carrier.

The two types of artificial myoglobins (Mbs) containing inherently distorted α-ethyl-2,4-dimethyldeuteroheme and undistorted 2,4-dimethyldeuteroheme were prepared to examine the influence of nonplanar heme deformation on the reactivity of Mb. In ferrous deoxy proteins, the paramagnetic proton nuclear magnetic resonance spectra showed that the deformed heme caused a 3.2-ppm lower-field shift of the proximal histidine signal, indicating a larger iron displacement from the heme plane upon the nonplanar deformation. The Mb with the nonplanar heme exhibited a markedly lower oxygen affinity as compa…

Characterization of myoglobin toxicity in renal cortical slices from Fischer 344 rats.

Rhabdomyolysis is associated with acute renal failure. The following study first characterized myoglobin in vitro toxicity using renal cortical slices isolated from male Fischer 344 rats. This model provided interaction between various cells within the nephron and provides myoglobin access predominantly through the basolateral membrane. Second, this study examined the effect of deferoxamine (DFX) and glutathione on myoglobin toxicity to determine the role of radicals and iron. Renal cortical slices were incubated for 30-120 min with 0, 4, 10 or 12 mg/ml myoglobin. Myoglobin was pretreated with…
