Transcriptome profiling of immune-responsive genes in the intestine of Cynoglossus semilaevis Günther challenged with Shewanella algae.

To better understand gene expression in the intestine after Shewanella algae infection and provide insights into its immune roles in the tongue sole, Cynoglossus semilaevis, sequencing-based high-throughput RNA analysis (RNA-Seq) for the intestines between the control group and 12 h post-injection group was performed. After assembly, there was an average of 23,957,159 raw sequencing reads, and 23,943,491 clean reads were obtained after filtering out low-quality reads. Then, 383 differentially expressed genes (DEGs) in the intestines in response to S. algae infection were identified. Subsequent…

Interaction between in vivo bioluminescence and extracellular electron transfer in Shewanella woodyi via charge and discharge.

Extracellular electron transfer (EET) and bioluminescence are both important for microbial growth and metabolism, but the mechanism of interaction between EET and bioluminescence is poorly understood. Herein, we demonstrate an exclusively respiratory luminous bacterium, Shewanella woodyi, which possesses EET ability and electron communication at the interface of S. woodyi and solid substrates via charge and discharge methods. Using an electro-chemiluminescence apparatus, our results confirmed that the FMN/FMNH2 content and the redox status of cytochrome c conjointly regulated the bioluminescen…

Ampicillin promotes the biofilm formation of Shewanella putrefaciens through the c-di-GMP-regulated BpfAGD system.

Although many Shewanella strains are naturally resistant to some β-lactam antibiotics, research into the effect of these antibiotics on Shewanella biofilm formation is scarce. Shewanella putrefaciens is not only known as an important seafood spoilage bacterium but can also cause infection in several aquatic animals. In addition, it is a rare opportunistic human pathogen. The present study found that although some antibiotics from cephalosporins, carbapenems, and monobactams repress the biofilm formation of S. putrefaciens CN32, multiple penicillin antibiotics increase its biofilm formation. Fu…

Uniform and Pitting Corrosion of Carbon Steel by Shewanella oneidensis MR-1 under Nitrate-Reducing Conditions.

Despite observations of steel corrosion in nitrate-reducing environments, processes of nitrate-dependent microbially influenced corrosion (MIC) remain poorly understood and difficult to identify. We evaluated carbon steel corrosion by Shewanella oneidensis MR-1 under nitrate-reducing conditions using a split-chamber/zero-resistance ammetry (ZRA) technique. This approach entails the deployment of two metal (carbon steel 1018 in this case) electrodes into separate chambers of an electrochemical split-chamber unit, where the microbiology or chemistry of the chambers can be manipulated. This appro…

Genetic characterization of plasmid-mediated quinolone resistance gene qnrS2 in Pseudoalteromonas and Shewanella isolates from seawater.

Three qnrS2-containing isolates of Pseudoalteromonas and Shewanella were collected from the seawater samples of Qingdao in China during 2014. They displayed resistance to ampicillin, ciprofloxacin, kanamycin, nalidixic acid and sulfamethoxazole. The qnrS2 genes were identified in the chromosomes of Pseudoalteromonas strains E8 and S16, and in a 140-kb plasmid in Shewanella strain S14, respectively. In addition, two copies of qnrS2 were identified in the strain E8. Sequence analyses revealed that there was an identical DNA segment located in the downstream of qnrS2 in strain S14 and E8, coding …

Oxic microbial ferrihydrite reduction rates of Shewanella oneidensis and the potential for Fe mobilization in oxic sediments.

Microbially mediated reduction of ferrihydrite (Fe(III) oxyhydroxide) plays a crucial role in Fe cycling, and hence nutrient and contaminant cycling, in subsurface environments. This process is typically considered a strictly anaerobic process confined to anoxic microsites within oxic subsurface environments. However, recent findings suggest that microbes can also mediate ferrihydrite reduction under oxic conditions. Here, we quantified cell-specific rates of ferrihydrite reduction by the model organism Shewanella oneidensis MR-1 under oxic and anoxic conditions. We used these rates to assess …

The viable but non-culturable (VBNC) status of Shewanella putrefaciens (S. putrefaciens) with thermosonication (TS) treatment.

