Congenital intestinal stenosis and Hirschsprung's disease: two extremely rare pathologies in a newborn puppy.

BACKGROUND: Hirschsprung's disease (HSCR) is a common congenital malformation of the enteric nervous system (ENS). During fetal development, ganglion cells of the ENS are derived from neural crest cells that migrate to the bowel. These cells reside principally in two ganglionated plexus: 1) The myenteric plexus, extending from the esophagus to the anus, and 2) submucous plexus, extending from the duodenum to the anus. In large animal species, there is a third plexus called Henle's or Schabadasch's plexus. ENS ganglion cells play a key role in normal gastrointestinal motility, respond to sensor…

Schistosoma mansoni induce granulomatous inflammation and lesion to the enteric nervous system in mouse colon.

Schistosomiasis mansoni is a major health problem, mainly occurring in developing countries. A large proportion of infected individuals suffer from motility related gastrointestinal problems. In the present study the granulomatous inflammation induced by eggs deposition in the colon wall and the lesion to the enteric nervous system was investigated. Mice infected with S. mansoni 4-12 weeks prior to the experiment, and uninfected control mice were studied. The colon showed both a diffuse mucosal, submucosal and muscularies inflammation as well as a granulomatous reaction. The diffuse mucosal in

Epigastrius with omphalocele--report of a case.

An epigastrius (parasitic twinning in the epigastrium) was delivered via the vaginal route spontaneously and died 37 days later. The autosite had cardiac hypertrophy with VSD, PDA, and PFO and an omphalocele. The liver was partly conjoined. There was a small swelling at the lower part of the autosite's bifurcated sternum to which the parasitic left pelvis and lower extremity were attached. On the second day after birth, parasitectomy was performed. The amputated specimens consisted of a small left pelvic girdle with a free extremity, a scrotal mass, nipplelike structures and two small protuber

Quantitative analysis of enteric neurons containing choline acetyltransferase and nitric oxide synthase immunoreactivities in the submucosal and myenteric plexuses of the porcine colon.

The enteric nervous system (ENS) controls gastrointestinal functions. In large mammals' intestine, it comprises an inner (ISP) and outer (OSP) submucous plexus and a myenteric plexus (MP). This study quantifies enteric neurons in the ISP, OSP, and MP of the pig ascending (AC) and descending colon (DC) using the HuC/D, choline acetyltransferase (ChAT), and neuronal nitric oxide synthase (nNOS) neuronal markers in whole mount preparations with multiple labeling immunofluorescence. We established that the ISP contains the highest number of HuC/D neurons/mm2, which were more abundant in AC vs. DC,…

Enteric neuron density correlates with clinical features of severe gut dysmotility.

Gastrointestinal (GI) symptoms can originate from severe dysmotility due to enteric neuropathies. Current methods used to demonstrate enteric neuropathies are based mainly on classic qualitative histopathological/immunohistochemical evaluation. This study was designed to identify an objective morphometric method for paraffin-embedded tissue samples to quantify the interganglionic distance between neighboring myenteric ganglia immunoreactive for neuron-specific enolase, as well as the number of myenteric and submucosal neuronal cell bodies/ganglion in jejunal specimens of patients with severe G

Regional differences in the sympathetic innervation of the guinea pig large intestine by neuropeptide Y- and tyrosine hydroxylase-immunoreactive nerves of divergent extrinsic origin.

Region-specific patterns of nerves with immunoreactivity to neuropeptide Y (NPY) have been described previously in the submucous plexus of guinea pig large intestine. Because these may have functional significance, the possibility of similar, characteristic variations of NPY-like immunoreactivity (NPY-ir) in the myenteric plexus was explored. Regional differences were found in the occurrence and pattern of distribution of NPY-ir in the myenteric plexus of the guinea pig large intestine. NPY-ir was present rarely within neuron somata in any region of the large intestine, and NPY-ir nerve fibers…

Neuronal plasticity of the enteric nervous system is correlated with chagasic megacolon development.

Chagas' disease is one of the few functional gastrointestinal disorders for which a causative agent has been identified. However, some pathological aspects of the chagasic megasyndromes are still incompletely understood. Chagasic megacolon is characterized by an inflammatory process, organ dilatation and neuronal reduction in both plexuses of the enteric nervous system (ENS). Although some studies on the ENS in Chagas' disease have been performed, the process of neuronal destruction and neuronal regeneration still remains unclear. Our hypothesis is that the regeneration process of the ENS may

The innervation of the human antro-pyloric region: organization and composition.

Although the composition of the gastric innervation has been determined in animal models, relatively little known about the innervation of the human antro-pyloric region. We used immunocytochemical techniques to establish the localization and co-expression of neuropeptides and nitric oxide in the human antrum and upper duodenum. Our results demonstrate the existence of a clearly defined submucosal plexus in the antral region that is absent in rats and guinea pigs. The abundant innervation of the lamina propria contains 3 major nerve populations: VIP- and NOS-, SP- and CGRP-, and GRP-immunoreac

Immunohistochemical investigations of gut hormones in the colon of patients with Hirschsprung's disease.

The distributions of gut hormones in the colon of Hirschsprung's disease were investigated by the peroxidase-antiperoxidase (PAP) immunohistochemical method. Three colonic segments (ganglionic, oligoganglionic, and aganglionic) were stained by the unlabeled antibody enzyme method. The immunoreactivity of vasoactive intestinal polypeptide (VIP) was found to be reduced in the oligoganglionic and aganglionic segments. Antisera to substance P and met-enkephalin demonstrated immunoreactive cells and fibers in the ganglionic segment, whereas these cells and fibers were almost completely absent in th

Enteric nervous system and inflammatory bowel diseases: Correlated impacts and therapeutic approaches through the P2X7 receptor.

