Ultrasound assessment of peripheral nerve pathology in neurofibromatosis type 1 and 2.

OBJECTIVE: The neurofibromatoses (NF) type 1 and 2 are hereditary tumor predisposition syndromes caused by germline mutations in the NF1 and NF2 tumor suppressor genes. In NF1 and 2, peripheral nerve tumors occur regularly. For further characterizing nerve ultrasound was performed in patients with NF1 and 2. METHODS: Patients with established diagnosis of NF1 (n=27) and NF2 (n=10) were included. Ultrasound of peripheral nerves and cervical roots was performed during routine follow-up visits. Healthy volunteers were studied for comparison. RESULTS: In patients with NF1, median cross-sectional…

Back to the future: proceedings from the 2010 NF Conference.

The neurofibromatoses (NF) encompass the rare diseases NF1, NF2, and schwannomatosis. The NFs affect 100,000 Americans; over 2 million persons worldwide; and are caused by mutation of tumor suppressor genes. Individuals with NF1 in particular may develop tumors anywhere in the nervous system; additional manifestations can include learning disabilities, bone dysplasia, cardiovascular defects, unmanageable pain, and physical disfigurement. Ultimately, the NFs can cause blindness, deafness, severe morbidity, and increased mortality and NF1 includes a risk of malignant cancer. Today there is no tr…

CTF meeting 2012: Translation of the basic understanding of the biology and genetics of NF1, NF2, and schwannomatosis toward the development of effective therapies.

The neurofibromatoses (NF) are autosomal dominant genetic disorders that encompass the rare diseases NF1, NF2, and schwannomatosis. The NFs affect more people worldwide than Duchenne muscular dystrophy and Huntington's disease combined. NF1 and NF2 are caused by mutations of known tumor suppressor genes (NF1 and NF2, respectively). For schwannomatosis, although mutations in SMARCB1 were identified in a subpopulation of schwannomatosis patients, additional causative gene mutations are still to be discovered. Individuals with NF1 may demonstrate manifestations in multiple organ systems, includin…

Exploring the cucurbitacins in neurofibromatosis: Molecular mechanisms and pharmacological potential.

Neurofibromatosis (NF) operates as a hereditary tumor-predisposition disease that primarily results from NF1 and NF2 gene mutations which cause the formation of noncancerous and cancerous tumors that affect the nervous system. The dysregulated operation of crucial signaling pathways, which include JAK/STAT and PI3K/Akt/mTOR and Ras/MAPK, has a major effect on how tumors start growing and their following development. The field of targeted therapeutics has made progress yet the existing long-term treatment options remain insufficient which drives the search for new therapeutic compounds that can

Neurofibromatosis.

There are two types of neurofibromatosis (NF): NF1 (type 1, peripheral NF, or von Recklinghausen syndrome); NF2 (type 2, central or bilateral acoustic NF). The mode if inheritance is autosomal dominant.

Reevaluating hybrid neurofibroma/schwannoma: Predominance of schwannoma features despite CD34 positivity and initial neurofibroma diagnosis.

Schwannomas consist of both high-cellularity regions (Antoni A area) and hypocellular regions (Antoni B area) in histopathological findings. Neurofibromas characteristically consist of CD34 positive spindle cells with thin, wavy, nuclei and wavy collagen bands. Previous reports have described segments of schwannomas with neurofibroma features as hybrid tumors, although hybrid tumors were diagnosed based on partial CD34 positivity in many previous reports. On the other hand, the Antoni B area of some schwannomas was reported to be positive for CD34. Therefore, the definition of a hybrid tumor h

Schwannomatosis: a Realm Reborn: year one.

PURPOSE OF REVIEW: In 2022, an international consensus recommendation revised the nomenclature for neurofibromatosis type 2 ( NF2 ) and Schwannomatosis (SWN), now grouped under the umbrella term Schwannomatosis, and defined new diagnostic criteria. RECENT FINDINGS: This review describes the molecular criteria for diagnosis of schwannomatosis and the subsequent diagnosis strategy, while setting out the most recent advances in our understanding of the natural history, pathology, molecular biology and treatment of schwannomatosis-associated tumors, including schwannomas, meningiomas and ependymo

Submandibular nodal schwannoma: where did it come from?

SummarySchwannoma, a type of benign neurological tumour arising from Schwann cells, is commonly seen in the head and neck region. However, the incidence of cervical lymph node schwannoma is extremely rare, with only two cases reported in the literature. Schwannomatosis is a disease characterised by a constellation of multiple schwannomas seen in a patient, guided by at least one confirmational histological diagnosis. This is a case of a painless progressively enlarging submandibular swelling which is confirmed as a schwannoma based on histology. The patient is also diagnosed with schwannomatos

Management of neurofibromatosis type 2 and schwannomatosis associated peripheral and intraspinal schwannomas: influence of surgery, genetics, and localization.

INTRODUCTION: Peripheral and intraspinal schwannomas are common and clinically complex pathologies in patients with Neurofibromatosis Type 2 (NF2) and Schwannomatosis (SWNT). Functional preservation and pain relief are the major goals in treating these tumors. METHODS: This retrospective observational study investigates the clinical and functional outcome of 205 operated peripheral (n = 148, 72%) and intraspinal (n = 57, 28%) schwannomas in 85 patients (53 NF2, 32 SWNT) treated at our department between 2006 and 2017. Associated factors such as genetics, age, and location were evaluated. RES

Images in clinical medicine: Segmental neurofibromatosis.

A 4-year-old girl presented for evaluation of cutaneous nodules that had been present on the left side of her trunk since birth. Physical examination revealed numerous soft, nontender papules and nodules across the left upper abdomen and wrapping around to the back.

Incidental (malignancy) and coincidental (idiopathic polydactylous longitudinal erythronychia) conditions in patients with segmental neurofibromatosis.

Segmental neurofibromatosis (SNF) is an uncommon presentation of neurofibromatosis type 1 (NF-1). Although patients with SNF are at a lower risk for developing NF-l-associated complications, the estimated occurrence of related malignancies may be approaching the frequency observed in patients with NF-1. Idiopathic polydactylous longitudinal erythronychia also may be associated with SNF, though the frequency of this association remains to be determined.

Primary actinomycosis of the foot in a patient with neurofibromatosis: a case report.

Actinomycosis is a chronic and suppurative infection caused by an endogenous gram-positive bacterium. The usual sites of infection are the head and neck, thorax, and abdomen and are almost always endogenous in origin. Primary actinomycosis is rare and is usually associated with external trauma. The authors report a case of a primary actinomycosis presented as a mass on the foot that mimics neurofibroma in a 56-year-old Asian woman with neurofibromatosis. The patient has no history of preceding trauma. She was treated with excisional biopsy. Histopathology of the excised mass showed features of

[Neurofibromatosis of the lower cervical spine: an operative case report].

PURPOSE OF THE STUDY: We report a case of type 1 neurofibromatosis (von Recklinghausen's disease) of the lower cervical spine in a 13-year-old girl. CASE REPORT: There was no neurological deficit. Plain films showed dysplastic 82 degrees kyphosis centered on the C4-C5 disc. Surgical treatment consisted in anterior multilevel interbody grafting and plate osteosynthesis combined with posterior arthrodesis. Good bone fusion was obtained with acceptable cervical mobility. The residual cervical kyphosis was 18 degrees. DISCUSSION: An evaluation of the cervical spine should be proposed for patient