Although thermosonication (TS) treatment has been widely used in food sterilization, the viable but non-culturable (VBNC) of bacteria with TS treatment has still concerned potential food safety and public health. The molecular mechanism of VBNC status of bacteria with TS treatment is not clearly known. Therefore, in this study, we used Shewanella putrefaciens, which was a common putrefactive bacteria in aquatic products, to study the VBNC state of bacteria with TS treatment. Firstly, our results revealed that S. putrefaciens still could enter the VBNC state after TS treatments: 50 kHz, 300 W, …

Characterization of a novel phage SPX1 and biological control for biofilm of Shewanella in shrimp and food contact surfaces.

Shewanella putrefaciens, commonly found in seafood, forms tenacious biofilms on various surfaces, contributing to spoilage and cross-contamination. Bacteriophages, owing to their potent lytic capabilities, have emerged as novel and safe options for preventing and eliminating contaminants across various foods and food processing environments. In this study, a novel phage SPX1 was isolated, characterized by a high burst size (43.81 ± 3.01 PFU/CFU) and a short latent period (10 min). SPX1 belongs to the Caudoviricetes class, exhibits resistance to chloroform, and sensitivity to ultraviolet. It sh…

Roles of multiple KASIII homologues of Shewanella oneidensis in initiation of fatty acid synthesis and in cerulenin resistance.

It is fully established that the condensing reaction for the initiation of fatty acid synthesis is essential for viability of many bacteria. In model bacteria such as Escherichia coli, this reaction is exclusively catalyzed by β-ketoacyl-ACP synthase (KAS) III (encoded by fabH) and the FabH loss results in a fatty acid auxotroph. However, such a notion has been under the challenge of recent findings. In an attempt to resolve the conflicting results, in this study, we examined the physiological role of multiple KASIII enzyme homologues in Shewanella oneidensis, an excellent model for researchin…

Transcriptome analysis reveals a stress response of Shewanella oneidensis deprived of background levels of ionizing radiation.

Natural ionizing background radiation has exerted a constant pressure on organisms since the first forms of life appeared on Earth, so that cells have developed molecular mechanisms to avoid or repair damages caused directly by radiation or indirectly by radiation-induced reactive oxygen species (ROS). In the present study, we investigated the transcriptional effect of depriving Shewanella oneidensis cultures of background levels of radiation by growing the cells in a mine 655 m underground, thus reducing the dose rate from 72.1 to 0.9 nGy h-1 from control to treatment, respectively. Positive regulation of the Shewanella oneidensis OmpS38, a major porin facilitating anaerobic respiration, by Crp and Fur.

Major porins are among the most abundant proteins embedded in the outer membrane (OM) of Gram-negative bacteria, playing crucial roles in maintenance of membrane structural integrity and OM permeability. Although many OM proteins (especially c-type cytochromes) in Shewanella oneidensis, a research model for respiratory versatility, have been extensively studied, physiological significance of major porins remains largely unexplored. In this study, we show that OmpS38 and OmpA are two major porins, neither of which is responsive to changes in osmolarity or contributes to the intrinsic resistance…

ε-Poly-l-lysine-Functionalized 2D MoS2 Nanosheets for Photothermal Inhibition of Cold-Adapted Shewanella putrefaciens.

Psychrotrophic Shewanella putrefaciens, notorious for dominating cold chain spoilage through cold-adapted membrane dynamics and robust biofilm formation, challenges conventional antimicrobial strategies that lose potency under refrigeration and raise food safety concerns. We report an engineering approach that transforms ε-poly-l-lysine (ε-PL), a GRAS-listed food preservative, into a triple-duty agent: a green exfoliant for MoS2 nanosheets, a cationic surface modifier, and a synergistic antimicrobial partner. The engineered ε-PL@MoS2 nanosheets exploit a unique vulnerability of cold-adapted ba…

Differentially Expressed Infection-Associated Genes in a Shewanella algae isolate From a Deceased Northern Bottlenose Whale.

Diseases are an important but poorly understood influence on marine ecosystems. However, many pathogens of wild marine organisms remain poorly characterized, limiting our understanding of their ecology and potential impacts. This study investigates the potential virulence mechanisms of a Shewanella algae strain isolated from the brain tissue of a deceased northern bottlenose whale (Hyperoodon ampullatus). Using long-read Oxford Nanopore RNA Sequencing, differential gene expression analysis (DGEA) was conducted comparing growth in standard media, Tryptic Soy Broth (TSB) and TSB supplemented wit…

Severe Sepsis and Wet Gangrene Requiring Foot Amputation Caused by an Emerging Human Pathogen - Shewanella algae.

