OBJECTIVE: Alzheimer's disease (AD) molecular mechanisms remain unclear, and effective therapies are lacking. This study employed whole-transcriptome sequencing to investigate the expression profiles of circRNAs, lncRNAs, miRNAs, and mRNAs in the hippocampus of APP/PS1 transgenic mice, aiming to identify ceRNA regulatory networks and clarify their roles in AD pathogenesis.
METHODS: Differentially expressed RNAs were integrated into ceRNA networks, and programmed cell death-related pathways were analyzed using GO and KEGG. The computationally predicted circRNA_13083/miR-298-5p/CD14 axis was pr

BACKGROUND & AIMS: The immune tolerance induced by hepatitis B virus (HBV) is a major challenge for achieving effective viral clearance, and the mechanisms involved are not well-understood. One potential factor involved in modulating immune responses is mesencephalic astrocyte-derived neurotrophic factor (MANF), which has been reported to be increased in patients with chronic hepatitis B. In this study, our objective is to examine the role of MANF in regulating immune responses to HBV.
METHODS: We utilized a commonly used HBV-harboring mouse model, where mice were hydrodynamically injected wi

Post-infarction inflammation and adverse remodeling remain major therapeutic challenges in ischemic heart disease. However, how specific intercellular communication drives macrophage metabolic maladaptation during this process remains unclear. This study aimed to elucidate the mechanisms by which damaged cardiomyocytes epigenetically reprogram macrophage immunometabolism post-myocardial infarction. Here, we identify a novel inter-organelle and inter-cellular signaling axis wherein hypoxic cardiomyocyte-derived exosomal NEAT1 acts as a pivotal epigenetic rheostat of macrophage immunometabolism.

Gram-negative bacteria derived extracellular vesicles (EVs), also known as outer membrane vesicles, have attracted significant attention due to their pathogenic roles in various inflammatory diseases. We recently demonstrated that EVs secreted by the periodontopathogen Aggregatibacter actinomycetemcomitans (Aa) can cross the blood-brain barrier (BBB) and that their extracellular RNA cargo can promote the secretion of proinflammatory cytokines, such as IL-6 and TNF-α, in the brain. To gain more insight into the relationship between periodontal disease (PD) and neuroinflammatory diseases, we inv

Natural killer (NK) cells preferentially accumulate at maternal-foetal interface and are believed to play vital immune-modulatory roles during early pregnancy and related immunological dysfunction may result in pregnant failure such as recurrent miscarriage (RM). However, the mechanisms underlying the establishment of maternal-foetal immunotolerance are complex but clarifying the roles of decidual NK (dNK) cells offers the potential to design immunotherapeutic strategies to assist RM patients. In this report, we analysed RNA sequencing on peripheral NK (pNK) and decidual NK cells during early

Acinetobacter baumannii is a multidrug-resistant (MDR) nosocomial pathogen with high mortality rates, necessitating alternative preventive strategies. The RNA chaperone Hfq plays a key role in bacterial virulence, and its deletion attenuates pathogenicity. In this study, we evaluated an hfq deletion mutant (Δhfq) of A. baumannii ATCC 17978 as a live-attenuated vaccine candidate. Mice vaccinated intraperitoneally with 2.3 × 107 CFU of Δhfq, followed by a booster on day 28, exhibited 90 % survival after an intraperitoneal lethal challenge with MDR A. baumannii AB5075, whereas all unvaccinated co

Alterations during the early stages of embryo development have been associated with long-term effects on the fetus, neonate, and adult, but this has not been investigated in horses. In recent years, intracytoplasmic sperm injection (ICSI) has gained in commercial popularity in the equine population. Research suggests an association between ICSI-produced embryos and placental malformations, but there exists little understanding of the physiology involved. Therefore, we aim to produce a complete transcriptomic analysis of chorioallantois and provide potential pathways that may be impacted follow

BACKGROUND: Chronic atrophic gastritis (CAG) is a clinically refractory gastric disease often characterized by high recurrence rates and adverse drug reactions. Anwei decoction (AWD), a traditional Chinese medicine formula, has been shown to significantly improve clinical symptoms in patients with CAG, as demonstrated by a multicenter cohort study (overall effective rate: 82.5%, P < 0.01). However, the unclear molecular mechanisms and therapeutic targets of AWD limit its international acceptance.
AIM: To investigate the therapeutic mechanisms of AWD against CAG from an integrated perspective.

BACKGROUND: In the past years, several studies investigated how distinct immune cell subsets affects post-myocardial infarction repair. However, whether and how the tissue environment controls these local immune responses has remained poorly understood. We sought to investigate how antigen-specific T-helper cells differentiate under myocardial milieu's influence.
METHODS: We used a transgenic T cell receptor (TCR-M) model and major histocompatibility complex-II tetramers, both myosin-specific, combined with single-cell transcriptomics (single-cell RNA sequencing [scRNA-seq]) and functional ph

Regnase-1 is a CCCH-type RNA-binding protein that constantly degrades mRNA species via its ribonuclease domain to maintain immune quiescence. But how and by what mechanisms Regnase-1 regulates Foxp3+ Tregs remains poorly studied. Here, we generated a conditional knock-in strain in which a degradation-resistant Regnase-1 mutant (R111A) was selectively expressed in Foxp3+ Tregs as a transgene (Foxp3YFP-CreLSL-R111Afl/fl) to investigate the roles of Regnase-1 in Tregs. We found that all male Foxp3YFP-CreLSL-R111Afl/fl mice died of fatal autoimmune diseases by 4 weeks of age, which was accompanied

