Peptidoglycan recognition proteins in the brain: Role in neuroinflammation and behavioral consequences.

Peptidoglycan recognition proteins (PGRPs) constitute an evolutionarily conserved family of pattern recognition molecules that detect bacterial peptidoglycan. While their antimicrobial functions have been well characterized in peripheral immunity, recent discoveries have unveiled critical roles for PGRPs in central nervous system inflammation and behavior. Among the four mammalian PGRP family members, PGLYRP1 exhibits unique expression in brain microglia and demonstrates potent pro-inflammatory properties in neurological contexts. Recently, PGLYRP1 has been shown to function as a key amplifier

[New aspects of the pathogenesis of gout. Danger signals, autoinflammation and beyond].

Gout is caused by monosodium urate (MSU) crystal-induced inflammation of the joints and periarticular tissues. MSU crystals activate the NOD-like receptor (NLR) NALP3, which functions as a pattern recognition receptor (PRR). Activated NALP3 mediates interleukin-1b (IL-1b) generation from its inactive pro-form, resulting in the activation of further cells and an IL-8-mediated neutrophil influx into the joint. Based on these new findings on the pathophysiology of gout, an open pilot study has recently demonstrated successful treatment of gout with the soluble IL-1R antagonist anakinra in 10 pati

Newer indications for neuromonitoring in critically ill neonates.

Continuous neuromonitoring in the neonatal intensive care unit allows for bedside assessment of brain oxygenation and perfusion as well as cerebral function and seizure identification. Near-infrared spectroscopy (NIRS) reflects the balance between oxygen delivery and consumption, and use of multisite monitoring of regional oxygenation provides organ-specific assessment of perfusion. With understanding of the underlying principles of NIRS as well as the physiologic factors which impact oxygenation and perfusion of the brain, kidneys and bowel, changes in neonatal physiology can be more easily r

NLRP3 promotes endometrial receptivity by inducing epithelial-mesenchymal transition of the endometrial epithelium.

Endometrial receptivity is crucial for successful embryo implantation. It is regulated by multiple factors which include ovarian steroid hormones and the immune microenvironment among others. Nod-Like Receptor Pyrins-3 (NLRP3) is a key intracellular pattern-recognition receptor and a critical component of the inflammasome, which plays an essential role in the development of inflammation and of immune responses. However, the physiological functions of NLRP3 in the endometrium remain largely unclear. This study investigated the physiological and pathological significance of NLRP3 in human endome

Microglia-derived galectin-3 in neuroinflammation; a bittersweet ligand?

Galectins are soluble β-galactoside-binding proteins found in all multicellular organisms. Galectins may act as danger-associated molecular patterns in innate immunity and/or as pattern-recognition receptors that bind to pathogen-associated molecular patterns. Among different galectin family members, galectin-3 has been the focus of studies in neurodegenerative diseases in recent years. This lectin modulates brain innate immune responses, microglia activation patterns in physiological and pathophysiological settings in a context-dependent manner. Galectin-3 is considered as a pivotal tuner of

Oxytocin selectively enhances the recognition of infant sadness in postpartum depression.

BackgroundLittle is known about the role of oxytocin in the processing of positive emotional infant cues in mothers with PPD.ObjectivesWe therefore examined whether intranasal oxytocin influences postpartum mothers' processing of positive infant emotional cues, both visual (facial expressions) and auditory (laughter), and intended caregiving responses.DesignThis double-blind randomized controlled crossover study included 45 mothers, with 3- to 9-month-old infants, who exhibited clinically relevant scores on the Edinburgh Postnatal Depression Scale, with 35 mothers fulfilling the criteria of a

Targeting mast cells: Uncovering prolific therapeutic role in myriad diseases.

The mast cells are integral part of immune system and they have pleiotropic physiological functions in our body. Any type of abnormal stimuli causes the mast cells receptors to spur the otherwise innocuous mast cells to degranulate and release inflammatory mediators like histamine, cytokines, chemokines and prostaglandins. These mediators are involved in various diseases like allergy, asthma, mastocytosis, cardiovascular disorders, etc. Herein, we describe the receptors involved in degranulation of mast cells and are broadly divided into four categories: G-protein coupled receptors, ligand gat

SLV330, a cannabinoid CB1 receptor antagonist, ameliorates deficits in the T-maze, object recognition and Social Recognition Tasks in rodents.

