The content and distribution of phospholipids (PL) in red blood cells (RBC) of PK and of G6PD deficiency were found to differ in both disorders. RBC immaturity could not be excluded as reason for PL alterations in PK deficiency. The relative diminution of phosphatidylethanolamine in G6PD deficiency may be connected with the enzyme depletion in this disorder and its influence on the PL of RBC membranes. Moist or all fatty acid deviations seem to be conditioned by RBC immaturity. Nearly all PL fatty acid aberrations in G6PD deficiency RBC were observed also in PK deficiency RBC. Additional devia…

Glaucoma, the leading cause of irreversible blindness worldwide, is characterized by elevated intraocular pressure (IOP) resulting from impaired aqueous humor (AH) outflow through the trabecular meshwork (TM). We demonstrate altered phospholipids (PLs) levels and increased activity of the interconversion enzyme phosphatidylserine decarboxylase (PSD) in the TM of individuals with primary open-angle glaucoma. Elevating PSD alone is sufficient to raise IOP in normotensive mice, whereas depleting PSD restores IOP in glaucomatous mice. Although certain PL classes are generally altered in glaucoma, …

The phospholipase A and acyltransferase (PLAAT) family is a group of structurally related proteins that are conserved among vertebrates. In humans, the family comprises five members (PLAAT1-5), which share common domain structures, and functions as phospholipase A1/A2 and acyltransferase enzymes. Regarding acyltransferase activities, PLAATs produce N-acyl-phosphatidylethanolamines, which serve as the precursor of bioactive N-acylethanolamines (NAEs). Recent evidence strongly suggests that PLAAT proteins play a crucial role in maintaining homeostasis in various organelles, such as the endoplasm

Coronary heart disease, also known as ischemic heart disease, is induced by atherosclerosis, which is initiated by subendothelial retention of lipoproteins. Plasma lipoproteins, including high density lipoprotein, low density lipoprotein (LDL), very low density lipoprotein, and chylomicron, are composed of a surface monolayer containing phospholipids and cholesterol and a hydrophobic core containing triglycerides and cholesteryl esters. Phospholipids play a crucial role in the binding of apolipoproteins and enzymes to lipoprotein surfaces, thereby regulating lipoprotein metabolism. High LDL-ch…

Low phospholipid-associated cholestasis and cholelithiasis (LPAC) is a genetic disorder characterized by cholesterol gallbladder and intrahepatic stones. It is caused by a mutation of the gene ABCB4, which encodes the canalicular protein ABCB4/MDR3, a flippase that plays an essential role in the secretion of phosphatidylcholine into bile. Failure of this protein leads to secretion of bile that is poor in phospholipids and, hence, highly lithogenic, with potent detergent properties. This, in turn, leads to cholangiocyte luminal membrane injury and biliary lesions causing cholestasis. The diagno…

The antiphospolipid syndrome (APS) is an autoimmune disease characterized by recurrent fetal loss, thrombotic events (arterial or venous) and hemocytopenic disorders associated to high titers of circulating aPL. Two variants of the APS have been described. Primary APS is a clinical entity without evidence of any other autoimmune disease and secondary APS is a clinical disorder mainly associated with Systemic Lupus Erithematosus (SLE). aPL are a widely group of immunoglobulins directed against different components or proteins factors. In 1990 three groups of researchers identified that β(2)GP-I

Cardiac hypertrophy, congestive heart failure, diabetic cardiomyopathy and myocardial ischemia-reperfusion injury are associated with a disturbance in cardiac sarcolemmal membrane phospholipid homeostasis. The contribution of the different phospholipases and their related signaling mechanisms to altered function of the diseased myocardium is not completely understood. Resolution of this issue is essential for both the understanding of the pathophysiology of heart disease and for determining if components of the phospholipid signaling pathways could serve as appropriate therapeutic targets. Islet amyloid polypeptide (IAPP) is a 37-residue hormone that forms cytotoxic amyloid fibers in the endocrine pancreas of patients with type … embranes on IAPP fibers. Fibrillogenesis of IAPP is catalyzed by synthetic and human tissue-derived phospholipids, leading to >tenfold increases in the rate of fibrillogenesis. The molecular basis of this phenomenon includes a strong dependence on the concentration and charge density of the membrane. IAPP binds to lipid membranes of mixed anionic (DOPG) and zwi …