Repeated implantation failure (RIF) is a significant limiting factor in assisted reproduction. Chronic endometrial inflammation has been noted in RIF women, therefore we sought to investigate the potential association of endometrial Th17/Treg ratio and endometrial inflammation in these cases. Endometrial pipelle biopsies were obtained from volunteers, 29 women with RIF (failure to achieve pregnancy following at least 3 transfers of high-grade embryos in IVF-cycles) and 27 fertile women (at least one child) in total, at the secretory phase of the menstrual cycle. Using tissues from 17 fertile a

ETHNOPHARMACOLOGICAL RELEVANCE: The combination of Alisma and Atractylodes (AA), a classical traditional Chinese herbal decoction, may protect against cerebral ischaemia/reperfusion injury (CIRI). However, the underlying mechanism has not been characterized. Intriguingly, exosomal microRNAs (miRNAs) have been recognized as vital factors in the pharmacology of Chinese herbal decoctions.
AIM OF THE STUDY: The aim of the present study was to assess whether the neuroprotective effect of AA was dependent on the efficient transfer of miRNAs via exosomes in the brain.

BACKGROUND: Novel therapeutic strategies in ovarian cancer (OC) are needed as the survival rate remains dismally low. Although dendritic cell-based cancer vaccines are effective in eliciting therapeutic responses, their complex and costly manufacturing process hampers their full clinical utility outside specialized clinics. Here, we describe a novel approach of generating a rapid and effective cancer vaccine using ascites-derived monocytes for treating OC.
METHODS: Using the ID8 mouse ovarian tumor model and OC patient samples, we isolated ascites monocytes and evaluated them with flow cytome

Gastric cancer (GC) is a malignant neoplasm displaying highly cancer-related mortality globally. Although our previous studies have confirmed that vitamin D possessed a direct anti-cancer effect on GC cells, the regulatory role of vitamin D on gastric tumor microenvironment (TME) remains unexplored. This study aims to expound the modulation of vitamin D on TME especially on GC-associated fibroblasts (CAFs) and to further elucidate the essential role of the CAFs-derived exosomal ingredients in tumor-stroma crosstalk. Patient-derived primary CAFs enhanced the aggressive characteristics of GC cel

Pemphigus vulgaris is an autoimmune blistering disease caused by IgG targeting desmoglein 3 (Dsg3), an adhesion molecule of keratinocytes. Anti-Dsg3 IgG production is prevented in healthy individuals, but it is unclear how Dsg3-specific B cells are regulated. To clarify the immunological condition regulating Dsg3-specific B cells, a pathogenic anti-Dsg3 Ig (AK23) knock-in mouse was generated. AK23 knock-in B cells developed normally without undergoing deletion or acquiring an anergic phenotype in vivo. The knock-in B cells showed Ca2+ influx upon IgM cross-linking and differentiated into AK23-

BACKGROUND: Autoimmune and non-autoimmune thyroiditis frequently occur in persons with hepatitis C virus (HCV) infection. Treatment with interferon alpha (IFNα) is also associated with significant risk for the development of thyroiditis. To explore HCV-thyroid interactions at a cellular level, we evaluated whether a human thyroid cell line (ML1) could be infected productively with HCV in vitro.
METHODS AND RESULTS: ML1 cells showed robust surface expression of the major HCV receptor CD81. Using a highly sensitive, strand-specific reverse transcription polymerase chain reaction assay, positive

Helper T cells from a mutant mouse model, LAT Y136F, hyper-proliferate and cause a severe lymphoproliferative disease that kills the mice by six months of age. LAT Y136F mice carry a tyrosine to phenylalanine mutation in the Linker for Activation of T cells (LAT) gene. This mutation leads to a number of changes in T cells that result in altered cytokine production including increased IL-4 production, increased proliferation, and decreased apoptosis. Hyper-proliferation of the mutant T cells contributes to lymphadenopathy, splenomegaly, and multi-organ T cell infiltration. miRNAs are short non-

Hepatitis C virus (HCV) still lacks a licensed vaccine. The envelope glycoprotein E2 is a key neutralizing target, but its dense N-glycan shield can hinder epitope exposure. In this study, we revisit E2 glycan editing and examine whether single-site deletion preserves antigen integrity while improving immune responses in mice under a DNA immunization setting. Using a secreted E2 ectodomain (sE2384-661), we generated five N to D mutants at conserved sites (N1, N2, N4, N6, and N11) and evaluated them in a unified DNA immunization model with identical CpG content and delivery conditions across gr

BACKGROUND AND OBJECTIVE: Despite integrase strand transfer inhibitor (INSTI)-containing regimens now being considered a preferred option for both initial therapy and switching strategies in virologically suppressed patients, their effects on lymphocyte phenotypes and functions in the course of effective combination antiretroviral therapy (cART) are still unclear. Thus, we investigated the effect of a 24-week elvitegravir/cobicistat/emtricitabine/tenofovir disoproxil fumarate (EVG/c/FTC/TDF) regimen on the T cell compartment and HIV reservoirs in HIV-infected patients switching from a suppress

BACKGROUND: Sepsis-induced myocardial injury (SIMI) affects approximately 20%-55% of septic patients, with associated mortality ranging from 37% to 70%; however, evidence for magnesium sulfate (MgSO₄) as a targeted SIMI intervention and its molecular basis remain poorly defined.
METHODS: We integrated three evidentiary levels. A retrospective cohort of 2219 SIMI patients from the MIMIC-IV database underwent 1:1 propensity score matching (PSM; n = 992) and piecewise Cox regression (Day 10 landmark) to evaluate 28-day mortality.
