The Art of Medical Diagnosis: Lessons on Interpretation of Signs from Italian High Renaissance Paintings.

Medicine is struggling with the constantly rising incidence of breast cancer. The key to this fight is to be able to speed up diagnosis, as … tive status of medical analyses performed by medical historians by referring to Italian Renaissance paintings and historical-artistic interpretations. In this field, analyzing disputes between researchers as a clash of methodologies in the ways interpretation transforms signs into meaning is a critical methodological reflection. Medicine is a diverse scient …

Wavelength and pulse duration effects on laser induced changes on raw pigments used in paintings.

In this study, the reaction of widely used artist's pigments in raw form to pulsed laser radiation of different wavelengths and pulse duration was investigated. Vermilion, lead chromate and malachite (in the form of pellets) were irradiated using laser pulses of 500 fs at 248 nm, and pulses of 150 ps and 15 ns at 1064 and 213 nm. Optical microscopy, colorimetry, spectrofluorimetry, micro-Raman spectroscopy and X-ray photoelectron spectroscopy were employed to characterize the physicochemical changes induced to the pigments. Change of crystalline phase was identified for vermilion while reducti

Cell Painting PLUS: expanding the multiplexing capacity of Cell Painting-based phenotypic profiling using iterative staining-elution cycles.

Phenotypic changes in the morphology and internal organization of cells can indicate perturbations in cell functions. Therefore, imaging-based high-throughput phenotypic profiling (HTPP) applications such as Cell Painting (CP) play an important role in basic and translational research, drug discovery, and regulatory toxicology. Here we present the Cell Painting PLUS (CPP) assay, an efficient, robust and broadly applicable approach that further expands the versatility of available HTPP methods and offers additional options for addressing mode-of-action specific research questions. An iterative

Nature experiences affect the aesthetic reception of art: The case of paintings depicting aquatic animals.

Art is a promising pathway to raise emotional engagement with nature, while enabling an indirect exposure to nature through aesthetic experience. However, the precise relationships between aesthetic experiences of art and experiences of nature remain unclear. The aim of this observational study is to highlight the effect of nature experiences on the aesthetic reception art, based on Early Modern paintings (16th-18th century). By focusing on marine ecosystems, that are difficult to directly interact with, the results presented are intended to explore whether marine activities and fish consumpti…

Simultaneous identification of historical pigments Prussian blue and indigo in paintings by electrospray mass spectrometry.

A new analytical protocol for identification of Prussian blue (PB) and indigo was proposed. Pigments useful for dating of artworks were detected by flow injection analysis/electrospray ionization mass spectrometry after alkalization of their suspensions in water, decomposition of PB to iron (III) hydroxide and hexacyanoferrate (II) and reduction of indigo to soluble leucoindigo using sodium dithionite. Limits of detection (PB 47 pg, indigo 59 pg) complied with requirements for analysis of microsamples of historical paintings. Potential of the developed method was proven in analysis of blue sam…

Application of molecular techniques to the elucidation of the microbial community structure of antique paintings.

This paper uses molecular techniques to describe the microstructure and microbiological communities of sixteenth century artwork and their relationships. The microbiological populations, analysed by denaturing gradient gel electrophoresis (DGGE), were highly influenced by the chemical composition of the pictorial layers detected by energy-dispersive X-ray analysis. DGGE revealed that the diversity of microbial communities was lower in pictorial layers composed of pigments with metals, such as Pb, Cu and Hg, than in those found in pictorial layers without such compounds. The number of cultivabl

The relationship between the microclimate and efflorescence of revealed mural paintings and the later protection strategy.

Throughout history, there were many fine mural paintings concealed within ancient buildings, hidden beneath layers of plaster, wall or other structure. In recent decades, research and practical efforts had primarily focused on nondestructive diagnosis of these hidden murals and the removal of their surface coverings. However, limited attention had been given to the consequences of overlay removal on mural preservation. This study aims to address this gap by examining the revealed mural paintings in the Prince Shi's Palace as a case study, employing an analysis of mural ontology, investigation …

Probing and painting to get the full picture of macrophage polarisation.