A 69-year-old woman with type 2 diabetes mellitus, peripheral vascular disease, and other comorbidities presented with recurrent syncopal episodes. Cellulitic skin changes in her right lower extremity were noted, as well as a large hemorrhagic bulla on the dorsum of her right foot. Severe sepsis was determined to be the reason for her syncopal episodes. Blood cultures and the bulla aspirate culture were positive for Shewanella algae that was pan-sensitive to antibiotics. Her clinical status was stabilized with a regimen of intravenous fluids and broad-spectrum antibiotics. However, due to the …

Draft genome sequence of carbapenem-resistant Shewanella algae strain AC isolated from small abalone (Haliotis diversicolor).

OBJECTIVES: Shewanella algae is an environmental marine bacterium and an emerging opportunistic human pathogen. Moreover, there are increasing reports of strains showing multidrug resistance, particularly carbapenem-resistant isolates. Although S. algae has been found in bivalve shellfish aquaculture, there is very little genome-wide data on resistance determinants in S. algae from shellfish. The aim of this study was to determine the whole genome sequence of carbapenem-resistant S. algae strain AC isolated from small abalone in Taiwan. METHODS: Bacterial genomic DNA was sequenced using an Il…

Bio-nano hybrids: molecular interactions between iron oxide nanoparticles and outer-membrane extracellular cytochromes of Shewanella oneidensis.

Metal oxide nanoparticles (NPs) coupled with dissimilatory metal-reducing bacteria enable continuous redox cycling through extracellular electron transfer (ET), a process central to bioremediation and bioenergy applications. However, the molecular mechanisms governing ET at the outer membrane-iron oxide NP interface remain unclear. Here, atomistic molecular dynamics simulations were employed to elucidate how the extracellular cytochromes of Shewanella oneidensis cooperatively mediate iron oxide NP adsorption, a prerequisite for extracellular ET. Our study shows that MtrF and two OmcA decaheme …

Potential-driven balance of extracellular electron transfer activity and microstructure stability in Shewanella biofilm for uranium recovery.

Bioelectrochemistry enables uranium recovery from uranium-contaminated water. Within microbial electrolysis cells (MECs), electroactive biofilm growth is critical, but the impact of electrode potential on biofilm structure and function remains unknown. This study explored variations in micro-structure, extracellular electron transfer, and uranium recovery of Shewanella oneidensis MR-1 biofilms cultivated under different potential shocks from -0.2 to 0.6 V (vs Ag/AgCl) in MECs at -0.2 V. Across this range, uranium recovery exhibited a clear non-monotonic trend, peaking at 0.2 V and exhibiting t…

Chemical mining of heterotrophic Shewanella algae reveals anti-infective potential of macrocyclic polyketides against multidrug-resistant pathogens.

Heterotrophic Gamma-proteobacterium Shewanella algae MTCC 12715, associated with an intertidal red algae Hypnea valentiae, presented broad-spectra of antibacterial activities against pathogenic bacteria bringing about nosocomial infection. Bioassay-guided fractionation of the bacterial crude extract resulted in two undescribed macrocyclic polyketide analogs, with anti-infective activities against methicillin-resistant Staphylococcus aureus and vancomycin-resistant Enterococcus faecalis (MIC 3.1-5.0 µg/mL). In order to identify the polyketide biosynthetic machinery termed type-I polyketide synt…

Combination of rosemary extract and buffered vinegar inhibits Pseudomonas and Shewanella growth in common carp (Cyprinus carpio).

BACKGROUND: Aquaculture is the fastest growing food-production sector, and common carp (Cyprinus carpio) is one of the most cultivated fish species in the world. Due to its intrinsic characteristics, fish meat is highly susceptible to microbiological spoilage. Pseudomonas and Shewanella are the primary and secondary occurring microbiota during storage of fish meat, with significant contribution to spoilage with the formation of hydrolytic enzymes (lipases and proteases). RESULTS: With in vitro testing, we show that rosemary extract (Inolens4), buffered vinegar and their combination (SyneROX) …