Cannabinoid CB(1) receptor (CB(1)R) signaling has been suggested to play an important role in the regulation of memory and cognition. In the present study, our aim was to investigate whether the CB(1)R antagonist SLV330 (doses ranging from 0.3 to 10mg/kg, given orally, p.o.) could ameliorate impairments in distinct aspects of cognition using different disruption models in both mice and rats. Effects of SLV330 were tested on working memory deficits in the T-maze Continuous Alternation Task (T-CAT) in mice; episodic memory deficits in the Object Recognition Task (ORT) and Social Recognition Task

A Curious "Mickey Mouse" Sign: Physiological FAPI Uptake in Uterus Didelphys on 68 Ga-FAPI PET/CT.

A 42-year-old woman with invasive lobular carcinoma of the left breast underwent staging with 68 Ga-FAPI PET/CT, which demonstrated intense uptake in the primary breast lesion and ipsilateral nodal metastases. Incidentally, symmetric intense pelvic tracer uptake was identified, corresponding on fused PET/CT to 2 distinct uterine horns consistent with uterus didelphys. The characteristic bilateral rounded appearance on PET images produced a striking "Mickey Mouse" sign, reflecting physiological FAPI accumulation within duplicated uterine stromal tissue. Subsequent ultrasonography confirmed uter

The formyl peptide receptors FPR1 and FPR2 as targets for inflammatory disorders: recent advances in the development of small-molecule agonists.

Formyl peptide receptors (FPRs) comprise a class of chemoattractant pattern recognition receptors, for which several physiological functions like host-defences, as well as the regulation of inflammatory responses, have been ascribed. With accumulating evidence that agonism of FPR1/FPR2 can confer pro-resolution of inflammation, increased attention from academia and industry has led to the discovery of new and interesting small-molecule FPR1/FPR2 agonists. Focused attention on the development of appropriate physicochemical and pharmacokinetic profiles is yielding synthesis of new compounds with

It Knows My Emotions Better Than I Do: Digital Phenotyping and the Contest Over Symptom Interpretation.

Digital phenotyping is increasingly presented as a means of detecting mental distress through smartphones, wearables, and other sensor-rich personal devices. In this article, we argue that the central philosophical and clinical issue raised by digital phenotyping extends beyond predictive accuracy to interpretive authority, which concerns who gets to name what a feeling, behavioral pattern, or physiological fluctuation means. Psychiatry has always involved uncertainty, proxy reasoning, and the negotiation of testimony, but digital phenotyping intensifies these dynamics by translating lived sta

60 YEARS OF NEUROENDOCRINOLOGY: The posterior pituitary, from Geoffrey Harris to our present understanding.

Geoffrey Harris pioneered our understanding of the posterior pituitary, mainly with experiments that involved the electrical stimulation of the supraoptico-hypophysial tract. In the present essay, we explain how his observations included clues to the pulsatile nature of the oxytocin signal - clues that were followed up by subsequent workers, including his students and their students. These studies ultimately led to our present understanding of the milk-ejection reflex and of the role of oxytocin in parturition. Discoveries of wide significance followed, including: the recognition of the import

Deep learning for multimodal physiological signal based assessment of sleep disordered breathing.

Sleep disordered breathing (SDB) is commonly assessed using polysomnography (PSG), which records multiple physiological signals during sleep. Accurate automated assessment of SDB related breathing abnormalities remains challenging because respiratory events occur within complex and stage dependent sleep dynamics. To address this problem, this study proposes a multimodal physiological signal based deep learning framework for SDB oriented sleep assessment. The proposed framework treats SDB related respiratory event detection as the clinically targeted task and incorporates sleep stage classifica

Multivariate resting-state functional connectivity predicts response to cognitive behavioral therapy in obsessive-compulsive disorder.

