A quantitative examination of various factors influencing the accuracy of the P—K test was undertaken. The skin site on the back of most recipients tested did not contribute significantly to the variance observed. Coefficients of variation of a single P—K test of the order of 10–20 per cent were frequently demonstrable, for weals exceeding 50 mm2 in area. The synthesis of a series of [1,4]benzoxazine-2,3-diones and a new class of compounds, benzobisoxazinetetrones, is described. These compounds were evaluated for their effect in the rat mast cell (RMC) test passively sensitized in vitro with rat antiovalbumin serum and for their effect in inhibitory passive cutaneous anaphylaxis (PCA) in the rat. Some of these compounds are of the same potency level as disodium cromglycate in the RMC test and some are effective orally in PCA.

Radioimmunosorbent techniques using antisera to human immunoglobulin E (IgE) showed the presence of high levels (up to 15,840 U/ml) of IgE in serum and plasma from fowls immunised by repeated intraperitoneal injections of horse serum. An anti-inflammatory corticosteroid, dexamethasone, was found to be a potent suppressor of cell sensitization induced by IgE antibody in a model of type I allergic inflammation in vivo. Low doses (approximately less than 10(-10) mol) of dexamethasone, administered intradermally to rabbits 90 min before injection of IgE into the same sites, suppressed local oedema induced by a local challenge with antigen 72 h later. The effect of aqueous extract of Siegesbeckia glabrescence (Compositae) whole plants (SGWP) on systemic or local anaphylaxis was studied. SGWP inhibited compound 48/80-induced systemic anaphylaxis 100% with a dose of 1000 mg/kg. IgE responses to Fasciola hepatica extract in 6 strains of mice (C3H/HeN, CBA/JN, DBA/2N, BALB/cAnN, C57BL/6N and NZBWF1) were investigated. Serum anti-F. hepatica IgE antibodies were determined by passive cutaneous anaphylaxis (PCA). DBA/2N and BALB/cAnN mice showed higher IgE responses than other strains.
