Evolution of rod bipolar cells and rod vision.

Bipolar cells are vertebrate retinal interneurons conveying signals from rod and cone photoreceptors to amacrine and ganglion cells. Bipolar cells are found in all vertebrates and have many structural and molecular affinities with photoreceptors; they probably appeared very early during vertebrate evolution in conjunction with rod and cone progenitors. There are two types of bipolar cells, responding to central illumination with depolarization (ON) or hyperpolarization (OFF). In most vertebrate species, rod signals are conveyed to specialized rod bipolar cells, which sum signals from many rods

Thyroid hormone regulatory system in the mouse retina: integrated transcriptional and translational analysis of deiodinase enzymes, thyroid hormone receptors and thyroid hormone transporters.

BACKGROUND: Thyroid hormone (TH) signaling is involved in complex processes regulating development and energy balance, especially in organs with high-rate metabolism such as the retina. Despite evidence of TH involvement in regulation of retinal functions and disease susceptibility, a comprehensive analysis of the tissue distribution of the components of the retinal TH regulatory system is lacking. The aims of the present study were (i) to provide a detailed description of the retinal expression sites and localization patterns of deiodinases (DIO2 and DIO3), TH receptors (THRα and THRβ), and T

HDAC inhibition ameliorates cone survival in retinitis pigmentosa mice.

Cone photoreceptor cell death in inherited retinal diseases, such as Retinitis Pigmentosa (RP), leads to the loss of high acuity and color vision and, ultimately to blindness. In RP, a vast number of mutations perturb the structure and function of rod photoreceptors, while cones remain initially unaffected. Extensive rod loss in advanced stages of the disease triggers cone death by a mechanism that is still largely unknown. Here, we show that secondary cone cell death in animal models for RP is associated with increased activity of histone deacetylates (HDACs). A single intravitreal injection

Molecular and cellular impact of a C203R/C198R M-opsin mutation.

A C203R mutation in M-opsin is a cause of blue cone monochromacy (BCM) in human patients. The equivalent mutation in murine M-opsin is C198R since human M-opsin has an extra 5 amino acid residues in the amino terminal region that are not present in murine M-opsin. The mechanism by which the C203R/C198R mutation causes BCM is unclear, and the function and dysfunction of cone opsins in general are understudied compared to that of rhodopsin in rod photoreceptor cells. To better understand the dysfunction caused by a C203R/C198R mutation in M-opsin, the effect of the mutation was examined both in

Disruption of the basal body protein POC1B results in autosomal-recessive cone-rod dystrophy.

Exome sequencing revealed a homozygous missense mutation (c.317C>G [p.Arg106Pro]) in POC1B, encoding POC1 centriolar protein B, in three siblings with autosomal-recessive cone dystrophy or cone-rod dystrophy and compound-heterozygous POC1B mutations (c.199_201del [p.Gln67del] and c.810+1G>T) in an unrelated person with cone-rod dystrophy. Upon overexpression of POC1B in human TERT-immortalized retinal pigment epithelium 1 cells, the encoded wild-type protein localized to the basal body of the primary cilium, whereas this localization was lost for p.Arg106Pro and p.Gln67del variant forms of POC

Distinct and atypical intrinsic and extrinsic cell death pathways between photoreceptor cell types upon specific ablation of Ranbp2 in cone photoreceptors.

Non-autonomous cell-death is a cardinal feature of the disintegration of neural networks in neurodegenerative diseases, but the molecular bases of this process are poorly understood. The neural retina comprises a mosaic of rod and cone photoreceptors. Cone and rod photoreceptors degenerate upon rod-specific expression of heterogeneous mutations in functionally distinct genes, whereas cone-specific mutations are thought to cause only cone demise. Here we show that conditional ablation in cone photoreceptors of Ran-binding protein-2 (Ranbp2), a cell context-dependent pleiotropic protein linked t

Ultrastructure of remnant photoreceptors in advanced hereditary retinal degeneration.