Cognitive behavioral therapy (CBT) is an effective treatment for many with obsessive-compulsive disorder (OCD). However, response varies considerably among individuals. Attaining a means to predict an individual's potential response would permit clinicians to more prudently allocate resources for this often stressful and time-consuming treatment. We collected resting-state functional magnetic resonance imaging from adults with OCD before and after 4 weeks of intensive daily CBT. We leveraged machine learning with cross-validation to assess the power of functional connectivity (FC) patterns to

Visual adaptation provides objective electrophysiological evidence of facial identity discrimination.

Discrimination of facial identities is a fundamental function of the human brain that is challenging to examine with macroscopic measurements of neural activity, such as those obtained with functional magnetic resonance imaging (fMRI) and electroencephalography (EEG). Although visual adaptation or repetition suppression (RS) stimulation paradigms have been successfully implemented to this end with such recording techniques, objective evidence of an identity-specific discrimination response due to adaptation at the level of the visual representation is lacking. Here, we addressed this issue wit

Multimodal detection of sleep apnoea using electrocardiogram and oximetry signals.

A method for the detection of sleep apnoea, suitable for use in the home environment, is presented. The method automatically analyses night-time electrocardiogram (ECG) and oximetry recordings and identifies periods of normal and sleep-disordered breathing (SDB). The SDB is classified into one of six classes: obstructive, mixed and central apnoeas, and obstructive, mixed and central hypopnoeas. It also provides an estimated apnoea, hypopnoea and apnoea-hypopnoea index. The basis of the method is a pattern recognition system that identifies episodes of apnoea by analysing the heart variability,

Early online detection of upper airway obstructions in obstructive sleep apnoea syndrome (OSAS) patients.

The obstructive sleep apnoea syndrome (OSAS) is a diagnosis related to snoring and caused by a collapse in the upper airway. OSAS patients suffer from desaturated oxygen levels during sleep as well as daytime sleepiness. In this paper, we propose a system able to identify and detect respiratory disorders online based on monitoring the airflow amplitude from a sleeping OSAS patient. By the use of chi(2)-analysis and a Haar wavelet transform on signals performed offline, reference templates indicating the specific apnoea pattern for four different patients are constructed and used for similarity

Neural changes with tactile learning reflect decision-level reweighting of perceptual readout.

Despite considerable work, the neural basis of perceptual learning remains uncertain. For visual learning, although some studies suggested that changes in early sensory representations are responsible, other studies point to decision-level reweighting of perceptual readout. These competing possibilities have not been examined in other sensory systems, investigating which could help resolve the issue. Here we report a study of human tactile microspatial learning in which participants achieved >six-fold decline in acuity threshold after multiple training sessions. Functional magnetic resonance i

Lack of early pattern stimulation prevents normal development of the alpha (Y) retinal ganglion cell population in the cat.

Binocular deprivation of pattern vision (BD) early in life permanently impairs global motion perception. With the SMI-32 antibody against neurofilament protein (NFP) as a marker of the motion-sensitive Y-cell pathway (Van der Gucht et al. [2001] Cereb. Cortex 17:2805-2819), we analyzed the impact of early BD on the retinal circuitry in adult, perceptually characterized cats (Burnat et al. [2005] Neuroreport 16:751-754). In controls, large retinal ganglion cells exhibited a strong NFP signal in the soma and in the proximal parts of the dendritic arbors. The NFP-immunoreactive dendrites typicall

[The effect of anchor voices and visible speech in training in the GRABS scale of perceptual evaluation of dysphonia].

INTRODUCTION: Perceptual evaluation of voice quality remains a key standard for judgment of vocal impairment. The GRABS method has become a commonly-used scale for rating severity of dysphonia, but it has no published, standardised protocol to follow. Training is important for reaching good interrater agreement for its parameters; however, the references most often cited for the GRABS provide no guidelines for clinical administration, speech material or rating calibration. This study investigated the effect of anchors (standard reference voices) and visible speech (narrow band spectrogram) in
