Lectins from Synadenium carinatum (ScLL) and Artocarpus heterophyllus (ArtinM) Are Able to Induce Beneficial Immunomodulatory Effects in a Murine Model for Treatment of Toxoplasma gondii Infection.

Infection by Toxoplasma gondii affects around one-third of world population and the treatment for patients presenting toxoplasmosis clinically manifested disease is mainly based by a combination of sulfadiazine, pyrimethamine, and folinic acid. However, this therapeutic protocol is significantly toxic, causing relevant dose-related bone marrow damage. Thus, it is necessary to improve new approaches to investigate the usefulness of more effective and non-toxic agents for treatment of patients with toxoplasmosis. It has been described that lectins from plants can control parasite infections, whe

Multifunctional Therapeutic Potential of Phytocomplexes and Natural Extracts for Antimicrobial Properties.

Natural products have been known for their antimicrobial factors since time immemorial. Infectious diseases are a worldwide burden that have been deteriorating because of the improvement of species impervious to various anti-infection agents. Hence, the distinguishing proof of antimicrobial specialists with high-power dynamic against MDR microorganisms is central to conquer this issue. Successful treatment of infection involves the improvement of new drugs or some common source of novel medications. Numerous naturally occurring antimicrobial agents can be of plant origin, animal origin, microb

[Subcutaneous manifestations of a centrocytic non-Hodgkin lymphoma at the injection site of a mistletoe preparation].

HISTORY AND CLINICAL FINDINGS: A 73-year-old man, first diagnosed as having centrocytic Non-Hodgkin lymphoma 5 years previously, presented with subcutaneous nodes of the abdominal wall at precisely the sites of previous regular injections of a mistletoe preparation. These nodes had first appeared 5 weeks after the first injection. Injections were stopped and he reported to the out-patient clinic. Except for the visible and easily nodes in the anterior wall no other subcutaneous nodes were palpated. Prominent cervical lymphomas and swelling of the epicranial aponeurosis and lower lip had, accor

Effect of diets containing genetically modified potatoes expressing Galanthus nivalis lectin on rat small intestine.

Diets containing genetically modified (GM) potatoes expressing the lectin Galanthus nivalis agglutinin (GNA) had variable effects on different parts of the rat gastrointestinal tract. Some effects, such as the proliferation of the gastric mucosa, were mainly due to the expression of the GNA transgene. However, other parts of the construct or the genetic transformation (or both) could also have contributed to the overall biological effects of the GNA-GM potatoes, particularly on the small intestine and caecum.

Role of complement in the toxicity of dietary legumes.

On the basis of in vivo data Jayne-Williams (1) has proposed that the toxicity of dietary legumes is due to their content of lectins which are immunosuppressive. On the basis of in vitro data with cultured lymphocytes (2), it is now proposed that ingested lectins bind to cell surfaces and cause autologous complement components to bind to and destroy immunologically competent cells. The hypothesis throws a possible light on the aetiology of favism and Whipple's disease.

Sublethal effects of the insecticidal fusion protein ω-ACTX-Hv1a/GNA on the parasitoid Eulophus pennicornis via its host Lacanobia oleracea.

BACKGROUND: The neurotoxin peptide ω-ACTX-Hv1a, fused to the carrier molecule GNA, presents potential for insect control as a biopesticide, being orally toxic to insect pests from different orders. However, thorough evaluation is required to assure its safety towards non-target invertebrates. Effects of this novel biopesticide on the parasitoid Eulophus pennicornis via its host Lacanobia oleracea are presented. RESULTS: Hv1a/GNA did not cause mortality when injected or fed to fifth-stage L. oleracea, but caused up to 39% reduction in mean larval weight (P < 0.05) and increased developmental t

Influence of carbohydrates on the cytotoxicity of an aqueous mistletoe drug and of purified mistletoe lectins tested on human T-leukemia cells.

