STATEMENT OF PROBLEM: Staphylococcus aureus (S aureus) and Streptococcus mutans (S mutans) are pathogenic bacteria associated with denture-related stomatitis. Zinc (Zn), a widely used metallic antibacterial agent, can induce the production of reactive oxygen species (ROS) in cells, thereby inhibiting microbial growth and proliferation; studies, however, are lacking.
PURPOSE: The purpose of this in vitro study was to determine the antibacterial and antibiofilm function of Zn2+ against S aureus and S mutans and assess gene expression changes to clarify its inhibitory mechanisms.

In this study, we evaluated the impact of Epigalocatechin-3-gallate (EGCG) on S. mutans biofilm development for 24 and 46 h using high-resolution confocal laser scanning microscopy. EGCG treatment led to the formation of interspaced exopolysaccharide (EPS)-microcolony complexes unevenly distributed on the surface of hydroxyapatite disc, forming a thinner and less complex biofilm structure with significantly reduced biomass, matrix volume, and thickness compared to the NaCl treated group (negative control). At 46 h, the biofilm of the EGCG-treatment group failed to form the bacterial-EPS supers

Streptococcus mutans, the principal pathogen associated with dental caries, impacts individuals across all age groups and geographic regions. Beyond its role in compromising oral health, a growing body of research has established a link between S. mutans and various systemic diseases, including immunoglobulin A nephropathy (IgAN), nonalcoholic steatohepatitis (NASH), infective endocarditis (IE), ulcerative colitis (UC), cerebral hemorrhage, and tumors. The pathogenic mechanisms associated with S. mutans frequently involve collagen-binding proteins (CBPs) and protein antigens (PA) present on th…

Candida albicans (C. albicans) and Streptococcus mutans (S. mutans) are frequently detected in the plaque biofilms of children with early childhood caries. This study investigated the effects of sucrose and farnesol on biofilm formation by the oral pathogens S. mutans and C. albicans, including their synergistic interactions. Biofilm formation dynamics were monitored using the Cell Index (CI). The CI for S. mutans increased in the brain-heart infusion medium, peaking at 10 h; however, the addition of sucrose reduced the CI. For C. albicans yeast cells, the CI increased at sucrose concentration…

Streptococcus mutans (S. mutans) is the main cariogenic pathogen associated with dental caries. Orientin-2''-O-β-L-galactoside, orientin and vitexin are natural flavonoids compound. In this study, the antibacterial ability of these flavonoids and their mechanisms in inhibiting S. mutans biofilm formation were investigated. Inhibition zone and 2-fold-dilution tests showed that these flavonoids exerted inhibitory effects on S. mutans. Phenol sulfuric acid method and lactate dehydrogenase (LDH) test revealed that they could reduce EPS formation and stimulate S. mutans to release LDH. Moreover, cr…

Streptococcus mutans is a key bacterial species in the caries process, which affects >90% of the population worldwide. However, other acidogenic and aciduric/acidophilic species may contribute to disease development. In Sweden, a country with low prevalences of caries and S. mutans, a significant portion of caries-affected adolescents lack detectable levels of S. mutans. The objectives of the present study were 1) to characterize the tooth biofilm and saliva microbiota of adolescents with caries disease, with or without detectable S. mutans, from tooth biofilm and saliva samples and 2) to asse…

We previously reported that, before tooth eruption, over one-half of infants aged 6 mos were already infected with Streptococcus mutans. The aim of this investigation was to determine the colonization of S. mutans after tooth eruption in the same cohort of 111 infants (35 pre-term, 76 full-term). Our results showed that S. mutans colonization increased with increasing age, so that by 24 mos of age, 84% harbored the bacteria (p < 0.01). The mean and median ages of S. mutans colonization in dentate infants were 15.7 mos and 16.0 mos, respectively. Factors associated with S. mutans colonization w…

Proteins belonging to the LraI (for "lipoprotein receptor antigen") family function as adhesins in several streptococci, as a virulence factor for endocarditis in at least one of these species, and potentially as metal transporters in many bacteria. We have identified and characterized the chromosomal locus containing the LraI family gene (designated sloC) from Streptococcus mutans, an agent of dental caries and endocarditis in humans. Northern blot analysis indicated that sloC is cotranscribed with three other genes. As with other LraI operons, the sloA and sloB genes apparently encode compon…

The role of Streptococcus mutans in the initiation of dental caries has been recognized and attributed, at least in part, to its ability to colonize the tooth surface. Therefore, factors which prevent S. mutans attachment to hydroxyapatite (HA) are of considerable interest for the prophylaxis of this infectious disease. Chitosan, a chitin derivative by N-deacetylation, is an interesting candidate in this respect, since it stimulates the ordered regeneration of oral soft tissues, prevents the deleterious action of organic acid, and exhibits bactericidal action against several pathogens. In the …

The nucleotide sequence has been determined for the Streptococcus mutans wall-associated protein A (wapA) gene from serotype c strains Ingbritt and GS5. The nucleotide sequence for each wapA gene was virtually identical, although the gene from strain GS5 contained a 24 base pair deletion. A 29 amino acid signal peptide was specified by each wapA gene with a mature protein of 424 amino acids (Mr, 45,276) for strain Ingbritt and 416 amino acids (Mr, 44,846) for strain GS5. In the C-terminal region of the wall-associated protein A, considerable sequence similarity was found with the membrane anch…

Dental caries is a widespread and contagious chronic infectious condition. As the principal cariogenic bacterium involved in dental caries, Streptococcus mutans (S. mutans) possesses cariogenicity-related properties, including acidogenicity, aciduricity, and exopolysaccharide (EPS) synthesis. Our previously designed peptide, P-113-DPS, has demonstrated antibacterial effects on S. mutans; however, its detailed impact on its cariogenic virulence factors remains unclear. This study focused on assessing changes in these factors following treatment with P-113-DPS. Furthermore, it aimed to investiga…

Dental caries remain an unresolved public health issue. Streptococcus mutans, a principal microorganism in the mouth, is closely associated with the onset and progression of dental caries. S. mutans adheres to tooth surfaces and synthesizes extracellular polysaccharides via the enzyme glucosyltransferase (Gtf), utilizing sucrose and forming biofilms. GtfC is linked to soluble and insoluble glucan synthesis, a significant factor in the adhesion of S. mutans to tooth surfaces and bacterial cell aggregation within biofilms. Chalcones, primary precursors for flavonoid and isoflavonoid biosynthesis…

BACKGROUND: Dental caries is a bacterial-mediated infectious disease that affects the hard tissues of the tooth, with Streptococcus mutans being the primary cariogenic pathogen due to its robust biofilm-forming ability. Controlling biofilm formation is essential for caries prevention. Recent studies have indicated that D-amino acids, which are not incorporated into proteins, play regulatory roles in bacterial processes such as growth inhibition and biofilm dispersal. However, whether D-amino acids can inhibit the growth of S. mutans remains controversial. This study aimed to investigate the ef…

UNLABELLED: Streptococcus mutans, a key oral pathogen, utilizes quorum sensing to regulate biofilm formation-a major virulence factor in the development of dental caries (tooth decay). Our recent research uncovered a complex interplay between the CSP-ComDE quorum sensing pathway and the Type II DpnII restriction-modification (R-M) system in S. mutans. The DpnII R-M system methylates adenine at 5'-GATC-3' sites and cleaves unmethylated DNA, significantly influencing foreign DNA acquisition and gene expression. In this study, we investigated the impact of a ΔRM mutant, which lacks adenine methyl…
