Assessment of China's contributions to the Regional Network for Asian Schistosomiasis and Other Helminth Zoonoses: a questionnaire survey.

BACKGROUND: The Regional Network for Asian Schistosomiasis and Other Helminth Zoonoses (RNAS+) was established in 1998, which has developed close partnerships with Asian countries endemic for schistosomiasis and other helminthiasis in Asia. RNAS+ has provided an ideal regional platform for policy-makers, practitioners and researchers on the prevention, control and research of parasitic diseases in Asian countries. China, one of the initiating countries, has provided significant technical and financial support to the regional network. However, its roles and contributions have not been explored

Schistosoma mansoni P-glycoprotein levels increase in response to praziquantel exposure and correlate with reduced praziquantel susceptibility.

One potential physiological target for new antischistosomals is the parasite's system for excretion of wastes and xenobiotics. P-glycoprotein (Pgp), a member of the ATP-binding-cassette superfamily of proteins, is an ATP-dependent efflux pump involved in transport of toxins and xenobiotics from cells. In vertebrates, increased expression of Pgp is associated with multidrug resistance in tumor cells. Pgp may also play a role in drug resistance in helminths. In this report, we examine the relationship between praziquantel (PZQ), the current drug of choice against schistosomiasis, and Pgp express

Human subcutaneous dirofilariasis caused by Dirofilaria immitis in a Greek adult.

Human dirofilariasis is a zoonotic infection caused by worms of the genus Dirofilaria. Most reported cases involve Dirofilaria repens, and D. immitis infection has been rarely reported. Canines act as a reservoir for the infection, while human infections are sporadic. Human dirofilariasis has been widely reported in South Europe; however, the worldwide distribution constantly changes. We herein report an extremely rare case of subcutaneous trunk dirofilariasis in a 45-year-old immunosuppressed woman, caused by D. immitis. The parasitic infection was detected using ultrasonography. The infectio

Unique antigenic gene expression at different developmental stages of Trichinella pseudospiralis.

Parasite-induced and parasite-regulated larval capsule formation and host immunosuppression are two major characteristics that are unique in Trichinella spp. infections, but the molecule(s) and mechanism(s) that mediate these processes remain largely unknown. Trichinella pseudospiralis and Trichinella spiralis, are obviously different with respect to these two characteristics. A comparative study of these two species, in particular their antigen expression profiles at different developmental stages (the main molecules involved in the cross-talk or interaction between each parasite and its host

Some molecular insights into schistosome evolution.

Robust phylogenies based on molecular data for species within the genus Schistosoma have been generated in recent years. The considerable progress made in understanding the relationships between many of the 19 recognised species of Schistosoma is reviewed with particular attention being given to the detection and analysis of parasite variation as shown by studies on ribosomal RNA genes, mitochondrial DNA and RAPDs. For the most part, molecular phylogenies agree with observations based on morphological or life-history characteristics. It is clear that the parasites of man do not form a monophyl

One Health implications and first evidence of environmental contamination of helminths in soil from goat farms in Ratchaburi, Thailand.

Zoonotic helminths are responsible for the majority of helminthic infections occurring in humans globally. Environmental systems serve as a reservoir for zoonotic helminths, facilitating their transmission to humans and animals. Livestock farms may serve as hotspots for zoonotic transmission, increasing infection risk. Focusing on goat farms in Ratchaburi Province, Thailand, we aim to detect and identify zoonotic helminths present in the soil environment through morphological and molecular techniques. Soil samples (n = 270) were collected from 30 goat farms in Ratchaburi Province, and helminth

Genome-wide identification and functional characterization of GPCR family genes reveal their key roles in the vitellarium development and egg production in Schistosoma japonicum.

BACKGROUND: Schistosomiasis is a zoonotic parasitic disease of the genus Schistosoma. Current therapeutic approaches predominantly rely on the single drug praziquantel, highlighting the urgent need to identify additional effective drug targets. G protein-coupled receptors (GPCRs) play important roles in various biological processes and are considered pivotal in drug development. However, the current understanding of the function of GPCRs in schistosomes remains limited. METHODS: We conducted a systematic bioinformatics analysis of Schistosoma japonicum GPCRs using public genomic resources to

Exploring the Schistosoma mansoni adult male transcriptome using RNA-seq.

Schistosoma mansoni is one of the agents of schistosomiasis, a chronic and debilitating disease. Here we present a transcriptome-wide characterization of adult S. mansoni males by high-throughput RNA-sequencing. We obtained 1,620,432 high-quality ESTs from a directional strand-specific cDNA library, resulting in a 26% higher coverage of genome bases than that of the public ESTs available at NCBI. With a 15×-deep coverage of transcribed genomic regions, our data were able to (i) confirm for the first time 990 predictions without previous evidence of transcription; (ii) correct gene predictions;

The discovery of Paratestophis gelicolus gen. nov., sp. nov. from the rainbow water snake, Enhydris enhydris, in Thailand, with systematic update of Echinochasmidae Odhner, 1910.

A new genus and species of trematode, Paratestophis gelicolus gen. nov., sp. nov., is described from the large intestine of the rainbow water snake, Enhydris enhydris, collected from several provinces in southern Thailand. Morphological analyses reveal distinct characteristics that differentiate P. gelicolus gen. nov., sp. nov. from related echinochasmid taxa, specifically its elongated bottle-shaped body, presence of 22 collar spines, parallel testes and parasitism of snakes-features not observed in other echinochasmid genera. Prospecting for Zoonotic Pathogens by Using Targeted DNA Enrichment.

