Penetrating, Ultra-small Microelectrode Arrays (UMEAs) have been shown to evoke less foreign body response on implantation in neural tissue. In the present study, an acidic PLA(2), designated Bl-PLA(2), was isolated from Bothrops leucurus snake venom through two chromatographic steps: ion-exchange on CM-Sepharose and hydrophobic chromatography on Phenyl-Sepharose. Bl-PLA(2) was homogeneous on SDS-PAGE and when submitted to 2D electrophoresis the molecular mass was 15,000Da and pI was 5.4. A gelatinolytic metalloproteinase (gMP) from red sea bream ( Pagrus major ) skeletal muscle was highly purified by ammonium sulfate fractionation and column chromatographies including (diethylamino)ethyl (DEAE)-Sephacel, phenyl-Sepharose, and gelatin-Sepharose. An L-amino acid oxidase (BjarLAAO-I) from Bothrops jararaca snake venom was highly purified using a stepwise sequential chromatography on Sephadex G-75, Benzamidine Sepharose and Phenyl Sepharose. A motility inhibiting factor (MIF) in sperm plasma membrane of mammalian spermatozoa (goat) has been demonstrated. This factor has been purified to apparent homogeneity by Sepharose-6B affinity chromatography and DEAE-cellulose ion-exchange chromatography. The molecular weight of the isolated factor has been estimated as 98 kDa by molecular sieving and analytical HPLC. OBJECTIVE: To determine the kinds of proteoglycan (PG) monochain and their distribution in the cell layer and cultural medium of bovine trabecular meshwork (BTM) culture.
METHODS: (3)H-glycosamine was incorporated in BTM cells in the third passage. PGs were extracted by 4 mol/L guanidine hydrochloride. The aliquots were chromatographed on Sephadex G-50 DEAE-Sephacel, Sepharose 4B and Sepharose CL-6B gel respectively. A hemophilic patient treated with Factor VIII (F.VIII) concentrates developed a F.VIII inhibitory activity. The patient's plasma showed 2 anti-F.VIII/von Willebrand Factor (vWF) antibodies (Abs) of the IgG and IgA class respectively. An enzyme hydrolyzing sphingomyelin was purified from extracts of solid cultures of Aspergillus saitoi 7041 by fractionation with isopropanol followed by successive column chromatographies on DEAE-Sepharose CL-6B, butyl-Toyopearl 650 M, and phenyl-Sepharose CL-4B. Poly(A) polymerase [EC 2.7.7.19] was highly purified from beef liver nuclei by the use of column chromatographies on heparin-Sepharose 4B and Blue Dextran-Sepharose 4B.