The enteric nervous system (ENS) consists of thousands of small ganglia arranged in the submucosal and myenteric plexuses, which can be negatively affected by Crohn's disease and ulcerative colitis - inflammatory bowel diseases (IBDs). IBDs are complex and multifactorial disorders characterized by chronic and recurrent inflammation of the intestine, and the symptoms of IBDs may include abdominal pain, diarrhea, rectal bleeding, and weight loss. The P2X7 receptor has become a promising therapeutic target for IBDs, especially owing to its wide expression and, in the case of other purinergic rece

Nervous structure of Meckel's diverticulum in children.

Meckel's diverticulum, being considered as the most frequent malformation of the digestive tract, has been largely presented in scientific papers, but a complete physiopathological mechanism for its natural history has not been yet described. We have studied the nervous system and the differences observed in eight Meckel's diverticulums with enteric or ectopic gastric mucosa, using specific immunohistochemical markers. It has been noted a significantly higher density of myenteric nerve fibers in areas with enteric mucosa compared with the areas with gastric heterotopias, while the transition z

Neuronal intestinal and fibromuscular arterial dysplasias associated with intraluminal mucosal web.

A two-year-old boy presenting with the signs and symptoms of partial intestinal obstruction caused by a mucosal web in the ileum is presented. Histopathological evaluation following resection and primary anastomosis revealed neuronal intestinal dysplasia associated with fibromuscular arterial dysplasia limited to the segment proximal to the web and normal findings in the distal part. These findings and a brief review of the literature support the hypothesis that neuronal and fibromuscular dysplasias may develop on the basis of congenital obstructive lesions of the bowel.

Neuronal activity of submucosal plexus of pyloric and ileocecal sphincteric regions of the cat gastrointestinal tract.

Extracellular recording of neuronal activity of the submucosal plexus of isolated pyloric and ileocecal sphincteric regions of cat intestine revealed both spontaneously active and silent neurons. The spontaneously active neurons consisted of fast- and slow-burst-type units and single-spike units. The silent neurons generated trains of spikes in response to deformation of the ganglion by the recording electrode or in response to local application of acetylcholine (Ach, 6 X 10(-5) -6 X 10(-2) microM/l) on the ganglion surface. Interactions of the excitatory type were made manifest by the synchro

Acetylcholinesterase-positive nerves of the rhesus monkey bronchial tree.

The rhesus monkey lung was stained both by histological methods and histochemically for specific acetylcholinesterase (AChE). AChE-containing nerves in bundles were demonstrated in connective tissue of the hilum and in association with clusters of ganglion cells. These bundles become associated with the bronchial tree as they enter the lung parenchyma, and their numbers of myelinated fibres diminish as they pass scattered ganglion cells along the bronchial system. Extrachondral and subchondral plexuses of nerves were found to be interconnected and to contribute to the perimuscular varicose ner

Autoradiographic localisation of muscarinic receptors in guinea-pig intestine: distribution of high and low affinity agonist binding sites.

Muscarinic receptors mediate a variety of intestinal functions including smooth muscle contraction, ganglionic transmission and water and electrolyte secretion. In this study, we have used [3H]quinuclidinyl benzilate ([3H]QNB) in an in vitro autoradiographic method to map the distribution of muscarinic receptors in guinea-pig ileum, colon and caecum. In addition, the relative distribution of low and high affinity agonist binding sites was assessed by the addition of the muscarinic agonist, carbachol, to selectively inhibit the binding of [3H]QNB to the high affinity sites. Although quantitativ

Visualization of mechanical stress-mediated Ca2+ signaling in the gut using intravital imaging.

Mechanosensory systems have been implicated in the maintenance of gut homeostasis, but details on the related mechanisms are scarce. Recently, we generated a conditional Ca2+ biosensor yellow cameleon 3.60 (YC3.60)-expressing transgenic mouse model and established a five-dimensional (5D; x, y, z, time, and Ca2+) intravital imaging system for investigating lymphoid tissues and enteric epithelial cell responses. To validate this gut-sensing system, we visualized responses of enteric nervous system (ENS) cells in Nestin-Cre/YC3.60flox mice with specific YC3.60 expression. The ENS, including the m

[Chronic intestinal pseudo-obstruction due to intestinal neuronal dysplasia type B (IND B), concerning one case].

Intestinal neuronal dysplasia type B (IND B) is an infrequent disease due to hyperplasia of the parasympathetic submucous plexus which causes alteration of intestinal motility, giving rise to symptoms of constipation and subocclusive manifestations. The disease is difficult to diagnose. It requires high clinical suspicion and should include differential diagnosis of patients with repeated subocclusive manifestations in order to make an early and correct diagnosis and avoid complications derived from unnecessary surgery that worsens the prognosis. We present the case of a 33-year-old Moroccan m…

Subtypes of Enteric Neurons in Hirschsprung Disease: A Comparison of Fully Ganglionic and Transitional Zone.

BACKGROUND: Hirschsprung disease (HD) is characterized by aganglionosis of the distal bowel and an adjacent transitional zone with structural abnormalities of the enteric nervous system (ENS). While previous studies have focused primarily on neuronal density, it remains unclear whether the neurochemical composition of enteric neurons differs between transitional and fully ganglionic bowel. This study therefore aimed to characterize neuronal subtype composition in these regions of HD specimens. METHODS: Colonic specimens resected during pullthrough surgery from pediatric HD patients were analy
