Tight neurovascular coupling in the spinal cord during nociceptive stimulation in intact and spinal rats.

Functional magnetic resonance imaging (fMRI) is based on neurovascular coupling, which allows inferring neuronal activity from hemodynamic changes. Spinal fMRI has been used to examine pain processes, although spinal neurovascular coupling has never been investigated. In addition, fluctuations in mean arterial pressure (MAP) occur during nociceptive stimulation and this may affect neurovascular coupling. The objective of this study was to examine neurovascular coupling in the rat spinal cord during nociceptive stimulation while MAP was manipulated by cervical spinal transection, which prevents…

Interplay of cerebral autoregulation and neurovascular coupling evaluated by functional TCD in different orthostatic conditions.

BACKGROUND: Adequate cerebral blood flow (CBF) is mainly governed by neurovascular coupling (NC) which adapts local CBF to underlying cortical activity, and cerebral autoregulation (CA)that tends to maintain constant CBF despite changes in arterial blood pressure (BP). Since it was suggested that resistance vessels play an important role in both mechanisms, we investigated the irregulative interplay by performing a functional transcranial Doppler(f-TCD) test under different orthostatic conditions. METHODS: Fifteen healthy volunteers performed a visual reading test stimulation task after stabil…

Neurovascular coupling in simultaneous EEG-fNIRS studies: A scoping review of analytical frameworks and clinical applications.

Simultaneous electroencephalography (EEG) and functional near-infrared spectroscopy (fNIRS) have been used across cognitive and physiological states and in diverse clinical populations. However, the interpretation of cross-modal coupling remains difficult because findings from different analytical frameworks, populations, and experimental conditions have not been systematically mapped. This scoping review followed the Arksey and O'Malley framework and the Preferred Reporting Items for Systematic Reviews and Meta-Analyses extension for Scoping Reviews (PRISMA-ScR). Trifluoperazine reduces infarct size, restores neurovascular coupling and improves early outcomes in experimental acute ischemic stroke.

Cerebral edema and neurovascular dysfunction are reliable predictors of outcome after acute ischemic stroke (AIS), yet effective targeted therapies remain limited. We investigated the therapeutic potential of trifluoperazine (TFP), an FDA-approved antipsychotic and calmodulin inhibitor that modulates astrocytic aquaporin-4 expression. TFP administered after recanalization in a mouse model of AIS reduced infarct volume and improved early neurological recovery. Importantly, TFP restored neurovascular coupling and enhanced cerebral blood flow responses to spreading depolarizations, indicating imp…

Neurovascular Coupling Disruption and Glymphatic Dysfunction Are Associated With Visual Impairment in Pediatric Monocular Anisometropic Amblyopia.

PURPOSE: To investigate abnormalities in neurovascular coupling and the glymphatic metabolic clearance in children with monocular anisometropic amblyopia, as well as their associations with visual acuity (VA) status. METHODS: Multimodal magnetic resonance imaging scans were acquired from 33 children with anisometropic amblyopia and 31 age-matched normal controls. Neurovascular coupling was quantified as the spatial correlation between fractional amplitude of low-frequency fluctuations and cerebral blood flow across 90 cortical regions. Glymphatic metabolic clearance was evaluated using diffus…

Abnormal neurovascular coupling induces glymphatic dysfunction in a mouse model of familial hemiplegic migraine type 2.

Migraine is a prevalent primary headache disorder that significantly impairs daily life. Recent imaging studies have suggested that chronic … he glymphatic system was observed in FHM2 mice. Using two-photon recording, we observed exaggerated neurovascular coupling in the barrel cortex of FHM2 mice. This abnormal neurovascular coupling resulted in a decrease in AQP4 expression within the perivascular astrocyte endfoot. This alteration may be a significant mechanism contributing to impairment of the glymphatic …

Neurovascular coupling dynamics assessed via transcranial Doppler: a comparative study between motor paradigms in healthy individuals.

Introduction.Neurovascular coupling (NVC) represents multiple mechanisms that adapt cerebral blood flow to neural activity. This study hypothesized that two NVC paradigms active hand movement (AHM) and active elbow flexion (AEF) would elicit similar hemodynamic responses.Methods.Seventeen healthy subjects (9 females, mean age: 34 ± 3 years) performed both motor paradigms. Each session began with a 1.5 min rest (baseline), followed by 1 min of motor paradigm (T1), and a 1.5 min recovery (T2). Transcranial Doppler was used to monitor cerebral blood velocity (CBv) in middle cerebral artery. Arter…

Impaired Neurovascular Coupling and Increased Functional Connectivity in the Frontal Cortex Predict Age-Related Cognitive Dysfunction.