Partially and highly purified lectins from Viscum album L. (mistletoe) cause a dose-dependent decrease of viability of human leukemia cell cultures, MOLT-4, after 72 h treatment. The LC50 of the partially purified lectin was 27.8 ng/ml, of the highly purified lectin 1.3 ng/ml. Compared to the highly purified lectin a 140-fold higher protein concentration of an aqueous mistletoe drug was required to obtain similar cytotoxic effects on MOLT-4 cells. Cytotoxicity of the highly purified lectin was preferentially inhibited by D-galactose and lactose, cytotoxicity of the mistletoe drug and the parti

Dietary plant bioactive compounds: context-dependent roles as antinutrients or beneficial phytochemicals-a comprehensive review.

Dietary plant bioactive compounds have traditionally been classified as either beneficial phytochemicals or harmful antinutrients; however, emerging evidence suggests that most of these compounds exhibit dual functionality depending on the context. This review integrates current knowledge of the phytochemical-antinutrient continuum, highlighting that the biological effects of these compounds vary and are influenced by factors such as concentration, chemical structure, food matrix, processing conditions, and host-related variables, including the gut microbiota and nutritional status. Major comp

A Sustainable Approach for the Use of Plant Proteins as Functional Ingredients in Dairy Alternatives.

The increasing global demand for sustainable and health-promoting foods has positioned plant-based proteins as key components in the development of dairy alternatives. This review critically examines plant protein sources: legumes, cereals, pseudocereals, oilseeds, and nuts not merely by composition, but by their comparative suitability for specific dairy applications, evaluating protein quality using the Digestible Indispensable Amino Acid Score (DIAAS), digestibility, and source-specific antinutritional factors (phytates in cereals; tannins, trypsin inhibitors, and lectins in legumes and oil

RTB Lectin: a novel receptor-independent delivery system for lysosomal enzyme replacement therapies.

Enzyme replacement therapies have revolutionized patient treatment for multiple rare lysosomal storage diseases but show limited effectiveness for addressing pathologies in "hard-to-treat" organs and tissues including brain and bone. Here we investigate the plant lectin RTB as a novel carrier for human lysosomal enzymes. RTB enters mammalian cells by multiple mechanisms including both adsorptive-mediated and receptor-mediated endocytosis, and thus provides access to a broader array of organs and cells. Fusion proteins comprised of RTB and human α-L-iduronidase, the corrective enzyme for Mucopo

Molecular Basis of Citrus sunki Susceptibility and Poncirus trifoliata Resistance Upon Phytophthora parasitica Attack.

Coevolution has shaped the molecular basis of an extensive number of defense mechanisms in plant-pathogen interactions. Phytophthora parasitica, a hemibiothrophic oomycete pathogen and the causal agent of citrus root rot and gummosis, interacts differently with Citrus sunki and Poncirus trifoliata, two commonly favored citrus rootstocks that are recognized as susceptible and resistant, respectively, to P. parasitica. The molecular core of these interactions remains elusive. Here, we provide evidence on the defense strategies employed by both susceptible and resistant citrus rootstocks, in para

Canavalia brasiliensis lectin attenuates diabetes endothelial dysfunction in rats: structural basis and implication of muscarinic/insulin receptors.

In diabetes, the endothelium acquires tendency toward vasoconstriction, inflammation, and thrombosis, and are associated with the reduced bioavailability of nitric oxide (NO). Plant lectins from the Canavalia genus elicit endothelium-dependent vasorelaxation in isolated rat aorta involving the participation of NO and the lectin domain. This study aimed to investigate ex vivo and in vivo the relaxant effect of Canavalia brasiliensis lectin (ConBr) in aorta from type 1 diabetic rats. ConBr was added at phenylephrine-contractions or acetylcholine-relaxation in isolated artery. In vivo, ConBr was …

Leishmania donovani: assessment of leishmanicidal effects of herbal extracts obtained from plants in the visceral leishmaniasis endemic area of Bihar, India.