The outer layers of the retinas of pigmented rats affected with hereditary retinal degeneration (rdy) were studied at an advanced stage in the degenerative process, ie, in 200 day old animals. At this age, most of the photoreceptors that survived the degenerative process were cones. The chromatin pattern of their nuclei clearly differentiated them from rods, displaced pigment cells and/or presumptive macrophages that also were found in the outer nuclear layer. None of the cones encountered had outer segments although structures resembling outer segment discs were found with a single cilium. Co

Glucose uptake by GLUT1 in photoreceptors is essential for outer segment renewal and rod photoreceptor survival.

Photoreceptors consume glucose supplied by the choriocapillaris to support phototransduction and outer segment (OS) renewal. Reduced glucose supply underlies photoreceptor cell death in inherited retinal degeneration and age-related retinal disease. We have previously shown that restricting glucose transport into the outer retina by conditional deletion of Slc2a1 encoding GLUT1 resulted in photoreceptor loss and impaired OS renewal. However, retinal neurons, glia, and the retinal pigment epithelium play specialized, synergistic roles in metabolite supply and exchange, and the cell-specific map

Genetic and epigenetic control of photoreceptor development.

Advances in 'omics' technologies have uncovered new insights into photoreceptor development. Dynamic expression of and interaction among transcription factors in post-mitotic photoreceptor precursors guide cone and rod cell fate and maturation, as well as homeostasis. Together with genetic and epigenetic modifications, we are now putting together the pieces of this complex puzzle to elucidate the molecular mechanisms underlying photoreceptor differentiation. In this review, we discuss our current understanding of developmental pathways that result in the generation of cones and rods from a com

Mouse cone arrestin gene characterization: promoter targets expression to cone photoreceptors.

Cone arrestin (CAR) is a novel member of the arrestin superfamily expressed in retinal cone photoreceptors and the pineal gland. To understand the regulatory mechanisms controlling its cone- and pineal-specific expression, and to facilitate further functional studies using gene knockout approaches, we characterized the genomic organization and the 5'-flanking region of the mouse CAR (mCAR) gene. The mCAR gene is comprised of 17 exons and 16 introns, encoding five alternatively spliced transcripts. A 215-bp proximal promoter fragment containing a TATA box, an Sp1 site and four cone-rod homeobox

Phosphoinositide 3-Kinase Enhancer Protein: Insights into Its Expression and Functions in Retinal Cells.

This study explores the expression and localization of phosphoinositide 3-kinase enhancer (PIKE) in the retina, unveiling its potential role in retinal degenerative diseases. Phosphatidylinositol 3-kinase (PI3K) activation is pivotal for photoreceptor functions, linked to retinal degenerative disorders. PIKE, an enhancer of PI3K/Akt activities, remains unexplored in retinal physiology. Through immunohistochemistry, immunoblots, and real-time PCR, we examined PIKE in rod-dominant and cone-dominant retinas of wild-type mice and models of diabetes and retinal degeneration. PIKE was present in pho

C8ORF37 Is Required for Photoreceptor Outer Segment Disc Morphogenesis by Maintaining Outer Segment Membrane Protein Homeostasis.

C8ORF37 is a causative gene for three different clinical forms of incurable retinal degeneration. However, the completely unknown function of C8ORF37 limits our understanding of the pathogenicity of C8ORF37 mutations. Here, we performed a comprehensive phenotypic characterization of a C8orf37 KO mouse line, generated using CRISPR/Cas9 technology. Both C8orf37 KO male and female mice exhibited progressive and simultaneous degeneration of rod and cone photoreceptors but no non-ocular phenotypes. The major ultrastructural feature of C8orf37 KO photoreceptors was massive disorganization of the out

Lutein delays photoreceptor degeneration in a mouse model of retinitis pigmentosa.

Retinitis pigmentosa is a retinal disease characterized by photoreceptor degeneration. There is currently no effective treatment for retinitis pigmentosa. Although a mixture of lutein and other antioxidant agents has shown promising effects in protecting the retina from degeneration, the role of lutein alone remains unclear. In this study, we administered intragastric lutein to Pde6brd10 model mice, which display degeneration of retinal photoreceptors, on postnatal days 17 (P17) to P25, when rod apoptosis reaches peak. Lutein at the optimal protective dose of 200 mg/kg promoted the survival of

Evaluating Human Rod Photoreceptor Function Using Pixelwise Intensity-Based Optoretinography.