Macrophages play central roles in tissue repair and remodelling, host defence and immunopathology. Their ability to rapidly reprogram key functions in response to local cues makes them attractive targets for drug discovery. While this plasticity creates therapeutic opportunities, it also complicates high-throughput screening since multiple, transient functional states can coexist. To address this challenge, we developed a 384-well, single-cell pipeline that uses two well-established biomarkers, inducible nitric oxide synthase and Arginase 1, as a compact primary readout for macrophage state. Airborne particle emissions from photocatalytic coatings and paints.

Titanium dioxide (TiO2)-based photocatalytic materials are increasingly proposed for indoor applications due to their ability to degrade volatile organic compounds (VOCs) and inactivate microorganisms. However, the same oxidation processes may lead to the formation of secondary airborne particles, potentially including SOA. In this study, we investigated whether transparent photocatalytic coatings and photocatalytic paints can act as indoor sources of ultrafine particles (UFPs) under controlled indoor lighting conditions. Chamber experiments were conducted by monitoring particle number concent

Painting new pathways: Castilleja enters the genomic era.

Castilleja (Orobanchaceae), commonly known as Indian paintbrush, is a genus of approximately 200 hemiparasitic species found primarily across the Americas. Long studied for its taxonomic complexity, vibrant floral displays, and ecological interactions with host plants, Castilleja has recently emerged as a versatile research system spanning parasitic biology, specialized metabolism, conservation genetics, and pharmacology. The availability of whole-genome sequencing is transforming the field, revealing expanded gene families involved in host recognition, enabling genomic species delimitation of

A Bronze Age lip-paint from southeastern Iran.

A small chlorite vial, discovered among numerous artifacts looted and recovered in the Jiroft region of Kerman province, southeastern Iran, contains a deep red cosmetic preparation that is likely a lip-coloring paint or paste. Through analytical research involving XRD (X-ray diffraction), SEM-EDS (scanning electron microscopy-energy-dispersive spectroscopy), and HPLC-MS (high-performance liquid chromatography-mass spectrometry) analyses, the mineral components of the reddish substance were identified as hematite, darkened with manganite and braunite, and traces of galena and anglesite, mixed w

Optimizing the Cell Painting assay for image-based profiling.

In image-based profiling, software extracts thousands of morphological features of cells from multi-channel fluorescence microscopy images, yielding single-cell profiles that can be used for basic research and drug discovery. Powerful applications have been proven, including clustering chemical and genetic perturbations on the basis of their similar morphological impact, identifying disease phenotypes by observing differences in profiles between healthy and diseased cells and predicting assay outcomes by using machine learning, among many others. Here, we provide an updated protocol for the mo

Body Painting of the Horse and Cow to Learn Surface Anatomy.

Gross anatomy is considered one of the most challenging subjects in teaching veterinary medicine. The use of body painting is reported in teaching surface human anatomy, but such reports are scarce in veterinary medicine. The aim of this study was to describe a practical session for teaching surface anatomy using body painting with second-semester students of veterinary medicine. Two practical sessions using live animals (equine and bovine) were offered with a focus on the locomotor and nervous systems and splanchnology. Students believed that the body painting sessions helped them to understa

Site-Specific Labeling of Affimers for DNA-PAINT Microscopy.

Optical super-resolution techniques allow fluorescence imaging below the classical diffraction limit of light. From a technology standpoint, recent methods are approaching molecular-scale spatial resolution. However, this remarkable achievement is not easily translated to imaging of cellular components, since current labeling approaches are limited by either large label sizes (antibodies) or the sparse availability of small and efficient binders (nanobodies, aptamers, genetically-encoded tags). In this work, we combined recently developed Affimer reagents with site-specific DNA modification fo
