Preparation and characterization of a silk fibroin/calcium sulfate bone cement.

This study aimed to prepare and characterize a silk fibroin/calcium sulfate (SF/CS) bone cement. SF solutions of the following concentrations 3, 4.5, 6, 7.5, and 9 g/L were used to prepare bone cement samples with SF-to-CS ratios of 0.35, 0.40, 0.45, 0.50, and 0.55 mL of SF solutions (or water as control) per g of CS. Compressive strength, setting time, degradation when immersed in phosphate-buffered saline, X-ray diffraction, Fourier infrared spectroscopy, and scanning electron microscopy were used to characterize the SF/CS bone cement. Biocompatibility was determined using rat bone mesenchym…

Oriented nanofibrous silk as a natural scaffold for ocular epithelial regeneration.

The aim of this study was to develop nanofibrous silk substrates for limbal stem cell expansion that can serve as a potential alternative substrate to replace human amniotic membrane. The human limbal stem cell was used to evaluate the biocompatibility of substrates (random and oriented nanofibrous mats, and human amniotic membrane) based on their phenotypic profile, viability, proliferation, and attachment ability. Biocompatibility results indicated that all substrates were highly biocompatible, as limbal stem cells could favorably attach and proliferate on the nanofibrous surfaces. Microscop…

Spider genomes provide insight into composition and evolution of venom and silk.

Spiders are ecologically important predators with complex venom and extraordinarily tough silk that enables capture of large prey. Here we present the assembled genome of the social velvet spider and a draft assembly of the tarantula genome that represent two major taxonomic groups of spiders. The spider genomes are large with short exons and long introns, reminiscent of mammalian genomes. Phylogenetic analyses place spiders and ticks as sister groups supporting polyphyly of the Acari. Complex sets of venom and silk genes/proteins are identified. We find that venom genes evolved by sequential …

Study on the microstructure of African wild silk cocoon shells and fibers.

Silk fibers and cocoon shells from four African wild silkmoths Gonometa postica, Anaphe panda, Argema mimosae and Epiphora bauhiniae-were studied to gain insight into the structure-property-function relations and potential commercial application. The surface and cross-section of cocoon shells and fibers revealed the presence of prominent structural variations. Cocoon shells were multilayered and porous structures constructed from highly cross-linked fibers that are densely packed within the sericin/gum. Fibers had fibrillar sub-structures running along the fiber axis and with greater number an…

Optical characterization of the nanoscale organization of mineral deposits on silk films.

The alignment of deposited minerals in tissues such as bone and teeth plays a critical role in the mechanical properties of these tissues. Therefore, assessment of features that are characteristic of aligned biominerals could aid in the development of novel biomaterials and engineered tissues that can be used to replace damaged or defective human tissues. In this study, we demonstrate that light scattering spectroscopy can serve as a useful tool for the noninvasive characterization of mineralization on aligned organic substrates. Specifically, we used silk films with oriented and nonoriented s…

Conformational polymorphism, stability and aggregation in spider dragline silks proteins.

Spider silk is spun in a complex and unique process, thought to depend on a hydrophobic conversion of a predominantly disordered to a beta-sheet rich protein structures. To test this hypothesis we monitored the effect of cationic (DOTAC) and anionic (alkyl sulfate) detergents and of (ii) solvent polarity using a series of alcohols on the secondary structure transition in dilute solutions of native spidroin. Our results showed that the detergents hydrophilic head charge and hydrophobic tail length cooperatively induced either a transition to the beta-sheet rich form or a stable helical state. C…

Dehydrated silk fibroin matrices as versatile delivery systems for extracellular vesicles.

Extracellular vesicles (EVs) have emerged as promising alternatives to cell-based therapies due to their low immunogenicity, ability to carry therapeutic cargo, and enhanced stability. However, rapid clearance from the bloodstream and phagocytic uptake by macrophages after systemic or local administration limit their therapeutic efficacy, highlighting the need for delivery systems that enable sustained and localized EV release. In this study, a natural protein silk fibroin (SF) was selected as a biomaterial carrier due to its tunable degradation kinetics and biocompatibility. SF was produced i…

Silk fibroin scaffolds loaded with growth factors can reverse liver fibrosis.

Currently, there are no effective pharmacological interventions in clinical practice to reverse liver fibrosis. This study explores the therapeutic potential of regenerated silk fibroin (RSF) scaffolds loaded with dual growth factors for reversing hepatic fibrosis. A chronic liver fibrosis mouse model was induced using carbon tetrachloride (CCl4) combined with a high-fat diet. RSF scaffolds, with or without hepatocyte growth factor (HGF) and fibroblast growth factor-4 (FGF-4), were implanted onto the liver surface to assess antifibrotic efficacy. Liver function was evaluated using biochemical …

Silk fibroin based in situ antibacterial hydrogel for urethral defect repair.

