Characterization and genome sequencing of two Propionibacterium acnes phages displaying pseudolysogeny.

BACKGROUND: Propionibacterium acnes is a Gram positive rod inhabiting the human skin that also infects orthopaedic implants and is associated with acne vulgaris. Previously, one lytic bacteriophage, PA6, from P. acnes has been sequenced and partially characterized. We recently isolated several inducible phages from P. acnes classified as Siphoviruses based on morphology and partial genome sequencing. RESULTS: In this study we sequenced the inducible P. acnes phages PAD20 and PAS50, isolated from deep infection and from skin, respectively. The genomes of PAD20 and PAS50 are 29,074 and 29,017 b…

Antibiotic susceptibility of Propionibacterium acnes isolated from acne vulgaris in Korea.

Propionibacterium acnes plays an important role in the development of acne, and inflammatory lesions are improved by antibiotics. Long-term use of antibiotics may result in development of resistant strains and treatment failure. The aim of the present study was to investigate the isolation rate of P. acnes and to evaluate its antibiotic susceptibility to widely used antibiotics in acne in Korea. Among 46 patients, 31 P. acnes strains were cultured. Isolated P. acnes was measured for minimum inhibitory concentration (MIC) of tetracycline, doxycycline, minocycline, erythromycin and clindamycin u…

Experimental pulmonary granuloma mimicking sarcoidosis induced by Propionibacterium acnes in mice.

Propionibacterium acnes has been implicated as an etiologic agent of sarcoidosis since the isolation of this bacterium from sarcoid lesions. We experimentally produced a murine pulmonary granuloma model using P. acnes with several features that simulate sarcoidosis. Mice were sensitized with heat-killed P. acnes and complete Freund's adjuvant and were subsequently challenged with heat-killed P. acnes at 2-week intervals. P. acnes-challenged mice developed epitheloid cell granulomas in the lungs. These mice showed a pulmonary immune response characterized by an increased number of T-lymphocytes…

Inducible Siphoviruses in superficial and deep tissue isolates of Propionibacterium acnes.

BACKGROUND: Propionibacterium acnes is a commensal of human skin but is also known to be involved in certain diseases, such as acne vulgaris and infections of orthopaedic implants. Treatment of these conditions is complicated by increased resistance to antibiotics and/or biofilm formation of P. acnes bacteria. P. acnes can be infected by bacteriophages, but until recently little has been known about these viruses. The aim of this study was to identify and characterize inducible phages from P. acnes on a genetic and morphological basis. RESULTS: More than 70% (65/92) of P. acnes isolates inves…

Antibiotic-resistant Propionibacterium acnes suppressed by a benzoyl peroxide cleanser 6%.

Adding topical benzoyl peroxide (BPO) to antibiotics can reduce resistant Propionibacterium acnes in patients with acne receiving antibiotic therapy. Benzoyl peroxide often is formulated as a wash, but no published data exist regarding BPO wash formulation efficacy in reducing resistant strains of P acnes. This 3-week, open-label, single-center study evaluated the effects of BPO cleanser 6% on antibiotic-resistant P acnes populations. The study involved 30 healthy adults who were free of acne but had high facial P acnes populations (10,000 colonies/cm2 or more) resistant to erythromycin and te…

[Biotyping of Propionibacterium acnes strains isolated from patients with acne vulgaris].

Propionibacterium acnes is the main bacterial etiologic agent in acne vulgaris pathogenesis. The rate of antibiotic resistant P. acnes isolates is less than 10% in our country. The aim of this study was to biotype P. acnes strains which were isolated from acne vulgaris patients and to investigate the biotype distribution in our region. P. acnes isolates from 64 patients (18 male, 46 female; mean age 19 +/- 3.1 years) were included in the study and biotyped by sugar-specific pH changes using sorbitol, erythritol and ribose. Results were compared with the global acne grading scores and clinical …

Antimicrobial susceptibility of strains of Propionibacterium acnes isolated from inflammatory acne.

In the last decade, a significant increase in the antimicrobial resistance of clinical specimens of Propionibacterium acnes to first line antibiotics used for acne treatment, has been informed in Europe and in the USA. No information about strains isolated from Latin-American countries is available. The antimicrobial susceptibility of 53 strains of P. acnes isolated from skin specimens of inflammatory acne patients, at the clinical Hospital University of Chile was tested. All isolates were susceptible to penicillin, minocycline, and nadifloxacin. Erythromycin and clindamycin resistance was fou…

Propionibacterium acnes isolated from explanted intraocular lens in pseudophakic bullous keratopathy.