More than 60 zoonoses are linked to small mammals, including some of the most devastating pathogens in human history. Millions of museum-archived tissues are available to understand natural history of those pathogens. Our goal was to maximize the value of museum collections for pathogen-based research by using targeted sequence capture. We generated a probe panel that includes 39,916 80-bp RNA probes targeting 32 pathogen groups, including bacteria, helminths, fungi, and protozoans. Laboratory-generated, mock-control samples showed that we are capable of enriching targeted loci from pathogen D

Genetic Analysis of Small-Subunit Ribosomal RNA, Internal Transcribed Spacer 2, and ATP Synthase Subunit 8 of Trichuris trichiura Ancient DNA Retrieved from the 15th to 18th Century Joseon Dynasty Mummies' Coprolites from Korea.

Although parasitic infection by Trichuris trichiura is a very common intestinal helminthic disease worldwide, there is still insufficient information on the genetic characteristics of ancient T. trichiura in different spatiotemporal perspectives. Utilizing coprolite specimens obtained from 15th-18th century mummies dating to the Joseon Dynasty, we analyzed small-subunit ribosomal RNA, internal transcribed spacer 2, and ATP synthase subunit 8 of T. trichiura ancient DNA (aDNA). In BLAST and phylogenetic analyses, the T. trichiura aDNA sequences of this study belong to a separate cluster that is

Single-cell analysis of recruited ILC2 cell fate during transition from circulation to establishment of tissue residency.

Circulating group 2 innate lymphoid cells (ILC2s) often serve as a first line of defense against infection prior to local expansion of lung-resident ILC2s. The fate of circulating ILC2s and their relationship with lung-resident ILC2s is not well understood. Using reporter mice in combination with single-cell RNA sequencing (scRNA-seq) and Cellular Indexing of Transcriptomes and Epitopes sequencing (CITE-seq), the fate of circulatory ILC2s was followed during the course of primary Nippostrongylus brasiliensis helminth infection. Circulating ILC2s rapidly acquire tissue-resident gene expression

Rapid group-2 innate lymphoid cell mobilization from the intestine aids in early lung defense and repair.

Circulating group 2 innate lymphoid cells (ILC2s) comprise a third of all lung ILC2s shortly after helminth infection, providing an early source of type-2 cytokines. However, the origin of circulating ILC2s and their relationship with ILC progenitors and tissue-resident ILC2s remain unresolved. Single-cell RNA-sequencing and trajectory analysis of ILC2 and ILC2 progenitors in the lung, bone marrow (BM), and intestine draining lymph nodes (LNs) revealed that circulating ILC2s in the lung mirror a population of ILC2s in the mesenteric LNs. Conversely, lung-resident ILC2s closely resembled BM ILC

Alteration of the fecal microbiota in Chinese patients with Schistosoma japonicum infection.

Schistosoma japonicum infection causes pathological injury to the host. Multiple studies have shown that intestinal helminth infection causes dysbiosis for the gut microbial community and impacts host immunology. However, the effect of acute S. japonicum infection on the gut microbiome structure (abundance and diversity) is still unclear. We collected fecal samples from healthy and infected patients from a single hospital in Hunan Province, China. The bacterial community was analyzed using 16S ribosomal RNA gene sequencing of the V4 hypervariable region using the HiSeq platform. Compared with

Genetic and molecular analysis of spe-27, a gene required for spermiogenesis in Caenorhabditis elegans hermaphrodites.

Hermaphrodites with mutations in the spe-27 gene are self-sterile, laying only unfertilized eggs; mutant males are fertile. Hermaphrodites make spermatids that fail to activate to crawling spermatozoa so passing oocytes sweep them out of the spermatheca. These spermatids do activate and produce self-progeny if young mutant hermaphrodites are mated by fertile (or sterile) males. Spermatids isolated from either mutant males or hermaphrodites initiate activation in vitro when treated with proteases, but then arrest with spiky membrane projections that resemble those of a normal intermediate in ps

RNAi-mediated silencing of ARV1 in Setaria digitata impairs in-vitro microfilariae release, embryogenesis and adult parasite viability.

Setaria digitata is a nematode that resides in the peritoneal cavity of ruminants causing cerebrospinal nematodiasis disease affecting livestock and inflicting significant economic forfeitures in Asia. Further, this nematode can infect humans, causing abscesses, allergic reactions, enlarged lymph nodes, eye lesions and inflammation of the lungs. The 'ARE2 required for viability1' (ARV1) encodes for putative lipid transporter localized in the endoplasmic reticulum (ER) and Golgi complex membrane in humans and yeast. In the present study, the functional role of S. digitata ARV1 (SD-ARV1) was inv

Autophagy in C. elegans development.

Autophagy involves the sequestration of cytoplasmic contents in a double-membrane structure referred to as the autophagosome and the degradation of its contents upon delivery to lysosomes. Autophagy activity has a role in multiple biological processes during the development of the nematode Caenorhabditis elegans. Basal levels of autophagy are required to remove aggregate prone proteins, paternal mitochondria, and spermatid-specific membranous organelles. During larval development, autophagy is required for the remodeling that occurs during dauer development, and autophagy can selectively degra

RNAi pathways in parasitic protists and worms.

Tropical diseases caused by parasitic worms and protists are of major public health concern affecting millions of people worldwide. New therapeutic and diagnostic tools would be of great help in dealing with the public health and economic impact of these diseases. RNA interference (RNAi) pathways utilize small non-coding RNAs to regulate gene expression in a sequence-specific manner. In recent years, a wealth of data about the mechanisms and biological functions of RNAi pathways in distinct groups of eukaryotes has been described. Often, RNAi pathways have unique features that are restricted t