Due to urethral strictures post hypospadias and bacterial infection, desirable urethral repair remains a great challenge in clinic. In situ formed hydrogels have shown great capacity on promoting tissue regeneration and can be delivered to fill irregularities in injury sites. In this study, we developed an in situ antibacterial hydrogel system (SFPLMA) by combining methacrylated silk fibroin (SFMA) and methacrylated polylysine (PLMA). The rapid photo-crosslinking of the methacryloyl moieties in SFMA and PLMA enabled the SFPLMA hydrogel to effectively fill the congenital irregularities associat…

Biomimetic trilayered silk-based electrospun scaffolds for regeneration of dura mater.

Dura mater healing is essential to prevent cerebrospinal fluid (CSF) leaks in neurosurgical procedures. Drawing inspiration from the hierarchical structure of native dura mater, we have designed a biomimetic electrospun trilayered scaffold (TLS) utilizing silk fiber to replicate both the structure and function of the original tissue. The electrospun trilayered scaffold comprises three distinct layers: a skull-facing layer constructed from silk fibroin combined with strontium-doped bioactive glass, a gradient inert polyurethane middle layer and a brain-facing layer consisting of polyurethane in…

Precisely Printable Silk Fibroin/Carboxymethyl Cellulose/Alginate Bioink for 3D Printing.

Three-dimensional (3D) bioprinting opens up many possibilities for tissue engineering, thanks to its ability to create a three-dimensional environment for cells like an extracellular matrix. However, the use of natural polymers such as silk fibroin in 3D bioprinting faces obstacles such as having a limited printability due to the low viscosity of such solutions. This study addresses these gaps by developing highly viscous, stable, and biocompatible silk fibroin-based inks. The addition of 2% carboxymethyl cellulose sodium and 1% sodium alginate to an aqueous solution containing 2.5 to 5% silk …

Sudan Black B Pretreatment to Suppress Autofluorescence in Silk Fibroin Scaffolds.

Natural polymers are extensively utilized as scaffold materials in tissue engineering and 3D disease modeling due to their general features of cytocompatibility, biodegradability, and ability to mimic the architecture and mechanical properties of the native tissue. A major limitation of many polymeric scaffolds is their autofluorescence under common imaging methods. This autofluorescence, a particular challenge with silk fibroin materials, can interfere with the visualization of fluorescently labeled cells and proteins grown on or in these scaffolds, limiting the assessment of outcomes. Here, …

Dual-functional bioactive silk sericin for osteoblast responses and osteomyelitis treatment.

Sericin, a natural protein from silk cocoon, has been reported for various biological properties in the biomaterials field. Modified forms of sericin have been studied for bone tissue engineering, while its unmodified form has been scarcely reported. Therefore, the purpose of this study was to evaluate physical and biological properties of unmodified sericin for potential use in bone surgery. Sericin was extracted from silk cocoons using a chemical-free boiling method. Sericin extract showed distinct bands with molecular weight ranging from 25 to 42 kDa including smear bands. Fourier transform…

PLLA Porous Microsphere-Reinforced Silk-Based Scaffolds for Auricular Cartilage Regeneration.

Microtia, frequently encountered in plastic surgery practice, is usually corrected by auricular reconstruction with prostheses or autologous cartilages. In recent decades, however, cartilage tissue engineering has been emerging as a promising alternative for its minimal invasion and low immunogenicity. As a critical factor for tissue engineering, scaffolds are expected to be sufficiently porous and stiff to facilitate chondrogenesis. In this work, we introduce novel poly-l-lactic acid (PLLA) porous microsphere-reinforced silk-based hybrid (SBH) scaffolds with a multihierarchical porous structu…

Silk Fiber-Reinforced Hyaluronic Acid-Based Hydrogel for Cartilage Tissue Engineering.

A continuing challenge in cartilage tissue engineering for cartilage regeneration is the creation of a suitable synthetic microenvironment for chondrocytes and tissue regeneration. The aim of this study was to develop a highly tunable hybrid scaffold based on a silk fibroin matrix (SM) and a hyaluronic acid (HA) hydrogel. Human articular chondrocytes were embedded in a porous 3-dimensional SM, before infiltration with tyramine modified HA hydrogel. Scaffolds were cultured in chondropermissive medium with and without TGF-β1. Cell viability and cell distribution were assessed using CellTiter-Blu…

Robust Silk Fibroin/Graphene Oxide Aerogel Fiber for Radiative Heating Textiles.

Aerogel fibers with ultrahigh porosity and ultralow density are promising candidates for personal thermal management to reduce the energy waste of heating an entire room, and play important roles in reducing energy waste in general. However, aerogel fibers generally suffer from poor mechanical properties and complicated preparation processes. Herein, we demonstrate hierarchically porous and continuous silk fibroin/graphene oxide aerogel fibers (SF/GO) with high strength, excellent radiative heating performance, and thermal insulation performance through coaxial wet spinning and freeze-drying. …
