Determination of total sulfite in wine by ion chromatography after in-sample oxidation.

Sulfur dioxide (SO2) or sulfites are the most common preservatives used in winemaking. The level of total SO2 is subject to regulation. Currently, the regulatory determination of total SO2 (including sulfites) is done by the optimized Monier-Williams (OMW) method, which includes time-consuming distillation and titration steps. This paper describes the development and application of an alternative, rapid, straightforward, and reliable method for the determination of total sulfite in wine. In this method, a simple oxidation step using alkaline hydrogen peroxide (H2O2) solution is followed by ion…

Electrochemiluminescence determination of pipemidic acid using sulfite as energy transfer mediator.

An electrochemiluminescence (ECL) based on energy transfer from electro-generated triplet sulfur dioxide to pipemidic acid (PPA) was studied. A weak ECL from triplet sulfur dioxide (3)SO2 * was observed when sulfite was electrochemically oxidized in sulfuric acid solution on a Pt electrode. When PPA was present, the weak ECL was enhanced. The enhanced ECL was attributed to energy transfer from (3)SO2 * to PPA. Based on the enhanced ECL, a flow-injection (FIA) ECL method for the determination of PPA was proposed. The proposed method allowed the measurement of PPA over the range of 1.0x10(-7) to

Influence of Cu(II) on the interaction between sulfite and horseradish peroxidase in vitro.

This paper discussed the quantitative influence of Cu(II) on the interaction between horseradish peroxidase (HRP) and sulfite (SO3(2-)), which is a derivate of sulfite dioxide in human bodies, by using fluorescence spectrum and ultraviolet (UV) absorption spectrometry in vitro. The results show that under the conditions of physiological pH and room-temperature, Cu(II) can bind strongly with both the protein part and the ferroporphyrin part in HRP at a low concentration (10(-4) mol L(-1)), and the combination constants are 2.047 x 10(3) and 7.66 x 10(2) L mol(-1), respectively. Under the same c

Effects of sulfite on glutathione S-sulfonate and the glutathione status of lung cells.

A mechanistic study was performed to elucidate the biochemical events connected with the cocarcinogenic effect of sulfur dioxide (SO2). Glutathione S-sulfonate (GSSO3H), a competitive inhibitor of the glutathione S-transferases, forms in lung cells exposed in culture to sulfite, the hydrated form of SO2. Changes in glutathione status (total GSH) were also observed during a 1-h exposure. Some cells were pretreated with 1,3-bis(2-chloroethyl)-1-nitrosourea (BCNU) to inhibit glutathione reductase. In human lung cells GSSO3H formed in a concentration-dependent manner, while glutathione (GSH) incre

Molybdenum cofactor deficiency in a patient previously characterized as deficient in sulfite oxidase.

The metabolic status of a patient previously characterized as deficient in sulfite oxidase was reexamined applying new methodology which has been developed to distinguish between a defect specific to the sulfite oxidase protein and sulfite oxidase deficiency which arises as a result of molybdenum cofactor deficiency. Urothione, the metabolic degradation product of the molybdenum cofactor, was undetectable in urine samples from the patient. Analysis of molybdenum cofactor levels in fibroblasts by monitoring reconstitution of apo nitrate reductase in extracts of the Neurospora crassa mutant nit-

Functional changes in the lungs of guinea pigs exposed to sodium sulfite aerosols.

Guinea pigs were exposed head only for 1 hr to submicrometer sodium sulfite aerosols (mass median aerodynamic diameter = 0.36 micron, sigma g = 2.96) at 474, 669, and 972 micrograms SO3(2-)/m3. Respiratory mechanics were measured in unanaesthetized animals before, during, and after exposure. Dose-related increases in resistance and decreases in compliance were observed. At 972 micrograms SO3(2-)/m3, the sodium sulfite aerosol caused a 50% increase in resistance and a 19% decrease in compliance. These changes were still present 1 hr after the end of exposure. The results were used to assess the

Combined Application of Ultra-High-Pressure Homogenization and Non-Saccharomyces Yeasts to Reduce Sulfites and Improve Wine Quality.