Propionibacterium acnes is an anaerobic bacterium that has been reported to produce ocular infection or inflammation following surgery or penetrating trauma. It has been well documented as an etiologic agent associated with infections of prosthetic implants, including intraocular lenses (IOLs). P. acnes is part of the natural flora of the external eye and has biological properties that may promote, heighten, or prolong an inflammatory response. It not only resists in vitro degradation by reticuloendothelial cells, but also demonstrates adjuvant properties in vivo. In this study, we cultured th…

Growth of Propionibacterium acnes strains on semisynthetic medium; antigenic polysaccharide production.

A semisynthetic medium, containing yeast extract as the only non defined component and glucose, glycerol, Tween 80 and mineral salts, has been developed to grow the strains of Propionibacterium acnes Beck 2037, Gerrath 2038 and Vogel 2039, at a rate comparable to that of complex media. An average of 10(12) cells per 1 (equivalent to approximately 1.5 g/l dry weight) was usually achieved. These yields are appropriated for biochemical and immunological studies, e.g. cell wall preparation, polysaccharides isolation, etc. In this work, some properties of the cell walls (sugar and amino acid compos…

Calcium sulphate mixed with antibiotics does not decrease efficacy against Cutibacterium acnes (formerly Propionibacterium acnes), in vitro study.

BACKGROUND: This study explored the in vitro efficacy of antibiotics mixed with calcium sulfate (ACS) against Cutibacterium acnes (C. acnes). METHODS: C. acnes isolates from orthopaedic infection sites were tested for antimicrobial susceptibility with ACS. Minimal inhibitory concentrations (MIC) were determined with a gradient diffusion method (Etest® strips). RESULTS: When tested with Etest®, all 22 isolates were susceptible to penicillin, ceftriaxone, vancomycin, and two were resistant to clindamycin (MICs of 4 and 8 mg/L). Penicillin and rifampin had the largest inhibition zone diameters.

Propionibacterium (Cutibacterium) granulosum Extracellular DNase BmdE Targeting Propionibacterium (Cutibacterium) acnes Biofilm Matrix, a Novel Inter-Species Competition Mechanism.

Acne vulgaris is the most common dermatological disorder worldwide affecting more than 80% of adolescents and young adults with a global prevalence of 231 million cases in 2019. The involvement of the skin microbiome disbalance in the pathophysiology of acne is recognized, especially regarding the relative abundance and diversity of Propionibacterium acnes a well-known dominant human skin commensal. Biofilms, where bacteria are embedded into a protective polymeric extracellular matrix, are the most prevalent life style for microorganisms. P. acnes and its biofilm-forming ability is believed to…

Propionibacterium acnes strain populations in the human skin microbiome associated with acne.

The human skin microbiome has important roles in skin health and disease. However, bacterial population structure and diversity at the strain level is poorly understood. We compared the skin microbiome at the strain level and genome level of Propionibacterium acnes, a dominant skin commensal, between 49 acne patients and 52 healthy individuals by sampling the pilosebaceous units on their noses. Metagenomic analysis demonstrated that although the relative abundances of P. acnes were similar, the strain population structures were significantly different in the two cohorts. Certain strains were h…

Decrease in Diversity of Propionibacterium acnes Phylotypes in Patients with Severe Acne on the Back.

Propionibacterium acnes, a major member of normal skin microbiota, is subdivided into 6 phylotypes: IA1, IA2, IB, IC, II and III. This study investigated P. acnes subgroups on the face and back in patients with severe acne and in healthy controls. In 71.4% of patients with severe acne, P. acnes phylotypes were identical on the face and back, whereas this was the case in only 45.5% of healthy controls. The healthy group carried phylotypes IA1 (39.1%) and II (43.4%), whereas the acne group carried a high predominance of IA1 (84.4%), especially on the back (95.6%). In addition, the single-locus s…

Propionibacterium acnes and the Th1/Th17 Axis, Implications in Acne Pathogenesis and Treatment.

Acne vulgaris is one of the most commonly seen conditions and the immunological link is a topic of active research. Recently, the Th17 pathway has been found to play a pivotal role in acne. The adaptive immune response toward Propionibacterium acnes leads to activation of Th17 axis. Consequently, the Th17 cytokines (IL-17, IL-1 β, IL-6, and tumor growth factor, in turn, activate the various pathogenic steps in acne. Drugs such as Vitamin D3 and isotretinoin which target the Th17 pathway may offer an additional pathway for their therapeutic response.