PURPOSE: More than 50 inherited retinal diseases are known, with rod photoreceptor functional loss serving as an early indicator in nearly half of them. The "optoretinogram" is a relatively new assay that detects optical changes in cells in response to stimuli. This tool has excellent potential for providing insights into the earliest functional changes of rod photoreceptors, with the potential to assist in the early detection, monitoring, and treatment of retinal diseases. METHODS: In this work, we obtained intensity-based optoretinograms (iORGs) from rod photoreceptors using an adaptive opt

Light and electron microscopic observations on retinal neurons of red-tail shark (Epalzeorhynchos bicolor H. M. Smith, 1931).

The structure of photoreceptors (PR) and the arrangement of neurons in the retina of red-tail shark were investigated using light and electron microscopy. The PR showed a mosaic arrangement and included double cones, single cones (SC), and single rods. Most cones occur as SC. The ratio between the number of cones and rods was 3:1.39 (±0.29). The rods were tall that reached the pigmented epithelium. The outer plexiform layer (OPL) showed a complex synaptic connection between the horizontal and photoreceptor terminals that were surrounded by Müller cell processes. Electron microscopy showed that

PURPOSE: The cone-dominant, 13-lined ground squirrel (13-LGS) retina mimics the human central retina, but a thorough examination of retinal development in this species has not been reported. Here, the embryonic and postnatal development of the 13-LGS retina was studied to further characterize 13-LGS as a practical alternative animal model for investigating cone-based vision in health and disease. METHODS: The spatiotemporal expression of key progenitor and cell type markers was examined in retinas from defined embryonic and postnatal stages using immunohistochemistry. Loss of cone photoreceptors caused by chromophore depletion is partially prevented by the artificial chromophore pro-drug, 9-cis-retinyl acetate.

Inactivating mutations in the retinoid isomerase (RPE65) or lecithin:retinol acyltransferase (LRAT) genes cause Leber congenital amaurosis (LCA), a severe visual impairment in humans. Both enzymes participate in the retinoid (visual) cycle, the enzymatic pathway that continuously generates 11-cis-retinal, the chromophore of visual pigments in rod and cone photoreceptor cells needed for vision. We investigated human RPE65-LCA patients and mice with visual cycle abnormalities to determine the impact of chronic chromophore deprivation on cones. Young patients with RPE65 mutations showed foveal co

A mouse model of cone photoreceptor function loss (cpfl9) with degeneration due to a mutation in Gucy2e.

During routine screening of mouse strains and stocks by the Eye Mutant Resource at The Jackson Laboratory for genetic mouse models of human ocular disorders, we identified cpfl9, a mouse model with cone photoreceptor function loss. The mice exhibited an early-onset phenotype that was easily recognized by the absence of a cone-mediated b-wave electroretinography response and by a reduction in rod-mediated photoresponses at four weeks of age. By genetic mapping and high-throughput sequencing of a whole exome capture library of cpfl9, a homozygous 25 bp deletion within exon 11 of the Gucy2e gene

Characterization of zebrafish rod and cone photoresponses.

Zebrafish is a popular species widely used in vision research. The zebrafish retina has one rod and four cone subtypes (UV-, blue-, green-, and red-sensitive) with 40%-rod 60%-cone ratio, making it suitable for comparable studies of rods and cones in health and disease. However, the basic photoresponse properties of the four zebrafish cone subtypes have not been described yet. Here, we established a method for collecting flash photoresponses from zebrafish rods and cones by recording membrane current with a suction electrode. Photoreceptor subtypes could be distinguished based on their charact

Eye features and retinal photoreceptors of the nocturnal aardvark (Orycteropus afer, Tubulidentata).

The nocturnal aardvark Orycteropus afer is the only extant species in the mammalian order Tubulidentata. Previous studies have claimed that it has an all-rod retina. In the retina of one aardvark, we found rod densities ranging from 124,000/mm² in peripheral retina to 214,000/mm² in central retina; the retina of another aardvark had 163,000 - 245,000 rods/mm². This is moderate in comparison to other nocturnal mammals. With opsin immunolabelling we found that the aardvark also has a small population of cone photoreceptors. Cone densities ranged from about 300 to 1,300/mm² in one animal, and fro