In recent years, the wine industry has searched for alternatives to reduce the use of sulfites, with ultra-high-pressure homogenization (UHPH) emerging as a promising technology. The objective of this study was to evaluate the combined effect of UHPH and non-Saccharomyces yeasts (Lachancea thermotolerans and Metschnikowia pulcherrima) on wine quality. To this end, fermentations were carried out using control Verdejo must, must treated with UHPH, and must treated with 50 mg/L SO2, using pure cultures and co-inoculations. May Sulfites in Wine Affect Gut Microbiota? An In Vitro Study of Their Digestion and Interplay with Wine Polyphenols.

Sulfites are widely used in the wine industry, but their human health effects remain debated. This study is the first to investigate the interaction between wine sulfites and gut microbiota under simulated gastrointestinal conditions. Using the simgi model, red and synthetic wines─with and without SO2 (200 mg/L)─underwent gastrointestinal digestion and colonic fermentation with fecal microbiota from three healthy donors (n = 3). SO2-treated wines slightly modified gut microbiota composition, decreasing beneficial bacteria like Bacteroides and Ruminococcus, while increasing Coprococcus and pro-…

Heterojunction photocatalyst FeS2/g-C3N5 for activating sulfites to degrade tetracycline: A stable degradation system based on heterogeneous processes.

The UV/sulfite system is a promising source of •SO4- and/or •OH, but its application is largely limited by the use of UV light due to its high cost and high energy consumption. Graphite carbon nitride (g-C3N5), as a new photocatalytic material, has better visible light absorption capacity and narrower band gap than g-C3N4, which is expected to activate sulfite under visible light to solve this problem. Herein, a novel FeS2/CN heterojunction material based on g-C3N5 was constructed by hydrothermal in-situ synthesis method and successfully activated sulfite, which was confirmed by tetracycline d

Sodium Sulfite as an Oxygen-Depletion-Based Chemical Model of Tumor Hypoxia In Vitro.

Hypoxia is a major determinant of tumor progression, yet most chemical hypoxia models rely on HIF stabilization rather than true oxygen depletion. Here, we evaluated sodium sulfite (SS), a direct oxygen scavenger, as an oxygen-dependent in vitro hypoxia model. Cancer cell lines were treated with optimized SS concentrations, and dissolved oxygen levels were monitored for up to 96 h. Cell viability, cell death, cell cycle distribution, morphological alterations, and hypoxia-associated metabolic and redox responses were assessed and compared with conventional hypoxia mimetics. SS reduced dissolve

A biodegradable calcium sulfite nanoreactor for pH triggered gas therapy in combination with chemotherapy.

As a gasotransmitter, endogenous sulfur dioxide (SO2) plays an important role in cardiovascular regulation. In addition, excessive SO2 can react with overexpressed hydrogen peroxide (H2O2) in tumor cells to generate toxic radicals, which can induce severe oxidative damage to tumor cells and result in cell apoptosis. This highlights the potential of SO2 in oncotherapy. However, the limited availability of endogenous H2O2 and uncontrolled release of SO2 gas significantly impede the effectiveness of SO2 gas therapy. To address this challenge, a biodegradable calcium sulfite (CS) nanocarrier loade

Photoinduced transformation of ferrihydrite in the presence of aqueous sulfite and its influence on the repartitioning of Cd.

The photoinduced transformation of ferrihydrite is an important process that can predict the geochemical cycle of Fe in anoxic environments as well as the fate of trace elements bonded to Fe minerals. We report that the photooxidation of sulfite by UV irradiation produces hydrated electrons (super-reductants), which significantly promote ferrihydrite reduction to Fe(II), and SO3•- (a moderate oxidant), enabling its further oxidation to more crystalline Fe(III) products. The experimental results show that the concentration of sulfite was key in influencing the rate and extent of surface-bound F

A new approach for sulfite determination by headspace liquid-phase microextraction with an optical probe.

A new approach of headspace liquid-phase microextraction with an optical probe (HS-LPME-OP), which solves the problem of the extraction phase retention in the hole of the optical probe and provides the possibility of simpler, more precise and reliable online processing of the analytical signal, was used for sulfite determination. A 1 × 10-4 M 5,5'-dithiobis-(2-nitrobenzoic) acid (DTNB) solution was used as an acceptor phase. It was placed in a plastic vial fixed in the headspace above the analyte solution. An optical probe immersed in the acceptor phase was used to monitor the analytical signa

Determination of Sulfite in Food by Liquid Chromatography Tandem Mass Spectrometry: Collaborative Study.

