Rhesus monkeys with experimental Plasmodium knowlesi infections of varying duration were treated with sodium sulfathiazole to sterilize the infection and after differing lengths of time were reinoculated intraperitoneally with homologous strains of the plasmodium, for the purpose of determining whether there is any acquired immunity to malaria in hosts from whom all parasites have been removed. Two monkeys, one receiving sulfathiazole on the 2nd day of acute infection and the other on the 4th day, had no immunity at the time of reinoculation, 3 weeks and 10 weeks later, respectively. Both deve…

Plasmodium vivax vaccine discovery lags behind Plasmodium falciparum due to the absence of a reliable in vitro culture system. We developed a miniature 384-well assay for the evolutionarily related parasite species Plasmodium cynomolgi and Plasmodium knowlesi, deploying it for screening invasion inhibition efficacy of antibodies elicited against 8 P. vivax proteins. All 8 antibodies showed consistent inhibition across both P. knowlesi and P. cynomolgi species, correlating with inhibition in ex vivo P. vivax isolates. The use of simian malaria parasites and this assay represent a robust, high-t…

Malaria is caused by multiple different species of protozoan parasites, and interventions in the pre-elimination phase can lead to drastic changes in the proportion of each species causing malaria. In endemic areas, cross-reactivity may play an important role in the protection and blocking transmission. Thus, successful control of one species could lead to an increase in other parasite species. A few studies have reported cross-reactivity producing cross-immunity, but the extent of cross-reactive, particularly between closely related species, is poorly understood. P. vivax and P. knowlesi are

BACKGROUND: Plasmodium knowlesi and Plasmodium vivax are the predominant Plasmodium species that cause malaria in Malaysia and play a role in asymptomatic malaria disease transmission in Malaysia. The diagnostic tools available to diagnose malaria, such as microscopy and rapid diagnostic test (RDT), are less sensitive at detecting lower parasite density. Droplet digital polymerase chain reaction (ddPCR), which has been shown to have higher sensitivity at diagnosing malaria, allows direct quantification without the need for a standard curve. The aim of this study is to develop and use a duplex …

Sera of Gambian individuals and rhesus monkeys immune against infections with Plasmodium falciparum and plasmodium knowlesi, respectively, were reacted with triton X-100-solubilized membranes of infected erythrocytes. Indirect immune precipitation with Staphylococcus aureus, Cowan strain A, followed by dodecylsulfate-polyacrylamide gel electrophoresis, were used to identify interspecies plasmodial antigens that were immunogenic in vivo. Both types of sera specifically precipitated Plasmodium-specific antigens with Mrs of 125,000, 90,000, and 65,000 to 50,000 from membranes of P. knowlesi-infec…

Within the overlapping geographical ranges of P. knowlesi monkey hosts and vectors in Southeast Asia, an estimated 1.5 billion people are considered at risk of infection. P. knowlesi can cause severe disease and death, the latter associated with delayed treatment occurring from misdiagnosis. Although microscopy is a sufficiently sensitive first-line tool for P. knowlesi detection for most low-level symptomatic infections, misdiagnosis as other Plasmodium species is common, and the majority of asymptomatic infections remain undetected. Current point-of-care rapid diagnostic tests demonstrate in

BACKGROUND: Plasmodium knowlesi causes severe and fatal malaria, and incidence in Southeast Asia is increasing. Factors associated with death are not clearly defined.
METHODS: All malaria deaths in Sabah, Malaysia, from 2015 to 2017 were identified from mandatory reporting to the Sabah Department of Health. Case notes were reviewed, and a systematic review of these and all previously reported fatal P. knowlesi cases was conducted. Case fatality rates (CFRs) during 2010-2017 were calculated using incidence data from the Sabah Department of Health.
RESULTS: Six malaria deaths occurred in Sabah…

Plasmodium knowlesi a simian malaria parasite is currently affecting humans in Southeast Asia. Malaysia has reported the most number of cases and P. knowlesi is the predominant species occurring in humans. The vectors of P. knowlesi belong to the Leucosphyrus group of Anopheles mosquitoes. These are generally described as forest-dwelling mosquitoes. With deforestation and changes in land-use, some species have become predominant in farms and villages. However, knowledge on the distribution of these vectors in the country is sparse. From a public health point of view it is important to know the…

Malaria is the most important parasitic disease with global concern. Plasmodium knowlesi recently has emerged from its natural simian host as a significant cause of human malaria, particularly in Malaysian Borneo. Therefore, it has been added as the fifth human Plasmodium specie which is widely distributed in Southeast Asia. Recent developments of new molecular tools enhanced our understanding about the key features of this malaria parasite. Here, we review some of the ways in which molecular approaches might be used for epidemiology of P. knowlesi and finally lead to an efficient control of m…

Plasmodial lactate dehydrogenase, key enzyme of anaerobic glycolysis, has been shown to be a potential immunodiagnostic marker as well as a novel target for chemotherapy. We have cloned, overexpressed and immunochemically characterized the recombinant lactate dehydrogenase of Plasmodium knowlesi, the fifth human malaria parasite. The P. knowlesi lactate dehydrogenase (PkLDH) gene was PCR amplified and 0.9 kb PCR product was cloned into pGEM-T Easy vector. Sequencing and BLAST analysis revealed open reading frame of 316 amino acids of PkLDH showing 96.8% homology with Plasmodium vivax LDH and a…

Intraerythrocytic stages of cell-free Plasmodium knowlesi possess significant activities of heme biosynthetic enzymes, viz. delta-aminolevulinic acid synthase (delta-ALAS), delta-aminolevulinic acid dehydrase (delta-ALAD), ferrochelatase (FC), and tryptophan pyrrolase (enzyme representing free heme pool). delta-Aminolevulinic acid synthase and FC showed higher activities in schizont than in ring trophozoite stage. Uninfected monkey erythrocytes did not possess the above-mentioned enzyme activities; on the contrary, leucocytes showed detectable enzyme activities. delta-Aminolevulinic acid synth…

A culture chamber fitted with a polycarbonate sieve has been used to isolate Plasmodium knowlesi merozoites as they are released from schizonts. A 3 mum pore-size sieve allows passage of normal erythrocytes and red cells containing rings and trophozoites and can be used to concentrate schizonts from a mixed cell population. A 2 mum pore-size sieve retains normal and parasitized cells and provides uncontaminated merozoites in high yield (5 x 10(10) merozoites per ml schizonts). Merozoite viability diminishes rapidly during 30 min after isolation. These preparations should prove valuable for stu…

The growing proximity between non-human primates (NHPs) and human communities in Thailand has raised concerns about zoonotic disease transmission. Among these NHPs, Assamese macaques (Macaca assamensis) are understudied in Thailand, particularly regarding their role as reservoirs for zoonotic malaria. The limited data on Plasmodium infections in this species highlights the need for further investigation. Therefore, this study determined the prevalence and molecular characterization of zoonotic Plasmodium spp. in Assamese macaques from Chiang Rai Province, northern Thailand. In total, 133 blood