Impaired cerebrovascular function contributes to the genesis of age-related cognitive decline. In this study, the hypothesis is tested that impairments in neurovascular coupling (NVC) responses and brain network function predict cognitive dysfunction in older adults. Cerebromicrovascular and working memory function of healthy young (n = 21, 33.2±7.0 years) and aged (n = 30, 75.9±6.9 years) participants are assessed. To determine NVC responses and functional connectivity (FC) during a working memory (n-back) paradigm, oxy- and deoxyhemoglobin concentration changes from the frontal cortex using …

Inverse neurovascular coupling contributes to positive feedback excitation of vasopressin neurons during a systemic homeostatic challenge.

Neurovascular coupling (NVC), the process that links neuronal activity to cerebral blood flow changes, has been mainly studied in superficial brain areas, namely the neocortex. Whether the conventional, rapid, and spatially restricted NVC response can be generalized to deeper and functionally diverse brain regions remains unknown. Implementing an approach for in vivo two-photon imaging from the ventral surface of the brain, we show that a systemic homeostatic challenge, acute salt loading, progressively increases hypothalamic vasopressin (VP) neuronal firing and evokes a vasoconstriction that …

Neurovascular coupling in the human visual cortex is modulated by cyclooxygenase-1 (COX-1) gene variant.

Functional hyperemia, the brain's capability to alter microvascular blood flow in response to the metabolic demands of active neurons, is essential for sustained mammalian brain function. Pharmacological studies in mice suggest neurovascular coupling to centrally involve cyclooxygenase-1 (COX-1) metabolites such as prostaglandins. In humans, however, genetic variation of the COX-1 gene impacting the coupling of neural activity to hemodynamic responses (HRs) has not been investigated yet. In this study, we determined whether COX-1 genotype-dependent enzymatic function impacts HRs in humans. Spe…

Substance P regulates Tacr1 neurons, which control nitric oxide-mediated neurovascular coupling in the mouse cortex.

Neuronal activity-driven increases in cerebral blood flow (CBF), known as neurovascular coupling (NVC), are crucial for sustaining the metabolic demands of the brain. Our group has found that Tacr1 neurons, a subset of somatostatin neurons expressing the substance P (SP) receptor and neuronal nitric oxide synthase (nNOS), exert a disproportionately large regulation of CBF. Here, we use two-photon imaging to show that Tacr1 neurons regulate CBF through nitric oxide (NO) release and SP-receptor signaling . We identify astrocytic calcium (Ca2+) transients as a secondary response to vasodilation. …

A systematic review of fNIRS-based neurovascular coupling research: Methodological landscape and translational considerations for spaceflight.

BACKGROUND: The foundation of cognition is neurovascular coupling (NVC), which is defined as a connection between cerebral blood flow and neuronal activity. NVC can be affected by the situation the person is present in, for example, spaceflight conditions such as sleep deprivation, hypoxia, and microgravity. Therefore, monitoring the phenomena of NVC and how it is being affected under different conditions is very important. There are several tools available for the monitoring of NVC, and among them all, a promising tool is functional near-infrared spectroscopy (fNIRS) that offers benefits like…

Impact of neurovascular coupling and glymphatic function on cognitive function in asymptomatic cerebral small vessel disease.

INTRODUCTION: Cognitive impairment ties to neurovascular coupling (NVC) dysfunction, which may interact with the glymphatic pathway. This study examined the relationships between NVC, glymphatic clearance, and cognitive function in asymptomatic cerebral small vessel disease (CSVD). METHODS: In 231 asymptomatic CSVD patients, whole-brain NVC was assessed via voxel-wise ratios of cerebral blood flow to fractional amplitude of low-frequency fluctuations (CBF/fALFF) and spatial correlation; glymphatic function via the diffusion tensor imaging analysis along the perivascular space (DTI-ALPS) index…

Transient neurovascular coupling impairment in brain penetrating arterioles of streptozotocin treated mice post recurrent nonsevere hypoglycemia.

Hypoglycemia is a major complication of insulin therapy, particularly in type 1 diabetes mellitus. During hypoglycemia, the brain initiates vascular adaptations to maintain adequate blood flow. Here, we examined the effects of non-severe recurrent hypoglycemia (RH) on brain vascular diameter and neurovascular coupling (NVC)-the functional mechanism adapting local blood flow to neuronal energetic demand-in a streptozotocin (STZ)-induced hyperglycemia mouse model. We assessed stimulus-induced dilation of penetrating arterioles (PAs) in the somatosensory cortex of awake mice using in vivo two-pho…

Evaluation of Neurovascular Coupling Behaviors for FES-Induced Wrist Movements Based on Synchronized EEG-fNIRS Signals.

The neuronal activities and cerebral hemodynamic responses are closely linked via the neurovascular coupling (NC) mechanism. This mechanism is strongly associated with the brain state and alters with the pathological conditions such as motor dysfunctions. Clinically, functional electrical stimulation (FES) is being increasingly used for motor function restoration, but the impact of FES on NC behaviors is still lack of study, which would prevent the understanding of FES in improving neurorehabilitation performance. In this work, we studied the NC behaviors based on synchronously recorded electr…