Sulfites are added to a wide range of food and beverage products to prevent browning or oxidation. Although most of the population do not experience side effects from consuming sulfites, a small subset has been shown to experience an "allergic-like" response. For this reason, the U.S. Food and Drug Administration requires that sulfites be labeled on all products that contain more than 10 mg/kg (parts per million) sulfur dioxide. The current regulatory method, optimized Monier-Williams (OMW) Official Method 990.28, has been successful in quantifying sulfites in most matrixes, but is time-consum…

Development of a liquid chromatography-tandem mass spectrometry method for the determination of sulfite in food.

Sulfites are widely used food preservatives that can cause severe reactions in sensitive individuals. As a result, the U.S. FDA requires that sulfites be listed on the label of any food product containing >10 mg/kg (ppm) sulfite (measured as sulfur dioxide). Currently, the optimized Monier-Williams (MW) method (AOAC Official Method 990.28) is the most common approach for determining sulfite concentrations in food samples. However, this method is time-consuming and lacks specificity in certain matrices. An improved rapid, sensitive, and selective method has been developed using electrospray ion…

Sulfite exposure-induced hepatocyte death is not associated with alterations in p53 protein expression.

Although sulfite (SO3(2-)) is commonly used as an antimicrobial agent and preservative in foods, medicines and wine, it has also been listed as an important risk factor for the initiation and progression of liver diseases due to oxidative damage. In general, apoptosis that is induced by oxidative stress is triggered by increases in p53 and alterations in Mdm2 and Bcl-2. However, the level of involvement of the p53 signaling pathway, which has been shown to be upregulated in some animal studies, in hepatocyte death remains unclear. To examine the response of the p53 signaling pathway to stimula

Sulfite oxidase controls sulfur metabolism under SO2 exposure in Arabidopsis thaliana.

In the present study, the significance of sulfite oxidase (SO) for sulfite detoxification and sulfur assimilation was investigated. In response to sulfur dioxide (SO(2)) exposure, a remarkable expansion of sulfate and a significant increase of GSH pool were observed in wild-type and SO-overexpressing Arabidopsis. These metabolic changes were connected with a negative feedback inhibition of adenosine 5'-phosphosulfate reductase (APR), but no alterations in gas exchange parameters or visible symptoms of injury. However, Arabidopsis SO-KO mutants were consistently negatively affected upon 600 nL

Ion chromatographic measurement of sulfide, methanethiolate, sulfite and sulfate in aqueous and air samples.

Simultaneous measurement of sulfur species was investigated using ion chromatography. Sulfide, methanethiolate, sulfite and sulfate are representative of sulfur species. The aqueous sulfur species were simultaneously measured using a two-detector system: suppressor-type conductivity detector for sulfite and sulfate, and fluorometric or electrochemical detector for the reduced sulfur compounds. The major sulfur-containing gases, hydrogen sulfide, methyl mercaptan and sulfur dioxide are collected into aqueous solution as the species listed above. Collection of sulfur gases using a membrane-based

[The problem and improvement of colorimetric method for determination of sulfite in foods containing sulfur compounds].

The modified Rankine colorimetric method for measuring sulfite added to food as a food additive has a low determination limit and is little influenced by interfering substances from foods. However, it can give erroneous results for foods containing Liliaceae Allium. So, four different methods, alkaline titration, a colorimetric method, ion chromatography and qualitative analysis with potassium iodate-starch paper, were examined. It was found that the sodium azide used in the colorimetric method forms sulfur dioxide during bubbling and heating. The proposed colorimetric method can be applied to

Isolated sulfite oxidase deficiency: mutation analysis and DNA-based prenatal diagnosis.

Isolated sulfite oxidase deficiency is an autosomal recessive, neurological disorder resulting from a defect in SUOX, the gene encoding the enzyme that catalyzes the terminal reaction in the sulfur amino acid degradation pathway. In its classical, severe form, sulfite oxidase deficiency leads to intractable seizures, severe and progressive brain pathology and death at an early age. We report here on clinical features and mutational analysis of the genetic defect in a newborn with sulfite oxidase deficiency. Cultured fibroblasts from this patient exhibited no detectable sulfite oxidase activity