One obstacle faced in the effective control of visceral leishmaniasis (VL) is the limited number of available treatment options. Furthermore, control efforts have been hindered further by the emergence of Leishmania resistance to many of the available drugs. In this study, we investigated the anti-leishmanial properties of 30 medicinally important plants from the VL endemic area of Bihar, India and compared them to two available anti-leishmanial drugs (sodium antimony gluconate and amphotericin B) and two plant lectins (phytohemagglutinin and concanavalin A) on Leishmania donovani promastigote…

Effect of immunomodulation with galactoside-specific mistletoe lectin on experimental listeriosis.

BALB/c-mice (n = 10 per experimental group) were intravenously infected with 5 x 10(4) and 5 x 10(5) viable cells of Listeria monocytogenes SLCC 4013. About 50-80 h after this challenge, all mice of the untreated control groups succumbed to their infection. Pretreatment of experimental animals on the optimal immunoactive schedule with the galactoside-specific mistletoe lectin (ML-1; 1 ng/kg body weight; days 1, 4, 5, 6 before challenge), however, evidently reduced the lethality of listerial infection (survival rate 60%).

Pinellia ternata lectin induces mitochondrial damage and inflammation in RAW264.7 cells via mannose receptor-mediated endocytosis.

A lectin from Pinellia ternata tubers (PTL, 12 kDa) with potent pro-inflammatory activity binds to the plasma membrane and translocates into the cytoplasm. However, its cellular entry route and inflammatory toxicity mechanisms remain unclear. Here, confocal microscopy showed that fluorescein isothiocyanate (FITC)-labeled PTL (50 μg/mL) accumulated on the cell surface and was endocytosed by RAW264.7 cells time-dependently. Pharmacological inhibition revealed that PTL uptake was mediated by the mannose receptor C-type 2 (MRC2) and predominantly via clathrin-mediated endocytosis. After endocytosi

Phytolectin Potentiates Copper Sulfide Nanoparticles Anticandida Activity Through Enhanced ROS Production.

Fungal infections caused by Candida albicans are challenging to treat with antifungal drugs due to the emergence of antimicrobial-resistant strains, necessitating the development of innovative therapeutic methods. The present study demonstrates that cationic copper sulfide nanoparticles (cCuS) synthesized using [2-((N-(2-hydroxyethyl)palmitamido)methyl)-1-methylpyridin-1-ium iodide] exhibit increased efficacy against C. albicans when delivered using Jacalin, a lectin from jackfruit that binds to cell surface glycans. The requirement of cCuS to kill was reduced twofold when delivered through Ja

DVL, lectin from Dioclea violacea seeds, has multiples mechanisms of action against Candida spp via carbohydrate recognition domain.

Lectins are proteins of non-immunological origin with the ability to bind to carbohydrates reversibly. They emerge as an alternative to conventional antifungals, given the ability to interact with carbohydrates in the fungal cell wall inhibiting fungal growth. The lectin from D. violacea (DVL) already has its activity described as anti-candida in some species. Here, we observed the anti-candida effect of DVL on C. albicans, C. krusei and C. parapsilosis and its multiple mechanisms of action toward the yeasts. Additionally, it was observed that DVL induces membrane and cell wall damage and ROS

Glycosylation of random IgG distinguishes seropositive and seronegative rheumatoid arthritis.

The N-glycosylation of human immunoglobulins, especially IgGs, plays a critical role in determining affinity of IgGs towards their effector (pro- and anti-inflammatory) receptors. However, it is still not clear whether altered glycosylation is involved in only antibody-dependent disorders like seropositive rheumatoid arthritis (RA) or also in pathologies with similar clinical manifestations, but no specific autoantibodies like seronegative RA. The clarification of that uncertainty was the aim of the current study. Another study aim was the detection of specific glycan forms responsible for alt