Cardiolipin antibodies (CAb) were measured in 14 patients with primary pulmonary hypertension (PPH). These included 9 females and 5 males (mean age 36.8 +/- 9.6 years) with duration of PPH 4.9 +/- 4.1 years. No rise in CAb IgG was noted, whereas CAb IgM arose to 13.0-42.5 MPI in 3 patients. 2 lethal outcomes were registered in the group of patients with normal CAb levels and 2 in the group with elevated CAb (20% against 66.7%, respectively). The deaths were caused by progressive circulatory failure and venous occlusion, pulmonary artery embolism, respectively. It is suggested that IgM CAb incr

Alcoholic liver injury has been reported to be directed preferentially against the proteins of the cell membrane, sparing the phospholipids. However, antiphospholipid antibodies against certain cell membrane phospholipids are known to be associated with a variety of diseases. We undertook this investigation to determine whether antiphospholipid antibodies were present in the serum of patients with alcoholic liver disease. We investigated seventy long-term alcoholic patients (> 80 gm ethanol/day for > 1 yr) and 8 normal nonalcoholic controls by means of enzyme-linked immunosorbent assay to dete…

Phospholipid peroxidation markedly reduces the stability of mixed micellar systems composed of cholate, phosphatidylcholine and supersaturating levels of cholesterol. This suggests that lipid peroxidation is likely to play a significant role in the precipitation of cholesterol from gallbladder bile, thus in the pathogenesis of cholesterol gallstones. This conclusion is supported by studies of the nucleation time of cholesterol in gallbladder biles, which was significantly reduced by exposure to a stream of oxygen. This effect of phospholipid peroxidation on cholesterol solubility may occur in

Binding of phospholipase A2 from porcine pancreas and from Naja melanoleuca venom to vesicles of 1,2-di(tetradecyl)-rac-glycero-3-phosphocholine (diether-PC14) is studied in the presence and absence of 1-tetradecanoyl-sn-glycero-3-phosphocholine and myristic acid. The bound enzyme coelutes with the vesicles during gel filtration through a nonequilibrated Sephadex G-100 column, modifies the phase transition behavior of bilayers, and exhibits an increase in fluorescence intensity accompanied by a blue shift. Using these criteria it is demonstrated that the snake-venom enzyme binds to bilayers of

Spinach class II chloroplasts were treated with snake venom phospholipase A2 in the presence of bovine serum albumin, and separated by sucrose-density centrifugation. The treatment yielded phospholipid-depleted chloroplasts which had lost 82.6% of the original phospholipids. About 20% of the phospholipids of chloroplasts were resistant to enzyme attack. These results suggest that phospholipids exist in two states in chloroplast membranes. In spite of considerable phospholipid depletion, the chloroplast preparations retained a large portion of their photoactivities, i.e. light-induced electron …

Incorporation in vitro of (32)P-labeled lysolecithin (LPC) or lysophosphatidylethanolamine (LPE) into respectively lecithin (PC) and phosphatidylethanolamine (PE) of rabbit granulocytes and alveolar macrophages was compared in the absence and in the presence of ingestible particles. Maximal synthesis of PC by intact cells occurred at added LPC concentrations of less than 0.05 mmole/liter, i.e., at levels found in plasma. Accumulation of PC-(32)P proceeded linearly for at least 30 min and varied directly with cell concentration. While per cell granulocytes and macrophages converted comparable a

Lipid peroxidation in unilamellar liposomes of known cholesterol-phospholipid composition was monitored under conditions of autoxidation or as induced by a superoxide radical generating system, gamma-irradiation or cumene hydroperoxide. Formation of cholesterol oxidation products was indexed to the level of lipid peroxidation. The major cholesterol oxidation products identified were 7-keto-cholesterol, isomeric cholesterol 5,6-epoxides, isomeric 7-hydroperoxides and isomeric 3,7-cholestane diols. Other commonly encountered products included 3,5-cholestadiene-7-one and cholestane-3 beta, 5 alph
